Male factor infertility elucidated about half the couple of infertility and in around 50% of cases, its etiology remains unknown. The aim of this study was to investigate a predisposing genetic background for Yq deletions and male infertility and effectiveness of molecular genetic approaches have uncovered several etiopathogenetic factors, such as microdeletions of Yq chromosome. The Y chromosome microdeletions removing the azoospermia factor (AZF) regions, which are most common molecular genetic causes of oligospermia or azoospermia. However, with the analysis of Yq deletions, we are able to obtain a better understanding of the clinical significance of genetic anomaly and to the identifying of fertility candidate genes in the AZF regions. Molecular genetic approaches, becomes a routine diagnostic test, that provides an etiology for spermatogenic disturbances, and prognosis for testicular sperm retrieval according to the type of deletion.  相似文献

为探究睾丸注射法制备转基因动物的可能性,文章将携带有山羊心脏型脂肪酸结合蛋白(H-FABP)和绿色荧光蛋白标签的重组载体经脂质体包裹后随机打点注射小鼠睾丸。对实验小鼠进行睾丸切片、精子荧光检测以及精子DNA检测,证实外源基因在亲代小鼠体内成功表达。睾丸注射后小鼠与正常母鼠交配产生的F1代,以及F1代自交产生的F2代在不同水平均可检测到外源基因的成功表达,阳性率分别为4%和30.23%。研究结果说明睾丸注射是一种制备转基因动物行之有效的方法,且外源基因可以稳定遗传。该方法的完善和成熟对于动物转基因以及动物性状改良和育种具有理论和实践意义。  相似文献

目的:探究雷珠单抗与阿柏西普玻璃体腔注射治疗渗出性年龄相关性黄斑变性的临床疗效与安全性。方法:选取2015年5月~2018年5月我院收治的渗出性年龄相关性黄斑变性患者98例(98眼),采用随机数字表法将患者分为两组,A组玻璃体腔内缓慢注射0.05 m L(0.5 mg)雷珠单抗注射液,B组玻璃体腔内缓慢注射0.05 m L(2 mg)阿柏西普注射液。比较两组患者的视力改善情况、眼动脉血流动力学、黄斑中心凹视网膜厚度及不良反应的发生情况。结果:治疗前两组患者的视力比较无统计学差异(P0.05),治疗后,B组视力显著高于A组(P0.05);。两组患者治疗前后眼动脉血流动力学相关指标比较均无统计学差异(P0.05)。;两组患者治疗后的黄斑中心凹视网膜厚度均显著降低,且治疗3个月B组治疗3个月显著低于A组(P0.05);B组患者注射药物后结膜下大出血发生率显著低于A组(P0.05)。结论:阿柏西普可显著改善渗出性年龄相关性黄斑变性患者的视力,且安全性高,可能与其可显著改善患者的黄斑中心凹视网膜厚度有关,且不良反应发生率低,安全性好。  相似文献

目的:探讨卡前列素氨丁三醇注射液联合缩宫素用于剖宫产产后出血的预防效果及对纤维蛋白原(FBI)、D-二聚体(D-D)水平的影响。方法:选择2014年1月至2016年10月我院接诊的92例剖宫产产妇,通过随机数表法分为观察组(n=46)和对照组(n=46)。对照组在胎儿娩出后给予缩宫素的宫内注射及静脉滴注,观察组在胎儿娩出后,先给予缩宫素的宫内注射,再在子宫肌壁注射卡前列素氨丁三醇注射液。比较两组产后出血情况、宫缩持续时间、子宫底下降速度、恶露持续时间、FIB、D-D水平及不良反应的发生情况。结果:观察组产后2 h、24 h出血量及产后出血率明显低于对照组(P0.05),宫缩持续时间长于对照组(P0.05),子宫底下降速度快于对照组(P0.05),恶露持续时间明显短于对照组组(P0.05)。两组治疗前FIB、D-D水平比较差异无统计学意义(P0.05)。治疗后,两组FIB、D-D水平均较治疗前显著降低(P0.05),且观察组FIB、D-D水平均明显低于对照组(P0.05);两组恶心呕吐、面部潮红、胸闷、头痛发生率比较差异无统计学意义(P0.05)。结论:在剖宫产产妇中应用卡前列素氨丁三醇注射液联合缩宫素可降低产后出血发生率,且安全性高,可能与有效减少FIB、D-D表达有关。  相似文献

目的:研究负压封闭引流技术联合前列地尔注射液对重度烧伤患者的疗效观察。方法:选取2017年2月至2018年2月的67例患者。按照随机数表法分为观察组(n=36)和对照组(n=31),对照组采用负压封闭引流技术治疗,观察组采用负压封闭引流技术联合前列地尔注射液治疗。观察两组治疗疗效情况,临床治疗指标(创面愈合时间、创面愈合率、VAS评分、住院时间),C反应蛋白(CRP)、降钙素原(PCT)水平,不良反应发生率。结果:治疗后,观察组总有效率显著高于对照组[86.11%(31/36)vs 61.29%(19/31)](P0.05);创面愈合时间、创面愈合率、VAS评分、住院时间均显著低于对照组[(16.93±4.19)天vs(24.27±6.01)天,(92.72±5.03)%vs(81.34±2.48)%,(2.93±0.52)分vs(6.70±1.30)分,(24.09±7.43)天vs(46.98±10.72)天](P0.05);CRP、PCT水平均显著低于对照组[(27.93±3.81)mg/L vs(83.86±7.43)mg/L,(0.10±0.02)ng/L vs(0.43±0.06)ng/L](P0.05);不良反应总发生率显著低于对照组[8.33%(3/36)vs 38.70%(12/31)](P0.05)。结论:负压封闭引流技术联合前列地尔注射液对重度烧伤患者的疗效显著,可促进创面恢复,避免感染,并发症少,安全可靠。  相似文献

This study was implemented to figure out whether lncRNA HOTAIR/miR‐17‐5p/PTEN axis played a role that was opposite to Shenqifuzheng (SQFZ) injection in regulating the chemosensitivity of gastric cancer cells. The gastric cancer tissues were gathered and four gastric cancer cell lines were prepared, including BGC‐823, MGC‐803, SGC‐7901, and MKN28. Moreover, cisplatin, adriamycin, mitomycin, and 5‐fluoroura were managed as the chemo‐therapeutics, and SQFZ was prepared as a Chinese medicine. Striking distinctions of HOTAIR, miR‐17‐5p, and PTEN expressions were observed between gastric cancer tissues and para‐carcinoma normal tissues (< 0.05). MKN28 was associated with the highest resistance to cisplatin, adriamycin, mitomycin, and 5‐fluoroura among all the cell types, and SQFZ significantly improved the MKN28 cells’ sensitivity to the drugs (< 0.05). The over‐expressed HOTAIR and miR‐17‐5p, as well as under‐expressed PTEN tended to significantly facilitate the viability, EMT process and proliferation of MKN28 cells that were subject to treatment of chemo‐therapies (< 0.05). SQFZ could amplify the effects of si‐HOTAIR, miR‐17‐5p inhibitor, and pcDNA‐PTEN on boosting the chemosensitivity of gastric cancer cells (< 0.05). In addition, HOTAIR was also found to directly target miR‐17‐5p, and PTEN appeared to be subject to the modification of HOTAIR and miR‐17‐5p in its acting on the viability, proliferation, EMT process, and apoptosis of gastric cancer cells. The HOTAIR/miR‐17‐5p/PTEN axis could be regarded as the potential treatment targets for gastric cancer, and adjuvant therapy of SQFZ injection could assist in further improving the treatment efficacy of chemo‐therapies for gastric cancer.  相似文献

Aneurysmal subarachnoid hemorrhage (aSAH) is a severe medical condition associated with a significant cause of mortality throughout the world. Cisterna magna injection model is accepted widely to mimic clinical aSAH and is performed on small animal models to study aSAH during neurosurgery. Coherent light scattered from the surface of the rat brain is used to infer information about the variations in blood flow during this condition. We obtained speckle images from the exposed cortex during the entire experiment using an external tissue imaging system. Contrast and fractal analyses are carried out for the recorded speckle pattern time series. Correlation analysis based on Hurst exponent for these images is found to be a more sensitive tool in studying aSAH as compared to routinely used laser speckle contrast analysis for assessing the changes in blood flow velocity. Additionally, our studies provide improved blood flow detection sensitivity with image Hurst exponent in combination with computed fractal dimension, during an event of aSAH.  相似文献

We established a rapid procedure for obtaining transgenic mice by directly injecting an enhanced green fluorescent protein (EGFP)-expressing plasmid (pIRES-EGFP) into the ovaries of fertile mice. The frequency of transgenic mouse production was determined by pair-mating, and by polymerase chain reaction (PCR) and sequence analysis of DNA taken from the tails of the offspring. The mice that received the EGFP gene transmitted it to their offspring (F(1)). Genetic and PCR analyses of F(1) progeny confirmed that the inserted EGFP was stably inherited. Of six female F(1) mice, all were able to pass the foreign DNA on to the next generation (F(2)). In situ hybridization using paraffin-embedded sections of ovarian and testicular tissues from the F(1) and F(2) progeny showed that the introduced gene was expressed in the gonads of the animals. The chromosomal location of the injected DNA was determined by fluorescence in situ hybridization, and the frequency of multiple site versus single site insertions is 85.71% (18/21) analyzed by FISH. We anticipate great progress in murine genetic engineering using this technique.  相似文献

Five mutants of Lactococcus lactis subsp. cremoris 3107 resistant to phage TP901-1 were obtained after treatment with ethyl methanesulfonate. Two of the mutants were also resistant to phage phiLC3. The remaining three mutants were as sensitive as 3107. Mutants E46 and E100 did not adsorb the two phages. Mutants E119, E121 and E126 adsorbed phage phiLC3 as well as 3107 but phage TP901-1 with significantly reduced efficiency. All, except E46, could be lysogenized with phage TP901-BC1034, a derivative of TP901-1 harboring an erythromycin-resistance marker. However, the lysogenization frequency was 10(3)-10(4) fold higher for 3107 than for the mutants. Mitomycin C induction of lysogenized mutants 3107 indicated that phage propagation was not affected in these four mutants. Electron microscopy and analysis of total DNA of infected cells showed that DNA was liberated from the phage particle during infection of strain 3107 with TP901-1 and that intracellular phage DNA replication occurred. This was not the case for mutants E121 and E126. This strongly suggests that some step starting with triggering DNA release and ending with DNA injection is impaired during infection with TP901-1. As such impairment was not seen when infecting E119, E121 and E126 with phiLC3, we conclude that TP901-1 and phiLC3 either are differently triggered by their receptor or utilize different pathways of injection.  相似文献

摘要 目的:探讨杏芎氯化钠注射液联合依达拉奉对急性脑梗死(ACI)患者神经功能、脑血流灌注及氧化应激水平的影响。方法:随机将2018年1月至2019年7月我院收治的120例ACI患者分为两组,均给予常规神经内科治疗,对照组(60例)在常规治疗基础上给予依达拉奉静脉滴注2周,观察组(60例)在对照组基础上加用杏芎氯化钠注射液滴注两周。观察两组临床疗效和安全性,并比较治疗前后神经功能、认知功能、残疾程度、脑血流灌注以及氧化应激水平的差异。结果:观察组治疗总有效率均高于对照组(P<0.05)。观察组治疗后美国国立研究院脑卒中评定量表(NIHSS)、改良RANKIN量表(MRS)评分、脑缺血区平均通过时间(MTT)、峰值时间 (TTP)、丙二醛(MDA)、晚期氧化蛋白产物(AOPP)低于对照组(P<0.05),简易智能精神状态检查量表(MMSE)评分、脑血流量(CBF)、脑血容量(CBV)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC )高于对照组(P<0.05)。两组不良反应率比较无统计学差异(P>0.05)。结论:杏芎氯化钠注射液联合依达拉奉治疗可有效地改善ACI患者神经功能,增加缺血区脑血流灌注量,改善氧化应激,提高临床疗效。  相似文献
