本文利用激光散斑屈光测试系统详细探讨了夜近视(night myopia)的产生机制。夜近视在环境亮度降低到产生暗视觉时出现,并随亮度继续降低而增大,平均值为1.35D。实验证明眼睛球差和色差不是造成夜近视的主要因素。夜近视数值在暗适应过程中呈增大的趋势。在暗视觉时,由于视野中缺少适宜的刺激,眼睛处于暗焦(dark focus)休止状态,这是造成夜近视的主要原因。  相似文献

We describe here three different hamster cell mutants which are resistant to diphtheria toxin and which provide models for investigating some of the functions required by the toxin inactivates elongation factor 2 (EF-2). Cell-free extracts from mutants Dtx(r)-3 was codominant. The evidence suggests that the codominant phenotype is the result of a mutation in a gene coding for EF-2. The recessive phenotype might arise by alteration of an enzyme which modifies the structure of EF-2 so that it becomes a substrate for reaction with the toxin. Another mutant, Dtx(r)-2, contained EF-2 that was sensitive to the toxin and this phenotype was recessive. Pseudomonas aeruginosa exotoxin is known to inactivate EF-2 as does diphtheria toxin and we tested the mutants for cross-resistance to pseudomonas exotoxin. Dtx(r)-1 and Dtx(r)-3 were cross-resistant while Dtx(r)-2 was not. It is known that diphtheria toxin does not penetrate to the cytoplasm of mouse cells and that these cell have a naturally occurring phenotype of diphtheria toxin resistance. We fused each of the mutants with mouse 3T3 cells and measured the resistance. We fused each of the mutants with mouse 3T3 cells and measured the resistance of the hybrid cells to diphtheria toxin. Intraspecies hybrids containing the genome of mutants Dtx(r)-1 and Dtx(r)-3 had some resistance while those formed with Dtx(r)-2 were as sensitive as hybrids derived from fusions between wild-type hamster cells and mouse 3T3 cells.  相似文献

This is the first report of a complete mitochondrial genome sequence from a photosynthetic member of the stramenopiles, the chrysophyte alga Chrysodidymus synuroideus. The circular-mapping mitochondrial DNA (mtDNA) of 34 119 bp contains 58 densely packed genes (all without introns) and five unique open reading frames (ORFs). Protein genes code for components of respiratory chain complexes, ATP synthase and the mitoribosome, as well as one product of unknown function, encoded in many other protist mtDNAs (YMF16). In addition to small and large subunit ribosomal RNAs, 23 tRNAs are mtDNA-encoded, permitting translation of all codons present in protein-coding genes except ACN (Thr) and CGN (Arg). The missing tRNAs are assumed to be imported from the cytosol. Comparison of the C.synuroideus mtDNA with that of other stramenopiles allowed us to draw conclusions about mitochondrial genome organization, expression and evolution. First, we provide evidence that mitochondrial ORFs code for highly derived, unrecognizable versions of ribosomal or respiratory genes otherwise ‘missing’ in a particular mtDNA. Secondly, the observed constraints in mitochondrial genome rearrangements suggest operon-based, co-ordinated expression of genes functioning in common biological processes. Finally, stramenopile mtDNAs reveal an unexpectedly low variability in genome size and gene complement, testifying to substantial differences in the tempo of mtDNA evolution between major eukaryotic lineages.  相似文献

We report synchronized sexual reproduction between the chlorophyll c-containing algal endosymbiont and its dinoflagellate host in Peridinium balticum (Pyrrhophyta). This organism's importance lies in that it may represent an intermediate between primitive non-photosynthetic and advanced photosynthetic dinoflagellates. Fusion of the endosymbionts and their nuclei occurred concomitantly with syngamy of the host gametes. Significant morphological changes, including condensation of chromatin and crystalline rod formation, occurred in the symbiont nucleus during zygote development. These observations provide evidence that the endosymbiotic nucleus is not passive in sexual processes, as opposed to its reported passive state during mitosis. P. balticum may not only represent an intermediate in the evolution of chloroplast acquisition by dinoflagellates, but also, an intermediate in the evolution of the peridinian dinoflagellate sexual life history.  相似文献

本文用脑室灌注和Fura2测定细胞内游离钙技术观察了地塞米松(dexamethasone,DEX)对家兔乙二醇双(2氨基乙醚)四乙酸性发热效应和下丘脑细胞内游离钙浓度([Ca2+]i)的影响,借此深入探讨地塞米松解热作用的中枢机制。结果发现:脑室灌注乙二醇双(2氨基乙醚)四乙酸(06nmol)引起家兔结肠温度明显升高,静脉注射地塞米松(5mg/kg)显著抑制家兔乙二醇双(2氨基乙醚)四乙酸性发热,地塞米松(60~120μmol/L)并不影响下丘脑细胞内[Ca2+]i,而事先脑室灌注抑制基因转录的放线菌素D(3nmol)则完全取消了地塞米松对乙二醇双(2氨基乙醚)四乙酸性发热的解热作用。这些结果提示:地塞米松显著抑制家兔乙二醇双(2氨基乙醚)四乙酸性发热,其机制与地塞米松激活脑内某些基因的表达有关,而与下丘脑神经细胞跨膜钙离子流无关。  相似文献

Background

Proteomic studies of formalin-fixed paraffin-embedded (FFPE) tissues are frustrated by the inability to extract proteins from archival tissue in a form suitable for analysis by 2-D gel electrophoresis or mass spectrometry. This inability arises from the difficulty of reversing formaldehyde-induced protein adducts and cross-links within FFPE tissues. We previously reported the use of elevated hydrostatic pressure as a method for efficient protein recovery from a hen egg-white lysozyme tissue surrogate, a model system developed to study formalin fixation and histochemical processing.

Principal Findings

In this study, we demonstrate the utility of elevated hydrostatic pressure as a method for efficient protein recovery from FFPE mouse liver tissue and a complex multi-protein FFPE tissue surrogate comprised of hen egg-white lysozyme, bovine carbonic anhydrase, bovine ribonuclease A, bovine serum albumin, and equine myoglobin (55∶15∶15∶10∶5 wt%). Mass spectrometry of the FFPE tissue surrogates retrieved under elevated pressure showed that both the low and high-abundance proteins were identified with sequence coverage comparable to that of the surrogate mixture prior to formaldehyde treatment. In contrast, non-pressure-extracted tissue surrogate samples yielded few positive and many false peptide identifications. Studies with soluble formalin-treated bovine ribonuclease A demonstrated that pressure modestly inhibited the rate of reversal (hydrolysis) of formaldehyde-induced protein cross-links. Dynamic light scattering studies suggest that elevated hydrostatic pressure and heat facilitate the recovery of proteins free of formaldehyde adducts and cross-links by promoting protein unfolding and hydration with a concomitant reduction in the average size of the protein aggregates.

Conclusions

These studies demonstrate that elevated hydrostatic pressure treatment is a promising approach for improving the recovery of proteins from FFPE tissues in a form suitable for proteomic analysis.  相似文献

Hoechst dye 33258-CsCl density gradients were used to isolate two satellite DNA species from Synura petersenii Korsh. sensu lato, a member of the Synurophyceae. One satellite DNA was identified as the chloroplast genome. The chloroplast genome is the smallest (91.5 kb) published for any chromophyte and approximates the size of the smallest functional chlorophyte chloroplast genome (Codium fragile, 89 kb). The second satellite DNA was small (34.5 kb), and its origin is undetermined. The potential of using the S. petersenii chloroplast genome in comparative studies for evaluating organellar evolution and algal systematics is discussed.  相似文献
