The outer mitochondrial membrane isoform of mammalian cytochrome b5 (OM b5) is considerably more stable than its microsomal counterpart (Mc b5), whereas the corresponding apoproteins (OM and Mc apo-b5) exhibit similar stability. OM and Mc apo-b5 are also similar in that their empty heme-binding pockets (core 1) are highly disordered but that the remainder of each apoprotein (core 2) displays substantial hololike structure. Core 1 residue 71 is leucine in all known mammalian OM b5's and serine in the corresponding Mc proteins. Replacing Leu-71 in rat OM (rOM) b5 with Ser has been shown to (1) decrease apoprotein thermodynamic stability by >2 kcal/mol and (2) extend conformational disorder beyond core 1 and into core 2, as evidenced in part by loss of a near-UV circular dichroism signal associated with the side chain of invariant residue Trp-22. Herein we report identification of a conserved Mc b5 core 2 packing motif that plays a key role in stabilizing apoprotein conformation in the vicinity of Trp-22, thereby compensating for the presence of Ser at position 71: a pi-stacking interaction between the side chains of Trp-22 and His-15 that is extended by hydrogen bonding between the side chains of His-15, Ser-20, and Glu-11. The corresponding conserved packing motif in OM b5's differs in having arginine at position 15 and glutamate at position 20. We also present evidence indicating that the conserved Mc b5 packing motif noted above contributes to the unusually extensive secondary structure exhibited by bovine Mc apo-b5 in the urea-denatured state. 相似文献

The feasibility of pH-sensitive polymeric nanoparticles that effectively target the acidic extracellular matrix of tumors is demonstrated. Plasmid DNA was complexed with polyethyleneimine (PEI) and further with a pH-sensitive diblock copolymer, poly(methacryloyl sulfadimethoxine) (PSD)-block-PEG (PSD-b-PEG), to obtain naonparticles. The shielding/deshielding of nanoparticles was tested along with cell viability and transfection efficiency at physiological and tumor pH. The nanoparticles composed of DNA/PEI/PSD-b-PEG were 300 nm in size and showed low cytotoxicity and transfection at pH 7.4 due to shielding of PEI by PSD-b-PEG. The PSD-b-PEG bound to PEI/DNA complex decreased the interaction of PEI positive charges with cells and reduced the cytotoxicity by 60%. At pH 6.6, the nanoparticles demonstrated high cytotoxicity and transfection, indicating PSD-b-PEG detachment from the nanoparticles and permit PEI to interact with cells. PSD-b-PEG is able to discern the small difference in pH between normal and tumor tissues and hence has remarkable potential in drug targeting to tumor areas. 相似文献

依赖于DNA甲基化的基因表达调控在植物生长发育过程中发挥重要功能,而DNA甲基转移酶是调节DNA甲基化模式的功能蛋白之一。本研究采用RACE技术克隆了小麦甲基转移酶基因TaDnMT2的包含完整编码区的cDNA序列,并系统分析了该基因的结构特征及其在小麦生长发育过程中的表达特性。结果表明,TaDnMT2的cDNA序列为1321bp(GenBank登录号:JN642641),其中5′-和3′-UTR(非翻译区)分别为84和115bp、ORF(开放阅读框)1122bp;TaDnMT2编码的氨基酸序列包含2个S-腺苷甲硫氨酸结合域(I和X)、甲基转移酶活性位点(IV)、靶胞嘧啶结合位点(VI)、中和DNA骨架负电荷域(VIII)和靶位点识别区(IX)6个高度保守域,属于DNA甲基转移酶家族的DnMT2亚类;三维结构预测显示,TaDnMT2蛋白可以形成包括7个β-折叠和4个α-螺旋的特定空间结构。表达特性分析的结果表明,TaDnMT2基因在‘京841’小麦不同发育时期的叶中表达量均较高,且其在三叶龄期和五叶龄期的表达量受春化处理的影响;在种子发育过程中,该基因在授粉后5d的种子中表达量较高,随着种子发育进程的推进其表达水平呈逐渐下降趋势;在不同发育时期的根系中,TaDnMT2基因均具有较高水平的表达,且在分蘖期根系中的表达量最高。推断TaDnMT2基因可能在小麦生长发育过程中发挥重要功能。 相似文献

为阐明Bt杀虫蛋白对次要靶标害虫粘虫Mythimna separata (Walker) (鳞翅目: 夜蛾科)的生理学影响, 本研究分析比较了粘虫高龄幼虫在室内取食低剂量Cry1Ac杀虫蛋白6, 12, 24和36 h后, 其体内主要的解毒酶(酯酶和谷胱甘肽-S-转移酶)、 保护酶(超氧化物歧化酶、 过氧化氢酶和过氧化物酶)和中肠蛋白酶(总蛋白酶、 强碱性类胰蛋白酶、 弱碱性类胰蛋白酶和类胰凝乳蛋白酶)等活性的变化。结果表明, 取食Cry1Ac杀虫蛋白后, 粘虫幼虫体内相关酶活力呈现不同的变化趋势: (1)酯酶、 谷胱甘肽-S-转移酶、 过氧化物酶(POD)、 类胰蛋白酶和类胰凝乳蛋白酶活力较对照显著降低(P<0.05); (2)超氧化物歧化酶(SOD) 活力较对照显著升高(P<0.05); (3)过氧化氢酶(CAT) 活力于6, 12和24 h显著低于对照(P<0.05), 36 h时显著高于对照(P<0.05)。结果提示Cry1Ac杀虫蛋白主要通过抑制粘虫幼虫中肠解毒酶和蛋白酶的活性, 扰乱SOD, CAT 和POD 3种保护酶的动态平衡而干扰幼虫的正常生理代谢, 从而起到毒杀粘虫的作用。 相似文献

Studying rhizobia in the root nodules of Sphaerophysa salsula (Pall.) DC in the northwest of China, we obtained five strains classified as genus Rhizobium on the basis of their 16S rRNA gene sequences. The sequence similarity of strain CCNWQTX14T with the most related species was 99.0%. Further phylogenetic analysis of housekeeping genes (recA and atpD) suggested the five strains comprised a novel lineage within Rhizobium. The nifH and nodD gene sequences of CCNWQTX14T were phylogenetically closely related with those of Sinorhizobium kummerowiae and R. sphaerophysae, respectively. The five strains isolated from different places were also distinct from related Rhizobium species using ERIC fingerprint profiles. The DNA–DNA hybridization value was 41.8% between CCNWQTX14T and Rhizobium sphaerophysae CCNWGS0238T. Our novel strains were only able to form effective nodules on its original host Sphaerophysa salsula. Our data showed that the five Rhizobium strains formed a unique genomic species, for which a novel species Rhizobium

A novel aerobic, bacteriochlorophyll-containing bacteria strain JLT1202rT was isolated from Beibu Gulf in the South China Sea. Cells were gram-negative, non-motile, and short-ovoid to rod-shaped with two narrower poles. Strain JLT1202rT formed circular, opaque, wine-red colonies, and grew optimally at 3–4?% NaCl, pH 7.5–8.0 and 28–30?°C. The strain was catalase, oxidase, ONPG, gelatin, and Voges–Proskauer test positive. In vivo absorption spectrum of bacteriochlorophyll a presented two peaks at 800 and 877?nm. The predominant cellular fatty acid was C18:1 ω7c and significant amounts of C16:0, C18:0, C10:0 3-OH, C16:0 2-OH, and 11-methyl C18:1 ω7c were present. Strain JLT1202rT contained Q-10 as the major respiratory quinone and the genomic DNA G+C content was 76.3?mol%. Phylogenetic analysis based on 16S rRNA gene sequences of various species with validly published names showed that strain JLT1202rT fell within the genus Roseibacterium, family Rhodobacteraceae, sharing the highest similarity with Roseibacterium elongatum OCh 323T (97.9?% similarity), followed by Dinoroseobacter shibae DFL 12T (95.4?% similarity). The phylogenetic distance of pufM genes between strain JLT1202rT and R. elongatum OCh 323T was 9.4?%, suggesting that strain JLT1202rT was distinct from the only strain of the genus Roseibacterium. Based on the variabilities of phylogenetic and phenotypic characteristics, strain JLT1202rT stands for a novel species of the genus Roseibacterium and the name R. beibuensis sp. nov. is proposed with JLT1202rT as the type strain (=JCM 18015T?=?CGMCC 1.10994T). 相似文献
