随着同步辐射装置的建设与发展及各种建模方法的产生与完善,小角X-射线散射(small angle X-ray scattering,SAXS)法已经逐渐成为结构生物学中的一种重要的工具。SAXS可以用于研究溶液中生物大分子的结构及构象变化,蛋白质的组装、折叠等动态过程。本文对SAXS的基本原理、常用的研究技术和建模方法及其应用进行了综述。  相似文献

随着同步辐射光源(尤其是目前快速发展的第四代同步辐射光源)技术的进步,可用于实验的辐射通量越来越高,实验样品(特别是蛋白质等生物大分子样品)受到的辐照损伤也越来越严重。在全球现有的同步辐射装置上,蛋白质等生物大分子溶液专用小角X射线散射(SAXS)实验站的光子通量基本上都在1013 cps量级。在如此高的通量下,蛋白质等生物大分子溶液样品在实验测量中受到的辐照损伤极其严重。如果没有有效的辐照防护措施,蛋白质溶液样品在毫秒级辐照时间内便会辐照损伤,导致不能获取有效的实验数据。辐照损伤严重制约了SAXS实验技术在蛋白质溶液样品方面的应用。因而,认识蛋白质溶液样品辐照损伤的产生机理、影响因素、判断标准,以及有效降低辐照损伤程度、延缓辐照损伤产生时间的方法,对于蛋白质等生物大分子溶液的散射实验具有重要的指导意义。本文在简要概述生物大分子溶液样品辐照损伤产生机理、影响因素、辐照剂量等基本概念的基础上,重点综述了同步辐射SAXS实验中辐照损伤的判断标准和防护措施。此外,本文还对比了各种防护措施的优缺点,讨论了在建HEPS新光源中SAXS束线可用的散射数据采集时间,指出辐照损伤防护剂是有价值的研究方向。  相似文献

本文介绍了槲皮素铋的合成方法,并通过紫外光谱、红外光谱、核磁氢谱、同步热分析进行了表征。氢谱显示铋离子在3'-OH位置与槲皮素配位,同步热分析确证了这个结果。通过DPPH法测定了槲皮素铋清除自由基的能力,结果显示,槲皮素铋清除DPPH自由基的能力弱于槲皮素。  相似文献

研究了四川南川金佛山特产的多辐溲疏不同海拔和生境的5个群体的41个个体叶片过氧化物酶,过氧化氢酶,酯酶,淀粉酶,柠檬酸脱氢酶及谷氨酸脱氢酶同工酶谱带位置上的变异式样,来自同一群体的不同个体在谱带上表现出极大的相似性,而来源于不同群体的个体却表现出明显的趋异性,显示出酶谱带位置与群体生长的生境有显著的相关性,一定的群体具有某些特有的酶谱带,空间隔离限制了群体间个体的相互基因交流,在进化中那些适应新的  相似文献

在电子计算机上,用快速沃尔什变换(FWT)方法计算具有视差的立体图对的沃尔什(Walsh)谱。从立体图对的Walsh 谱观察到,根据视差规则投影到左右两平面的图形,其Walsh 谱的高峰位置有相对的位移。位移的大小随视差大小而变。视差越大,高峰位置平移越大,且成一定比例。位移方向是视该投影面靠近所观察的物体的远端和近端而定。离物体远端的一边高峰位置向低频移,离物体近端的一边高峰位置向高频移。由于Walsh 变换在工程技术上较易于实现,在计算速度上,快速沃尔什变换(FWT)比快速富氏变换(FFT)快。今后如果需要得到立体图对的空间频谱,利用快速沃尔什变换就更方便。  相似文献

现有扩散光学断层成像(diffuse optical tomography,DOT)系统常采用高档光探测器和放大器,令系统成本居高不下、性价比较低.本文旨在使用全新技术降低系统成本、增加成像深度,研发一套可实用于人脑成像的新型扩散光学断层成像系统.为此采用了成本较低的新型光探测器和一款自主设计研制的光电二极管前置放大器.相较于实验室原有DOT系统,新系统整体成本节约了40%以上.其次,设计了一款具有弹性并可适应多种不规则待测物(subject)外形的无光纤穿戴式脑-机接口(brain computer interface,BCI)装置.本文首次将手持式3D激光扫描仪应用于待测物外形结构和光源-探测器位置信息的精确获取.最后综合应用上述技术装备于人脑形状仿体成像实验中,新系统的目标成像深度提升至35 mm.验证结果说明本系统已具备了实际人脑功能成像能力.  相似文献

群落结构如何响应环境变化是生态学研究长期关注的核心问题之一。粒径谱由个体大小和多度构建而来,与营养级转换速率相关、反映生态系统过程动态以及表征生态系统稳定性,可以将其视为一个综合的功能多样性指标用于预测和表征群落的组成以及生态系统功能如何响应环境压力。粒径谱研究最初始于水生生态系统,近年来被引入到土壤动物群落生态学的研究中。简要回顾粒径谱的概念由来及理论基础,分析比较了当前粒径谱研究中的4种易混淆类型,介绍了常用的两类土壤动物粒径谱构建方法及其生态学意义,梳理了土壤动物粒径谱对环境梯度响应与生态化学计量学相结合的研究进展,并指出了应用粒径谱研究土壤动物群落的难点及限制条件。未来,在基础理论研究方面,土壤动物粒径谱应关注个体大小与营养级位置及能量利用关系;在应用方面,土壤动物粒径谱可结合传统的分类方法广泛应用于指示环境污染、生态恢复、保育生物以及土地利用变化等。  相似文献

目的设计螺旋CT机的多排多层探测器,提高影像质量。方法将常规多排探测器改进为多排多层探测器,输出端接超高倍光电耦合放大器,可成倍提高探测器与采集系统的信噪比。结果多排多层探测器比同样排数的单层探测器输出的信噪比高、省时、剂量少,且图像质量高。结论多排多层探测器输出的数据量大幅增加,使CT系统的成像质量显著提高。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
