斑茅割手密复合体(GXAS07-6-1)及其与甘蔗F_1的GISH分析

斑茅割手密复合体(GXAS07-6-1)是广西蔗茅属斑茅和广西甘蔗属割手密的属间杂种,聚集了双亲的优点。本研究利用基于Alu-like的PCR鉴定方法对GXAS07-6-1及甘蔗与GXAS07-6-1的3份F_1材料进行真实性鉴定,基于基因组原位杂交技术(GISH)对父本GXAS07-6-1及其3份F_1染色体组成及核型进行分析。研究结果表明:3份F_1材料为GXAS07-6-1的真杂种;父本GXAS07-6-1的染色体众数为62条,其中30条来自蔗茅属斑茅,32条来自甘蔗属割手密,核型分类属于1B,其染色体按"n+n"方式传递;GXASF_108-2-17、GXASF_108-2-22、GXASF_108-2-32的染色体数目为78~80条,其中69~71条来自甘蔗属,9~11条来自蔗茅属斑茅,3份F_1的核型分类分别属于2B、1B、1B,染色体传递方式均为"n+n"。父本GXAS07-6-1及3份F_1材料中均未发现有染色体的交换或易位现象。甘蔗与斑割复合体杂交,蔗茅属斑茅染色体在亲子间传递过程存在丢失现象。     

Chromosome Observations of Eight Species Endemic to China

A report of chromosome numbers for eight species endemic to China is made in the paper, including first counts for 4 genera and 4 species and first karyotypic analyses of two species. Sinojohnstonia chekiangensis (Migo) W. T. Wang (Boraginaceae) 2n=24*; Coptis chinenis Franch (Ranunculaceae) 2n=18**; Dichocarpum dalzielii (Drumm. et Hutch.) W. T. Wang et Hsiao (Ranunculaceae) 2n=24*; Eomecon chionantha Hance (Papaveraceae) 2n=18; Camptotheca acuminata Dcne. (Nyssaceae) 2n=44; Calycanthus chinensis Cheng et S. Y. Chang (Calycanthaceae) 2n=22**; Eucommia ulmoides Oliv. (Eucommiaceae) n=17; Pinellia pedatisecta Schott (Araceae) 2n=26; The previous reports of chromosome numbers of the same groups are compared with our own (See Table 1). The vouchers for the present study are preserved in the Herbarium of Futan University.     

Studies on Karyotypes of 5 Species of Rohdea and Tupistra

Five species of the genera Rohdea and Tupistra of Liliaceae were cytotaxonomically investigated in this work. The materials were collected from Jiangxi, Zhejiang, and Sichuan provinces. The results are shown as follows. 1. Rohdea Roth There are two forms of Rohdea japonica (Thunb.) Roth. The karyotype formula of the cultivated form is 2n=38=28m+8sm+2st (Plate, 1), while that of the wild one is 2n=38=26m+10sm+2st (Plate, 1). They both belong to Stebbins' (1971) karyotype classification 2 B. 2. Tupistra Ker-Gawl. The karyotype of Tupistra chinensis Baker from Mt. Lushan, Jiangxi,is 2n=38=24m+14sm(Plate, 2), which differs from that of Campylandra watanabei(=T. chinensis) from Taiwan, which is 2n = 38 = 32m + 4sm + 2st (Chang et Hsu, 1974). The chromosome numbers and karyotypes of the three species from Nanchuan, Sichuan, are reported here for the first time. The karyotype formulas of T. delavayi Franch. and T. jinshanensis Z. L. Yang et X. G. Luo are both 2n= 38 = 26m + 12sm (Plate, 2-3), and that of T. wattii (C. B. Clarke) Hook. f. is 2n= 38= 28m + 10sm (Plate, 3). They all belong to 2B. 3. In Convallarieae (s. 1.) the chromosome numbers of all the genera, except for Theropogon, are 2n=38 or both 2n=38 and 2n=36 in Aspidistra. The karyotypes of Rohdea and Tupistra are most similar, the differenc in karyotype between two genera is only presence or absence of a pair of subterminal chromosomes. The karyotype of Rohdea is similar to that of Convallaria rather than to those of Aspidistra and Speirantha, while the karyotype of Tupistrais similar to that of Reineckia rather than to those of the other genera. 应用形态、染色体和分子特征研究我国雷氏按蚊的分类鉴别(双翅目:蚊科)

为确定可靠的雷氏按蚊Anopheles lesteri Baisas et Hu.1936分类鉴别特征,对采自不同地区的雷氏按蚊进行了形态、染色体和分子特征的观察和分析。检视了辽宁、广东现场标本,江苏实验室品系的成蚊、虫卵特征,描述了染色体核型,并测定了采自广东、辽宁、河南和云南4省的现场标本,以及江苏、海南和广西实验室品系的核糖体DNA间隔2区（ITS2）和D3序列。结果显示成蚊、虫卵形态变异较大,不具备稳定、明确的鉴别特征;染色体核型具有多态现象,性染色体X、Y分别有3个和2个类型,可分为染色体型A与B;唯有ITS2分子序列具有客观、稳定的种间差异,系雷氏按蚊可靠、可行的鉴别特征。     

A new view on the scenario of karyotypic stasis in Epinephelidae fish: Cytogenetic,historical, and biogeographic approaches

Epinephelidae (groupers) is an astonishingly diverse group of carnivorous fish widely distributed in reef environments around the world, with growing economic importance. The first chromosomal inferences suggested a conservative scenario for the family. However, to date, this has not been validated using biogeographic and phylogenetic approaches. Thus, to estimate karyotype diversification among groupers, eight species from the Atlantic and Indian oceans were investigated using conventional cytogenetic protocols and fluorescence in situ hybridization of repetitive sequences (rDNA, microsatellites, transposable elements). Despite the remarkable persistence of some symplesiomorphic karyotype patterns, such as all species sharing 2n=48 and most preserve a basal karyotype (2n=48 acrocentrics), the chromosomal diversification in the family revealed an unsuspected evolutionary dynamic, where about 40% of the species escape from the ancestral karyotype pattern. These karyotype changes showed a relation with the historical biogeography, likely as a byproduct of the progressive occupancy of new areas (huge diversity of adaptive and speciation conditions). In this context, oceanic regions harboring more recent clades such as those of the Indo-Pacific, exhibited a higher karyotype diversity. Therefore, the karyotype evolution of Epinephelidae fits well with the expansion and geographic contingencies of its clades, providing a more complex and diverse scenario than previously assumed.     

12种酢浆草属植物核型分析

【目的】为了从细胞学角度明确12种酢浆草属（Oxalis）植物核型特征与亲缘关系,探究酢浆草属植物染色体的多样性,进一步为酢浆草种质资源鉴定与杂交育种亲本选择提供参考。【方法】应用根尖压片法对酢浆草属12个物种植物有丝分裂中期的染色体形态、数目及其核型进行观察分析。【结果】结果表明：（1）7个物种的染色体数目为首次报道,5种已报道物种中,兔耳酢浆草（O. fabaefolia）和黄花酢浆草（O. pes-caprae）染色体数目与前人报道一致,其余3种,与先前报道存在差异,其中2n=26为酢浆草属植物中首次报道的染色体数目,共发现7种染色体基数,其中x=13为首次报道,倍性范围从2x~6x;实际染色体大小范围为0.27~2.23 μm,着丝点位置为中部着丝点区（m）和亚中部着丝点区（sm）,核型类型共4种,核型不对称系数范围为56.31%~65.40%。（2）12种酢浆草属植物中兔耳酢浆草最为进化,扁平酢浆草（O. compressa）最为原始。（3）根据染色体核型相似性可将12个酢浆草属植物划分为4组。【结论】研究认为,12个酢浆草属物种具有广泛的核型多样性,核型分类结果与前人形态学分类不完全一致。     

吸虱亚目非典型线粒体基因组研究进展

吸虱是真兽类哺乳动物体表的专性寄生虫,在全世界广泛分布,物种数量高达540种。近年来随着分子生物学的飞速发展,在NCBI中已收录15种吸虱线粒体基因组序列,其独特的非典型线粒体基因组发生剧烈的裂化现象,形成数目不等的微环染色体。本文综述了15种吸虱线粒体基因组的结构和组成、RNA基因、非编码区以及对吸虱祖先线粒体核型推测的方法和结果。探讨了不同种属间以及与其它昆虫的差异,提出今后对吸虱亚目线粒体基因组研究的展望。     

中国桤木属植物的细胞学研究(Ⅰ)

对桦木科桤木属自然分布于中国西南地区的4个种进行了细胞学研究,结果分别为:川滇桤木K(2n)=56=30m+24sm+2st;桤木K(2n)=56=38m+16sm+2M;蒙自桤木K(2n)=56=28m+26sm+2st;毛桤木K(2n)=56=42m+14sm;其中川滇桤木的核型属于2A型,其余均为2B型;上述种类染色体数目均为2n=56。4种材料的核型均为首次报道。     

莲C0t-1 DNA的荧光原位杂交分析

为分析中国莲Cot-1DNA在其中期染色体上的分布,从中国莲基因组DNA中分离出Cot-1DNA,将基因组和所分离的Cot-1DNA用生物素标记后作探针,对中国莲染色体进行原位杂交。杂交结果用耦联有荧光素Cy3的生物素抗体检测,发现在每对染色体上均显示出特定的荧光原位杂交带。同时分析了FISH和GISH信号分布的异同。基于Cot-1DNA荧光原位杂交带型及染色体型,构建了中国莲核型。     

Cytogenetic and genetic data support Crossodactylus aeneus Müller, 1924 as a new junior synonym of C. gaudichaudii Duméril and Bibron, 1841 (Amphibia, Anura)

The nominal anuran species Crossodactylus gaudichaudii Duméril and Bibron, 1841 and Crossodactylus aeneus Müller, 1924 are indistinguishable based on adult and larval morphology, being subject of taxonomic doubts. Here, we describe the karyotypes of C. gaudichaudii and C. aeneus, using classical and molecular cytogenetic markers. In addition, we used sequences of the H1 mitochondrial DNA to infer their phylogenetic relationships by Maximum Likelihood (ML) and Maximum Parsimony (MP) approaches and species delimitation test (by bPTP approach). The karyotypic data do not differentiate C. gaudichaudii and C. aeneus in any of the chromosome markers assessed. In both phylogenetic analyses, C. gaudichaudii and C. aeneus were recovered into a strongly supported clade. The species delimitation analysis recovered the specimens assigned to C. gaudichaudii and C. aeneus as a single taxonomic unit. Taken the cytogenetic and genetic results together with previous studies of internal and external morphology of tadpoles and biacoustic pattern, C. gaudichaudii and C. aeneus could not be differentiated, which supports the hypothesis that they correspond to the same taxonomic unit, with C. aeneus being a junior synonym of C. gaudichaudii.
