Young intact plants of maize (Zea mays L. cv INRA 508) were exposed to 2 to 4 kilopascals partial pressure oxygen (hypoxic pretreatment) for 18 hours before excision of the 5 millimeter root apex and treatment with strictly anaerobic conditions (anoxia). Hypoxic acclimation gave rise to larger amounts of ATP, to larger ATP/ADP and adenylate energy charge ratios, and to higher rates of ethanol production when excised root tips were subsequently made anaerobic, compared with root tips transferred directly from aerobic to anaerobic media. Improved energy metabolism following hypoxic pretreatment was associated with increased activity of alcohol dehydrogenase (ADH), and induction of ADH-2 isozymes. Roots of Adh1 mutant plants lacked constitutive ADH and only slowly produced ethanol when made anaerobic. Those that were hypoxically pretreated acclimated to anoxia with induction of ADH2 and a higher energy metabolism, and a rate of ethanol production comparable to that of nonmutants. All these responses were insensitive to the presence or absence of NO3. Additionally, the rate of ethanol production was about 50 times greater than the rate of reduction of NO3 to NO2. These results indicate that nitrate reductase does not compete effectively with ADH for NADH, or contribute to energy metabolism during anaerobic respiration in this tissue through nitrate reduction. Unacclimated root tips of wild type and Adhl mutants appeared not to survive more than 8 to 9 hours in strict anoxia; when hypoxically pretreated they tolerated periods under anoxia in excess of 22 hours.  相似文献

Seedlings of alcohol dehydrogenase 1 null mutants (Adh1-) of Zea mays L., which fail to synthesize alcohol dehydrogenase 1 (ADH1) isozymes, were hypoxically acclimated by 18 h of exposure to an atmosphere of 4% (v/v) O2 in N2 at 25[deg]C. Their ability to tolerate subsequent anoxia by exposure to anaerobic (O2-free) conditions was compared with that of unacclimated seedlings that were transferred immediately from an atmosphere of 40% (v/v) O2 to anaerobic conditions. Only 10% of the root tips of unacclimated seminal roots survived 6 h of anoxia, whereas 70% of the hypoxically acclimated root tips were viable at 24 h. During anoxia, acclimated root tips had enhanced ADH activity compared with unacclimated root tips, through induction of Adh2. Despite this, enzyme activity was still only about 5% that of acclimated, wild-type root tips and about half that of unacclimated, wild-type root tips. During anoxia, acclimated Adh1- root tips showed a higher rate of anaerobic respiration and ethanol production, greater concentrations of ATP and total adenylates, and a greater adenylate energy charge compared with unacclimated root tips. These results suggest that although enhanced ADH activity may have raised fermentation rates in acclimated Adh1- tissues and thereby contributed to energy metabolism and viability, the high levels of ADH activity inducible in acclimated, wild-type maize root tips appear to be in excess of that required to increase rates of fermentation.  相似文献

When root tips of fully aerobic, intact maize (Zea mays L.) seedlings are made anaerobic, viability normally is only 24 hours or less at 25°C. We find that viability can be extended to at least 96 hours if seedlings are given a hypoxic pretreatment for 18 hours by sparging the solution with 4% O2 in nitrogen (v/v) before anoxia. Fully aerobic root tips (sparged with 40% O2) had very low alcohol dehydrogenase (ADH) activity (per gram root fresh weight), and the level remained low under anoxia. In hypoxically pretreated roots, however, high levels of ADH activity were induced, and activity rose further during the initial 24 hours of anoxia, and then remained high at about 20 times that of controls in 40% O2. ADH activity in roots in solution sparged with air (21% O2) was about three times that in 40% O2. Improved viability of hypoxically pretreated root tips was associated with maintenance of a high energy metabolism (ATP concentration, total adenylates, and adenylate energy charge). Roots that were not pretreated lost 94% of the total adenylates and ATP at 24 hours of anoxia. The relation between induced ADH activity, energy metabolism, and improved anoxia-tolerance in acclimated maize root tips is discussed.  相似文献

We tested the hypothesis (J.-H. Xia and P.H. Saglio [1992] Plant Physiol 100: 40-46) that the enhanced ability of maize (Zea mays) root tips to survive anoxia, elicited by a 4-h exposure to 3% O2 ("acclimation"), is due to less cytoplasmic acidosis early in anoxia. Cytoplasmic pH and fermentation reactions were monitored in excised and intact (attached) maize root tips by simultaneous in vivo 13C- and 31P-NMR spectroscopy. We demonstrate that both excised and intact acclimated root tips have significantly higher cytoplasmic pH values under anoxia. This reduction in cytoplasmic acidosis is greater in intact root tips. Remarkably, cytoplasmic pH does not change when root tips are transferred from 3% O2 to anoxia. The earlier observation of considerable lactate efflux and lowered intracellular lactate in excised, acclimated root tips (ibid.) was extended to intact seedlings. The predominant fermentation end product retained in the cells of acclimated root tips is alanine. We discuss the relationship between cytoplasmic pH and levels of intracellular lactate and alanine in sugar-replete roots, and the role of cytoplasmic pH in determining survival under anoxia.  相似文献

A hypoxic pre-treatment (HPT) can improve the anoxic survival of flooding sensitive plants. Here, we tested whether a 4-d HPT of wheat plants (Triticum aestivum L.) would improve their anoxic resistance, and if so, why. We found that the metabolic adjustment during prolonged HPT involved an increased lactate excretion rate, the up-regulation of glycolytic and fermentative enzymes as well as the accumulation of various sugars. Therefore, HPT wheat roots could sustain a 3 times higher ethanolic fermentation rate during an anoxic period compared to non-pre-treated (NHPT) roots. Nevertheless, the enhanced fermentation rate provided temporary relief to the energy crisis only, and both NHPT and HPT plants died after 5d of anoxia in illumination. Comparison of different low oxygen incubation systems using excised roots or roots of intact plants revealed striking differences. The benefits of intact shoots, oxygen transport as well as additional sugar supply enabled a more stable energy supply of anoxia-treated NHPT and HPT roots. However, the height of the fermentation rate was correlated with a high ATP content during dark anoxic incubation, but not in illumination.  相似文献

Summary Under anoxia, rice coleoptiles have a remarkable capacity to grow and to preserve undamaged mitochondrial structure and functions. The transfer of aerobically grown intact seedlings to anaerobic conditions resulted in the appearance of unusual mitochondria in coleoptiles as well as in leaves and roots. These mitochondria become filled with stacks of extended cristae, but, obviously, are not affected structurally (only in the root cortex the cells are damaged after a longer period without oxygen). On the contrary, the mitochondria and other organelles of excised coleoptiles, roots and leaves disintegrate after a relatively short exposure to an oxygen-free environment. The degeneration can be avoided if the excised organs are supplied with glucose. Then the mitochondrial fine structure resembles that of intact plants kept under anaerobic conditions.The observations suggest that the capacity of rice coleoptiles to grow under anoxia and to preserve undamaged mitochondria and other organelles is not caused by the resistance of the cell organelles to oxygen deficiency, but rather by the ability of the seedling to transport organic compounds easily, even under the exclusion of oxygen, from the grain to the coleoptile where they can be utilized by glycolysis.  相似文献

1. Incubation of hepatocytes from fed or starved rats with increasing glucose concentrations caused a stimulation of lactate production, which was further increased under anaerobic conditions. 2. When glycolysis was stimulated by anoxia, [fructose 2,6-bis-phosphate] was decreased, indicating that this ester could not be responsible for the onset of anaerobic glycolysis. In addition, the effect of glucose in increasing [fructose 2,6-bisphosphate] under aerobic conditions was greatly impaired in anoxic hepatocytes. [Fructose 2,6-bisphosphate] was also diminished in ischaemic liver, skeletal muscle and heart. 3. The following changes in metabolite concentration were observed in anaerobic hepatocytes: AMP, ADP, lactate and L-glycerol 3-phosphate were increased; ATP, citrate and pyruvate were decreased: phosphoenolpyruvate and hexose 6-phosphates were little affected. Concentrations of adenine nucleotides were, however, little changed by anoxia when hepatocytes from fed rats were incubated with 50 mM-glucose. 4. The activity of ATP:fructose 6-phosphate 2-phosphotransferase was not affected by anoxia but decreased by cyclic AMP. 5. The role of fructose 2,6-bisphosphate in the regulation of glycolysis is discussed.  相似文献

Hypoxic pretreatment (3 kPa oxygen) of maize (Zea mays L.) root tips improved their survival time in a subsequent anoxic incubation from 10 h to more than 3 d, provided that glucose was added to the medium to sustain metabolism. The glycolytic flux (lactate + ethanol) was the same in both pretreated and untreated root tips during the 1st h after transfer to anoxia. It was only after 2 h that it declined sharply in untreated tips, but was sustained in pretreated ones. Right after the transition from normoxia to anoxia of untreated root tips, the only fermentative product detected was lactic acid, which accumulated in a 7:1 proportion after 30 min in tissue and medium, respectively. It took 10 min before ethanol could be detected and 20 min for it to be produced at its maximum rate at the expense of lactate production, which slowed down. In contrast, in hypoxically pretreated root tips, ethanol was produced at a maximum rate right after the transfer to anoxia. Concurrently, low amounts of lactic acid were produced that accumulated in a 1:1 proportion after 30 min in tissue and medium, respectively. This large efflux of lactic acid could account for the higher cytoplasmic pH values always found in pretreated tissues. The presence of cycloheximide during pretreatment abolished this difference, suggesting that the greater efficiency of lactate efflux was linked to protein synthesis. The role of lactate in cytosolic pH regulation and in sensitivity to anoxia is discussed.  相似文献

Prior exposure of roots of intact wheat seedlings for 15–30h to hypoxia (0016-006 mol m 02) greatly increased their toleranceto subsequent anoxia, as assessed by the ability of the rootsto elongate upon return to air. Such hypoxically pretreatedroots had 2–4-fold higher activities of pyruvate decarboxylase(PDC) and 35–l7-fold higher activities of alcohol dehydrogenase(ADH) in their 0–1 mm apices and 0–5 mm root tipsthan in apices and tips of roots pretreated in air (026–031mol m3 02). The ADH/ PDC ratio increased I 3–5-fold duringhypoxic pretreatment. Furthermore, the rate of alcoholic fermentationby 0–5 mm tips of the hypoxically pretreated roots was14-4-fold faster than in tips from aerobically pretreated roots.No consistent difference between 02 pretreatment was found foralcoholic fermentation by tissues taken between 10 and 20 mmfrom the root tip. The observed activities of PDC and rates of alcoholic fermentationindicate that alcoholic fermentation is usually rate-limitedby PDC in 0–1 mm apices and 0–5 mm tips of wheatroots. Comparisons with data in the literature indicate thatwheat has at most a small Pasteur effect, which may explainwhy wheat is more intolerant to anoxia than rice. Exogenous glucose delayed the loss of elongation potential inboth aerobically and hypoxically pretreated roots. In the absenceof glucose, more than 85% of aerobically pretreated roots hadlost their elongation potential after 9 h anoxia, compared with30% in the presence of glucose. After 21 h anoxia nearly allaerobically pretreated roots had lost their elongation potential,compared with 10% and 0% of hypoxically pretreated roots inthe absence and presence, of glucose, respectively. The protective effect of glucose was presumably not due to anendogenous sugar deficiency; at the start of anoxia, 0–1mm apices of aerobically pretreated roots contained sufficientsugar for 23 h of their measured rate of ethanol synthesis yet,85% of these apices had lost their elongation potential afteronly 9 h of anoxia. It is suggested that in wheat roots, lowrates of synthesis of ethanol and hence of ATP, lead to injuryof cells, in turn generating a requirement for exogenous glucose,despite high endogenous sugar concentrations. Tolerance of anoxia in maize root tips is greatly improved when seedlings are pretreated with 2 to 4 h of hypoxia. We describe the patterns of protein synthesis during hypoxic acclimation and anoxia. We quantified the incorporation of [(35)S]methionine into total protein and 262 individual proteins under different oxygen tensions. Proteins synthesized most rapidly under normoxic conditions continued to account for most of the proteins synthesized during hypoxic acclimation, while the production of a very few proteins was selectively enhanced. When acclimated root tips were placed under anoxia, protein synthesis was depressed and no "new" proteins were detected. We present evidence that protein synthesis during acclimation, but not during subsequent anoxia, is crucial for acclimation. The complex and quantitative changes in protein synthesis during acclimation necessitate identification of large numbers of individual proteins. We show that mass spectrometry can be effectively used to identify plant proteins arrayed by two-dimensional gel electrophoresis. Of the 48 protein spots analyzed, 46 were identified by matching to the protein database. We describe the expression of proteins involved in a wide range of cellular functions, including previously reported anaerobic proteins, and discuss their possible roles in adaptation of plants to low-oxygen stress.  相似文献

Elongation by stems of overwintered tubers of Potamogeton pectinatus (L.) is strongly promoted over several days by oxygen-free conditions. Characteristics of the respiration underpinning this unusual response were examined. Anaerobic plants produced ethanol and CO(2) in approximately equimolar amounts, indicating that glycolysis coupled to alcoholic fermentation was the principal CO(2)-producing respiratory pathway. Rates of CO(2) evolution by aerobic and anaerobic whole plants (shoot and tuber) were similar, suggesting a rate of glycolysis three times that of aerobic plants, i.e. a strong Pasteur effect. In the shoot alone, anaerobic CO(2) production was twice the aerobic rate indicating a 6-fold increase in the rate of glycolysis in this tissue. Anoxic stems contained more sucrose at a stronger concentration than slower-growing aerobic stems or anaerobic leaves, demonstrating that sugar supply to the site of most rapid growth exceeded demand in the absence of oxygen. Concentrations of potentially toxic acetaldehyde in the external medium were small (approximately 0.2 mol m(-3)) during anoxia and on return to aerated conditions. Lactic acid was undetectable under anaerobic conditions and in vivo (31)P-NMR analysis of shoots revealed a cytoplasmic acidification of only 相似文献

We tested the hypothesis that H+ extrusion contributes to cytoplasmic pH regulation and tolerance of anoxia in maize (Zea mays) root tips. We studied root tips of whole seedlings that were acclimated to a low-oxygen environment by pretreatment in 3% (v/v) O2. Acclimated root tips characteristically regulate cytoplasmic pH near neutrality and survive prolonged anoxia, whereas nonacclimated tips undergo severe cytoplasmic acidosis and die much more quickly. We show that the plasma membrane H+-ATPase can operate under anoxia and that net H+ extrusion increases when cytoplasmic pH falls. However, at an external pH near 6.0, H+ extrusion contributes little to cytoplasmic pH regulation. At more acidic external pH values, net H+ flux into root tips increases dramatically, leading to a decrease in cytoplasmic pH and reduced tolerance of anoxia. We present evidence that, under these conditions, H+ pumps are activated to partly offset acidosis due to H+ influx and, thereby, contribute to cytoplasmic pH regulation and tolerance of anoxia. The regulation of H+ extrusion under anoxia is discussed with respect to the acclimation response and mechanisms of intracellular pH regulation in aerobic plant cells.  相似文献

The extent of phosphate uptake measured by the relative changes in cytoplasmic Pi, vacuolar Pi, ATP, glucose-6-phosphate, and UDPG was determined using in vivo31P nuclear magnetic resonance spectroscopy. Maize (Zea mays) root tips were perfused with a solution containing 0.5 or 1.0 millimolar phosphate at pH ~6.5 under different conditions. In the aerated state, phosphate uptake resulted in a significant increase (>80%) in vacuolar Pi, but cytoplasmic Pi only transiently increased by 10%. Under N2, the cytoplasmic Pi increased ~150% which could be attributed to a large extent to the breakdown of ATP, sugar phosphates and UDPG. Vacuolar Pi increased but only to the extent of ~10% of that seen under aerobic conditions. 2-deoxyglucose pretreatment was utilized to decrease the level of cytoplasmic Pi. When pretreated with the 2-deoxyglucose, the excised maize roots absorbed phosphate from the perfusate with a significant increase in the cytoplasmic Pi. The increase could only be traced to external phosphate since the concentrations of other phosphorus containing species remained constant during the uptake period. With 2-deoxyglucose pretreatment, phosphate uptake under anaerobic conditions was substantially inhibited with only the vacuolar phosphate showing a slight increase. When roots were treated with carbonyl cyanide m-chlorophenyl hydrazone, no detectable Pi uptake was found. These results were used to propose a H+-ATPase related transport mechanism for phosphate uptake and compartmentation in corn root cells.  相似文献

The effects of in vitro anoxia on the release of glutamate in isolated nerve terminals were studied. The extra-synaptosomal concentration of glutamate ([Glu]ext) under aerobic conditions was 2.3 microM and increased to 4.9 microM after 10 min of anoxia. However, when synaptosomes were incubated in the presence of lactate plus pyruvate instead of glucose, to prevent anaerobic glycolysis, anoxia induced an eightfold increase in the [Glu]ext. The accumulation of glutamate in the external medium during anoxia was Ca2+ independent and insensitive to a significant reduction of the Ca(2+)-dependent release of the amino acid. These results indicate that a Ca(2+)-independent efflux of cytoplasmic glutamate occurs during in vitro anoxia in isolated nerve terminals.  相似文献

Incubation of 3-d-old seedlings of Oryza sativa L. cv Arborio under anaerobic conditions, leads to a large increase in the titer of free putrescine while aerobic incubation causes a slight decrease. After 2 days, the putrescine level is about 2.5 times greater without oxygen than in air. The rice coleoptile also accumulates a large amount of bound putrescine and, to a lesser extent, spermidine and spermine (mainly as acid-soluble conjugates). Accumulation of conjugates in the roots is severely inhibited by the anaerobic treatment. Feeding experiments with labeled amino acids showed that anoxia stimulates the release of 14CO2 from tissues fed with [14C]arginine and that arginine is the precursor in putrescine biosynthesis. After 2 d of anoxia, the activity of arginine decarboxylase was 42% and 89% greater in coleoptile and root, respectively, than in the aerobic condition. The causes of the differences in polyamine metabolism in anoxic coleoptiles and roots are discussed.  相似文献

The radial movement of oxygen in excised corn and jack bean roots was measured with a platinum wire electrode embedded in the root tissue. Measurements were made with the roots exposed to air and with the roots immersed in nutrient solution in the presence and absence of millimolar sodium azide. Effective rates of oxygen diffusion in the root tissue were also measured from 5 to 30 C and compared to the respiration rates of similar root segments over the same temperature range. Under conditions which allow the roots to exude freely, the interior of the root operates under an oxygen deficit. Inhibition of respiratory oxygen uptake by low temperature or azide treatment increased the flux of oxygen to the root interior.  相似文献

Radial oxygen loss (ROL) has been suggested to be a major process to protect plants exposed to root anaerobic stress. In the present study, we aimed to test the importance of root porosity and radial oxygen loss on the aquatic macrophyte resistance to sediment anoxia. We expected that species living in eutrophic environments characterized by anaerobic conditions in sediments exhibited higher root porosity and radial oxygen loss than species restrained to oligotrophic environments. In this way, we compared the responses to sediment anoxia of two hydrophyte species growing under meso-eutrophic conditions in the field (Myriophyllum spicatum L. and Vallisneria spiralis L.) and three species growing under oligotrophic conditions (Potamogeton coloratus Horne, Elodea canadensis Michx and Sparganium emersum Michx.). Under laboratory conditions, ROL, root porosity, plant metabolism (aerobic respiration, photosynthesis, root fermentative activity) and plant growth were analysed after 3?months of acclimation in anaerobic sediments and compared with control values obtained from aerobic sediments. The results showed that two meso-eutrophic species (M. spicatum and V. spiralis) survived in anaerobic sediments and maintained similar photosynthesis rates than those measured under aerobic conditions. In contrast, the three oligotrophic species (P. coloratus, E. canadensis and S. emersum) suffered net biomass loss and depressed their photosynthesis rates under anaerobic conditions. All variables associated with plant tolerance to anaerobic conditions (maintenance of photosynthesis, aerobic respiration and growth rate, and limitation of root fermentative activity) were positively linked to root porosity and ROL. According to our hypothesis, species that could survive to anaerobic conditions were the species able to increase their root porosity and ROL under these conditions. Thus, in ecological studies, it would be useful to use the root porosity and ROL plasticity as biological traits in order to model the distribution of macrophytes in river floodplains.  相似文献

Nitrate reduction, nitrite reduction, and ammonium assimilationwere measured over c. 24 h in excised sterile barley roots,in air or under low oxygen tensions. Partial anoxia had relativelylittle effect, but the pathway of nitrogen assimilation wasseverely inhibited during complete anoxia, when the uptake ofnitrate ceased. Much of the nitrate which was present in theroots at the time of excision was apparently unavailable forassimilation. None of the reactions of the pathway served inplace of oxygen as an electron acceptor under anaerobic conditions.The concentration of nitrate in the external solution duringgrowth and during the experimental treatments had no directeffect on anaerobic ethanol formation, although an indirecteffect was noted which was due to variations in the carbohydratecontent of the tissue.  相似文献
