六种选矿药剂对藻类的毒性比较

本文评价了六种选矿药剂对藻类的毒性效应,它们对斜生栅藻的毒性大小排列顺序是:二号油>Fu>0145>Yx>MPA>S-808,96h-EC_(50)值分别为41.2,50.1,82.0,177.8,198.2和900ppm。六种选矿药剂对藻类毒性最大的是二号油,毒性最小的是S-808。对藻类细胞形态的观察结果表明,0145对藻类的细胞形态有轻度的致畸效应,在其它五种药剂的培养物中,均未发现畸变细胞。在室温下存放10d后MPA,0145和二号油,毒性明显下降,其下降速率的顺序是:MPA>0145>二号油。藻类对S-808具有净化脱色作用,100ppm浓度的S-808溶液经藻类作用32d后,其色度可减少48％,作用62d和93d,色度分别降低54％和58％。0145抑制藻类的光合放氧,经50,100和200ppm浓度的0145处理4h后,与对照相比,藻类的光合放氧速率分别下降15、34和36％。     

人体和动物模型的体表物理信息地形图的研究

对人体头面、躯干、四肢、耳廓各局部几十个及整个人体等体表部位正、背面等210个部位进行超微弱冷光和温度测量,输入电子计算机,经特殊的自编程序处理,获得十分清晰的,由3000多数据构成的各个局部或人体整体的冷光和温度地形图。 对家兔左、右耳廓、胸腹部、背部都分别观察32个部位的冷光与体表温度,经计算机分析处理,每观察区域获得约由2000个数据构成的精确的冷光、温度地形分市图。并可见不同生理、病理状态及不同病程家兔体表冷光、温度等地形图呈有规律的改变。 此外,我们还编制了以体表左右相应对称部位差值为分析数据进行地形图分析的程序,用以人体和动物体表物理信息对称规律的研究。 本工作以图形的形式显示物理参量在体表的广泛的分布规律,以揭示机体内部的不同生理、病理状态。本方法定位准确、直观醒目,为研究体表信息及机体生命活动规律提供了与逐点直接测量方法相互补充的有益的新手段。     

人α1/158Ⅴ型(α1c型)干扰素基因在大肠杆菌中的表达及其产物的初步纯化

采用DNA聚合酶链反应(PCR)技术,将本实验室从中国人胎肝细胞染色体DNA中发现和分离的IFN-α1/158V基因的原始克隆,改造成适于进行非融合蛋白原核表达的结构形式,并在大肠杆菌中获得高效表达。测得重组IFN-α1/158V的抗病毒活性为1.9×10~7单位/升菌液。随后又采用以单克隆抗体亲和层析为主的纯化流程对表达产物进行初步纯化,获得了在SDS-聚丙烯酰胺凝胶电泳上呈现单一条带的纯化产物。     

The TIS11 primary response gene is a member of a gene family that encodes proteins with a highly conserved sequence containing an unusual Cys-His repeat.

The TIS11 primary response gene is rapidly and transiently induced by both 12-O-tetradecanoylphorbol-13-acetate and growth factors. The predicted TIS11 protein contains a 6-amino-acid repeat, YKTELC. We cloned two additional cDNAs, TIS11b and TIS11d, that contain the YKTELC sequence. TIS11, TIS11b, and TIS11d proteins share a 67-amino-acid region of sequence similarity that includes the YKTELC repeat and two cysteine-histidine containing repeats. TIS11 gene family members are not coordinately expressed: (i) unlike TIS11, the TIS11b and TIS11d mRNAs are detectable in quiescent Swiss 3T3 cells and are not dramatically induced by 12-O-tetradecanoylphorbol-13-acetate; (ii) cycloheximide superinduction does not occur for TIS11b and TIS11d; and (iii) unlike TIS11, TIS11b expression is extinguished in PC12 pheochromocytoma cells.     

Molecular cloning and characterization of the gene encoding the adenine methyltransferase M.CviRI from Chlorella virus XZ-6E.

The gene encoding the DNA methyltransferase M.CviRI from Chlorella virus XZ-6E was cloned and expressed in Escherichia coli. M.CviRI methylates adenine in TGCA sequences. DNA containing the M.CviRI gene was sequenced and a single open reading frame of 1137 bp was identified which could code for a polypeptide of 379 amino acids with a predicted molecular weight of 42,814. Comparison of the M.CviRI predicted amino acid sequence with another Chlorella virus and 14 bacterial adenine methyltransferases revealed extensive similarity to the other Chlorella virus enzyme.

The effect of alpha-hANF on contractile force and action potential duration (APD) of isolated myocardium in guinea pig was investigated. At the level of 20 nmol/L and 40 nmol/L alpha-hANF, the contraction amplitude and rate of contraction phase decreased to a remarkable extent. And at 40 nmol/L, APDs were shortened significantly 20 min after alpha-hANF. The results suggest that there exists a direct inhibitory effect of alpha-hANF on myocardial contractile force and action potential duration.     

Isozymes of phosphorylase kinase in rabbit skeletal muscle. Functional implications of differences in phosphorylase kinase and phosphorylase activities in individual muscle fibers

Immunological and microanalytical methods were used to investigate the two isozymes of phosphorylase kinase, enzyme w and enzyme r, in psoas major and tibialis anterior muscles. Peptide mapping experiments indicated that the alpha subunit of enzyme w and alpha' subunit of enzyme r were structurally very similar. Both subunits were completely immunoprecipitated from muscle extracts with an antibody specific for the beta subunit of the kinase, indicating that alpha and alpha' subunits are completely assembled with beta subunits in adult muscle fibers. The relative amounts of enzymes w and r in single fibers were determined from amounts of alpha and alpha' subunits, which were detected by immunoblotting. Phosphorylase kinase and phosphorylase activities were measured in the same fibers, as well as in individual fibers from diaphragm and soleus muscles. Slow oxidative fibers were found to contain low levels of enzyme r, but almost no enzyme w. Considerably more enzyme r was present in fast oxidative-glycolytic fibers. Fast glycolytic fibers contained the most enzyme w, and the highest levels of enzyme r were found in a subgroup of such fibers. Interestingly, more than half of the fast glycolytic fibers analyzed contained both isozymes. In these fibers phosphorylase was positively correlated with enzyme w, but negatively correlated with enzyme r. Total kinase activity ranged 30-fold from the highest in one of the psoas fibers to the lowest in one of the soleus fibers and was closely correlated with the phosphorylase levels. In psoas and soleus fibers, calculated absolute maximal rates for phosphorylase b to a conversion varied almost 2,500-fold.     

慢性蜜蜂麻痹病病毒核酸与蛋白质组分的研究

从自然感染的意大利麻痹病蜂(APis mellifera)头部,经二次差速离心与蔗糖梯度离心获得纯化的慢性蜜蜂麻痹病病毒(CBPV)。纯化的CBPV制备物感染正常蜜蜂,4天后出现典型的麻痹症状,接着死亡,平均死亡率分别为95％与100％。SDS-聚丙烯酰胺凝胶电泳分析,二次差速离心初步纯化的病毒制备物含有多条蛋白带,而蔗糖密度梯度纯化的病毒制备物仅含有单一的多肽带。5％、7.5％与10％SDS-聚丙烯酰胺凝胶电泳分析,均检测出一种病毒蛋白质,分子量大约为24,200道尔顿,而且不同凝胶浓度检测的蛋白质分子量相近。慢性蜜蜂麻痹病病毒核酸也用SDS-聚丙烯酰胺凝胶电泳分析,结果表明,凝胶中有5条带,对核酸酶敏感,证明该病毒含有5个单股RNA组分。对慢性蜜蜂麻痹病病毒的基因组结构进行了讨论。     

激光、血卟啉对肿瘤细胞DNA单链断裂及其重接的影响

用简化的Kohn氏碱洗脱法,观察了光敏剂血卟啉衍生物(HPD)对小鼠S-180肿瘤细胞DNA单链断裂及其重接修复的影响。激光HPD能导致S-180细胞DNA单链断裂明显增加,而且这种断裂随着保温时间的延长,继续增多。在本实验条件下没有观察到HPD对X线的增敏作用,HPD不能增加X线所致的DNA单链断裂,也不能影响其重接。单链断裂重接动力学的实验进一步证明了这个论点。
