为了全面了解古尔班通古特沙漠荒漠肉苁蓉居群分布的遗传多样性特点,本研究通过ISSR分子标记技术,利用Nei和Shannon等多样性指数对古尔班通古特沙漠中5个居群166个个体的荒漠肉苁蓉遗传多样性、荒漠肉苁蓉种群和种内的遗传多样性进行分析。在供试材料中,8个引物共扩增出144个多态位点,多态位点百分率达100%,5个居群的多态位点百分率差异在46.53%~77.78%之间。在物种水平上,Nei基因多样度(h)为0.260 4,Shannon多样性指数(I)是0.411 0。遗传变异分析表明,物种水平的居群间遗传分化系数Gst为0.222 2,居群间的基因流Nm为1.750 7。研究显示古尔班通古特沙漠中荒漠肉苁蓉多态位点比例高,各居群基因交流较多,不同居群间遗传变异并不明显,这些对肉从蓉资源有效地保护和利用具有重要意义。  相似文献

采用ISSR 分子标记技术, 对西双版纳分布的兰科濒危植物流苏石斛( Dendrobium fimbriatum) 5 个居群共114 个个体的遗传多样性进行了研究。从100 条引物中筛选出了12 条用于扩增, 共检测到117 个位点, 其中105 个为多态位点。分析结果表明, 流苏石斛居群水平遗传多样性较低。在物种水平上, 流苏石斛多态位点百分率PPB 为89 .74% , Nei′s 基因多样性指数H 为0 . 3227 , Shannon′s 多样性信息指数Hsp 为0 . 4779 ; 在居群水平上, 各个居群的多态位点百分率PPB 差异较大( 6.84% ~ 39.32% ) , 平均值为23.93% , Nei′s 基因多样性指数H 为0 . 0871 , 各个居群的Shannon′s 多样性信息指数Ho 平均为0.1290。AMOVA 分析的结果显示, 流苏石斛的遗传变异大多数存在于居群间, 占总遗传变异的74 . 79%。基于Nei′s遗传多样性分析得出的居群间遗传分化系数Gst = 0 . 7443。各居群间的Nei′s 遗传一致度( I) 范围为0 . 5882~0 . 8331。Mantel 检测发现, 居群间的遗传距离和地理距离之间无显著的正相关关系( r= 0.2419, P=0.2416) 。鉴于流苏石斛的遗传多样性现状和居群遗传结构, 我们建议对流苏石斛居群所有个体实施及时的就地保护, 同时建立迁地保护居群, 促进基因交流。  相似文献

利用ISSR分子标记对新疆白梭梭4个居群,105个个体进行了遗传多样性的比较分析。在供试材料中,11个引物共扩增出171个多态位点,多态位点百分率为84.85%,4个居群的多态位点百分率差异在33.92%.40.35%之间。Shannon多样性指数(I)为0.3518,物种水平的Nei基因多样度(h)为0.3482。遗传变异分析表明,物种水平的居群间遗传分化系数Gst为0.6238,居群间的基因流Nm为0.3016。遗传分析表明吐鲁番居群和甘家湖居群的遗传距离最近。  相似文献

华中特有珍稀植物裸芸香的AFLP遗传多样性分析   总被引:2,自引:1,他引:1  
采用选择性扩增片段多态性(AFLP)方法对华中特有单种属植物裸芸香(Psilopeganum sinense)的8个自然居群的遗传多样性进行了检测与分析。结果表明:裸芸香的遗传多样性较低,且居群内遗传多样性显著低于物种水平遗传多样性。筛选出的5对引物共得到180个位点,76个为多态位点,多态位点百分率为42.2%,8个居群多态位点百分率为:3.3%~16.7%,居群平均多态位点百分率为9.4%;8个居群Nei多样性指数为0.01987~0.06987,Shannon’s多样性指数为0.0197~0.0816。居群间分化系数Gst=0.5069,居群间基因流为0.2432,不足以维持居群间的基因交流及现有的遗传结构。AMOVA分析表明总遗传变异的13.17%存在于4个地理区域之间,50.45%存在于地理区域内的居群间,36.38%的遗传变异存在于居群内个体间。NTSYS分析表明遗传距离与地理距离不存在相关关系。UPGMA聚类结果表明长江南北两岸的居群并没有产生明显分化。最后,分析了裸芸香的濒危原因并提出了有效的保育措施。  相似文献

卵叶海桑 (Sonneratiaovata)是海桑科濒危红树植物 ,在我国仅分布于海南文昌清澜自然保护区内。采用简单序列重复区间扩增 (ISSR)分子标记技术对该天然居群和东寨港红树林自然保护区引种的人工居群共 3个居群 3 9个个体进行了遗传变异分析。 1 1个引物共扩增出 1 85条带 ,其中 1 2 7条具多态性 ,多态位点百分率为 68.65 %。在居群水平上相对较低 ,多态位点百分率 3 6.76%~ 5 4.5 9% ,平均值为 47.2 1 %。Nei的基因多样性、Shannon信息指数在物种水平上分别为 0 .1 41 1和 0 .2 2 92 ;在居群水平上平均值分别为 0 .1 2 0 9和0 .1 91 0。Nei的遗传分化系数Gst表明 :87.5 8%遗传变异分布在居群内 ,1 2 .42 %的遗传变异分布在居群间。居群间的遗传一致度达 0 .970 7。东寨港迁地保护的人工居群有效地保护了卵叶海桑的遗传多样性。  相似文献

利用ISSR分子标记技术对濒危植物连香树10个居群的遗传多样性和遗传变异进行了分析,结果表明:连香树物种水平遗传多样性较高,多态位点百分率(PPB)达到69.59%,Nei’s基因多样性指数(H)和Shannon信息指数(I)分别为0.231 3和0.351 4;而在居群水平上,多态位点百分率(PPB)为30.61%,Nei’s基因多样性指数(H)和Shannon信息指数(I)分别为0.115 6和0.173 3。遗传变异分析表明,居群间遗传分化程度高,遗传分化系数(GST)为0.500 3,居群间基因流Nm为0.527 3。Mantel检测,居群间的遗传距离和地理距离之间不存在显著的相关性。生境的片断化使居群间的基因流受阻,可能是导致居群间高遗传分化和居群水平低遗传多样性的主要原因。  相似文献

孑遗植物水杉的遗传多样性研究   总被引:36,自引:5,他引:31  
采用随机扩增多态DNA(RAPD)方法对孑遗植物水杉 (Metasequoiaglyptostroboides)的 8个居群 (包括 6个孑遗居群和 2个人工栽培居群 )的 48个个体进行了遗传多样性研究。用 16个随机引物共扩增出清晰谱带 5 7条。孑遗居群平均多态位点百分率为 38.6 %。各居群多态位点百分率差异显著 ( 10 .5 3%~ 33 .33 % ) :湖北利川的小河和桂花 2个居群较高 ,分别是 33 .33%和 31.5 8% ;湖南龙山洛塔居群较低 ,为 10 .5 3 %。人工栽培居群中 ,美国密西根州和湖北武汉 2个居群的多态位点百分率也较低 ,仅分别为 10 .5 3 %和 8.77%。用POPGENE 1.31版软件进行数据处理后结果如下 :孑遗居群的平均Nei’s基因多样性为 0 .130 5 ,Shannon’s遗传多样性信息指数为 0 .192 1,水杉孑遗居群遗传距离与地理距离的相关系数r =0 .2 346 6 (P =73 .2 7% ) ,基因分化系数Gst=0 .10 82 ,表明居群间有一定分化。用UPGMA方法聚类可知 ,2个人工居群与湖北利川居群优先聚类。鉴于目前水杉孑遗居群生境人为破坏严重、遗传多样性较低和原生母树分布分散 ,建议在湖北利川小河和桂花野生水杉最适生境建立水杉遗传多样性保护点 ,将现有的 2 5 0多株原生母树分别采种育苗定植于保护点 ,以达到最大限度地保存现有水杉遗传多样性的目的。  相似文献

利用ISSR标记对全国10个刺槐居群子代100个个体的遗传多样性进行了比较分析,从65个随机引物中筛选出10个多态性引物进行扩增,共检测到91个位点,多态位点数(AP)为85,多态位点百分率(P)为93.41%.刺槐在种级水平的遗传多样性参数略高于居群水平,多态位点百分率(P)分别为95.60%、69.01%,Shannon′s信息指数(I)分别为0.6145、0.3733,Nei′s基因多样性指数(H)分别为0.4337、0.2514.居群间的遗传分化指数Gst、Nei′s基因多样性指数和Shannon′s信息指数统计结果,均显示出中国刺槐居群内遗传多样性大于居群间遗传多样性.利用PopGen32软件对10个居群进行聚类分析可知,10个刺槐群体可分为三大类,亲缘关系和地理分布呈一定的相关性,但没有形成明显的地理变异模式.  相似文献

采用ISSR标记对中国北方地区的8个山荆子居群进行了遗传多样性分析,以探讨山荆子在苹果属植物演化过程中的作用.结果显示:(1)从55个ISSR引物中,筛选出11个多态性引物,共扩增出71个位点,其中多态性位点69个,多态位点达97.47%.(2)在物种水平上,有效等位基因数Ne=1.467 0,Nei's基因多样性(H)为0.284 6,Shannon信息指数(I)为0.439 9,多态位点百分率(PPB)=80.22;基因流(Nm)为1.659 1,居群间基因分化系数(Gst)为0.231 6,Φst值为0.236 0.(3)聚类分析结果显示,在遗传距离为0.155 5、0.136 9、0.125 8处8个山荆子居群分别聚为两大类、三亚类、四小类,但居群聚类位置与地理位置无明显的相关性.(4)山荆子的居群内遗传多样性贡献率占76.40%,而居群间的贡献率占23.60%,说明山荆子的遗传多样性主要分布在居群内,但居群间的遗传分化程度也较高.由此推测,山荆子的起源中心在华北和东北范围内,其中灵空山(山西)和塞罕坝(河北)居群可近似为核心居群;ISSR标记与花部的一些性状可能有连锁关系.  相似文献

利用ISSR分子标记技术,对濒危植物夏蜡梅10个居群的遗传多样性进行了分析.结果表明:夏蜡梅种的遗传多样性较高,多态位点百分率为73.08%,Shannon指数为0.3097,Nei指数为0.1987;而居群水平的遗传多样性较低,多态位点百分率平均为23.65%,Shannon指数平均为0.1251,Nei指数平均为0.0839.AMOVA分子差异分析显示:居群间遗传分化程度高,57.11%的变异存在于居群间,42.89%存在于居群内,基因分化系数(Gst)为0.5779;居群间的基因流为0.3651.生境的片断化使居群间的基因流受阻,可能是导致居群间高遗传分化和居群水平低遗传多样性的主要原因.10个居群间的平均遗传距离为0.1471.利用UPG-MA法对10个居群进行聚类,结果是天台县内的2个居群、临安市内的8个居群各组成一大类群.  相似文献

《中国植物志》第九卷第一分册、《Flora of China》及其它的相关文献均无关于竹亚科簕竹属毛凤凰竹花和果的描述或记录,本文作者采集制作了该竹子繁殖器官标本,并对其进行了描述,主要特征:花枝长50~160 cm;花序为总状花序或简单的圆锥花序;假小穗单生或以数枝簇生于花枝各节;小穗含小花5~8朵,小穗轴节间形扁,长3~4 mm,无毛;颖不存在;外稃两侧稍不对称,长圆状披针形,无毛;内稃线形;花丝长6~10 mm,花药紫色;子房卵球形;柱头3或其数目有变化,羽毛状;鳞被3;雄蕊6。花期3~4月。成熟颖果为细狭的长圆形,长0.8~1.0 cm,直径2.0~2.5 mm,棕褐色,果皮较厚。花标本采自厦门市园林植物园竹类植物区。  相似文献

以版纳省藤的萌蘖芽为试材,从取材、材料的处理、外植体的诱导分化、芽的增殖、生根以及影响试管苗形成几个重要因素等方面探讨了版纳省藤组织培养和快速繁殖的技术和方法,获得了生长素与细胞分裂素对芽的诱导分化及芽的增殖最佳配比浓度。以改良MS为基本培养基,在增殖培养基中添加0.5mg·L-1NAA、0.5mg·L-1KT和0.1mg·L-1BA,产生的有效苗最多;在生根培养基中添加1.5~2.0mg·L-1IBA生长素有利于版纳省藤组培苗生根。  相似文献

A new chlorarachniophytan alga, Gymnochlora stellata Ishida et Y. Hara gen. et sp. nov., has been isolated from Anae Island in Guam. It is a green, star-shaped, unicellular, amoeboid organism with several filopodia that do not form a reticulopodial network. Neither zoospores nor walled coccoid cells have been observed throughout the life cycle. The chloroplast ultrastructure is similar to those of described species; however, the pyrenoid matrix, which is invaded by many tubular structures originating from the inner membrane of the chloroplast envelope, is unique. A classification system is proposed for the Chlorarachniophyta. In this system, the ultrastructural features of the pyrenoid and the location of the nucleomorph in the periplastidial compartment are used as generic criteria, while the morphological features of the vegetative cells and life cycle patterns are used for species criteria. The described species, except for Cryptochlora perforans Calderon-Saenz et Schnetter, are also reassessed under the new system, and consequent nomenclatural requirements for the genus Chlorarachnion are dealt with in this paper. The taxonomic rank of a previously described species, Chlorarachnion globosum Ishida et Y. Hara, is elevated and Lotharella globosa (Ishida et Y. Hara) Ishida et Y. Hara gen. nov. et comb. nov. is proposed.  相似文献
