The effects of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) (30 mg/kg subcutaneously per day for 8 days) to C57BL/6N mice were studied on tyrosine hydroxylase (TH), L-3,4-dihydroxyphenylalanine decarboxylase (DDC), and monoamine oxidase (MAO) activities in the striatum, and TH, DDC, dopamine-beta-hydroxylase (DBH), and MAO activities in the hypothalamus. Treatment with MPTP led to a large decrease in TH activity and a parallel decrease in DDC activity in the striatum, as compared with the saline controls. In contrast, MPTP administration did not cause a decrease of the activities of TH, DDC, and DBH in the hypothalamus. There was also no reduction in MAO activities of striatum and hypothalamus. These data indicate that MPTP administration to mice results in specific degeneration of the dopaminergic nigrostriatal pathway and that DDC in the mouse striatum may mainly be localized in the dopaminergic neurons with TH.  相似文献

Protein-tyrosine kinases (PTKs) play an integral role in T cell activation. Stimulation of the T cell antigen receptor (TCR) results in tyrosine phosphorylation of a number of cellular substrates. One of these is the TCR zeta chain, which can mediate the transduction of extracellular stimuli into cellular effector functions. We have recently identified a 70 kd tyrosine phosphoprotein (ZAP-70) that associates with zeta and undergoes tyrosine phosphorylation following TCR stimulation. Here we report the isolation of a cDNA clone encoding ZAP-70. ZAP-70 represents a novel PTK and is expressed in T and natural killer cells. Moreover, tyrosine phosphorylation and association of ZAP-70 with zeta require the presence of src family PTKs and provide a potential mechanism by which the src family PTKs and ZAP-70 may interact to mediate TCR signal transduction.  相似文献

The ground cricket Dianemobius nigrofasciatus overwinters as an egg in Japan, being univoltine in Hokkaido and northern Honshu and bivoltine farther south. In Hokkaido, however, this cricket is heard singing in winter in several fumarolic fields covered with moss and grasses locally known as “bokke”. In such warm “islets” the adult density was high in early summer and again in autumn, indicating that the cricket is bivoltine in contrast to the univoltine life cycle outside the bokke habitats in Hokkaido. Eggs laid by females collected at regular intervals from a bokke habitat showed a clear seasonal cycle of diapause incidence. At 26°C, the bokke strains produced non‐diapause eggs under long days and diapause eggs under short days as in the southern bivoltine populations, although the critical day‐length was longer than in the south. Several strains derived from non‐bokke habitats in Hokkaido and northern Honshu produced high percentages of diapause eggs under long days as well as short days as expected for the univoltine life cycle. Winter adults singing in bokke habitats could be either survivors of the autumn generation or individuals derived from eggs laid in autumn and then matured in response to the high soil temperature. In the laboratory, the proportion of egg diapause in short days was decreased by selection only for several generations. Phylogenetic trees of bokke and non‐bokke populations, based on both the nucleotide sequence of the mitochondrial COI gene and four allozyme loci, suggest that bokke populations have not been isolated from non‐bokke populations for an evolutionarily significant time.  相似文献

Natural carotene sample obtained from palm oil was proved to suppress the promoting stage of two-stage carcinogenesis of mouse skin, and also inhibit the proliferation of human malignant tumor cells, such as neuroblastoma GOTO cells, stomach cancer HGC-27 cells, and pancreatic cancer PANC-I cells. Among the major constituents of palm carotene, alpha-carotene showed stronger anti-proliferative effect than beta-carotene. The present results indicate that further investigation for not only beta-carotene but also other kinds of natural carotenes, such as alpha-carotene, should be carried out.  相似文献

Staurosporine, an antitumor-promoting agent, suppressed phorbol ester-enhanced phospholipid synthesis. The inhibitory effect of staurosporine was found to be dominant in the synthesis of phosphatidylcholine and phosphatidylethanolamine. The manner of this inhibitory action by staurosporine was similar to that of various kinds of antitumor-promoting agents, which have the ability to interact with Ca2(+)-calmodulin complex, although the effective dose of staurosporine was 1,000 times lower than these calmodulin-interacting agents. Furthermore, staurosporine was proved to interact directly with Ca2(+)-calmodulin complex. Thus, it is possible that staurosporine showed inhibitory effect on phospholipid metabolism via the modulation of Ca2(+)-calmodulin system.  相似文献

The effect of thyrotropin-releasing hormone (TRH) and lithium on myo-inositol metabolism has been assessed in rat cerebral cortex, cerebellar cortex, and sciatic nerves. Sprague-Dawley male rats were injected subcutaneously with 10 mEq/kg of LiCl and intraperitoneally with 10 mg/kg of TRH-tartrate, alone or in combination. Either lithium or TRH alone had little effect on the myo-inositol concentration in cerebellar cortex, whereas the combination of lithium and TRH significantly lowered the level. The myo-inositol level of cerebellar cortex reached its nadir (70% of values in untreated control rats) 30 min after addition of TRH and then returned to the control level at 90 min. In cerebral cortex, both lithium alone and lithium plus TRH significantly reduced the myo-inositol level. No effect was seen on the myo-inositol concentration in sciatic nerves with these regimens. These results suggested that the pharmacological dose of TRH activated phosphatidylinositol turnover in rat cerebellar cortex and subsequently reduced the myo-inositol level in the presence of lithium.  相似文献

N-(6-Phenylhexyl)-5-chloro-1-naphthalenesulfonamide (SC-9) activated Ca2+-activated, phospholipid-dependent protein kinase (protein kinase C). SC-9 acted as a substitute for phosphatidylserine, which is one of the endogenous factors in activating protein kinase C. SC-9 was also effective in regulating the physiological functions at the whole-cell level. For example, SC-9 stimulated hexose transport activity in mouse fibroblasts, a protein kinase C-regulated cellular function. Thus, SC-9 may be useful to study the molecular basis of the regulation of protein kinase C activity, and the biological significance of this enzyme.  相似文献
