药敏药片对国际三株标准菌试验,抑菌圈直径范围均符合判定标准。与进口药敏药片符合率为100%。与药敏纸片片间差比较试验,药片抑菌圈=1~2mm,精密度高,符合国际要求,而纸片抑菌圈=9~13mm,均匀度检测明显不合格。药片变异系数CV=1.31~2.92,纸片CV=14.78~21.67表明药片变量离散度小,片间差很小;而纸片变量离散度明显大,故纸片片间差很大。  相似文献

本文报导省市级三所医院同时用药敏药片和药敏纸片对国际标准质株做抑菌试验,其结果:药片片重差变异系数cv=2.5,纸片cv=14.4.抽样五种抗菌素抑菌环直径差变异系数分别为:药片平均cv=3.07,而纸片平均cv=6.85;均匀度L药片为2-3mm纸片为3-5mm.表明药片片间差小,均匀度、准确度合格,药物含量均匀,精密度好于纸片.  相似文献

应用ELISA和PCR法检测502例乙肝病人血清,401例HBsAg阳性血清中,有114例(28.4%)抗-HCV和HCVRNA双项阳性,25例(6.2%)HCVRNA单项阳性;21例(5.2%)抗-HCV单项阳性。将HBsAg乙肝病人分成HBVDNA,HBeAg阳性组和HBVDNA,HBeAg阴性组。前者抗-HCV阳性率为11.6%~20.5%,HCVRNA阳性率为16.2%~20.5%。后者抗-HCV阳性率为20.2%~55.6%,HCVRNA阳性率为23%~60.3%。结果说明长期携带HBV者和慢性乙肝病人均可重叠HCV感染。HBVDNA阳性组抗-HCV和HCVRNA阳性率明显高于HBVDNA阳性组  相似文献

重组大肠杆菌Escherichia coliHMS174(pTZ18UPHB) 含有携带聚羟基烷酸(PHA) 合成基因( phaCAB)** 的质粒pTZ18UPHB,是很有潜力的PHA 生产菌,但存在着质粒不稳定和不能合成3羟基丁酸(3HB) 与3羟基戊酸(3HV) 共聚物[P(3HBco3HV)] 的缺陷。将RK2 质粒上的par DE 基因引入pTZ18UPHB 构成质粒pJMC2 ,该质粒可以在宿主E.ColiHMS174 中稳定遗传。将培养基中的磷酸盐浓度降至18 m mol/L,发现E.Coli HMS174(pJMC2) 能够以丙酸为前体合成P(3HBco3HV) ,其中3HV 在共聚物中的含量为5 % ~8 % 。在5L自动发酵罐中分批补料培养E.Coli HMS174(pJMC2) ,培养基初始磷酸盐浓度为15 m mol/L,30 h 后每升培养液中干菌体可达42-5 g,P(3HBco3HV) 占干重的70 % ,其中3HV 在共聚物中的含量为4-9 % 。  相似文献

将编码柯萨奇B3病毒(CVB3)衣壳蛋白VP1和VP2的基因,分别克隆到具有7.5k启动子的痘苗病毒表达载体pGJP5上;将CVB3衣壳蛋白全基因克隆到具有T7启动子的痘苗表达载体pTM1上,并筛先到相应的重组痘苗病毒VVP1、VVP2和VVP/4/2/3/1。VVP1和VVP2稳定表达产物为CVB3衣壳蛋白VP1和VP2,而VVP4/2/3/1为一无分泌性的多聚蛋白,且这三种表达产物均属无分泌性  相似文献

以甲基藉精(MV)O-1mmol/L在1500μmol m^-2a^-1光下处理C3植物花生、水稻和C4植物玉米、甘蔗的叶圆片30mm,O2^2+产生速率随MV浓度提高而加快MV浓度超过10μmol/L,光合放氧出现负植并持续增大。光氧化作用降低叶绿素荧光参数FV/Fm,ΦPSⅡ和qp,而qN则或提高(C3植物,MV10μmol/L)或几平下)。热耗散系数KD的变化与QN相似。秘C4植物相比,C3  相似文献

利用DNA重组技术,将Ⅲ型中国株HCVE2/NS1基因片段插入真核表达载体,然后转染哺乳动物细胞NIH3T3以表达E2糖蛋白.检测显示来自3月以上培养的细胞克隆中表达产物分子量为70kD,经Westernblot证实该表达产物能与抗HCV阳性血清进行特异性反应.以上表明首次在哺乳动物细胞中成功表达Ⅲ型中国株HCV的E2糖蛋白,并建立相应的稳定表达细胞系.  相似文献

应用逆转录-聚合酶链反应(RT-PCR)技术,从HCV感染者血清中扩增编码HCV病毒蛋白酶的NS2-NS3cDNA片段,在其5′和3′端分别引入EcoRⅠ和XbaⅠ限制性内切酶位点,定向克隆至真核表达载体pcDNA3,构建重组载体pcDNA-NS23,重组表达载体经限制性内切酶消化鉴定.用SP6和T7通用引物对目的基因片断进行序列分析.序列同源性分析结果表明,与HCV-J、HC-C2有高度的同源性,与HCV-1、HCV-J6、HCV-J8同源性差,提示所克隆的基因属HCVⅡ型.该区内重要的功能位点如Zn2+依赖性金属蛋白酶催化中心、丝氨酸蛋白酶催化中心等均高度保守  相似文献

本文报道云南热带地区尾孢菌属及其近似属真菌45种,其中有一个新种:番木瓜短胖孢(CercosporidiumpapayaeY.L.Guo,sp.nov.),寄生在番木瓜科(Caricaceae)番木瓜(CaricapapayaL.)叶上。短胖孢属真菌在番木瓜科植物上是首次报道。文中为新种提供了拉丁文简介、描述并附图。模式标本保藏在中国科学院微生物研究所真菌标本室(HMAS)。寄生在番木瓜(CaricapapayaL.)上的CercosporamamaonisViegas&Chupp与本种近似,二者都具有子座,分生孢子梗紧密簇生,但前者分生孢子梗(5~25×2~3μm)和分生孢子(10~45×2~3μm)均短而窄。番木瓜尾孢(CercosporapapayaeHansf.)与本种的区别在于无子座,分生孢子梗色泽深(中度褐色),分生孢子梗长(50~200×3.5~6μm),分生孢子无色,针形,窄(20~75×3~5μm)。  相似文献

The distribution of microorganisms in soil aggregates with different diameters was determined using a “washing and sonic vibration” method. In humic rendzina the number of bacteria, actinomycetes and fungi located in aggregates measuring 3 to 1 and ≤0.5 mm was greater than in those of 7 to 5 mm.Pseudomonas were more numerous in aggregates of ≤0.5 mm than in those of 3 to 1 mm and spore-forming aerobic bacteria—in aggregates measuring 3 to 1 mm than in those of 7 to 5 mm. The number of microorganisms growing on asparagine agar andArthrobacter-Corynebacterium increased as 7-5<3-1<0.5 and 7-5<0.5<3-1 mm, respectively. In podzolic loess spore-forming aerobic bacteria inhabited preferentially aggregates measuring 7 to 5 mm,Arthrobacter-Corynebacterium aggregates of ≤0.5 mm. The number of bacteria was greater in aggregates of 3 to 1 than in those measuring ≤0.5 mm. Aggregates of various diameters differed also in the number of some microorganisms both in the outer and inner parts and the partition ratio of microorganisms between these parts. Differences were more numerous in the humic rendzina aggregates.  相似文献

The effectiveness of treating bacterial infections is seriously threatened by the emergence of bacterial resistance to chemical treatment. Growth of microbes in biofilm is one of the main causes of resistance to antimicrobial drugs. Quorum sensing (QS) inhibition, which targets the QS signalling system by obstructing cell-cell communication, was developed as an alternative treatment by creating innovative anti-biofilm drugs. Therefore, the goal of this study is to develop novel antimicrobial drugs that are effective against Pseudomonas aeruginosa by inhibiting QS and acting as anti-biofilm agents. In this study, N-(2- and 3-pyridinyl)benzamide derivatives were selected to design and syntheses. Antibiofilm activity was revealed by all the synthesized compounds and the biofilm was visibly impaired, and the OD595nm readings of solubilized biofilm cells presented a momentous difference between the treated and untreated biofilms. The best anti-QS zone was observed for compound 5d and found to be 4.96 mm. Through in silico research, the physicochemical characteristics and binding manner of these produced compounds were examined. For the purpose of understanding the stability of the protein and ligand complex, molecular dynamic simulation was also carried out. The overall findings showed that N-(2- and 3-pyridinyl)benzamide derivatives could be the key to creating effective newer anti-quorum sensing drugs that are effective against different bacteria.  相似文献

Ninety clinical isolates of Pseudomonas aeruginosa were examined for susceptibility to carbenicillin by the broth dilution and disc diffusion methods. Inhibition zone diameters varied at given minimal inhibitory concentration levels of the antibiotic. Nevertheless, the results obtained allowed the proposal of the following tentative criteria for the interpretation of inhibition zones. Pseudomonadaceae yielding zones of inhibition measuring at least 10 and 16 mm in diameter around 25-and 100-mug discs, respectively, are sensitive to this antibiotic when examined by the standardized Bauer-Kirby method of disc susceptibility testing. Isolates characterized by zones of less than 100 mm in diameter around 25-mug discs should be tested with 100-mug discs before they are reported as sensitive or resistant to carbenicillin.  相似文献

Nasal mucous velocity was estimated by following the motion of radiopaque discs of Teflon by means of a fluoroscopic image intensifier. From 5 to 10 discs were deposited on the superior surface of the inferior turbinate with a forceps. No local anesthesia was employed and the subjects experienced no discomfort. The linear velocity of the discs was obtained by playing the videotape onto a television monitor, measuring distance with a ruler, and dividing by elapsed time. Duplicate runs of 1-2 min, 15 min apart were very reproducible but runs at 4-h intervals or daily over a 5-day period had a coefficient of variation of 30%. Average nasal velocity for individual ranged from 0 to 22.5 mm/min and group means ranged from 6. 8 to 10.8 mm/min. There was no statistically significant difference in nasal mucous velocity between young and elderly subjects nor was there a sexual difference. The saccharin test of nasal mucous transport was unsatisfactory because of inability to repeat the test more often than 1-2 h and its propensity to produce mild discomfort in a significant number of subjects. Saccharin times did not correlate significantly with values of nasal mucous velocity.  相似文献

Background

The present study reports the antibacterial capacity of alkaloid compounds in combination with Methicillin and Ampicillin-resistants bacteria isolated from clinical samples. The resistance of different bacteria strains to the current antibacterial agents, their toxicity and the cost of the treatment have led to the development of natural products against the bacteria resistant infections when applied in combination with conventional antimicrobial drugs.

Method

The antibacterial assays in this study were performed by using inhibition zone diameters, MIC, MBC methods, the time-kill assay and the Fractional Inhibitory Concentration Index (FICI) determination. On the whole, fifteen Gram-positive bacterial strains (MRSA/ARSA) were used. Negative control was prepared using discs impregnated with 10 % DMSO in water and commercially available Methicillin and Ampicillin from Alkom Laboratories LTD were used as positive reference standards for all bacterial strains.

Results

We noticed that the highest activities were founded with the combination of alkaloid compounds and conventional antibiotics against all bacteria strains. Then, results showed that after 7 h exposition there was no viable microorganism in the initial inoculums.

Conclusion

The results of this study showed that alkaloid compounds in combination with conventional antibiotics (Methicillin, Ampicillin) exhibited antimicrobial effects against microorganisms tested. These results validate the ethno-botanical use of Cienfuegosia digitata Cav. (Malvaceae) in Burkina Faso. Moreover, this study demonstrates the potential of this herbaceous as a source of antibacterial agent that could be effectively used for future health care purposes.  相似文献

In this paper, a wild boar head was taken as the bionic research object for the development of new ridgers, a kind of plough. The reverse engineering technology was adopted to obtain the surface geometrical information of the head. Several three-dimensional (3D) point clouds of the head were captured first using a non-touch laser scanner, and an integrated point cloud was generated by aligning these point clouds using UG/Imageware. Then, the digital surface model of the head was rebuilt by means of CATIA. The characteristic curves of the surface model were analyzed. The results show that the average error between the rebuilt surface and the point cloud is -0.431 ram. The max curvature of the ridge on the neb of the head is 0.187 mm^-1, and the max and rain Gauss curvatures on the surface are 0.008 mm^-2 and -0.002 mm^-2. These geometrical information are the essential parameters for biomimetics study of the ridger.  相似文献

Chicken skeletal muscle taken from embryos in ovo was examined by thin-section electron microscopy. Measurements of filament diameters reveal three nonoverlapping groups of filaments: thin (actin myofibrillar) filaments with mean diameters of 5.3 +/- 0.6 nm (S.D.), thick (myosin myofibrillar) filaments with mean diameters of 15 +/- 1.4 nm, and intermediate filaments with mean diameters of 9.3 +/- 0.9 nm. During muscle development these diameters do not change. By counting the number of filaments observed in the sarcoplasm at different stages, we find that the spatial density of intermediate filaments decreases during avian myogenesis in ovo, from 91 intermediate filaments/micron 2 at 6 days to 43 intermediate filaments/micron 2 at 17 days in ovo. Initially randomly arranged, some intermediate filaments become associated with Z discs, sarcoplasmic reticulum, nuclear membrane, and the sarcolemma between 6 and 10 days in ovo. These associated intermediate filaments course both parallel and transverse to myofibrils, forming lateral connections between myofibrillar Z discs and longitudinal connections from Z disc to Z disc within myofibrils. Intermediate filaments also appear to connect Z discs with the nuclear membrane. The intermediate filament associations persist through day 17 of development, after which the presence of cytoskeletal filaments is obscured by the densely packed myofibrils and membranes. Intermediate filament distribution becomes anisotropic during development. A greater proportion of intermediate filaments in the immediate perimyofibrillar area are oriented parallel to myofibrils than in other areas, so that the majority of the intermediate filaments nearest the myofibrils course parallel to them. The longitudinal intramyofibrillar intermediate filaments persist throughout development, as shown by their existence in KI-extracted adult myofibrils.  相似文献
