Impact and management of the eriophyoid mite Trisetacus juniperinus on the evergreen cypress Cupressus sempervirens

Abstract 1 In the Mediterranean region, the eriophyoid mite Trisetacus juniperinus causes considerable damage to the evergreen cypress, Cupressus sempervirens L., particularly in nurseries and young stands, disturbing the apical growth of the tree. 2 The impact of mites on survival and apical growth of two commercial clones of cypress (Agrimed and Bolgheri), as well as the results of differently timed pesticide applications to suppress mite population on newly grafted trees, were evaluated. 3 Mites easily infested clonal scions from rootstocks that were previously infested in the nursery, inducing tip deformation and disturbance of the growth. Apical growth was significantly lower in infested than in control trees 2 years following the graft. 4 Deformed apical buds were left early by mites, which dispersed in the crown and may have incurred high mortality. This is interpreted as a defensive reaction of the cypress to the mite attack, which involves costs resulting in reduced apical growth in both clones. However, mites partly overcame tree defences in the Bolgheri clone. 5 Healthy rootstock and graft material should be used to limit damage and maintain plant growth because natural infestations rarely occur. In case of attack, a pesticide should be applied as soon as possible because precocious mite infestation has a log‐lasting effect on tree growth, with considerable economic damage.     

人细胞差异表达基因cDNA消减杂交体系的建立

为克隆肺腺癌分化相关基因, 采用诱导分化与消减杂交相结合的策略, 建立了全反式维甲酸(RA)诱导前后人肺腺癌细胞系的cDNA消减文库, 得到124个cDNA消减克隆. 经加减法杂交差异筛选、DNA和RNA印迹、cDNA全序列测定和生物学功能分析, 分离到3个在人肺腺癌细胞系分化过程中由RA激活而特异表达的新的cDNA序列这一策略和技术路线适用于分离细胞中呈过量表达或表达抑制基因的cDNA克隆, 并具有反映细胞分化过程中基因表达动态变化特征和相对简便适用的特点.     

Oral administration of antigen does not influence the proliferation and IFN-γ production of responsive CD8+ T cells but enables to establish T cell clones with different lymphokine production profile.

Feeding of a whole casein diet, which abolished the α_s1-casein-specific proliferation and IFN-γ productivity of CD⁴⁺ T cells, did not affect the proliferative response of CD8⁺ T cells with regard to the antigen dose response, cell dose response, kinetics of the proliferation and epitope specificity, as well as IFN-γ production. To assess the characteristics of the CD8⁺ T cells, we established α_s1-casein-specific CD8⁺ T cell clones from both casein-fed and control mice. The established clones produced different amount of IFN-γ and IL-10, and one clone derived from the casein-fed mice produced a remarkable amount of IL-10. The clones from casein-fed mice produced considerable amounts of TGF-β, while those from control mice produced only small amounts. The possible role of CD8⁺ T cells in oral tolerance is discussed. Characterization of isolates and clones of leishmania by analysis of kinetoplast DNA

The genetic characterization of pathogenic isolates of Leishmania was attempted by analysis of the molecular properties of kinetoplast DNA (kDNA) minicircles. Unit minicircle size is not conserved during speciation of Leishmania since the minicircles of strains and clones of L t major are smaller (700 bp) than those found in certain strains of L mexicana ssp (820 bp), L donovani (850 bp) or L t tropica (900 bp). Schizodeme analysis of minicircles reveals a high degree of sequence divergence in kDNA of Leishmania with the degree of microheterogeneity varying between species. This sequence divergence allows the discrimination of species, strains, and clones of Leishmania into schizodemes. Southern blot hybridization experiments reveal that at high stringency overall minicircle sequence homology is conserved among clones and strains of one species (L t major) but not between different species. This property of minicircle DNA permits the use of kDNA probes as a species-specific diagnostic test for the identification of unknown Leishmania isolates. The properties of kDNA from an L t tropica strain LRC-L32 (a “recidiva” organism) are so diverged from those of L t major strains as to support the classification [22,23] of L t tropica and L t major as separate species of Leishmania rather than subspecies of L tropica.     

Two Possibly Distinct Prostaglandin E1 Receptors in N1E-115 Clone: One Mediating Inositol Trisphosphate Formation, Cyclic GMP Formation, and Intracellular Calcium Mobilization and the Other Mediating Cyclic AMP Formation

Prostaglandin E1 (PGE1)-mediated transmembrane signal control systems were investigated in intact murine neuroblastoma cells (clone N1E-115). PGE1 increased intracellular levels of total inositol phosphates (IP), cyclic GMP, cyclic AMP, and calcium ([Ca2+]i). PGE1 transiently increased inositol 1,4,5-trisphosphate formation, peaking at 20 s. There was more than a 10-fold difference between the ED50 for PGE1 at cyclic AMP formation (70 nM) and its ED50 values at IP accumulation (1 microM), cyclic GMP formation (2 microM), and [Ca2+]i increase (5 microM). PGE1-mediated IP accumulation, cyclic GMP formation, and [Ca2+]i increase depended on both the concentration of PGE1 and extracellular calcium ions. PGE1 had more potent intrinsic activity in cyclic AMP formation, IP accumulation, and cyclic GMP formation than did PGE2, PGF2 alpha, or PGD2. A protein kinase C activator, 4 beta-phorbol 12 beta-myristate 13 alpha-acetate, had opposite effects on PGE1-mediated IP release and cyclic GMP formation (inhibitory) and cyclic AMP formation (stimulatory). These data suggest that there may be subtypes of the PGE1 receptor in this clone: a high-affinity receptor mediating cyclic AMP formation, and a low-affinity receptor mediating IP accumulation, cyclic GMP formation, and intracellular calcium mobilization.     

大鼠肾脏组织cDNA文库的制备和葡萄糖转运蛋白cDNA克隆的筛选

 大鼠的肾脏组织经匀浆后,提取纯化总RNA和mRNA合成cDNA,进行甲基化,加人工接头,最后连入载体(λgt11)和外壳蛋白包装步骤制成肾脏组织的cDNA文库。插入载体的cDNA长度为0.5kb到8kb之间。经反复稀释,测定出该文库的效价为1.5×10~7pfu/mL。     

Variability in the transmission efficiency of potyviruses by different clones ofAphis gossypii

A sample of seventy-twoAphis gossypii Glover (Homoptera: Aphididae) clones was collected in south-eastern France. The efficiency of these clones to transmit a potyvirus (papaya ringspot virus T-strain) was assessed in controlled conditions. In a first screening, the virus was transmitted by all clones and a 3.5-fold difference between the most and least efficient clones was obtained. During subsequent trials, which were carried out to confirm the differences in the transmission efficiency of these clones, only one clone proved to be more efficient than the others. This difference appeared consistent over a 1-year period, and was also confirmed with 4 other related potyviruses.

Résumé Un échantillon de 72 clones deA. gossypii a été collecté dans le Sud-Est de la France. L'efficacité de transmission d'un potyvirus (PRSV-T) a été mesurée en conditions contr?lées pour chacun de ces clones. Un premier screening a permis de montrer que tous les clones transmettaient ce virus, et qu'un rapport de 3,5 existait entre l'efficacité de transmission du clone le plus efficace et celle du clone le moins efficace. Au cours des essais ultérieurs destinés à confirmer ces différences, un seul clone s'est montré significativement plus efficace que les autres. Cette différence s'est maintenue pendant la période d'essais (1 an). Elle s'est reproduite avec 4 autre potyvirus apparentés.
