PRL-1, -2, and -3 represent a novel class of protein-tyrosine phosphatase with a C-terminal prenylation motif. Although PRL-1 has been suggested to be associated with the nucleus, the presence of three highly homologous members and the existence of a prenylation motif call for a more detailed examination of their subcellular localization. In the present study, we first demonstrate that mouse PRL-1, -2, and -3 are indeed prenylated. Examination of N-terminal epitope-tagged PRL-1, -2, and -3 expressed in transiently transfected cells suggests that PRL-1, -2, and -3 are present on the plasma membrane and intracellular punctate structures. Stable Chinese hamster ovary cells expressing PRL-1 and -3 in an inducible manner were established. When cells were treated with brefeldin A, PRL-1 and -3 accumulated in a collapsed compact structure around the microtubule-organizing center. Furthermore, PRL-1 and -3 redistributed into swollen vacuole-like structures when cells were treated with wortmannin. These characteristics of PRL-1 and -3 are typical for endosomal proteins. Electron microscope immunogold labeling reveals that PRL-1 and -3 are indeed associated with the plasma membrane and the early endosomal compartment. Expression of PRL-3 is detected in the epithelial cells of the small intestine, where PRL-3 is present in punctate structures in the cytoplasm. When cells are treated with FTI-277, a selective farnesyltransferase inhibitor, PRL-1, -2, and -3 shifted into the nucleus. Furthermore, a mutant form of PRL-2 lacking the C-terminal prenylation signal is associated with the nucleus. These results establish that the primary association of PRL-1, -2, and -3 with the membrane of the cell surface and the early endosome is dependent on their prenylation and that nuclear localization of these proteins may be triggered by a regulatory event that inhibits their prenylation.  相似文献

目的 观察酪酸梭菌——婴儿型双歧杆菌二联活菌制剂对受试人群肠道菌群的影响。方法 检测受试者服用前后的肠道菌群并计数。结果 肠杆菌、肠球菌、拟杆菌的数量无明显变化 ,双歧杆菌、乳杆菌的数量明显增加 ,差异有非常显著性 (P<0 .0 1) ,结论 酪酸梭菌——婴儿型双歧杆菌二联活菌制剂具有一定的调节人体肠道菌群、增殖双歧杆菌和乳杆菌的作用。  相似文献

The t haplotypes (t) are recent evolutionary derivatives of an alternate form of the mouse t complex region located at the proximal end of chromosome 17. This variant form of approximately 1% of the mouse genome is a source of mutations altering numerous sperm functions crucial for fertilization. Males that carry two t haplotypes (t/t) are invariably sterile. t haplotypes contain four inversions relative to the wild-type t complex (+), so that in matings involving a +/t heterozygote, t is usually transmitted as a single unit. However, rare recombinants have been recovered, which carry only part of the t genotype and express only some of the t-dependent phenotypes. Use of these partial t haplotypes in genetic crosses has resulted in the general location of the two major t male sterility factors, S1 and S2, within inversions 1 and 4, respectively. Since sterility can result from a plethora of sperm defects, we have made a detailed study of various functional parameters of sperm from mice carrying S1 or S2 heterozygously or homozygously or in combination. Both S1 and S2 contain mutations altering sperm functions, including motility, capacitation, binding to the zona pellucida, binding to the oocyte membrane, and penetration of the zona pellucida-free oocyte. Therefore it seems clear that each of these factors contains multiple genes contributing to sterility. Furthermore, our results indicate that genes within S1 interact with genes in S2 for all sperm functions examined. However, S1 and S2 genes affecting motility interact in a purely additive fashion, while S1 and S2 genes affecting most other sperm characteristics interact in a synergistic manner. Additionally, the patterns of synergism between S1 and S2 for abnormalities in capacitation, sperm-oolemma binding, and zona-free oocyte penetration are nearly identical. This suggests that these three defects are caused by mutation of the same gene within each sterility factor. These findings will not only be instrumental in matching the various t haplotype sperm defects to candidate genes for S1 and S2, but will facilitate a more comprehensive understanding of the cellular and genetic mechanisms underlying t haplotype male sterility.  相似文献

不同地区小菜蛾种群羧酸酯酶的毒理学性质研究   总被引:2,自引:0,他引:2  
在1995~1997年对湖北武汉、河北张家口地区小菜蛾Plutella xylostella(L.)种群的抗药性进行了研究。结果表明对阿维菌素的抗性和台湾敏感种群相比,武汉种群抗性为4.3倍,张家口种群抗性为1.8倍;对马拉硫磷的抗性武汉和张家口种群分别为2.2和2.9倍;对氟铃脲的抗性分别为3.2和0.5倍;对溴氰菊酯的抗性分别为2.4和1.7倍。对羧酸酯酶(Care)的研究结果表明,三个种群幼虫CarE对a-乙酸萘酯或β-乙酸萘酯(a或β-NA)水解活性差异显著,但成虫Care活性没有明显差异。武汉和张家口种群幼虫CarE对a-NA和β-NA的亲和力没有明显差异,但是武汉种群幼虫Care对底物的亲和力高于张家口种群。敏感品系Care对a—NA的亲和力明显高于对β-NA,相差约3倍。不同类型的抑制剂对小菜蛾幼虫CarE的抑制能力不同。增效磷和对氧磷对敏感品系CarE水解a-NA具有明显的抑制作用,分别比对武汉种群Care的抑制作用大4.577倍(SVl)和2.576倍(对氧磷)。  相似文献

β-葡萄糖苷酶是纤维素酶的重要组分之一,它不仅可水解纤维二糖和寡糖,更可解除纤维二糖对β-1,4-内切葡聚糖酶和外切葡聚糖酶的抑制,提高水解速率和程度.利用SephadexG-150和DEAE-SephadexA-50层析法从黑曲霉变异株L-22中分离提纯了β-葡萄糖苷酶,该酶是由两个分子量相同的亚基组成的二聚体,每个亚基分子量为203kD.该酶最适pH为4.8,pH稳定范围在3.6~6.4;最适温度是60℃,温度稳定范围为4~60℃;酶分子含糖量为8.35%.它是一个酸性β-葡萄糖苷水解酶,专一性地水解β-糖苷键.而不水解α-糖苷键,对短链底物表现了相对高的活力.用动力学分析和共价化学修饰方法探讨了与该酶活力有关的必需基团.由pH对lgVm和lgVm/Km的影响,推测出酶活性部位至少有两个可解离基团为酶活性所必需,它们在酶-底物复合物中的pKes1和pKes2的值分别为4.0和5.6,在游离酶中的pK值分别为4.2和5.9.由此可初步判断这两个可解离基团可能为组氨酸和含羧基的氨基酸,它们与酶的催化和底物结合可能有关.  相似文献

森林脑炎(TBE)病毒属黄病毒科,基因姐RNA含有单个开放阅读框架,5′端编码病毒的结构蛋白,3′端编码非结构蛋白。翻译成聚蛋白后,通过细胞和病毒编码的蛋白酶裂解产生单个的病毒蛋白。成熟的病毒是由两个相关的E和M膜蛋白脂质包膜所包围的立体对称的核衣壳组成。包膜E蛋白在病毒的感染周期中对细胞的识别和穿入细胞具有极其重要的功能,同时E蛋白诱导保护性的免疫反应,E蛋白内某一位点单个氨基酸的改变可引起病毒毒力的改变。因此,对TBE病毒分子生物学的研究有助于了解病毒与宿主细胞相互作用的机理,为病毒感染的特异性诊断、疫苗的研制和抗病毒药物的设计提供理论依据。  相似文献

豚鼠耳蜗分离外毛细胞经fluo-3和fura-red染色后,用激光扫描共聚焦显微镜监测其胞内游离钙的空间分布和动态变化,以fluo-3/fura-red荧光比值大小指示游离钙浓度的高低。外毛细胞胞内游离钙呈不均匀分布,荧光比值分别为细胞质1.71±0.85,质膜1.61±0.75,表皮板1.47±0.65及胞核1.39±0.66,显示细胞质游离钙浓度最高而胞核游离钙浓度最低。静息状态下连续扫描时荧光比值变化小于0.1,而在受到机械刺激后荧光比值增加幅度达0.3—1.2。实验结果表明,fluo-3和fura-red双发射比例法能更准确反映听毛细胞胞内钙的空间分布和动态变化,第二信使钙可能参与了听毛细胞的机械-电换能过程,研究了换能过程中听毛细胞钙信号的时空模式。  相似文献

Insect glutathione S‐transferases (GSTs) play important roles in detoxifying toxic compounds and eliminating oxidative stress caused by these compounds. In this study, detoxification activity of the epsilon GST SlGSTE1 in Spodoptera litura was analyzed for several insecticides and heavy metals. SlGSTE1 was significantly up‐regulated by chlorpyrifos and xanthotoxin in the midgut of S. litura. The recombinant SlGSTE1 had Vmax (reaction rate of the enzyme saturated with the substrate) and Km (michaelis constant and equals to the substrate concentration at half of the maximum reaction rate of the enzyme) values of 27.95 ± 0.88 μmol/min/mg and 0.87 ± 0.028 mmol/L for glutathione, respectively, and Vmax and Km values of 22.96 ± 0.78 μmol/min/mg and 0.83 ± 0.106 mmol/L for 1‐chloro‐2,4‐dinitrobenzene, respectively. In vitro enzyme indirect activity assay showed that the recombinant SlGSTE1 possessed high binding activities to the insecticides chlorpyrifos, deltamethrin, malathion, phoxim and dichloro‐diphenyl‐trichloroethane (DDT). SlGSTE1 showed higher binding activity to toxic heavy metals cadmium, chromium and lead than copper and zinc that are required for insect normal growth. Western blot analysis showed that SlGSTE1 was induced in the gut of larvae fed with chlorpyrifos or cadmium. SlGSTE1 also showed high peroxidase activity. All the results together indicate that SlGSTE1 may play an important role in the gut of S. litura to protect the insect from the toxic effects of these compounds and heavy metals.  相似文献
