探讨红莲型细胞质雄性不育(HL-CMS)水稻不育系‘粤泰A’(‘YTA’)和保持系‘粤泰B’(‘YTB’)中3个腺苷酸转位酶(ANT)基因在三叶期根、茎、叶以及不同发育时期的幼穗中的表达模式。结果表明:ANT1和ANT2在‘YTA’和‘YTB’三叶期的根、茎、叶中的表达量都较高,而在生殖生长不同时期的幼穗中表达量较低。‘YTA’中ANT1在不同时期的幼穗中表达量都较低,而ANT2的表达量到穗生长期II的幼穗才较低。‘YTB’中ANT1在生殖生长的穗生长期III的幼穗有很高的表达。ANT3的表达水平在研究的各组织中的表达都较低,在‘YTB’穗生长期V的幼穗中表达最高。相对于‘YTA’,‘YTB’中只有ANT3在穗生长期III和穗生长期V的幼穗中呈现明显的优势表达;而相对于‘YTB’,‘YTA’中ANT2和ANT3在穗生长期I、ANT2在穗生长期VI幼穗具有明显的优势表达。3个ANTs基因在HL-CMS不育系‘YTA’和保持系‘YTB’不同组织及发育时期的表达模式的差异,暗示它们可能与HL-CMS水稻不育系和保持系的发育调控有关。  相似文献

Background

Cytoplasmic male sterility (CMS) has often been associated with abnormal mitochondrial open reading frames. The mitochondrial gene orfH79 is a candidate gene for causing the CMS trait in CMS-Honglian (CMS-HL) rice. However, whether the orfH79 expression can actually induce CMS in rice remains unclear.

Results

Western blot analysis revealed that the ORFH79 protein is mainly present in mitochondria of CMS-HL rice and is absent in the fertile line. To investigate the function of ORFH79 protein in mitochondria, this gene was fused to a mitochondrial transit peptide sequence and used to transform wild type rice, where its expression induced the gametophytic male sterile phenotype. In addition, excessive accumulation of reactive oxygen species (ROS) in the microspore, a reduced ATP/ADP ratio, decreased mitochondrial membrane potential and a lower respiration rate in the transgenic plants were found to be similar to those in CMS-HL rice. Moreover, retarded growth of primary and lateral roots accompanied by abnormal accumulation of ROS in the root tip was observed in both transgenic rice and CMS-HL rice (YTA).

Conclusion

These results suggest that the expression of orfH79 in mitochondria impairs mitochondrial function, which affects the development of both male gametophytes and the roots of CMS-HL rice.  相似文献

To elucidate reactive oxygen species (ROS) metabolism of cotton cytoplasmic male sterility and the effects of restorer gene on the metabolism of ROS, the metabolism changes in the production and scavenging of ROS and gene expression related to ROS-scavenging enzymes were investigated in the anther mitochondria of CMS line, maintainer line and hybrid F1. During the abortion preliminary stage (sporogenous cell division stage), anthers of CMS line had a little higher superoxide (O2) production rate and hydrogen peroxide (H2O2) and malondialdehyde (MDA) contents than those of maintainer or hybrid F1. Simultaneously, a little higher ROS contents might serve as a signal to increase the activity of superoxide dismutase (SOD) in anthers of CMS line to reduce the ROS damage to the anther development. But at the abortion peak (pollen mother cell meiosis stage), anthers of CMS line had extraordinarily higher ROS contents and lower ROS-scavenging enzymic activities compared with the hybrid F1, during which the ROS contents and ROS-scavenging enzymic activities in hybrid F1 were approximate to those of maintainer line. The expression of Mn-sod and apx mRNA in anther of CMS line was obviously inhibited when ROS produced with a great deal during anther abortion, however the gene expression in hybrid F1 kept normal with the maintainer. Excessive accumulation of O2·−, H2O2 and MDA, significant reduction of ROS-scavenging enzymic activities and lower gene expression level of ROS-scavenging enzyme were coinstantaneous with male cells death in anthers of CMS line. But when the restorer gene was transferred into CMS line, excessive production of ROS could be eliminated in the anthers of hybrid F1. The restorer gene likely plays an important role in keeping the dynamic balance between the production and elimination of ROS.  相似文献

红莲型水稻细胞质雄性不育花粉总蛋白质初步比较分析   总被引:4,自引:0,他引:4  
采用固相pH梯度/SDS-PAGE双向电泳对红莲型细胞质雄性不育水稻的不育系(YTA)和保持系(YTB)二核期花粉总蛋白质进行了分离,通过银染显色,获得了分辨率和重复性较好的双向电泳图谱。用PDQuest2DE软件可识别约1500个蛋白质点,其中差异表达的蛋白质点数为120。将其中15个差异点采用基质辅助激光解析电离飞行时间质谱(matrix assisted laser desorption/ionizaton time of flight mass spectrometry,MALDI-TOF-MS)进行了肽质指纹图分析,通过采用Mascot软件对MSDB数据库查询,其中7个蛋白质点得到了鉴定。YTA相对于YTB有部分参与物质和能量代谢的蛋白质缺失或表达量降低,这些蛋白质分别是水稻线粒体H -转运ATPase(H -ATPase)α链、盐诱导型膜联蛋白、线粒体NAD -依赖型苹果酶和磷酸核糖焦磷酸合成酶等。这些蛋白质的表达下调或缺失可能与线粒体提供能量不足而导致的花粉不能正常发育有关。线粒体电压依赖性阴离子通道(VDAC)这一重要蛋白质在YTA中的上调表达有可能与花粉败育过程中细胞的程序性死亡相关。  相似文献

钙在高等植物中被称为第二信使,与植物的有性生殖有关。为了研究水稻(Oryza sativa L.)花药中钙的定位与花粉败育的关系,利用焦锑酸钾沉淀法研究了非花粉型细胞质雄性不育系G37A及其保持系G37B花药的发育过程及其细胞中Ca^2+ 的分布变化。研究发现,在2个材料间花药中钙的分布存在大量差异。G37B的可育花药在花粉母细胞时期及二分体时期,很少看到有Ca^2+的沉积;而在单核花粉时期,Ca^2+沉积急速地增加,主要定位在绒毡层细胞、花粉外壁外层及乌氏体的表面;随后花药壁上沉积的Ca^2+减少而花粉的外壁外层仍然有很多Ca^2+沉积物。相反,G37A的不育花药在花粉母细胞时期和二分体时期有大量的Ca^2+沉积在小孢子母细胞和花药壁,中间层和绒毡层特别多。在二分体时期之后,不育花药的Ca^2+沉积减少,特别是绒毡层内切向质膜附近的Ca^2+几乎消失。但是同时期的可育花药中,有大量的Ca^2+沉积在绒毡层。不育花药的Ca^2+沉积在开花几天后消失。根据研究结果推测在不育花药发育早期中更多的钙离子与花粉败育有一定的关系。  相似文献

Exploring novel source of cytoplasmic male sterility (CMS) is essential to stablize the productivity of hybrid rice. Dongxiang wild rice ( Oryza rufipogon Griff. ) has been recorded as the northest distributed wild rice in China that is resistant to several biotic or abiotic stresses. A male sterile line M01A defective of anther was identified in the F3 population from an interspecific cross between Dongxiang wild rice and cultivated rice ( Oryza sativa L. ssp. indica ). Crosses and successive backcrosses were made between M01A and a variety of breeding materials and 19 progeny families were obtained. Among the families, some were defective of anthers, and some have twisted and degenerated anthers without microsporogenesis or with a few typical aborted pollen. These resuits implicate that the male sterility of M01A was genetically regulated by the interaction between the nucleus and the cytoplasm. Only one cross produced male-fertile hybrid in which the paternal parent contains a part of the genome of Dongxiang wild rice, which implies that Dongxiang wild rice itself could be the source of the fertility restorer to M01A.  相似文献

采用双向凝胶电泳对水稻红莲型细胞质雄性不育的不育系小孢子发育单核期和二核期花药总蛋白进行了分离,通过银染显色,获得了分辨率和重复性较好的双向电泳图谱,且单核期和二核期花药总蛋白质在双向电泳胶上分布的图谱十分相似。PDQuest 2DE图像分析软件在等电点(pI)3.0~10.0、分子量(M.W.)9.0~98.0 kD之间可识别约1 800个蛋白质点。比较分析发现单核期和二核期花药中共有241个差异表达的蛋白质点,其中仅在单核期中表达的点数为125,仅在二核期中表达的为13点;表现为表达量差异的105点,其中在二核期表达下调的点数为70点,表达上调的为33点。还对蛋白质点集中的区域(pI 4.5~8.0,M.W.25.0~70.0 kD)中的41个差异蛋白质点进行了分子量和等电点分析。  相似文献

对转gst基因棉花恢复系浙大强恢"配制的杂种F1(三系杂交棉)的成熟花药和花粉育性进行了研究.结果表明,在花药的长、宽、鲜重和成熟花粉粒的育性方面,浙大强恢"所配的F1和保持系DES-HAB277"接近,无显著性差异,但比受体恢复系DES-HAF277"所配的F1分别提高47.7%、61.8%、28.5%和39.6%.以不育系DES-HAMS277"和保持系DES-HAB277"的花药为对照,对浙大强恢"和受体恢复系所配的F1的小孢子发生进行了细胞学观察发现,不育系小孢子败育主要发生在造孢细胞增殖期和小孢子母细胞形成期,且在减数分裂期彻底败育,不能形成四分体;受体恢复系所配的F1在小孢子发生和雄配子形成的各个发育时期都有部分败育,平均败育率约为20%,且主要发生在小孢子母细胞减数分裂期和小孢子单核期;而浙大强恢"所配的F1与保持系一样,花药的发育、小孢子的发生以及雄配子的形成均正常.研究结果从细胞形态学方面证明gst基因对三系杂交棉具有防止部分小孢子败育和提高花粉育性的功能.  相似文献

利用酶联免疫测定技术研究了水稻细胞质雄性不育系珍汕97A及其保持系珍汕97B在幼穗发育过程中叶片、幼穗和花药中内源IAA、GA1+4、ABA和iPAs含量的动态变化。结果表明,1)从幼穗发育的雌雄蕊形成期到三核花粉期,保持系叶片中IAA水平高于不育系,并在二核花粉期最高;在幼穗和花药中也以保持系为高。2)保持系与不育系叶片中GA1+4含量变化趋势相似,均为先升后降,但从单核到三核花粉期以保持系为高;在幼穗和花药中也都以保持系为高。3)不育系叶片中ABA水平在幼穗发育早期明显高于保持系,中期与保持系相近,后期又高于保持系;在幼穗和花药中也都以不育系为高。4)保持系叶片、幼穗和花药中iPAs含量始终显著高于不育系。5)保持系叶片中IAA+GA1+4+iPAs与ABA之比值也始终高于不育系。提示不育系叶片、幼穗和花药中IAA、GA1+4、iPAs和ABA含量出现了异常,且IAA、GA1+4和iPAs亏缺以及ABA盈积可能与水稻细胞质雄性不育发生有关。  相似文献

采用固相pH梯度-SDS PAGE 双向电泳对红莲型细胞质雄性不育水稻(YTA)的不育系和保持系(YTB)单核期花粉总蛋白质进行了分离,通过银染显色,获得了分辨率和重复性较好的双向电泳图谱。Image Master 2D V5.0 软件可识别约1800个蛋白质点,其中差异表达的蛋白质点数为85。将其中16个差异点采用基质辅助激光解析电离飞行时间质谱(matrix assisted laser desorption/ionizaton time of flight mass spectrometry, MALDI-TOF-MS)进行了肽质指纹图分析,通过采用Mascot 软件对MSDB数据库查询,其中9个蛋白质点得到了鉴定。YTA相对于YTB有部分参与碳代谢和淀粉合成的酶缺失或表达量降低,这些蛋白质分别是ADP-葡萄糖磷酸转移酶(AGPase),UDP-葡萄糖醛酸脱羧酶,乙酰辅酶A合成酶和二氢硫辛酸脱氢酶等。其中AGPase是参与淀粉合成的蛋白,与花粉发育密切相关。乙酰辅酶A合成酶和二氢硫辛酸脱氢酶是细胞内合成乙酰辅酶A的重要酶,而乙酰辅酶A是进入TCA循环的重要底物,乙酰辅酶A的缺乏可以导致TCA循环不能顺利进行,从而不能提供小孢子发育所需要的大量能量。YTA相对于YTB部分参与碳水化合物代谢的重要酶缺失或表达量降低,有可能导致因线粒体提供的能量不足,淀粉合成受阻,因而花粉不能正常发育。   相似文献

Summary Development of anthers in cytoplasmic male sterile (CMS) Petunia diverges from the normal sequence of events early in meiosis. Quantitative and qualitative changes in morphology, proteins and free amino acid contents correlate with this divergence. In anthers of the fertile line (5719), total protein content increases, and SDS-PAGE protein patterns change as the anthers mature. Enhanced levels of three polypeptides with molecular weights of 64,000, 63,000 and 45,000 daltons characterize premeiosis in fertile anthers. Protein levels and patterns from anthers of the CMS line (5707) show little alteration during anther development. Protein synthesis seems to be at least partially blocked in the CMS microspore. The 63,000 and 45,000 dalton proteins are not present, and the absence of any unique protein(s) in the CMS line argues against a virus as the causal agent of CMS in Petunia. Analysis of free amino acids from anthers of the fertile line shows levels of proline and pipecolic acid 2–3 and 10–20 fold higher, respectively, than in the CMS line. The amino acids incorporated into proteins show no such differences; analysis of protein hydrolysates shows similar levels of each amino acid in both fertile and CMS lines at every developmental stage examined.  相似文献

红莲型CMS是在我国杂交水稻生产中被广泛利用的雄性不育细胞质之一。为了同时定位红莲型CMS育性恢复主效和微效QTL,利用红莲型CMS不育系粤泰A(YTA)与“Lemont/特青”RIL群体测交,结合1张含有198个DNA分子标记的高密度遗传图谱,对测交F1群体的小穗育性和花粉育性进行复合区间作图。在对YTA的育性恢复性方面,该。RIL群体的2个亲本之间具有明显差异,特青的恢复性较强,其测交F1的小穗育性和花粉育性分别为72%和51%;而Lemont测交F1的小穗育性和花粉育性分别为32%和9%。复合区间作图定位到4个育性恢复QTL,分别位于水稻第1、2和10号染色体上,单个QTL的贡献率在5%~24%之间。其中,除1个QTL的增效基因来源于Lemont外,其余3个QTL的增效基因均来源于特青。效应最大的QTL为qRF-10-1,该QTL位于10号染色体RM258-C16标记区间,对小穗育性表型变异的贡献率为24%,对花粉育性的贡献率为17%,且该QTL被检测到的LOD值显著较高,因此是1个主效QTL,其增效基因来源于特青。除了主效QTLqRF-10-1外,其它3个QTL对性状的贡献率均在10%以下(5%~8%)。由此表明,该RIL群体对红莲型CMS的育性恢复由1个主效QTL控制,并受其它几个微效QTL的影响。该QTL定位结果与小穗育性在测交F1群体中呈连续的双峰分布的结果相一致。与主效QTL qRF-10-1紧密连锁的SSR标记为RM258,该主效QTL可作为分子标记辅助育种的操作目标之一,用于杂交稻分子育种中培育红莲型CMS的强恢复系。  相似文献

In indica rice, the HongLian (HL)-type combination of cytoplasmic male sterility (CMS) and fertility restoration (Rf) is widely used for the production of commercial hybrid seeds in China, Laos, Vietnam and other Southeast Asian countries. Generally, any member of the gametophytic fertility restoration system, 50% of the pollen in hybrid F(1) plants displays recovered sterility. In this study, however, a HL-type hybrid variety named HongLian You6 had approximately 75% normal (viable) pollen rather than the expected 50%. To resolve this discrepancy, several fertility segregation populations, including F(2) and BC(1)F(1) derived from the HL-CMS line Yuetai A crossed with the restorer line 9311, were constructed and subjected to genetic analysis. A gametophytic restoration model was discovered to involve two non-allelic nuclear restorer genes, Rf5 and Rf6. The Rf5 had been previously identified using a positional clone strategy. The Rf6 gene represents a new restorer gene locus, which was mapped to the short arm of chromosome 8. The hybrid F(1) plants containing one restorer gene, either Rf5 or Rf6, displayed 50% normal pollen grains with I(2)-KI solution; however, those with both Rf5 and Rf6 displayed 75% normal pollens. We also established that the hybrid F(1) plants including both non-allelic restorer genes exhibited an increased stable seed setting when subjected to stress versus the F(1) plants with only one restorer gene. Finally, we discuss the breeding scheme for the plant gametophytic CMS/Rf system.  相似文献

细胞质雄性不育水稻不育系珍汕97A和其保持系珍汕97B,处于不育期的光(温)敏核不育水稻W6154s和培矮64s的花药中类脂褐素(LFLP)含量随花粉发育或败育而增高.不育花药中LFLP的形成速率比可育花药快,三核期的珍汕97A和不育期W6154s的花药,其LFLP比相应具育性花药高24%.用抗氧化剂GSH、BHT和N2处理离体的单核期花药,发现GSH可降低珍汕97A和不育期的W6154s的LFLP含量.结果认为,水稻雄性不育与膜脂过氧化作用的荧光产物类脂褐素的积累有关.  相似文献

To clarify the time and cause of pollen abortion, differences on the microsporogenesis and tapetum development in the anthers of male fertile maintainer line and cytoplasmic male sterile (CMS) line pepper were studied using transmission electron microscopy. The results showed that CMS line anthers appeared to have much greater variability in developmental pattern than male fertile maintainer line ones. The earliest deviation from normal anther development occurred in CMS line anthers at prophase I was cytomixis in some microspore mother cells (MMCs), and vacuolisation in tapetal cells. Then, MMCs in CMS line anthers developed asynchronously and a small part of ones at the different stage degenerated in advance appearing to have typical morphological features of programmed cell death (PCD). Most MMCs could complete the meiosis, but formed non-tetrahedral tetrad microspores with irregular shape and different size and uncertain number of nuclei, and some degenerated ahead of time as well. Tapetal cells in CMS line anther degenerated during meiosis, and were crushed at the tetrad stage, which paralleled the collapse of pollens. Pollen abortion in CMS line anthers happened by PCD themselves, and the premature PCD of tapetal cells were closely associated with male sterility.  相似文献

One of the cytoplasmic male sterility (CMS) types used for hybrid rice (Oryza sativa L.) production in China is the Honglian (HL)-CMS. Previous studies suggested that pollen abortion of the sterile plants was resulted from a special programmed cell death (PCD) program started at meiosis in the microspores. To elucidate the molecular basis of the pollen abortion, we compared the biochemical and physiological properties such as content of reactive oxygen species (ROS), ATP, NADH, total glutathione and ascorbate acid, the activities of dehydroascrbate reductase, glutathione reductase, ascorbate peroxides and superoxide dismutase, and the integrity of mitochondrial genome DNA isolated from an HL-CMS line, Yuetai A and its maintainer line, Yuetai B. Our results indicated that the mitochondria of the HL-CMS line suffered from a serious oxidative stress during microspores development. Oxidative stress induced by abnormal increased ROS at meiosis stage resulted in the depletion of ATP and NADH, and the degradation of mitochondrial genomic DNA. This suggests that the presence of redox signal originated in mitochondria affects the rest of the cell. Therefore, it is possible that the abortion of premature microspores in HL-CMS line is induced by the chronic oxidative stress in mitochondria in the early stage of pollen development.  相似文献

利用RAPD技术,从248个随机寡核苷酸引物(10-mer)中筛出18个引物对红莲型杂交稻组合红莲2号及其亲本(T-07A、T-07B、YD6-05),另6个红莲型胞质不育系的骨干恢复和汕优63及其亲本共14份水稻材料进行分析。共检测到173个多态性标记。聚类分析结果表明:不育系与保持系间因核背景相似,遗传差异很小;杂种(F1)的基因型更倾向于恢复系;恢复系与保持系间遗传距离的相对较大,但各恢复系之间的遗传距离较小。利用这些标记能有效地地区交组合中不育系,保持系、恢复系和杂种(F1)。  相似文献

采用双向电脉技术对不同光周期条件下,光敏感核不育水稻(农垦58s)的可育与不育花药蛋白的变化进行分析,发现花粉发育的不同阶段中,不育花药具有四个特异蛋白pI6.2/bMW70KD,pI6.2/MW68KD,pI6.2/MW38KD和pI7.4/MW37KD.对游离组蛋白的分析表明.长日照诱导的不育花药中游离组蛋白的相对百分率均明显低于短日照下的可育花药.据此推测长日照诱导不百花药蛋白质组成和代谢变化.不育花药中游离组蛋白含量低,可能受DNA合成数量少的影响.  相似文献

将T4 RNA连接酶和AFLP技术特点相结合构建了适于mtRNA的差异显示方法 ,并比较了水稻 (OryzasativaL .)红莲型细胞质雄性不育系、保持系和杂种一代mtRNA的差异。在 4组引物对的选择扩增产物中共找到 6个差异片段 ,其中差异条带DTA为不育系仅有 ,条带DAB为不育系和保持系特有 ,而条带DBF1、DBF2 、DBF3 、DBF4 为保持系和F1杂种共有。这表明水稻红莲型不育系粤泰A与杂种一代泰优 2号mtRNA的差异大于保持系粤泰B和杂种一代泰优 2号mtRNA的差异。Northern杂交证实条带DTA在不育系、保持系和杂种一代的转录确有差异 ,表明它与红莲型细胞质雄性不育有关。条带DTA全长 2 5 9bp ,尽管未发现有同源序列和新的开放阅读框 ,但它可作为探针从cDNA文库中筛选全基因序列 ,进而寻找与细胞质雄性不育有关的开放阅读框架。  相似文献
