Glycogen phosphorylase in the vegetative mycelium ofFlammulina velutipes converts glycogen to α-glucose 1-phosphate (G1P) in the colony during fruit-body development. Glycogen may contribute to the synthesis of trehalose as the starting material in the vegetative mycelium during the fruiting process of the colony, and the trehalose produced is translocated into the fruit-bodies as the main carbohydrate substrate for their development. Trehalose phosphorylase activity in the vegetative mycelium was at a relatively high level until fruit-body initiation, suggesting the turnover of this disaccharide during the vegetative stage of the colony development. Trehalose phosphorylase activity in the stipes showed a peak level at the early phase of fruit-body development, suggesting the continuing phosphorolysis of trehalose by this enzyme. The stipes also showed a high specific activity of phosphoglucomutase at a sufficient level to facilitate the conversion of G1P to α-glucose 6-phosphate (G6P). In the pilei a large amount of G1P remained until the growth of the fruit-bodies ceased. Trehalase activities in the stipes and pilei were at a very low level, and this enzyme may not contribute to the catabolism of trehalose in the fruit-body development.  相似文献

Arabinogalactan-proteins (AGPs) are highly diverse plant proteoglycans found on the plant cell surface. AGPs have large arabinogalactan (AG) moieties attached to a core-protein rich in hydroxyproline (Hyp). The AG undergoes hydrolysis by various glycoside hydrolases, most of which have been identified, whereas the core-proteins is presumably degraded by unknown proteases/peptidases secreted from fungi and bacteria in nature. Although several enzymes hydrolyzing other Hyp-rich proteins are known, the enzymes acting on the core-proteins of AGPs remain to be identified. The present study describes the detection of protease/peptidase activity toward AGP core-proteins in the culture medium of winter mushroom (Flammulina velutipes) and partial purification of the enzyme by several conventional chromatography steps. The enzyme showed higher activity toward Hyp residues than toward proline and alanine residues and acted on core-proteins prepared from gum arabic. Since the activity was inhibited in the presence of Pefabloc SC, the enzyme is probably a serine protease.  相似文献

The role of lipids and carbohydrates in the adaptation of Flammulina velutipes to hypothermia (5 to −5°C) in natural environments has been studied for the first time. The main changes are were found to occur in membrane lipids: the levels of sterols and glycolipids decreased, and the proportion of phospholipids with a high degree of nonsaturation (2.2) increased, which was due to predominance of two fatty acids, linoleic (35% of the total) and linolenic (50%). Phosphatidylcholine became the major phospholipid. Under hypothermic conditions, glycerol, known to have antifreeze properties, accumulated in the cell cytosol along with arabitol and trehalose. 菌柄是金针菇等食用菌的主要商品部位,但其生长机制仍不明确。本研究对金针菇伸长期和成熟期菌柄进行了转录组联合蛋白组分析,结果显示,两样本显著性差异表达基因和蛋白分别为721个和61个,均以上调表达为主。GO(gene ontology)功能聚类分析表明:有72.41%的差异表达基因富集在催化活性(catalytic activity)条目下。细胞组分(cell part)和绑定结合(binding)条目同时富集了较多的差异表达基因和蛋白。KEGG通路富集分析显示:碳水化合物代谢通路(carbohydrate metabolism)和氨基酸代谢通路(amino acid metabolism)富集的差异表达基因较多。差异表达蛋白富集较多的通路是单环菌素生物合成(monobactam biosynthesis,ko00261)、链霉素生物合成(streptomycin biosynthesis,ko00521)和有机含硒化合物代谢(selenocompound metabolism,ko00450)等。内质网蛋白质加工(protein processing in endoplasmic reticulum,ko04141)和MAPK信号通路(MAPK signaling pathway-yeast,ko04011)在转录组和蛋白组的KEGG富集分析中均为差异通路。本研究联合转录组和蛋白组数据筛选了40个金针菇菌柄发育中差异表达基因,为深入研究揭示食用菌菌柄发育过程提供候选基因。  相似文献

Qualitative and quantitative characteristics of the alterations in the lipid composition of the membrane of the basidial fungus Flammulina velutipes in the course of surface culture development were investigated. Modifications of the lipid composition were shown to be timed to specific ontogeny stages, such as changes in the growth rate of the colonies, the appearance of differentiated vegetative cells, and the formation of generative structures. A slowdown of growth correlated with an alteration in the ratio of major classes of phospholipids, namely, with a decrease of phosphatidylcholine relative content and an increase in phosphatidylethanolamines. The differentiation of vegetative cells of the mycelium proceeded along with modifications of molecular composition of glycoceramides. In the course of the first week of growth, the surface culture of F. velutipes produced monohexosylceramides with epoxidized methyl sphingadienine as a sphingoid base. Later on, along with culture growth and specialization of mycelium cells, molecular species with methyl sphingadienine, common for basidiomycetes, start to prevail among the fungal glycoceramides. The formation of fruit bodies is accompanied by enrichment of molecules of phospholipids, mainly, the phosphatidylcholines, with unsaturated fatty acids.  相似文献

金针菇的多酚氧化酶(PPO)、过氧化物酶(POD)和过氧化氢酶(CAT)活性在全株中呈现区域化分布。菌盖的酶活性最低,菌柄上部酶活性稍高,菌柄中部较高,菌柄下部活性最高。用硫脲、亚硫酸盐、CaCl2、低CO2、低温等方法处理金针菇,对以上三种酶的活性均有抑制作用。高02、H2O2和温度的提高均促进三种酶活性。高N2(减压后充N2)不能抑制三种酶活性。  相似文献

Our previous study revealed that, when exposed to light from below, fruit-bodies of humus-borne agarics grow straight downward both in the ‘Coprinus Type’ and ’Non-Coprinus Type’ species and that, in the latter, gills tilt by gravitropism, caps swell and wave, and stipes twist. The present study revealed that fruit-bodies of some lignicolous agarics also grew straight downward. Among themFlammulina velutipes andHypsizygus marmoreus fruit-bodies showed almost the same behaviour as the ‘Non-Coprinus Type’, butPleurotus ostreatus (assumedPolyporus Type) fruit-bodies did not show the gill tilting and cap waving. Rather, it redifferentiated to produce new gills or new fruit-bodies, possibly by gravimorphogenesis, or formed spiral stipes. The biochemical mechanism underlying the development of fruiting bodies in Flammulina velutipes, an edible mushroom, was investigated using the YBLB colorimetric assay to distinguish between the normal strain (FVN-1) and the degenerate strain (FVD-1). In this assay, the color of the YBLB medium (blue-green) inoculated with FVN-1 exhibiting normal fruiting body development changed to yellow, while the color of the medium inoculated with FVD-1 changed to blue. In this study, we found that this color difference originated from extracellular laccase produced by FVN-1. Moreover, FVN-1 exhibited considerably higher extracellular laccase activity than FVD-1, under conditions facilitating fruiting body formation. Overall, these findings suggest that extracellular laccase is involved in the fruiting body development process in F. velutipes.  相似文献
