Mineralogical and geochemical characteristics of tailings from a gold mine in northeastern Thailand were investigated in relation to acid mine drainage (AMD) and the release potentials of toxic elements. The tailings can be divided into upper tailings and lower tailings. The upper tailings usually contain pyrrhotite, pyrite ± chalcopyrite, calcite, quartz, andradite and diopside. The lower tailings mainly contain goethite, quartz, chlorite, muscovite, calcite and hematite ± pyrrhotite. These assemblages clearly relate to the original types of gold deposit prior to mining and mineral processing. The upper tailings are defined as potential acid forming (PAF), whereas the lower tailings are classified as non-acid forming (NAF). Regarding heavy metals, apart from high Mn level, the other heavy metals appear to have low concentrations in the upper tailings. On the other hand, the lower tailings contain high contents of As, Cu and Pb, which appear to be higher than the National Total Threshold Limit Concentrations. Goethite, the main mineral assemblage in the lower tailings, reveals characteristic of arsenic adsorbent. As a result, the tailing pond is recommended to be covered to prevent the oxidizing processes of the upper tailings; otherwise, AMD generation may take place soon after the mine closure. Land reclamation and monitoring plans must be planned very well and carried out with great care since arsenic contamination has been reported in steam water close to the tailing dam.  相似文献

Age-related macular degeneration is the leading cause of blindness in the elderly. The Y402H polymorphism in complement factor H promotes disease-like pathogenesis, and a Cfh+/− murine model can replicate this phenotype, but only after two years. We reasoned that by combining CFH deficiency with cigarette smoke exposure, we might be able to accelerate disease progression to facilitate preclinical research in this disease. Wild-type and Cfh+/− mice were exposed to nose-only cigarette smoke for three months. Retinal tissue morphology and visual function were evaluated by optical coherence tomography, fundus photography and autofluorescence, and electroretinogram. Retinal pigment epithelial cell phenotype and ultrastructure were evaluated by immunofluorescence staining and transmission electron microscopy. Cfh+/− smoking mice showed a dome-like protruding lesion at the ellipsoid zone (drusen-like deposition), many retinal hyper-autofluorescence spots, and a marked decrease in A- and B-wave amplitudes. Compared with non-smoking mice, wild-type and Cfh+/− smoking mice showed sub-retinal pigment epithelium complement protein 3 deposition, activation of microglia, metabolic waste accumulation, and impairment of tight junctions. Microglia cells migrated into the photoreceptor outer segment layer in Cfh+/− smoking mice showed increased activation. Our results suggest that exposing Cfh+/− mice to smoking leads to earlier onset of age-related macular degeneration than in other animal models, which may facilitate preclinical research into the pathophysiology and treatment of this disease.  相似文献

Stromal cell-derived factor-1 (SDF-1) and CXC chemokine receptor 4 (CXCR4) have been found to be tightly correlated with the progression of prostate cancer (PC). In this study, we investigated the effects of an SDF-1α/CXCR4 inhibitor, AMD3100, on cell progression and metastasis potential of human PC cells. Human PC cell lines (LNCaP, PC3, and DU145) were cultured to detect SDF-1α/CXCR4, which showed higher SDF-1α and CXCR4 expression than the normal human prostate epithelial cell line, RWPE-1. AMD3100 was confirmed to be an inhibitor of SDF-1α, and to detect the effect of SDF-1α/CXCR4 inhibition on PC, PC cells were treated with AMD3100 or/and CXCR4 siRNA. The results suggested that inhibition of the SDF-1α/CXCR4 pathway could promote the E-cadherin level but inhibit the levels of invasion and migration of vimentin, N-cadherin and α5β1 integrin. Finally, tumor formation in nude mice was conducted, and the cell experiment results were verfied. These data show that AMD3100 suppresses epithelial–mesenchymal transition and migration of PC cells by inhibiting the SDF-1α/CXCR4 signaling pathway, which provides a clinical target in the treatment of PC.  相似文献

用低浓度的放线菌素D(AMD)处理人的外周血淋巴细胞,可以导致细胞周期各阶段核仁形成区(NOR)的银可染性显著减少,表明了放线菌素D对rDNA的转录活性具有明显的抑制作用。本实验采用IBAS图象分析系统,对经不同浓度的放线菌素D处理过的外周血淋巴细胞进行形态学测量和计算机统计分析,其结果阐述了细胞中NOR的银可染性(即rDNA的转录活性指标)与AMD浓度梯度的相关性,并以此建立了相应的回归方程。本文结果表明,间期的银染核仁面积是检测rDNA转录活性较为敏感的指标。  相似文献

C-X-C motif chemokine 12/C-X-C chemokine receptor type 4 (CXCL12/CXCR4) signaling is involved in ontogenesis, hematopoiesis, immune function and cancer. Recently, the orphan chemokine CXCL14 was reported to inhibit CXCL12-induced chemotaxis – probably by allosteric modulation of CXCR4. We thus examined the effects of CXCL14 on CXCR4 regulation and function using CXCR4-transfected human embryonic kidney (HEK293) cells and Jurkat T cells. CXCL14 did not affect dose–response profiles of CXCL12-induced CXCR4 phosphorylation, G protein-mediated calcium mobilization, dynamic mass redistribution, kinetics of extracellular signal-regulated kinase 1 (ERK1) and ERK2 phosphorylation or CXCR4 internalization. Hence, essential CXCL12-operated functions of CXCR4 are insensitive to CXCL14, suggesting that interactions of CXCL12 and CXCL14 pathways depend on a yet to be identified CXCL14 receptor.  相似文献

Background

An increasing body of studies has assessed the contribution of Val62Ile polymorphism to age-related macular degeneration (AMD) risk, but the exact association still remains uncertain. This meta-analysis was undertaken in order to further characterize the potential association between Val62Ile polymorphism and AMD risk in four different ethnic populations.

Methods

A meta-analysis was performed using data available from 16 case–control studies evaluating correlation between the Val62Ile polymorphism and AMD in Caucasian, Chinese, Japanese and South Korean populations. Data extraction and study quality assessment were performed in duplicate. Summary odds ratios (ORs) and 95% confidence intervals (CIs) of allele contrast and genotype contrast were estimated using the random-effects model. The Q-statistic test was used to identify heterogeneity, and the funnel plot was adopted to evaluate publication bias.

Results

Sixteen studies involving a total of 11,400 subjects based on the search criteria were included in the meta-analysis. In overall populations, the Val62Ile polymorphism seemed to be associated with AMD (ORAA vs. GG = 0.40, 95% CI = 0.28–0.59; ORAA + GA vs. GG = 0.72, 95% CI = 0.64–0.80; ORAA vs. GC + GG = 0.50, 95% CI = 0.36–0.70; ORA vs. G = 0.68, 95% CI = 0.58–0.78; ORGA vs. GG = 0.71, 95% CI = 0.65–0.77). Similarly, subgroup analysis also revealed that this polymorphism was related to AMD in all ethnicities.

Conclusions

This meta-analysis suggested that Val62Ile polymorphism was associated with susceptibility to AMD.  相似文献

目的探讨AMD3100阻断SDF-1/CXCR4轴后,对局灶脑缺血/再灌注大鼠缺血半暗带血管再生的影响。方法将SD大鼠随机分为假手术组(S组)、模型组(IR组)、AMD3100组(IRA组)、生理盐水组(IRN组)。采用线栓法制备大鼠局灶脑缺血/再灌注模型,缺血2h后将IR、IRA和IRN组分为再灌注12h,1、3和7d四个亚组。HE染色观察局灶脑缺血/再灌注后大脑皮质病理变化。免疫组化法检测CD31在缺血半暗带表达。荧光定量PCR检测外周血中AC133mRNA表达。结果与IRN组比较,IRA 12h外周血中AC133mRNA显著升高,第1d升高达峰值(P0.01),IRA 3dAC133mRNA表达比IRA1d显著减少(P0.05);与IRN组比较,IRA组CD31阳性血管密度在第1d无显著变化(P0.05),第3和7d血管密度显著减少(P0.01);IRA 7d梗死区由大量坏死神经细胞和泡沫细胞填充,坏死较严重。结论持续注射AMD3100能动员干/祖细胞快速进入外周血,但可能抑制局灶脑缺血/再灌注大鼠缺血半暗带血管再生,加重梗死区坏死。  相似文献

Lysosome-associated membrane protein-2 (LAMP2), is a highly glycosylated lysosomal membrane protein involved in chaperone mediated autophagy. Mutations of LAMP2 cause the classic triad of myopathy, cardiomyopathy and encephalopathy of Danon disease (DD). Additionally, retinopathy has also been observed in young DD patients, leading to vision loss. Emerging evidence show LAMP2-deficiency to be involved in oxidative stress (ROS) but the mechanism remains obscure. In the present study, we found that tert-butyl hydroperoxide or antimycin A induced more cell death in LAMP2 knockdown (LAMP2-KD) than in control ARPE-19 cells. Mechanistically, LAMP2-KD reduced the concentration of cytosolic cysteine, resulting in low glutathione (GSH), inferior antioxidant capability and mitochondrial lipid peroxidation. ROS induced RPE cell death through ferroptosis. Inhibition of glutathione peroxidase 4 (GPx4) increased lethality in LAMP2-KD cells compared to controls. Cysteine and glutamine supplementation restored GSH and prevented ROS-induced cell death of LAMP2-KD RPE cells.  相似文献

The diversity and activity of dissimilatory Fe(III)-reducing bacteria was investigated in acidic, ochre-precipitating springs on Mam Tor, East Midlands, UK. The springs at this acid rock drainage site are located below a 3000 year old landslip, where biooxidation of exposed pyrite-containing minerals has resulted in the production of metal-laden acidic waters. A diverse microbial community was found downstream in the sediments dominated by Fe(III) minerals, and included close relatives to known acidophilic (Acidimicrobium and Acidiphilium) and neutraphilic (Geobacter and Pelobacter) Fe(III)-reducing bacteria. Analysis by XRD and TEM confirmed the presence of both amorphous and well-defined Fe(III) mineral phases in the sediments including lepidocrocite, goethite and schwertmannite. Microcosm-based experiments demonstrated that the bioavailable Fe(III) was reduced under anaerobic conditions, concomitant with sulphate release. XRD analysis suggested that schwertmannite (an iron sulphate hydroxide) was utilized preferentially by the Fe(III)-reducing bacteria, leading to the release of sulphate. Although the microcosms contained sufficient concentrations of naturally occurring electron donor to sustain significant levels of Fe(III) reduction, this process was stimulated by the addition of glycerol and complex electron donors. Thus, the acidic Fe(III)-containing sediments contain a diversity of DIRBs that can be stimulated by the addition of electron donor as a first step in the reversal of acid rock and acid mine drainage contamination.  相似文献
