Nitric oxide plays an important role in various biological processes, such as neurotransmission, blood pressure control, immunological responses, and antioxidant action. The control of its local concentration, which is crucial for obtaining the desired effect, can be achieved with exogenous NO-carriers. Coordination compounds, in particular ruthenium(III) and (II) amines, are good NO-captors and -deliverers. The chemical and photochemical properties of several ruthenium amine complexes as NO-carriers in vitro and in vivo have been reviewed. These nitrosyl complexes can stimulate mice hippocampus slices, promote the lowering of blood pressure in several in vitro and in vivo models, and control Trypanosoma cruzi and Leishmania major infections, and they are also effective against tumor cells in different models of cancer. These complexes can be activated chemically or photochemically, and the observed biological effects can be attributed to the presence of NO in the compound. Their efficiencies are explained on the basis of the [RuIINO+]3+/[RuIINO0]2+ reduction potential, the specific rate constant for NO liberation from the [RuNO]2+ moiety, and the quantum yield of NO release.  相似文献

以琼脂粉为基质制备金属螯合载体,并用于固定重组腈水解酶。研究发现:制备金属螯合载体最合适的金属离子为Zn2+。当Zn2+离子浓度0.3 mol/L、给酶量15.6 mg/g、固定化pH 8.0、固定化温度40℃时,制得的固定化酶活性最高。固定化酶最适反应温度为50℃、最适反应pH为7.0。当扁桃腈浓度为10 mmol/L、反应1 h时,固定化酶最大产率为0.041 mmol/(g·h);在反应12 h时,产物e.e.值可达到99%以上。固定化酶重复使用8次以后,酶活力仍保持在45%。  相似文献

The trans-Golgi network (TGN) is one of the main, if not the main, sorting stations in the process of intracellular protein trafficking. It is therefore of central importance to understand how the key players in the TGN-based sorting and delivery process, the post-Golgi carriers (PGCs), form and function. Over the last few years, modern morphological approaches have generated new insights into the questions of PGC biogenesis, structure and dynamics. Here, we present a view by which the “lifecycle” of a PGC consists of several distinct stages: the formation of TGN tubular export domains (where different cargoes are segregated from each other and from the Golgi enzymes); the docking of these tubular domains onto molecular motors and their extrusion towards the cell periphery along microtubules; the fission of the forming PGC from the donor membrane; and the delivery of the newly formed PGC to its specific acceptor organelle. It is now important to add the many molecular machineries that have been described as operating at the TGN to this “morphofunctional map” of the TGN export process.  相似文献

The ability of holothurian triterpene glycosides (cucumarioside A2-2 from Cucumaria japonica, cucumarioside G1 from C. fraudatrix, frondoside A from C. frondosa, and holotoxin A1 from Apostichopus japonicus) to form supramolecular lipid-saponin complexes was studied. TEM demonstrated that all the studied compounds form supramolecular cholesterol-saponin complexes (nanoparticles) in aqueous medium. The complexes formed by cucumarioside A2-2, holotoxin A1, and frondoside A had a tubular structure and fundamentally differed in the structure from the particles produced by cucumarioside G1. The morphology of the nanoparticles formed by cucumarioside A2-2, holotoxin A1, and cucumarioside G1 changed depending on the fraction of cholesterol in the lipid-saponin system; however, this pattern was not observed for frondoside A. At the same molar fraction of cholesterol in the lipid-saponin system, cucumarioside A2-2 formed the particles with the most pronounced tubular structure; the cholesterol-saponin complexes of holotoxin A1 had a less pronounced tubular structure, whereas the structure of frondoside A particles was extremely heterogeneous. Comparative analysis of the morphology of the described supramolecular complexes and specific structural features of the glycosides demonstrated that the structure of the corresponding nanoparticles depended on the degree of branching of the carbohydrate moiety in the glycoside molecule and the complexation with cholesterol was determined by the specific features of aglycone structure. Thus, the feasibility of producing new generation antigen carriers using the complexes in question was proved.  相似文献

以磁性金属螯合琼脂糖微球为载体,利用金属螯合配体(IDACu2+)与蛋白质表面供电子氨基酸相互作用的原理,定向固定了木瓜蛋白酶。固定化最适条件为Cu2+1.5×10-2mol/g载体、固定化时间4h、固定化pH7.0、给酶量30mg/g载体。固定化酶的最适反应温度70℃、最适反应pH8.0,固定化酶的热稳定性明显高于溶液酶,固定化酶活力回收为68.4%,且有较好的操作稳定性,载体重复使用5次后固定化酶酶活为首次固定化酶79.71%。  相似文献

Charge carrier dynamics in organolead iodide perovskites is analyzed by employing time‐resolved photoluminescence spectroscopy with several ps time resolution. The measurements performed by varying photoexcitation intensity over five orders of magnitude enable separation of photoluminescence components related to geminate and nongeminate charge carrier recombination and to address the dynamics of an isolated geminate electron–hole pair. Geminate recombination dominates at low excitation fluence and determines the initial photoluminescence decay. This decay component is remarkably independent of the material structure and experimental conditions. It is demonstrated that dependences of the geminate and nongeminate radiative recombination components on excitation intensity, repetition rate, and temperature, are hardly compatible with carrier trapping and exciton dissociation models. On the basis of semiclassical and quantum mechanical numerical calculation results, it is argued that the fast photoluminescence decay originates from gradual spatial separation of photogenerated weakly bound geminate charge pairs.  相似文献

The interactions of model proteins with porous matrices in biosensors are considered. The viscoelastic properties of casein and albumin were assessed by a dynamic method of a piezoquartz resonator by applying thin layers of the studied solutions to a piezocrystal. The experimental data on the viscoelastic characteristics of protein solutions of various concentrations were compared with the characteristics of their tangential motion in the porous carriers of cellulose nitrate. It was demonstrated that the parameters of dynamic viscosity correlated with the motion time of the protein solutions in the porous polymeric carrier.  相似文献

Net nitrate uptake, 36ClO?3/NO?3 influx and 36Cl? influx into Pisum sativum L. cv. Feltham First seedlings have been examined following growth in culture medium containing different combinations of chloride and nitrate. When young (6 days old) seedlings, that had been grown in the absence of N were used, nitrate accumulation stimulated net nitrate uptake and 36ClO?3/NO?3 influx (r2= 0.99) while chloride accumulation inhibited nitrate uptake and 36ClO?3/NO?3 influx (r2= 0.65). When nitrate was provided during growth there was no effect of chloride pretreatment on net nitrate uptake and there was little effect of total [NO?3+ Cl?]i on 36ClO?3/NO?3 influx (r2= 0.26). A direct effect of Cl? on 36ClO?3/NO?3 influx was only found when seedlings had been starved of N for more prolonged periods (14 days). When moderate chloride was supplied during growth, 36Cl? influx was insensitive to nitrate or chloride accumulated, but significantly correlated with loge [NO?3+ Cl?]i (r2= 0.75). When trace amounts of Cl? were supplied during growth 36Cl? influx was inhibited by (a) NO?3 in the external medium and (b) Cl? pretreatment, but was insensitive to NO?3 pretreatment. The sensitivity of 36Cl? influx to external nitrate was not found following Cl? pretreatment in the absence of nitrate. The possibility that there are two populations of chloride carriers which differ in their sensitivity to external nitrate is discussed. Tentative schematic models to account for the regulation of nitrate and chloride uptake are proposed in the context of current hypotheses for regulation of ion transport and control systems theory.  相似文献

采用内装塑料片、塑料丝、海绵块的中空鱼网状塑料球为填料的生物滴滤池,对某垃圾压缩站产生的低浓度大风量的含氨臭气进行了近1年的连续脱臭试验。研究了有关的净化效果与生物膜特性。在进口氨气浓度0.8mg/m3~1.5mg/m3,风量8000m3/h,停留时间2.5s,氨气去除率为90%以上,达到国家一级排放水平。系统添加营养液时净化效果从75%提高到90%。3种填料的生物膜量、脱氢酶活性及硝化活性速率的排列顺序是:塑料片>塑料丝>海绵,并从工艺及填料方面探讨进一步提高净化效果的技术措施。  相似文献
