利用改进的磁珠富集法从林麝(Moschus berezovskii)的基因组中分离到10个新的(AC)n重复微卫星座位,并分析其在24个林麝个体(取样于中国四川金凤山群体)中的多态性。其中,5个位点在24个林麝个体中具有4—13个等位基因,观察杂合度和期望杂合度分别是0.429—0.957和0.587—0.902,平均多态信息含量是0.730。表明这5个微卫星位点具有高度的多态性,可以用于林麝遗传多样性研究,对林麝的保护具有很重要的意义。  相似文献

爆杖花(Rhododendron spinuliferum)是中国西南地区特有的观赏和药用植物。为了研究爆杖花和碎米花之间的杂交物种形成过程,本研究利用FIASCO方法对爆杖花进行微卫星引物开发,从100对引物中筛选出28个微卫星标记,其中22个为多态。利用爆杖花两个居群共24个个体对22个多态性位点进行分析,结果显示:每个位点具有2~5个等位基因,平均为3.4个,其观测杂合度和期望杂合度分别为0.083~0.792和0.153~0.744。对筛出的28个微卫星标记在碎米花的两个自然居群中也做了检测,结果显示:有22个微卫星标记成功扩增,其中20个有多态性;每个多态位点有2。6个等位基因,平均为3.2个,其观测杂合度和期望杂合度分别为0.000~0.833和0.117~0.736。开发的微卫星标记可用于爆杖花及其近缘物种的居群遗传学分析和杂交物种形成研究。  相似文献

南天竹(Nandina domestica)是一种具有较高观赏价值的常绿灌木。本研究从南天竹基因组中开发和筛选出10个微卫星位点,能在该物种中进行稳定PCR扩增且具一定的多态性。采用4个南天竹群体24个个体进行检测发现,每个位点的等位基因数为2~6,期望杂合度和表观杂合度分别为0.153~0.778和0~1。本研究结果为后续对南天竹遗传多样性的研究打下了基础。另外,这10个位点在小檗科4个其他物种中也有一定的通用性。  相似文献

异型花是青藏高原特有的一年生草本植物。为了更好地利用和保护这一重要的植物资源,本研究开发了异型花的多态性微卫星标记。首先,基于限制性位点相关DNA测序(RAD-seq)技术,开发了异型花14个多态性微卫星位点和2个单态性微卫星位点的引物;随后,对异型花的3个自然居群的57个个体的14个位点进行了分析。结果显示:14个位点观察到的等位基因数为4~7个,平均为5.071个;期望杂合度范围为0.003~0.312 (平均值为0.194),而观测杂合度范围为0~0.281 (平均值为0.047)。其中,仅有3对引物在龙胆科的椭圆叶花锚中具有通用性。本研究开发的多态标记对研究异型花种群结构和遗传多样性具有重要意义。  相似文献

猪个体基因杂合度对生长性状的影响   总被引:5,自引:0,他引:5  
逐个测定 139头F2 代杂交猪 (Susscrofa) (英系大白猪×梅山猪 )的初生重、180日龄活重 (LWT180 )、日增重(ADG)、相对生长率 (RGR)和KR值等 5个生长性状 ,对每一个体分别进行 2 4个微卫星座位的基因分型。对性别、家系和标记与生长性状的关系进行分析 ,结果显示 :它们与这些生长性状没有显著关联 (P >0 0 5 )。以 2 4个中性微卫星座位的杂合度来表示个体基因杂合度 ,比较猪生长性状在各个体基因杂合度水平间的差异 ,并分析其变化规律。结果表明 :初生重从个体基因杂合度水平为 1( 1 5 185kg)时至杂合度水平为 2 ( 1 30 6 3kg)时达性状最低值 ,之后随基因杂合度水平的增加而增加 ;180日龄活重、日增重、相对生长率和KR值等 4个重要经济性状随个体基因杂合度的增加呈近似于正弦函数曲线变化 ,在个体基因杂合度水平 2 (杂合度为 0 5 796 )时的性状值最高 ,之后随着个体基因杂合度的增加而降低。同一性状在不同的个体基因杂合度水平表型值不同 ,在某一特定的杂合度水平时达最高 ,提示在猪选配时将个体基因杂合度控制在恰当的水平 ,可以使目的性状的表型值最高。  相似文献

为了探索东方亚种数量正在减少的原因和制定科学、有效的保护措施,利用微卫星DNA对大鸨东方亚种(Otistardadybowskii)的遗传多样性进行了研究。应用3个大鸨指名亚种的微卫星位点和13个波斑鸨的微卫星位点扩增了47个个体的基因组DNA,筛选出8个具有多态性的微卫星。其中有3个微卫星的多态性较低,其余5个微卫星的多态性较高。各位点的观察杂合度为0.0435-1.0000,平均杂合度(h)为0.6595;各位点多态信息含量(PIC)为0.0416-0.8520,平均为0.5497;有效等位基因数(E)为1.04-7.46,平均为3.61。4个位点符合HardyWeinberg平衡,4个位点偏离了HardyWeinberg平衡。多方面比较发现,大鸨东方亚种遗传多样性很低,且低于指名亚种,这可能由于其种群较小、历史遗传瓶颈作用、生境破碎化、分布地域紧缩等原因造成的。  相似文献

茶花鸡是我国具有独特遗传特性的地方家禽品种,为了进一步阐明其群体遗传变异和遗传结构状况,采用了33个家鸡特异性的微卫星标记对该鸡种自然群体中30个个体进行了多态性电泳检测。33个微卫星座位共检测到105个等位基因,所有座位都呈现出多态性,每个座位的等位基因数在2~5个之间,平均每个座位等位基因数3.20个。群体平均杂合度和平均多态信息含量分别为0.612 9和0.527 6。结果表明,茶花鸡自然群体遗传多样性较丰富。  相似文献

Through an improved enrichment protocol, 20 microsatellite loci were isolated and characterized in an endangered animal, forest musk deer (Moschus berezovskii). In which, six loci showed high polymorphism with five to 13 alleles in 48 sampled individuals. The observed and expected heterozygosities were 0.362–0.955 and 0.784–0.906, respectively. The average polymorphism information content (PIC) was 0.8108 in these six microsatellites and they could be the valuable genetic markers for further investigation of forest musk deer species.  相似文献

This study reported the isolation and characterization of eight polymorphic microsatellite loci in endangered forest musk deer Moschus berezovskii. An improved enrichment protocol was used to isolate microsatellites, and polymorphism was explored with samples from wild musk deer population collected in Miyalo of Sichuan Province in China. Approximately 70% of clones from the genomic library constructed in current study contained dinucleotide (AC) repeats. Eight microsatellite loci amplified were highly polymorphic within forest musk deer population. The number of alleles per locus ranged from 6 to 14, and the observed and expected heterozygosities ranged from 0.41 approximately 1.0 and from 0.8 approximately 0.9, respectively. The average polymorphic information content (PIC) value for these markers was 0.82. This demonstrated that the eight microsatellite loci developed here are highly polymorphic, and can be used as genetic markers for further investigation of musk deer. Also, the results showed that the musk deer distributed in Miyalo had a relatively higher level of genetic variation.  相似文献

The Miyaluo captive forest musk deer population (Sichuan Province, China) is one of the largest captive breeding populations in the world. In order to evaluate the genetic quality and provide available genetic management strategy, seven polymorphism microsatellite loci were applied to assess the genetic variation of the Miyaluo forest musk deer. The results indicated that a total of 168 alleles were detected from these seven microsatellite loci in 361 individuals, and the number of the alleles per locus ranged from 12 to 41 with a mean of 24. The average observed heterozygosity, expected heterozygosity, and PIC were 0.782, 0.854, and 0.837, respectively. Considering the results of the loci Hardy–Weinberg equilibrium test, the comparison of the common allele frequency as well as the private allele between the adults and juveniles, we concluded that the heterozygosity and the genetic diversity of the Miyaluo captive breeding population are increasing due to the input of new individuals from other populations. However, the frequency of some alleles declined sharply, and some were even lost indicating that there is a risk for diversity loss. Thus, we proposed an improved management and breeding strategy for the captive breeding population of the forest musk deer.  相似文献

A total of 15 polymorphic microsatellite loci were isolated from forest musk deer (Moschus berezovskii). These loci showed high levels of allelic diversity, presenting 6–14 alleles per locus in a group with 31 individuals. The mean observed and expected heterozygosities were 0.82 and 0.68, respectively. The average polymorphic information content value, cumulative discrimination power, overall probability of exclusion with both unknown parents or with only unknown sire for these markers were 0.78, 1.0, 0.9999 and 1.0, respectively, indicating that this suit of microsatellite loci would be a powerful tool for the genetic determination of the forest musk deer.  相似文献

Forest musk deer (Moschus berezovskii) are rare as a result of poaching for musk and habitat loss. Some captive populations of forest musk deer have been established for decades in China. However, little genetic information is available for conservation management. In this paper, genetic variations, population structures, and the genetic bottleneck hypothesis were examined using 11 microsatellite loci from captive populations in Miyalo, Jinfeng and Maerkang in Sichuan Province, China. Estimates of genetic variability revealed substantial genetic variation in the three populations. A total of 142 different alleles were observed in 121 forest musk deer and the effective number of alleles per locus varied from 6.76 to 12.95. The average values of observed heterozygosity, expected heterozygosity, and Nei's expected heterozygosity were 0.552, 0.899 and 0.894 respectively. The overall significant (P < 0.001) deficit of heterozygotes because of inbreeding within breeds amounted to 34.5%. The mean FST (P < 0.001) showed that approximately 90.2% of the genetic variation was within populations and 9.8% was across populations. The UPGMA diagram, based on Nei's unbiased genetic distance, indicated that the three populations were differentiated into two different groups and it agreed with their origin and history. Bottleneck tests indicated that all three populations have undergone a population bottleneck, suggesting a small effective population size. Acknowledging that the genetic structure of populations has crucial conservation implications, the present genetic information should be taken into account in management plans for the conservation of captive forest musk deer.  相似文献

To investigate the karyotypic relationships between Chinese muntjac (Muntiacus reevesi), forest musk deer (Moschus berezovskii) and gayal (Bos frontalis), a complete set of Chinese muntjac chromosome-specific painting probes has been assigned to G-banded chromosomes of these three species. Sixteen autosomal probes (i.e. 6-10, 12-22) of the Chinese muntjac each delineated one pair of conserved segments in the forest musk deer and gayal, respectively. The remaining six autosomal probes (1-5, and 11) each delineated two to five pairs of conserved segments. In total, the 22 autosomal painting probes of Chinese muntjac delineated 33 and 34 conserved chromosomal segments in the genomes of forest musk deer and gayal, respectively. The combined analysis of comparative chromosome painting and G-band comparison reveals that most interspecific homologous segments show a high degree of conservation in G-banding patterns. Eleven chromosome fissions and five chromosome fusions differentiate the karyotypes of Chinese muntjac and forest musk deer; twelve chromosome fissions and six fusions are required to convert the Chinese muntjac karyotype to that of gayal; one chromosome fission and one fusion separate the forest musk deer and gayal. The musk deer has retained a highly conserved karyotype that closely resembles the proposed ancestral pecoran karyotype but shares none of the rearrangements characteristic for the Cervidae and Bovidae. Our results substantiate that chromosomes 1-5 and 11 of Chinese muntjac originated through exclusive centromere-to-telomere fusions of ancestral acrocentric chromosomes.  相似文献

1986年10月-1987年5月在四川盆地西北缘,利用粪堆计数法,对保护区和非保护区的4种不同生境及不同海拔区间林麝种群密度进行了调查研究。得到保护地区原生林、次生乔木林、次生灌木林、人工林4种生境中林麝种群密度(M±SE)分别为3.94±1.57、1.55±0.23、0.61±0.22、0.00头/平方公里,其中最佳生境林麝密度为9.88头/平方公里;非保护地区次生乔木林中林麝种群密度(M±SE)为0.15±0.09头/平方公里,非常显著地低于保护地区次生乔木林(P<0.005),此区已丧失了利用价值。按年平均增长率49.80%计算,在无任何人为猎取的情况下,要恢复到1.5-2.5头/平方公里(正常栖息密度),约需6-7年。而后,可按每年自然繁殖总量的60%或总数量的20%猎取。同时得到决定林麝水平分布的关键因子是植被类型、乔灌木密度及基底硬度;决定林麝垂直分布的主要因子是植被垂直分布和人为干扰程度。  相似文献

Ten microsatellite DNA loci developed for the white-toothed shrew (Crocidura russula) were tested for PCR amplification and for utility in linkage studies in the house musk shrew, Suncus murinus. Four primer pairs successfully yielded PCR amplicons and showed polymorphism between two mutant strains, BAN-kc,oeb and WZ. Cloning and sequencing of the PCR amplicons of all the four loci confirmed the presence of microsatellite sequences. Alleles segregating in an F2 resource population constructed from the two strains ranged between two and five. Linkage analysis of the four loci together with 18 other polymorphic markers and three mutant loci resulted in five linkage groups containing three newly mapped microsatellite loci. This study reports the first microsatellite markers being registered in this species.  相似文献

通过对圈养林麝(Moschusberezovskii)外周血淋巴细胞CD4~+、CD8~+亚群的检测,探讨林麝细胞免疫功能状态,并探索应用流式细胞仪分析其淋巴细胞亚群的方法,为研究林麝重大疾病的病理机制及诊断方法提供科学依据。本研究选取健康林麝和患呼吸道疾病林麝各5头,以双色流式细胞术检测其外周血淋巴细胞CD4~+、CD8~+亚群的含量,并进行比较。结果显示,羊源CD4、CD8的流式荧光抗体能够标记林麝细胞并有效检测;患病林麝与健康林麝相比,外周血CD4~+细胞含量无差异(P 0.05),CD8~+细胞含量则显著降低(P 0.01),CD4~+/CD8~+比值显著增高(P 0.01)。结果表明,患呼吸系统炎性疾病的林麝其外周血淋巴细胞CD8~+亚群变化显著,检测淋巴细胞亚群对林麝疾病的诊断有重要意义。  相似文献

Olfactory receptors (ORs) are encoded by OR genes. The OR genes in forest musk deer (Moschus berezovskii), which rely on olfaction for reproductive and social communication, are poorly understood. In this study, we analyzed the genome sequence of the forest musk deer to obtain its olfactory subgenome and compared it to other species. A total of 1378 OR‐related sequences were detected in the forest musk deer genome including 864 functional genes, 366 pseudogenes and 148 partial genes. These OR genes were classified into Class I and Class II and were further classified into 18 families and 244 subfamilies through sequence identity. Comparative analyses of the OR genes’ protein sequences in species from different orders (forest musk deer, human, mouse and dog) showed that 12 clusters were specific to forest musk deer. However, when compared to other Artiodactyl species (i.e. cattle, yak and pig) only two clusters were specific to forest musk deer. The odor identification potential of the OR genes in the forest musk deer was focused mainly on floral, woody, lemon, sweet and fatty odors. We also found that OR genes specific to forest musk deer were involved in the identification of spearmint and caraway. Our work is the first genome‐wide analysis of OR genes in forest musk deer. These findings will assist with better understanding the relationship between behavior and olfaction in the forest musk deer and the characteristics of the olfactory subgenome in Artiodactyl mammals.  相似文献
