The distribution of the Golgi enzyme beta1, 6-N-
acetylglucosaminyltransferase (core 2 GlcNAc-T for short) has been
investigated in several tissue and cell systems by combining the potentials
of a polyclonal antibody and a novel, sensitive fluorescent enzyme assay.
In normal rat tissues, levels of the protein were found to vary and as a
general trend did not correlate with enzyme activities. Additionally, we
observed tissue-specific core 2 GlcNAc-T forms of various size: 75 kDa
(liver), 70 kDa (spleen), 60 kDA (heart), and 50 kDa (heart and lung).
These forms might arise from differential protein modifications;
alternatively, the smaller form may be a product of proteolytic cleavage,
given the presence of a catalytically inactive 50 kDa species in rat serum.
Chinese hamster ovary (CHO), MDAY-D2, PSA- 5E, and PYS-2 cell lines
consistently displayed a 70 kDa enzyme. When induced to retrodifferentiate
in the presence of butyrate + cholera toxin, CHO cells exhibited a 21-fold
increase in enzyme activity, while protein levels remained constant. A
similar trend was observed in the embryonal endoderm cell lines PSA-5E and
PYS-2, where an approximately 100-fold difference in core 2 GlcNAc-T
activity was found notwithstanding unchanged amounts of the protein and
identical mRNA levels, as evidenced by RT-PCR. In contrast, levels of core
2 GlcNAc-T activity in MDAY-D2 cells correlated well with protein
expression. Taken together, these observations demonstrate that core 2
GlcNAc-T expression may be subjected to multiple mechanisms of regulation
and suggest that in at least some instances (i.e., PSA-5E and PYS-2 cells)
expression may be regulated exclusively via posttranslational mechanism(s)
of control.

A model mineralizing system was subjected to magnetic resonance microscopy to investigate how water proton transverse (T2) relaxation times and magnetization transfer ratios can be applied to monitor collagen mineralization. In our model system, a collagen sponge was mineralized with polymer-stabilized amorphous calcium carbonate. The lower hydration and water proton T2 values of collagen sponges during the initial mineralization phase were attributed to the replacement of the water within the collagen fibrils by amorphous calcium carbonate. The significant reduction in T2 values by day 6 (p < 0.001) was attributed to the appearance of mineral crystallites, which were also detected by x-ray diffraction and scanning electron microscopy. In the second phase, between days 6 and 13, magnetic resonance microscopy properties appear to plateau as amorphous calcium carbonate droplets began to coalesce within the intrafibrillar space of collagen. In the third phase, after day 15, the amorphous mineral phase crystallized, resulting in a reduction in the absolute intensity of the collagen diffraction pattern. We speculate that magnetization transfer ratio values for collagen sponges, with similar collagen contents, increased from 0.25 ± 0.02 for control strips to a maximum value of 0.31 ± 0.04 at day 15 (p = 0.03) because mineral crystals greatly reduce the mobility of the collagen fibrils. 相似文献

Peridinium balticum (Pyrrhophyta) exists as a symbiosis between a nonphotosynthetic dinoflagellate host and a chlorophyll c-containing alga. It is hypothesized that P. balticum is an evolutionary link between primitive nonphotosynthetic and advanced photosynthetic dinoflagellates. This study documents pre- and postfertilization events of sexual reproduction in this unusual dinoflagellate for the first time. Light microscopy and TEM observations showed that gametes resemble vegetative cells except in the organization of their chloroplasts. Fusion of gametes occurred in a specific orientation, i.e., apical to sulcal area. The presence of an intact membrane between fusing pairs prior to plasmogamy was suggestive of enzymatic digestion of plates during fertilization. Rupture of this membrane triggers plasmogamy and karyogamy of the host, followed by that of the algal symbiont. A discussion of the cellular processes involved in gamete formation, fertilization, and zygote development is presented. The results of this investigation demonstrate that a synchronous sexual reproduction cycle has evolved for the symbiont and its dinoflagellate host. 相似文献

Mitochondrial gene sequences, in contrast to their plastid counterparts, have been rarely exploited for use in determining algal phylogenetic relationships. Their utilization would prove advantageous particularly for the assessment of evolutionary relationships among protistan lineages that include photo synthetic species and their colorless heterotrophic dependents that contain only a reduced plastid genome as well as ancestral nonphotosynthetic relatives. Recent studies that have examined mitochondrial protein (coxl, cox2, cob, and nad5) rather than ribosomal RNA genes confirm that these algal sequences are sufficiently conserved to be used routinely in algal systematics. In this investigation, we sequenced the mitochondrial nad4L gene, encoding a small (ca. 99-amino acid) polypeptide subunit of the NADH dehydrogenase complex, from two chromophytic algae and an oomycete to determine its suitability for use in determining algal and protistan evolutionary relationships. Our analysis demonstrated that nad4L could clearly resolve these three organisms as a distinct clade, the stramenopiles, to the exclusion of terrestrial plants/chlorophytes and eufungi. However, the position of deeply rooted species within the stramenopiles proved tenuous and would best be examined in conjunction with the analysis of longer mitochondrial protein gene sequences. 相似文献

Extant chromophytic algae have been suggested to have originated via the engulfment of a photo synthetic alga by a colorless protist. The dinoflagellate Peridinium foliaceum (Stein) Biecheler contains a reduced chlorophyll c–containing endosymbiont and, thus, represents an evolutionary intermediate stage in the establishment of chloroplasts. Although the exact phylogenetic relationship of the symbiont to extant algal species is unknown, it had been suggested that the P. foliaceum symbiont was either a diatom or a chrysophyte. Identification of the closest living relative of the P. foliaceum symbiont would provide a free-living model system with which the photosynthetic symbiont could be compared. Nucleotide sequence analysis of rbcL and rbcS (encoding the large and small subunits ofribulose-1,5-bisphosphate carboxylase/oxygenase) by the P. foliaceum symbiont was performed to provide insights into its identity. Cloned restriction fragments from a chloroplast DNA library were screened, and clones encoding the rbcLS operon were sequenced. Parsimony phylogenetic analysis was performed for each gene. Our data strongly suggest that the symbiont originated from a photosynthetic diatom. 相似文献

目的:评价PBL教学模式在生物化学中应用的教学效果,对生物化学理论教学的方式进行探讨。方法:用PBL模式进行教学,采用理论考试的形式对教学效果进行评价。结果:PBL教学组的学生在学习态度和积极性等方面好于传统教学组;在考试成绩方面PBL教学组病例分析考试成绩好于传统教学组,而基础知识考试成绩没有差别。结论:PBL模式能提高学生的分析问题和解决问题的综合应用能力,而基础知识的掌握程度与传统教学相比没有区别。 相似文献

Two critical innovations had a profound influence upon the evolutionary history of plants: the nutritionally dependent embryo and apoplastic phloem loading processes. Both depend upon the ability of the plant cell membrane to transport sugars. The evolutionary origin of sugar transport by plants is, therefore, of special phylogenetic importance. Recent evidence suggests that hexoses such as glucose are the main form of sugar transported apoplastically across the placental junction between gametophyte and sporophyte of the moss Polytrichum (Renault et al., 1992, Plant Physiology 100: 1815–1822). There is also considerable evidence that hexose transport may be involved in apoplastic phloem loading in Arabidopsis and other flowering plants. Results of numerous molecular, biochemical, immunofluorescence, and ultrastructural studies indicate that bryophytes and charophycean algae are related to the ancestors of vascular plants. This report demonstrates that the charophyte Coleochaete orbicularis exhibits enhanced growth in the presence of glucose under conditions in which inorganic carbon sources are limiting. Computer image analysis was used to demonstrate that nonaerated cultures of C. orbicularis grown for 7 weeks in an inorganic medium supplemented with 1% glucose produce as much as 13.6 times the biomass of clonal cultures grown for the same length of time in the same medium without glucose. Furthermore, addition of 1% glucose to nonaerated cultures yielded chlorophyll a concentration 20 times higher after 4 weeks growth than cultures grown under the same conditions without added glucose. This and other evidence derived from comparative growth measurements strongly suggest that Coleochaete takes up (or loads) glucose. Experimental results also suggest that hexose uptake by charophytes (such as Coleochaete) that typically grow in low alkalinity waters might provide a supplementary source of organic carbon when dissolved carbon dioxide levels are low. This capability could have served as an evolutionary precursor to hexose import by plant embryos, as well as leptom and phloem loading in bryophytes and vascular plants. 相似文献
