De novo purine nucleotide biosynthesis: cloning of human and avian cDNAs encoding the trifunctional glycinamide ribonucleotide synthetase-aminoimidazole ribonucleotide synthetase-glycinamide ribonucleotide transformylase by functional complementation in E. coli.

The trifunctional enzyme encoding glycinamide ribonucleotide synthetase (GARS)-aminoimidazole ribonucleotide synthetase (AIRS)-glycinamide ribonucleotide transformylase (GART) was cloned by functional complementation of an E. coli mutant using an avian liver cDNA expression library. In E. coli, genes encoding these separate activities (purD, purM, and purN, respectively) produce three proteins. The avian cDNA, in contrast, encodes a single polypeptide with all three enzyme activities. Using the avian DNA as a probe, a cDNA encoding the complete coding sequence of the trifunctional human enzyme was also isolated and sequenced. The deduced amino acid sequence of the human and avian polyproteins show extensive sequence homologies to the bacterial purD, purM, and purN encoded proteins. Avian and human liver RNAs appear to encode both a trifunctional enzyme (G-ARS-AIRS-GART) as well as an RNA which encodes only GARS. The trifunctional protein has been implicated in the pathology of Downs Syndrome and molecular tools are now available to explore this hypothesis. Initial efforts to compare the expression of GARS-AIRS-GART between a normal fibroblast cell line and a Downs Syndrome cell line indicate that the levels of RNA are similar.

甘肃东乡几种早中新世哺乳动物化石

本文记述了在甘肃东乡椒子沟发现的几种早中新世哺乳动物化石:Gomphotherium sp、Dzungariotheriura orgosense、Rhinocerotidae gen. indet.和Paraentelodon macrognathus sp. nov.,着重讨论了全北区猪齿兽类的系统关系。根据化石的性质判断,东乡椒子沟化石点的地质时代应与欧洲Burdigalian早期,亦即MN_3相当或稍早。笔者将原定为上新世临夏组的第一、二岩性段划出,命名为椒子沟组,其时代为早中新世。     

人体小卫星DNA探针的制备

根据人体小卫星DNA核心顺序,化学合成长23碱基寡核苷酸探针,筛选人体基因组文库,旨在获得能用作遗传分析探针的小卫星顺序。结果得到15个含小卫星的阳性重组子。随机取其一(C_(35.9))作探针,试做群体分析。所有个体均可检出多条杂交带。其中某些带具有多态性。在一定检测条件下,检出的DNA图谱在有限的个体内具有个体特异性。结果表明筛选文库得到的小卫星顺序可用于小卫星多态性的检测。其它小卫星探针的筛选和应用性研究正在进行。     

c-kit protein, a transmembrane kinase: identification in tissues and characterization.

The proto-oncogene c-kit encodes a transmembrane kinase which is related to the receptors for colony-stimulating factor type 1 and platelet-derived growth factor, as well as to the immunoglobulin superfamily. Antibodies specific for the kinase domain of the P80 gag-kit protein of the Hardy-Zuckerman 4 feline sarcoma virus were prepared. These kit-specific antibodies were used to identify and characterize the c-kit protein in cat brain tissue. The c-kit protein product displays an autophosphorylating activity in immune complex kinase assays, and, in turn, this activity was used to identify the c-kit protein in different tissues. In cat brain, a single 145-kilodalton (kDa) glycoprotein was detected. Its N-linked carbohydrates were found to be sensitive to digestion with the endoglycosidases (neuraminidase, endoglycosidase F, and endoglycosidase H), indicating hybrid and/or complex and high-mannose structures. A partial purification of the c-kit protein was achieved by wheat germ agglutinin affinity chromatography, and the autophosphorylating activity of the partially purified c-kit protein was characterized and found to be specific for tyrosine. The kit antibodies cross-react with the murine c-kit protein product, and variant c-kit proteins in different mouse tissues were identified, with sizes of about 145 kDa (brain), 160 kDa (spleen), and 150 kDa (testis).     

An in-vivo measurement and analysis of viscoelastic properties of the spinal cord of cats

An in-vivo experimental technique was employed to determine the linear and nonlinear characteristics of viscoelastic properties of the spinal cord of anesthetized cats. The stress relaxation and recovery curves were reproducible in a group of cat experiments. The data of linear viscoelastic properties were used to develop a power law model with Boltzmann's convolution integral. The model was capable of predicting a prolonged stress relaxation and recovery curve. For larger deformation, the results were quantified using a nonlinear analysis of viscoelastic response of the spinal cord under the uniaxial experiment.     

Dinocrocuta gigantea头骨的发现

本文记述了甘肃和政县晚中新世地层中发现的一个完整的巨鬣狗头骨;讨论了它的分类地位,修正了 Schlosser 1903年建种时的一些鉴定错误.巨鬣狗是鬣狗科中一个十分特化的成员,应代表一个独立的属.根据命名规则,我们采用 N. schmidt-Kittler 1976年所创的 Dinocrocuta. 因此,这个种应订正为 Dinocrocuta gigantea (schlosser, 1903).     

广西黄皮属六种叶片的比较解剖观察

<正> 芸香科黄皮属(Clausena)植物全球约25种,分布于东半球热带、亚热带地区;我国记载有9种,产长江以南各省区,以两广、云南种类较多;广西现知6种,民间大多作药用,黄皮和小叶黄皮在我国南部广泛栽培,果实供生食或加工,黄皮的优良品种为岭南佳果之一。目前正在挖掘其潜在的药用价值及进行化学成分和挥发油的研究。中国人民解放军181医院药理室,用大鼠进行动物实验,证明黄皮叶有降血脂作用。在研究黄皮属的分类时,某些种的形态特征很接近,叶的形状、大小变化大,在无花果的情况下更难区分,笔者试图从叶的解剖构造探讨种间的差异,为植物分类和生药鉴定提供依据,供临床用药及有关方面参考。     

一种高生产力和生态协调的亚热带针阔混交林——杉木火力楠混交林的研究

 通过八年杉木针阔混交林的综合定位研究,筛选出一种高生产力和生态协调的人工林一以8杉木2火力楠为优势的混交林。林分的蓄积量和乔木层贮存的能量分别比杉木纯林高13.7%和11.3%,杉木火力楠混交林提高了林分的光能利用率,改善了林内小气候;增加了林地有机质的含量;促进了土壤中有益微生物的繁衍和土壤理化性质的改良,提高了土壤肥力和蓄水保水能力;增强了林分对害虫自我抑制能力。
