汉阳陵文物表面硫酸盐形成原因微生物学证据   总被引:2,自引:0,他引:2  
用仅含硫源的无机盐培养基对采自汉阳陵13号和15号遗址坑文物表面的土样进行细菌富集筛选实验,并对分离的菌株进行生理生化特性研究、16SrDNA序列分析及硫代谢能力测试。分离得到5株细菌:TD1、TD2、TD3、TD15及PL13。其中TD1属于Alcaligenes,TD3属于Bacillus;硫代谢能力测试结果显示TD2、TD15和PL13代谢产生硫酸盐的速率远远小于TD3和TD1,TD3氧化硫源生成硫酸盐的速率是TD1的1.4倍。实验证实这些菌株均能氧化S、S2O32-和S2-生成硫酸盐从而获得能量,这表明微生物氧化土壤中硫化物是汉阳陵文物表面硫酸盐增加的原因,从而为汉阳陵文物表面硫酸盐的形成原因提供了微生物学依据。  相似文献

Ethyl glucuronide (EtG) has been shown to be a suitable marker of excessive alcohol consumption. Determination of EtG in hair samples may help to differentiate social drinkers from alcoholics, and this testing can be widely used in forensic science, treatment programs, workplaces, military bases as well as driving ability test to provide legal proof of drinking. A method for determination of EtG in hair samples using large volume injection-gas chromatography-tandem mass spectrometry (LVI-GC/MS/MS) was developed and validated. Hair samples (in 1 mL deionized water) were ultrasonicated for 1h and incubated overnight; these samples were then deproteinated to remove impurities and derivatisated with 15 μL of pyridine and 30 μL of BSTFA. EtG was detected using GC/MS/MS in multiple-reaction monitoring mode. This method exhibited good linearity: y=0.0036 x+0.0437, R2=0.9993, the limit of detection and the limit of quantification were 5 pg/mg and 10 pg/mg, respectively. The extraction recoveries were more than 60%, and the inter-day and intra-day relative standard deviations (RSD) were less than 15%. This method has been applied to the analysis of EtG in hair samples from 21 Chinese subjects. The results for samples obtained from all of those who were teetotallers were negative, and the results for the other 15 samples ranged from 10 to 78 pg/mg, except for one negative sample. These data are the basis for interpretation of alcohol abuse.  相似文献

A LC–MS/MS method was developed and validated for the determination of dehydroandrographolide succinate (DAS), a traditional Chinese medicine derivative used for the treatment of pneumonia, upper respiratory tract infection, and chronic bronchitis. Following protein precipitation, DAS was detected by ion transition at m/z 531.2/99.0 in multiple reaction monitoring mode with negative electrospray ionization-tandem mass spectrometry. The limit of detection was 0.5 ng/mL, and the lower limit of quantification was 10 ng/mL in human plasma. Good linearity was maintained over the range of 10–5000 ng/mL, and the correlation coefficient was better than 0.99. The accuracy ranged from 95.3% to 113%, RSD from 0.928% to 6.47%, for the within- and between-run analysis at all QC levels. The recovery ranged from 85.5% to 93.4% and the matrix effect from 107% to 119%. No significant carryover and good stability were found during method validation. The developed method was successfully applied to the determination of DAS in human plasma in a pharmacokinetic study following intravenous infusion of potassium sodium DAS to nine healthy volunteers.  相似文献

黑曲霉Aspergillus niger发酵豆渣产β-葡萄糖苷酶(BGL)。粗酶液依次经过乙醇沉淀和DEAE-Sepharose Fast Flow离子交换层析,获得了两种电泳纯的β-葡萄糖苷酶BGL1和BGL2,它们的相对分子质量分别为102kDa、97kDa,总酶活回收率达到48%。在pH2.5及温度为55℃时,两种BGL水解京尼平苷的速度均达到最大;BGL水解活性受到Na+的激活但不受K+的影响,但Mg2+、Ba2+、Cu2+、Fe2+、Zn2+和Hg+等离子对BGL活性有不同程度的抑制作用(其中对BGL1的抑制普遍强于BGL2),而且同一种离子对不同BGL活性的影响性质不同,Ca2+显著激活BGL1而对BGL2无影响,Fe3+显著抑制BGL1而激活BGL2。在同样条件下,BGL对对硝基苯-β-D吡喃葡萄糖苷(pNPGlu)、对硝基苯-β-D吡喃半乳糖苷(pNPGal)和水杨苷均有催化活性,其中对pNPGlu的Km值最小,BGL1、BGL2分别为0.764和1.934mmol/L;而BGL对京尼平苷水解的催化效率最高,BGL1、BGL2的Kcat/Km值分别为4.28×104和1.04×105L/mol·s。BGL1、BGL2活性的pH稳定范围相近,分别为pH2.0-7.0、pH2.0-8.5,但它们的热稳定性差异较大,55℃时BGL2保温60min,酶活保留了60%,而BGL1的酶活半衰期只有10min。  相似文献

Abstract 1. Climate change has been occurring at unprecedented rates and its impacts on biological populations is beginning to be well documented in the literature. For many species, however, long‐term records are not available, and trends have not been documented. 2. Using museum specimens from southern USA, we show that the stream‐dwelling beetle Gyretes sinuatus has shown an 8% increase in body size and change in body shape (fineness ratio) from 1928 to 1988. Any directional morphological change observed over time could be an indicator of a microevolutionary response. 3. During these 60 years, there have also been changes in temperature, precipitation, and location of collection sites. Unlike the global trend, mean annual temperature in the region has decreased, and furthermore, total annual precipitation has increased. By investigating how these various ecological and geographical variables may affect body size and shape, we can examine which pressures may promote larger and/or thinner beetles. 4. Results indicate that mean annual temperature was the most predictive variable for the change in size and shape. We suggest there is an adaptive role for temperature on body size and shape of stream dwelling organisms. 5. We found that museum specimens can be invaluable resources of information when collection date and location information is available. We promote the use of such specimens for future studies of the morphological response to climate change.  相似文献

As part of global climate change, variation in precipitation in arid ecosystems is leading to plant adaptation in water-use strategies; significant interspecific differences in response will change the plant composition of desert communities. This integrated study on the ecophysiological and individual morphological scale investigated the response, acclimation and adaptation of two desert shrubs, with different water-use strategies, to variations in water conditions. The experiments were carried out on two native dominant desert shrubs, Tamarix ramosissima and Haloxylon ammodendron, under three precipitation treatments (natural, double and no precipitation, respectively), in their original habitats on the southern periphery of Gurbantonggut Desert, Central Asia, during the growing season in 2005. Changes in photosynthesis, transpiration, leaf water potential, water-use efficiency, above-ground biomass accumulation and root distribution of the two species were examined and compared under the contrasting precipitation treatments. There were significant interspecific differences in water-use strategy and maintenance of photosynthesis under variation in precipitation. For the phreatophyte T. ramosissima, physiological activity and biomass accumulation rely on the stable groundwater, which shields it from fluctuation in the water status of the upper soil layers caused by precipitation. For the non-phreatophyte H. ammodendron, efficient morphological adjustment, combined with strong stomatal control, contributes to its acclimation to variation in precipitation. On account of its positive responses to increased precipitation, H. ammodendron is predicted to succeed in interspecific competition in a future, moister habitat.  相似文献

Sulfate reduction rates and biogeochemical parameters of fish farm sediments across the Mediterranean were investigated in the order to evaluate the potential effects of organic matter inputs on habitat quality for the common seagrass Posidonia oceanica. Four study sites were selected in Spain, Italy, Greece and Cyprus to represent the Mediterranean basin. P. oceanica was found in immediate vicinity of all the farms, which were located at physically exposed sites about 1 km from the shore lines. Organic matter accumulation, sulfate reduction rates and sulfur pools were measured in depth profiles along transects from the farms in both bare and vegetated sediments. Results show that although the organic matter accumulation was minor at the sites (POC < 2.8% DW), the sulfate reduction rates were high, in particular at the largest farm in Italy (up to 212 mmol m−2 d−1), similar to rates found at shallower, temperate fish farm sites, where higher sedimentation rates can be expected. Sulfate reducing bacteria in these low-organic, carbonate-rich Mediterranean sediments respond strongly to organic matter loadings and cause habitat degradation. Sulfate reduction rates measured in the P. oceanica sediments were among the highest recorded (7.8–42.0 mmol m−2 d−1) similar to rates found in degrading meadows impacted by organic matter loadings. As sulfate reduction rates were correlated with the sedimentation rates along the transects rather than organic matter pools this suggests mineralization processes were controlled by organic matter loading in fish farm sediments. The vegetated sediments near the net cages were more reduced due to accumulation of sulfides compared to control sites, which is a possible contributing factor to the observed seagrass decline in the farm surroundings. It is recommended that Mediterranean fish farms are placed in areas with rapid dispersal of particulate waste products to minimize organic matter loading of the sediments and thereby preserve habitat quality for benthic fauna and flora.  相似文献
