The effects of temperature and photoperiod on the ability of wheat (Triticum aestivum L.) seedlings to show induced responses (increased accumulation of hydroxamic acids, Hx) upon infestation by the bird cherry-oat aphid, Rhopalosiphum padi L. were evaluated under laboratory conditions. Induction of Hx was significantly higher at lower temperatures. No such clear trend was found for the photoperiod effect. The significant effect of environmental conditions on growth rate of seedlings and the significant negative correlation between growth rate prior to infestation and induction of Hx suggested that environmental effects on induced responses were at least partially mediated through their effect on plant growth rate. After statistically uncoupling the effect of environmental conditions from the effect of plant growth rate, the effect of temperature on induction of Hx was no longer significant. Therefore, the temperature effect was mediated by plant growth rate.  相似文献

Abstract. Few empirical data exist to examine the influence of regional scale environmental gradients on productivity patterns of plant species. In this paper we analyzed the productivity of several dominant grass species along two climatic gradients, mean annual precipitation (MAP) and mean annual temperature (MAT), in the Great Plains of the United States. We used climatic data from 296 weather stations, species production data from Natural Resource Conservation Service rangeland surveys and a geographic information system to spatially integrate the data. Both MAP and MAT were significantly related to annual above-ground net primary production (ANPP). MAP explained 54% to 89% of the variation in ANPP of two C4 short-grasses, Bouteloua gracilis and Buchloë dactyloides, and two C4 tall-grasses, Andropogon gerardii and Schizachyrium scoparium (= Andropogon scoparius). MAT explained 19% to 41% of the variation in ANPP of two C4 grasses, B. gracilis and B. dactyloides, and 41% to 66% of the variation in ANPP of two C3 grasses, Agropyron smithii and Stipa comata. ANPP patterns for species along both gradients were described by either linear, negative exponential, logistic, normal or skewed curves. Patterns of absolute ANPP (g/m2) for species differed from those of relative ANPP (%) along the MAP gradient. Responses were similar for species with common functional characteristics (e.g. short-grasses, tall-grasses, C3, C4). Our empirical results support asymmetric responses of species to environmental gradients. Results demonstrate the importance of species attributes, type of environmental gradient and measure of species importance (relative or absolute productivity) in evaluating ecological response patterns.  相似文献

The decrease in respiration rate following thyroidectomy is preceded by changes in the lipid composition of the mitochondrial membrane (Hoch, F.L., Subramanian, C., Dhopeshwarkar, G.A. and Mead, J.F. (1981) Lipids 16, 328–334) and in concert, changes in the kinetic parameters of the adenine nucleotide translocase (Mak, I.T., Shrago, E. and Elson, C.E. (1981) Fed. Proc. 40, 398). To demonstrate that physiological adaptation also involves this sequence of events, rats were housed at 8°C for 3–4 weeks. Cold adaptation resulted in a modest (5%) increase in the unsaturation index for the mitochondrial fatty acids comprised of a significant increase in arachidonic acid and a reciprocal decrease in linoleic acid. Phospholipid analysis indicated that cold adaptation increased the mitochondrial phosphatidylethanolamine and reciprocally decreased the phosphatidylcholine content. Concomitantly, cold adaptation resulted in 25–30% increases in rat liver mitochondrial respiratory activities without changing the respiratory control or ADP/O ratios. The kinetic parameters of the adenine nucleotide translocase were determined by the back-exchange method (Pfaff, E. and Klingenberg, M. (1968) Eur. J. Biochem. 6, 66–79). At 0–4 and 10°C, the Vmax and Km of the cold-adapted rat liver adenine nucleotide translocase were not distinguishable from the control values. The Ki values determined by Dixon plot studies for atractylate and palmitoyl-CoA were also comparable between the two groups. However, at 25 and 37°C, cold-adapted rat liver adenine nucleotide translocase exhibited a 20% increase in Vmax and a 20% decrease in Km for external ADP. The results suggest that one adaption to a cold environment involves hormone-mediated changes in the lipid composition in the mitochondrial membranes which in turn modulate the adenine nucleotide translocase and subsequent respiratory activities.  相似文献

Modulation of [3H]muscimol binding by picrotoxin, pentobarbitone, and etomidate was investigated in rat cerebellar and cerebral cortical membranes. In cerebellum, at 37 degrees C in the presence of chloride ions (150 mM), picrotoxin and picrotoxinin decreased specific [3H]muscimol binding to 43 +/- 3% of control, with an EC50 of 1.2 +/- 0.1 microM. [3H]Muscimol saturation experiments in the presence and absence of picrotoxin indicated that the picrotoxin effect was primarily due to a loss of high-affinity muscimol sites with KD approximately equal to 10 nM. Pentobarbitone enhanced specific [3H]muscimol binding to 259 +/- 3% of control, with EC50 = 292 +/- 37 microM, and etomidate increased binding to 298 +/- 18%, with EC50 = 7.1 +/- 1.0 microM. The influence of temperature and chloride ion concentration on these effects was investigated by comparing experiments at 37 and 0 degrees C in the presence or absence of chloride at constant ionic strength. The results indicate that studies at 0 degrees C underestimate the coupling between GABA receptors and barbiturate sites and that they greatly overestimate the importance of chloride ions in this phenomenon. In cerebral cortical membranes (37 degrees C, 150 mM Cl-), the effect of picrotoxin was similar to that observed in cerebellum, whereas the effects of pentobarbitone and etomidate were greater, but occurred at higher concentrations.  相似文献

Effects of temperature and thermoperiod on larval development and the induction of diapause were investigated in the European corn borer, Ostrinia nubilalis. Developmental threshold temperatures characteristic of the first four larval instars were estimated under both thermoperiods and constant temperatures. Threshold values were similar under the two conditions, but the 4th-larval instar was shown to display a significantly lower developmental threshold temperature than was characteristic of the earlier instars. Although developmental times (days per instar) were not greatly affected by fluctuating temperature regimes, the quantity of growth (weight, head width) during the 4th and 5th (last) stadia was found to be much greater under thermoperiodic regimes than under comparable constant temperature conditions. By means of thermoperiodic regimes having identical mean temperatures but different cryophase durations, it was demonstrated that the induction of diapause is dependent on the duration of the cryophase rather than on the mean temperature of the thermoperiod. To be effective, the cryophase must be colder than the insect's thermoperiodic response threshold. It was also demonstrated that thermoperiodic responses that are readily produced under continuously dark rearing conditions are not manifested under continuous light.  相似文献

The effects of incubation of yeast spheroplasts at elevated temperature (40°C) on a number of activities involved in protein biosynthesis have been examined in preparations obtained from wild-type cells (wt A364A) and a temperature-sensitive mutant (ts 7–45) derived from it. With wild-type cells, preincubation of spheroplasts at the elevated temperature had little or no effect on the following: (1) the ribosomal subunit-polysome pattern; (2) the translation of exogenous natural mRNA in postpolysomal extracts devoid of endogenous mRNA; (3) the translation of poly(U) in postpolysomal extracts; (4) the incorporation of methionine into 40 S preinitiation and 80 S initiation complexes; (5) the synthesis of Met-tRNA in postribosomal (cytosol) extracts; and (6) the formation of eIF-2·GTP·Met-tRNAf ternary complex in the cytosol. With temperature-sensitive spheroplasts that had not been preincubated at the elevated temperature, the concentration of free, native 40 S subunits appeared to be lower and that of 60 S subunits higher than in wild-type cells; translation of exogenous natural mRNA in postpolysomal extracts was somewhat lower than in wild-type preparations, but all of the other reactions and components measured were comparable to those in wild-type preparations. Preincubation of temperature-sensitive spheroplasts at 40°C resulted in: (1) a further decrease in the level of 40 S subunits; (2) disaggregation of polysomes; (3) loss of ability to translate natural mRNA but not poly(U); (4) decreased ability to form 40 S preinitiation intermediates; and (5) production of an activity, found in the cytosol, that inhibited Met-tRNA synthetase reversibly. The inhibitor had the characteristics of a protein and did not appear to be a proteinase, nuclease, or nucleotidase.  相似文献

Sodium-dependent d-glucose uptake into proteoliposomes reconstituted from dimyristoylphosphatidylcholine (DMPC) and hog kidney brush border membrane extract is strongly affected by temperature and the physical state of the membranes. This dependence is defined by a nonlinear Arrhenius plot with a break point at 23°C, a temperature not significantly different from the phase transition temperature of the pure lipid (24°C). The transport process is characterized by different activation energies: 35.1 kcal/mol below and 5.5 kcal/mol above the transition temperature. The shift in the break point for the d-glucose transport activity from 15°C, in the brush border membranes, to 23°C in the reconstituted system leads us to conclude that the lipids surrounding the sodium/d-glucose cotransport system can exchange readily with the bulk lipid used for reconstitution. The results thus provide no evidence for the presence of an annulus of specific lipids surrounding the transport system.  相似文献

Mitochondrial, microsomal and pellicular membranes were isolated from Tetrahymena cells grown at 39°C or 15°C, and phospholipids, in turn, were separated from total lipids extracted from these membranes. The effect of growth temperature on their solid-to-fluid phase transition temperature was examined by wide-angle X-ray diffraction. The transition temperatures of phospholipids from mitochondria, microsomes and pellicles were 21, 19 and 26°C for cells grown at 39°C and ?8, ?3 and 6°C for cells grown at 15°C, respectively. All phospholipids were found in a completely fluid state at these growth temperatures. From a comparison between the phospholipids and total lipids from pellicles of cells grown at 39°C, a triterpenoid alcohol, tetrahymanol, caused the transition temperature to increase. The alignment of tetrahymanol in membranes was examined with pellicle's total lipid oriented in a sample holder.  相似文献

The kinetics of bacteriorhodopsin action are investigated by means of the temperature pulse method. The temperature jump is provided by a short laser pulse (wavelengths λ1 = 2.94 μm or λ2 = 10.6 μm). The kinetics of evolution of the spectral intermediates M-412 and O-640 are investigated. A very fast increase in the O-640 form is observed as a result of the temperature jump. The experimental results are most consistent with the physical model based on electron resonance tunneling and permitting two protons transported per cycle. A branched scheme of the bacteriorhodopsin photocycle and corresponding mathematical equations are discussed.  相似文献
