Summary. We investigated the behavior of migration of Amoeba proteus in an isotropic environment. We found that the trajectory in the migration of A. proteus is smooth in the observation time of 500-1000 s, but its migration every second (the cell velocity) on the trajectory randomly changes. Stochastic analysis of the cell velocity and the turn angle of the trajectory has shown that the histograms of the both variables well fit to Gaussian curves. Supposing a simple model equation for the cell motion, we have estimated the motive force of the migrating cell, which is of the order of piconewton. Furthermore, we have found that the cell velocity and the turn angle have a negative cross-correlation coefficient, which suggests that the amoeba explores better environment by changing frequently its migrating direction at a low speed and it moves rectilinearly to the best environment at a high speed. On the other hand, the model equation has simulated the negative correlation between the cell velocity and the turn angle. This indicates that the apparently rational behavior comes from intrinsic characteristics in the dynamical system where the motive force is not torquelike.  相似文献

Using the patch-clamp technique, we observed profound oscillations of the whole-vacuole outward current across the tonoplast of Mesembryanthemum crystallinum L. (common ice plant). These current oscillations showed a clear voltage dependence and appeared at membrane potentials more positive than 90–100 mV. This paper describes the oscillations in terms of two separate mechanisms. First, the Mesembryanthemum vacuolar membrane shows a negative slope conductance at membrane potentials more positive than 100–120 mV. The fact that the oscillations and the negative slope conductance show a similar threshold potential suggests that (part of) the same mechanism is involved in both phenomena. The second mechanism involved is the voltage drop across the series resistance. As a result, the potential actually experienced by the vacuolar membrane deviates from the command potential defined by the patch-clamp amplifier. This deviation depends in an Ohmic manner on the current magnitude. We suggest that the interplay of the negative slope conductance and the voltage drop across the series resistance can cause a positive feedback which is responsible for the current oscillations. The hexose part of glycolysis has been studied in the freshwater Chinese crab Eriocheir sinensis exposed to high pressure (101 ATA, i.e. 1000 m depth) at 14°C and in normoxic conditions. Glycolytic fluxes (from glucose, JA and from Glucose 6 Phosphate, JB) have been determined using NADH depletion during the conversion of dihydroxy acetone phosphate into α-glycerol phosphate. Measurements have been performed at 14 and 19°C. Pressure exposure induces an increase of glycolytic flux and a decrease of the time needed for the transition from aerobic to anaerobic glycolysis. As a consequence pressure-exposed crabs have a higher potential to increase glycolytic flux than control animals at atmospheric pressure. It is concluded that high pressure known to alter numerous enzymes individually, can also modify an overall metabolic pathway.  相似文献

The objective of this study was to examine the role of dopamine (DA) receptors in the nucleus accumbens (ACB) in controlling feedback regulation of mesolimbic somatodendritic DA release in the ventral tegmental area (VTA) of Wistar rats using ipsilateral dual-probe in vivo microdialysis. Perfusion of the ACB for 60 min with the DA uptake inhibitor GBR-12909 (10-1,000 microM) or nomifensine (10-1,000 microM) dose-dependently increased the extracellular levels of DA in ACB and concomitantly reduced the extracellular levels of DA in the VTA. Coperfusion of 100 microM nomifensine with either 100 microM SCH-23390 (SCH), a D1 antagonist, or 100 microM sulpiride (SUL), a D2 receptor antagonist, produced either an additive (for SCH) or a synergistic (for SUL) elevation in the extracellular levels of DA in the ACB, whereas the reduction in the extracellular levels of DA in the VTA produced by nomifensine alone was completely prevented by addition of either antagonist. Application of 100 microM SCH or SUL alone through the microdialysis probe in the ACB increased the extracellular levels of DA in the ACB, whereas the extracellular levels of DA in the VTA remained unchanged. Overall, the results suggest that (a) increasing the synaptic levels of DA in the ACB activates a long-loop negative feedback pathway to the VTA involving both D1 and D2 postsynaptic receptors and (b) terminal DA release within the ACB is regulated directly by D2 autoreceptors and may be indirectly regulated by D1 receptors, possibly on interneurons and/or through postsynaptic inhibition of the negative feedback loop.  相似文献

Motility disorders due to tail defects are often seen in clinical andrology. Sperm motility should be assessed with regard to the morphology of the flagellum. Since suitable longitudinal sections are rarely obtained by routine transmission electron microscopy (TEM) and in view of the importance of dense fibres in modulating sperm motility and providing tensile strength, a detailed, study of human sperm flagellum by negative staining andTEM was attempted. The study was undertaken in two groups of men (I) fertile and (II) asthenozoospermic. The study revealed that outer dense fibres extend to 50–60% of the principal piece. Normal dense fibres were seen in 83% sperms and 23% sperms in groupsI andII respectively. The characteristics seen were variation in diameter, breakage or degradation with lacking or extended endpiece. The negative staining method provides an easy and useful analytical tool for identifying the defects of dense fibres and quantifying them.  相似文献

The main goal of this paper is to investigate a cure rate model that comprehends some well‐known proposals found in the literature. In our work the number of competing causes of the event of interest follows the negative binomial distribution. The model is conveniently reparametrized through the cured fraction, which is then linked to covariates by means of the logistic link. We explore the use of Markov chain Monte Carlo methods to develop a Bayesian analysis in the proposed model. The procedure is illustrated with a numerical example.  相似文献

The amount of sample available for clinical and biological proteomic research is often limited and thus significantly restricts clinical and translational research. Recently, we have integrated pressure cycling technology (PCT) assisted sample preparation and SWATH‐MS to perform reproducible proteomic quantification of biopsy‐level tissue samples. Here, we further evaluated the minimal sample requirement of the PCT‐SWATH method using various types of samples, including cultured cells (HeLa, K562, and U251, 500 000 to 50 000 cells) and tissue samples (mouse liver, heart, brain, and human kidney, 3–0.2 mg). The data show that as few as 50 000 human cells and 0.2–0.5 mg of wet mouse and human tissues produced peptide samples sufficient for multiple SWATH‐MS analyses at optimal sample load applied to the system. Generally, the reproducibility of the method increased with decreasing tissue sample amounts. The SWATH maps acquired from peptides derived from samples of varying sizes were essentially identical based on the number, type, and quantity of identified peptides. In conclusion, we determined the minimal sample required for optimal PCT‐SWATH analyses, and found smaller sample size achieved higher quantitative accuracy.  相似文献

目的:研究个性化护理干预对乳腺癌手术患者负性情绪和睡眠质量的影响。方法:选取2013 年1 月-2015 年1 月我院收治 的乳腺癌手术患者100 例,根据术后干预方式不同,将患者分为研究组和对照组,每组各50 例。对照组患者给予常规护理,研究 组患者给予个性化护理。应用焦虑自评量表(SAS)评价患者的焦虑情况,应用抑郁自评量表(SDS)评价患者的抑郁情况,应用匹 兹堡睡眠质量指数(PSQI)评价患者的睡眠质量,并比较两组患者对护理的满意度。结果:研究组护理满意度显著高于对照组,差 异具有统计学意义(P<0.05);两组干预前SAS、SDS 和PSQI 评分无统计学差异(P>0.05);干预后两组SAS、SDS 和PSQI 评分 均显著降低,且研究组低于对照组,差异具有统计学意义(P<0.05)。结论:个性化护理干预能显著改善乳腺癌手术患者的负面情 绪,提高患者的睡眠质量。  相似文献

A high-performance liquid chromatography–tandem mass spectrometry (HPLC–MS–MS) method, using back-flush column-switching was developed for total drug concentrations of ropivacaine in serum and drainage blood in the measuring range 0.1–10 μg/mL. Samples were diluted with internal standard (2H7-ropivacaine) and extraction buffer, centrifuged and injected directly onto a BioTrap 500 MS extraction column. Using a time programmed six-port valve switch, ropivacaine was back-flushed onto a Zorbax SB-Aq analytical column, gradient eluted and finally detected after electro spray ionisation and multiple reaction monitoring (MRM) of the transitions m/z 275 → m/z 126 and m/z 282 → m/z 133 for ropivacaine and 2H7-ropivacaine, respectively. Accuracy (bias-%) was −1.5 to 5.8% and intermediate precision (C.V.) was 1.4–3.1%. The low sample amount required (10 μL), high specificity and short run time (6 min) makes it very suitable for determination of ropivacaine. Using the same methodology as described above and 200 μL ultrafiltrate, the free drug concentrations of ropivacaine in serum could be precisely determined with a C.V. below 3%. The method was used to investigate the safety of reinfusion of drainage blood after knee and hip arthroplasty when ropivacaine (Naropin®) was used for local analgesia. 目的:通过模型加压兔子肾脏,监测加压前后肾功能变化,论证压力对肾功能的影响,根据实验结果给多囊肾临床治疗提供借鉴。方法:将成年新西兰兔随机分为三组(A,B,C),A组兔子切除一侧肾脏,另一侧肾脏通过模型固定圆形气泡予肾脏加压;B组兔子切除一侧肾脏,另一侧肾脏通过模型固定塑料圆球予肾脏加压;C组兔子切除一侧肾脏,另一侧肾脏不予加压处理。分别监测实验组和对照组兔子术前术后肾功的变化(血肌酐、尿素)。对不同加压肾脏标本进行HE、masson和六氨银染色,了解肾脏加压后的病理改变。结果:实验组A行加压术后肾功相对术前与对照组C相比无明显差异;实验组B行加压术后,肌酐、尿素升高明显,与术前、实验组A、对照组C比较均有明显差异(P<0.01)。实验组C一侧肾切除后相比术前肾功能无明显差异。病理示:肾脏加压后,肾小球、肾小管、肾血管继发一系列损害肾功的病理改变。结论:对肾脏施加的压力足够大的时候,压力致使肾脏发生相应病理改变,肌酐、尿素升高,肾功能明显下降。因此传统的囊肿去顶减压术,仍是有意义的对症治疗手段。  相似文献
