Abstract: Lichen-forming ascomycetes and their green algal photobionts completely die off within approximately 3 years of storage at room temperature. Macroscopically this is recognizable as a colour change, the green shades of the chlorophylls being lost. In fluorescent light microscopy preparations an increase in fungal autofluorescence and a significant decrease in chlorophyll autofluorescence in the Trebouxia cells was observed. In transmission electron microscopy preparations of Xanthoria parietina and its green algal photobiont, Trebouxia arboricola, the fungal membrane systems were found to be largely broken down whereas the shrivelled algal protoplast failed to rehydrate after storage at room temperature. When stored in the desiccated state at - 20 °C, both partners of the symbiosis stayed fully viable for up to 13 years, their colouration and chlorophyll fluorescence being unchanged. Viability was measured as ascospore ejection and germination rates in Xanthoria parietina, soredium germination rates in Xanthoria fallax, Hypogymnia physodes and Parmelia sulcata, and autospore formation rate in Trebouxia cells (green algal photobiont), which had been isolated from the thalli after rehydration. Thallus fragments of Xanthoria parietina were shown to grow normally after one week of storage in LN2 without any cryoprotectant. In the desiccated state deep-frozen samples can be repeatedly brought to room temperature and back to - 20 °C without any loss of viability. Cryopreservation is therefore a suitable mode of long-term storage of viable lichen thalli for experimental studies or transplant experiments.  相似文献

为了探明乌鲁木齐南部山区树生地衣种类及其群落结构特征, 应用多元分析中的主成分分析及聚类分析方法, 对分布在乌鲁木齐南部山区森林生态系统中的树生地衣植物群落进行数量分类, 并对其群落结构的物种多样性、相似性和均匀度等群落参数进行了比较系统的研究。结果表明, 分布在乌鲁木齐南部山区的树生地衣共有39种, 隶属于5目13科26属。根据多元分析结果, 将该地区的树生地衣划分为3种类型: (1) 小茶渍(Lecanora hageni (Ach.) Ach.) + 蜈蚣衣(Physcia stellaris (L.) Nyl.) + 柳茶渍(L. saligna (Schrad.) Zahlbr.)群落; (2) 斑面蜈蚣衣(Physcia aipolia (Humb.) Furm.) + 对开蜈蚣衣(Ph. dimidiata (Arn.) Nyl.) + 喇叭石蕊(Cladonia pyxidata (L.) Hoffm.)群落; (3) 拟石黄衣(Xanthoria fallax (Hepp) Arnold.) + 丽石黄衣(X. elegans (Link.) Th. The bright yellow wall lichen, Xanthoria parietina , is usually inhabited by oribatid mites (Acari) which do not only find shelter, but also graze on selected areas of the thallus. As X. parietina does not produce symbiotic vegetative propagules and its compatible photobiont, unicellular green algae of the genus Trebouxia , are rare outside lichen thalli, we tested the hypothesis of dispersal of viable Trebouxia cells via acarine faeces. The lichenivorous mites, Trhypochtonius tectorum and Trichoribates trimaculatus , were isolated from thalli of X. parietina and cultured in the laboratory on a lichen diet. Light microscopic investigations of faecal pellets from mites that had been feeding on X. parietina indicated gut passage of intact ascospores and photobiont cells. In a series of experiments, viable algal and fungal cells contained in such faecal pellets were cultured. The taxonomic affiliation of these isolates was identified using molecular techniques, i.e. comparative investigations of nuclear ribosomal gene data (ITS 1 and 2, 5.8S rDNA) in the algal and fungal partners, and of the species-specific hydrophobin gene sequence in the fungal partner. Our culturing experiments demonstrated that the faecal pellets of both lichenivorous mites, upon feeding on X. parietina , contain viable ascospores and photobiont cells ( Trebouxia arboricola ) and thus might be a common and successful mode of vegetative short- and long-distance dispersal of this and numerous other lichen-forming ascomycetes and their photobionts. Summary Cryotechniques, such as low temperature scanning electron microscopy (LTSEM) and freeze-substitution for transmission electron microscopy (TEM), were applied to two cyanobacterial and three green algal macrolichens in order to locate free water and to visualize drought-induced structural alterations at the mycobiont—photobiont interface. The following species were examined:Peltigera canina/Nostoc punctiforme, Sticta sylvatica/Nostoc sp. (both Peltigerales),Parmelia sulcata/Trebouxia impressa, Hypogymnia physodes/Trebouxia sp. (both Lecanorales), andXanthoria parietina/Trebouxia arboricola (Teloschistales). In all species free water was confined to the symplast and the apoplast. No intercellular water reservoirs were found in the gas-filled thallus interior. Thalline fluctuations in water content reflect fluctuations in apoplastic and symplastic water. All the taxonomically diverse lichen photobionts have access to water and dissolved nutrients via the fungal apoplast only. Drought stress (i.e., water content 20%/dw and below) caused dramatic shrinkage and deformation in all cell types. At any level of hydration the fungal and algal protoplast maintained close contact with the cell wall. This applied to the cyanobacterial photobionts and their murein sacculus and gelatinous sheath too. Although the cytoplasm of both partners was strongly condensed in desiccated lichens the cellular membrane systems, usually negatively contrasted, were very well preserved. The significance of these data is discussed with regard to the functioning of the symbiotic relationship.  相似文献

Little is known about the nature of the association between mycobionts and photobionts in isolated lichen communities. Here we studied the photobiont diversity of different Caloplaca species in a fog-induced community in the Atacama Desert. We compared nrDNA ITS sequences of both symbionts, photobionts and mycobionts, along with morphological characters of the different lichen thalli, to investigate the diversity and to assess the degree of selectivity and specificity of photobiont species in a community of Caloplaca species. Specimens of six fungal species (C. orthoclada, C. fernandeziana, and four undescribed species) were sampled along an altitudinal gradient on a coastal bluff with a strong fog presence, 60 km south of Iquique, Chile. The photobiont species in this community belong to three species of the genus Trebouxia in the strict sense: T. arboricola, T. decolorans, and T. gigantea. Most of the fungal species were lichenized with photobionts belonging to different haplotypes of T. arboricola and T. decolorans, although the algae of three specimens, associated with two fungal species (C. orthoclada and C. sp1), were related to representatives of T. gigantea. These results indicate that members of the genus Caloplaca in northern Chile have moderate photobiont selectivity and appear to be selective to members of the T. arboricola group. Also, at high altitudes, changes in the photobiontal haplotype composition were observed in comparison to lower altitudes, probably generated by a higher water availability given higher fog condensation and precipitation in the upper areas of the bluff. This may suggest that ecological factors, such as altitude and water availability could result in a local shift of the associated photobiont and specialization as a product of local adaptation.  相似文献

Surface alkanes and fatty acids from the thalli of the lichen Xanthoria parietina, its photobiont Trebouxia sp., and its mycobiont were analysed by GC-MS. The green alga Trebouxia sp. synthesized mainly unsaturated fatty acids such as (Z,Z,Z)-9,12,15-18 : 3 (Z,Z)-9,12-18 : 2 and (Z)-9-18 : 1, and light alkanes C8-C15 (up to 83% of total n-alkanes). However, the mycobiont contained mainly saturated fatty acids such as hexadecanoic (16 : 0) and octadecanoic acid (18 : 0), and also very long-chain n-alkanes C22-C34. Dehydroabietic acid was found in both lichen and mycobiont. The occurrence of different amounts of n-alkanes and fatty acids in the photobionts and mycobionts of X. parietina was shown for the first time. Lichens collected from different locations in the Jerusalem hills contained n-alkanes ranging in concentration from 187 to 211 mg x (g dry wt)-1; n-alkane concentrations in the photobiont and mycobiont were 17-24 and 215-262 mg x (g dry wt)-1, respectively.  相似文献

Chaenotheca phaeocephala and C. subroscida have been characterized by differences in their morphology and ecology. They are, however, morphologically plastic and sometimes seem to intergrade. It has been suspected that the differences in morphology between these two taxa could possibly be phenotypic and caused by their occurrence in different habitats and/or by the association with different photobionts. To test if there is a genetic difference between the species ITS1–5.8S-ITS2 rDNA sequences of six specimens of the mycobiont of C. phaeocephala and five of C. subroscida were compared. Further the ITS1–5.8S-ITS2 rDNA sequences of the photobiont of one specimens of C. phaeocephala and six of C. subroscida were compared to each other and to other species of Trebouxia. The mycobiont sequences of both C. phaeocephala and C. subroscida were monophyletic, substantially different and the groups had strong supports. They also proved to be associated with different Trebouxia species, C. phaeocephala with T. arboricola and Chaenotheca subroscida with T. jamesii. The morphologically based concepts of C. phaeocephala and C. subroscida thus not merely represent phenotypic variation but also are associated with considerable genetic differences in the extremes of the ITS1–5.8S-ITS2 rDNA region.  相似文献

Abstract: The photobiont inventory of a stand of the Acarosporetum sinopicae, a lichen community comprising saxicolous, chalcophilous lichens, has been analysed. Investigated lichen species were Acarospora rugulosa, A. sinopica, Bellemerea diamartha, Lecanora polytropa, L. subaurea, Lecidea silacea, L. lapicida, Rhizocarpon geographicum, and Umbilicaria cylindrica. For all these lichen species this is the first record of the photobionts, except for L. lapicida. The photobionts were cultured axenically and investigated using light microscopical and molecular methods (ITS-sequence analyses). Every lichen species contained only one photobiont species. All photobionts belong toTrebouxia jamesii , but two different subspecies were found with the morphological differences corresponding to molecular differences. The new subspecies T. jamesii subsp. angustilobata is described, differing from the typical T. jamesii by a crenulate chloroplast but identical to the latter taxon in respect to the pyrenoid structure in the light microscope. These results are discussed with respect to the photobiont inventory of the Physcietum adscendentis, analysed in an earlier study.  相似文献

Recent DNA sequence analyses have revealed the diversity of algal partners in lichen symbioses. Although morphologically similar, different genetic lineages of photobionts are detected in wide geographic ranges of the same lichen fungal species. We studied the photobiont of the genus Trebouxia, which are known as partners of diverse lichen-forming fungal species in the Mediterranean region. We studied the phylogeny of these algae with a multilocus dataset including three loci: ITS, rbcL, and actin type I gene. The two lineages found, informally named Trebouxia sp. 1 and Trebouxia sp. 2, are related to Trebouxia arboricola/decolorans. The cultivation under axenic conditions succeeded only for one of them so far. We used light microscopy, confocal laser scanning microscopy and transmission electron microscopy for phenotypic characterisation. The ultrastructural characters currently used to describe species in the genus do not support the segregation of Trebouxia sp.1 from Trebouxia arboricola. The preferential presence in Mediterranean climates of these strains suggests eco-physiological adaptation. Despite their asexuality in long living lichen symbioses, coccoid algal lichen partners have apparently diversified genetically and physiologically.  相似文献

The identification of damaged lichens is often difficult due to changes in the morphology of regenerating specimens. We examined the Ultrastructure of grazing damages to four species of calcicolous lichens ( Aspicilia calcarea, Physcia adscendens, Tephromela atra and Xanthoria parietina ) and free-living cyanobacteria (family Chroococcaceae) caused by individuals of four species of land snails ( Chondrina clienta, Balea perversa, Clausilia bidentata and Helicigona lapicida ). We also investigated the radular structure of the four lichen-feeding snails to examine whether differences in radular morphology result in species-specific grazing damages. Individuals of all four snail species removed the cyanobacteria layer covering the limestone or lichen surfaces. The four lichen species were grazed to a different extent by the different snail species. SEM-images showed that B. perversa left distinct depressions on the thalli of A. calcarea , whereas H. lapicida grazed off the thalli of this lichen rather evenly. Both snail species left visible radular traces on the lichen surface. In contrast, Ch. clienta left shallow depressions without radular traces on the thalli of A. calcarea. In Tephromela atra , grazing damages were observed on both thallus and ascocarp. Ascocarps of T. atra were partly grazed by B. perversa. Helicigona lapicida grazing on T. atra removed more or less evenly the entire lichen tissue including the ascocarps. In foliose lichens, grazing by Ch. clienta, B. perversa and Cl. bidentata resulted in depressions of different depths, while H. lapicida removed entire pieces of the thalli. In general, radular traces were less distinct in foliose lichens than in crustose lichens.  相似文献

Unicellular algae of genus Trebouxia are the most frequent symbiotic photobionts found in lichen species adapted to extreme environments. When lichenised, they cope well with freezing temperature of polar regions, high-mountains environments and were successfully tested in open-space experiments. Trebouxia sp. is considered potential model species for exobiological experiments. The aim of this paper is to evaluate cryoresistence of Trebouxia sp. when isolated from lichen thalli and cultivated on media. In our study, six algal strains were exposed to repeated freezing/thawing cycles. The strains of Trebouxia sp. (freshly isolated from lichen Lasallia pustulata), Trebouxia erici, Trebouxia asymmetrica, Trebouxia glomerata, Trebouxia irregularis, and Trebouxia jamesii from culture collection were cooled from 25 to -40 °C at two different rates. The strains were also shock frozen in liquid nitrogen. After repeated treatment, the strains were inoculated and cultivated on a BBM agar for 7 days. Then, cell viability was assessed as relative share of living cells. Potential quantum yield of photochemical reactions in PS II (F(V)/F(M)), and effective quantum yield of photochemical reactions in PS II (Φ(PSII)) were measured. While the slow cooling rate (0.5 °C min(-1)) did not cause any change in viability, F(V)/F(M), and Φ(PSII), the fast cooling rate (6.0 °C min(-1)) caused species-specific decrease in all parameters. The most pronounced interspecific differences in cryoresistance were found after shock freezing and consequent cultivation. While T. asymmetrica and T. jamesii exhibited low viability of living cells (18.9% and 34.7%) and full suppression of photosynthetic processes, the other strains had viability over 60%, and unaffected values of F(V)/F(M), and Φ(PSII). This indicated a high degree of cryoresistance of T. glomerata, T. erici, T. irregularis and Trebouxia sp. strains. These strains could be used for detailed investigation of underlying physiological mechanisms and as models for astrobiological tests taken in the Earth facilities.  相似文献

根据对新疆博格达山岩面生地衣群落20个样点(20m×20m)调查的数据,以各地衣种的盖度为指标结合双向指示种分析方法(TWINSPAN)和除趋势对应分析法(DCA)对博格达山岩面生地衣群落进行数量分类并分析了群落结构特征及其多样性和相似性。采用典范对应分析法(CCA)对各群落的物种分布格局与环境因子的关系进行研究。结果表明,TWINSPAN分析和DCA排序将分布在博格达山的37种岩面生地衣分为以下5个群丛。群丛1:斑纹网衣(Lecidea tessellate Florke)+粉芽盾衣(Peltula euploca(Ach.)Poelt)+杜瑞氏黄梅(Xanthoparmelia durietzii Hale)群丛,有25个种,总覆盖度为30.145%,多样性为4.025;群丛2:袋衣(Hypogymnia physodes(L.)Nyl.)+白边平茶渍(Aspicilia sublaqueata(H.Magn.)J.C.Wei)+砖孢胶衣(Collema subconveniens Nyl.)群丛,有17个种,地衣总盖度为15.885%,多样性为3.196;群丛3:聚茶渍(Lecanora accumulate H.Magn.)+丽石黄衣(Xanthoria elegans(Link)Th.Fr.)+亚洲平茶渍(Aspicilia asiatica(H.Magn.)Yoshim.)群丛,有30个种,地衣总盖度为37.87%,多样性为4.357;群丛4:中华石果衣(Endocarpon sinense H.Magn.)+伴藓大孢蜈蚣衣(Physconia muscigena(Ach.)Poelt.)+垫脐鳞衣(Rhizoplaca melanophthalma(DC.)LeuckertPoelt)群丛,有24个种,地衣总盖度为30.458%,多样性为3.912;群丛5:石胶衣(Collema flaccidum(Ach.)Ach.)+短绒皮果衣(Dermatocarpon vellereum Zschacke)+绿黑地图衣(Rhizocarpon viridiatrum(Wulfen)Korber.)群丛,有18个种,地衣总盖度为19.331%,多样性为3.515。CCA排序结果反映,该地区岩面生地衣的分布与海拔高度、光照强度、岩石pH和人为干扰有关,其中影响最大的因素是海拔高度,其次为光照强度和干扰。坡向和岩石大小对地衣种类分布的影响不显著。  相似文献

Lipid metabolism was studied in different photobiont species from lichens by following incorporation of radiolabel from [1-14C]acetate. In four algal photobionts, Coccomyxa mucigena, C. peltigera variolosae, Trebouxia aggregata, T. erici, polar lipids were mainly (73-90%) labelled while triacylglycerols were the most highly labelled non-polar lipid class. A rhamnose-containing lipid was found in two Coccomyxa species, representing about 11% of the polar lipids of C. mucigena. All the major algal glycosyl- and phospho-glycerides were labelled with monogalactosyldiacylglycerol and phosphatidylglycerol, respectively, being the main labelled lipids in the polar lipid classes. The photobionts were grown in media differing in their phosphate content by one hundred-fold. Low phosphate levels caused only a small decrease in the proportion of phosphoglyceride labelling--mainly in phosphatidylglycerol. However, total lipid labelling was reduced (by 83.3 and 76.6% in two Coccomyxa spp. and 62.1 and 27% in two Trebouxia spp.) for the green algae. By comparison, variations in phosphate availability had no significant effect on a Nostoc sp. Examination of the algal species by electron microscopy revealed phosphorus-containing granules. This reserve of phosphorus explains why the algal photobionts were able to maintain the proportion of phosphoglyceride labelling well and may be an important adaptive mechanism for lichens.  相似文献

A lichen biodiversity study conducted in the Khanty-Mansi Autonomous Okrug (district) reports 958 species in 199 genera and 72 families. Cetrelia alaskana (Llano) W. L. Culb. et C. F. Culb. is new to West Siberia; Graphis elegans (Borrer ex Sm.) Ach., Porpidia hydrophila (Fr.) Hertel et A. J. Schwab, and Sclerococcum sphaerale (Ach.) Fr. are new to Siberia; Acarospora fusca B. de Lesd., Cliostomum tenerum (Nyl.) Coppins et S. Ekman, Pertusaria corallina (L.) Arnold, and P. stalactiza Nyl. are new to Russia. High diversity of epilithic lichens is ecologically due to a variety of ecotopes that include mountain habitats on the eastern spurs of the Subarctic and Northern Urals in addition to plain ones.  相似文献

Lichens are a symbiosis consisting of heterotrophic, fungal (mycobiont) and photosynthetic algal or cyanobacterial (photobiont) components. We examined photobiont sequences from lichens in the Ross Sea Region of Antarctica using the internal transcribed spacer region of ribosomal DNA and tested the hypothesis that lichens from this extreme environment would demonstrate low selectivity in their choice of photobionts. Sequence data from three targeted lichen species (Buellia frigida, Umbilicaria aprina and Umbilicaria decussata) showed that all three were associated with a common algal haplotype (an unnamed Trebouxia species) which was present in all taxa and at all sites, suggesting lower selectivity. However, there was also association with unique, local photobionts as well as evidence for species-specific selection. For example, the cosmopolitan U. decussata was associated with two photobiont species, Trebouxia jamesii and an unnamed species. The most commonly collected lichen (B. frigida) had its highest photobiont haplotype diversity in the Dry Valley region, which may have served as a refugium during glacial periods. We conclude that even in these extreme environments, photobiont selectivity still has an influence on the successful colonisation of lichens. However, the level of selectivity is variable among species and may be related to the ability of some (e.g. B. frigida) to colonise a wider range of habitats.  相似文献

With the aim of understanding how some lichens can survive intensive fertilization we investigated two green algal ( Trebouxia) lichens, Hypogymnia physodes (L.) Nyl. and Platismatia glauca (L.) W. Culb., and compared control (Ctr), and intensively fertilized (F) thalli. We measured total N, proteins and amino acids to assess lichen N status. Chlorophyll a indicated photosynthetic capacity and photobiont mass, ergosterol the metabolic demands of the fungus, and chitin the fungal biomass. For carbon status we measured glucose, the photobiont ( Trebouxia) export product ribitol, and the mycobiont-specific carbohydrates arabitol and mannitol. The F-thalli had 2-3 times higher protein and N concentrations, 5-10 times higher chlorophyll a concentrations, while ergosterol and chitin were doubled. The ribitol concentrations were 4-5 times higher in the F-thalli, while the fungal carbohydrates did not increase to the same extent. The amino acid arginine had increased 60-fold. The F-thalli also had a relatively higher N investment in the photobiont in relation to mycobiont tissue compared to the Ctr-thalli, probably resulting in an increased capacity for carbon assimilation, most possibly required for maintaining the higher nutrient status of the F-thalli. Arginine accumulation possibly avoided toxic effects of accumulated NH4+, albeit binding a significant fraction of assimilated carbon.  相似文献

Reactive oxygen species (ROS) are normally produced in respiratory and photosynthetic electron chains and their production is enhanced during desiccation/rehydration. Nitric oxide (NO) is a ubiquitous and multifaceted molecule involved in cell signaling and abiotic stress. Lichens are poikilohydrous organisms that can survive continuous cycles of desiccation and rehydration. Although the production of ROS and NO was recently demonstrated during lichen rehydration, the functions of these compounds are unknown. The aim of this study was to analyze the role of NO during rehydration of the lichen Ramalina farinacea (L.) Ach., its isolated photobiont partner Trebouxia sp. and Asterochloris erici (Ahmadjian) Skaloud et Peksa (SAG 32.85 = UTEX 911).  相似文献
