本文通过X射线照射SMMC-7721、BEL-7402和HepG2三种肝癌细胞后,以克隆形成试验检测其存活分数,结果显示在梯度剂量X射线0、2、4、6、8、10 Gy照射下SMMC-7721、BEL-7402、HepG2三种细胞克隆存活分数逐渐下降,其中SMMC-7721在三种肝癌细胞系中对辐射最敏感,BEL-7402辐射抗性在三种肝癌细胞系中最高。Western blot检测发现PER1在SMMC-7721中的表达水平明显显著高于BEL-7402和HepG2(P<0.05)。过表达PER1蛋白以后,BEL-7402接受5 Gy X射线照射后凋亡明显增多,同时,western blot和RT-qPCR试验结果发现,X射线照射过表达PER1的BEL-7402细胞,抗凋亡蛋白Bcl-2表达明显降低,凋亡执行蛋白Caspase-3断裂明显增多。研究结果表明PER1蛋白的高水平表达可以促进X射线诱导的凋亡,增强肝癌细胞的辐射敏感性。  相似文献

目的:研究白花蛇舌草豆甾醇(stigmasterol from Hedyotis diffusa willd.,SHD)对人肝癌细胞SMMC-7721、BEL-7402的体外抑制作用,对肝癌H22的体内抑制作用及对其增殖周期、凋亡的影响。方法:MTT法评价SHD对人肝癌细胞SMMC-7721、BEL-7402的抑制率变化规律。昆明雄性小鼠60只,随机取10只为正常对照组,余接种H22瘤株,随机分为模型对照组、5-FU阳性对照组(30mg/kg)和高中低剂量SHD给药组(剂量分别为15、30、60mg/kg),腹腔给药10 d后,比较各组瘤重抑制率、H22细胞周期分布、凋亡率。结果:SHD对SMMC-7721、BEL-7402细胞具有体外抑制作用;SHD显著抑制H22肿瘤,增加G0-G1期细胞比例,降低G2/M期细胞比例,促进肿瘤细胞凋亡。结论:SHD在体外、体内均具有抑制肝癌细胞的作用,此作用与阻滞肿瘤细胞增殖周期,促进肿瘤细胞凋亡有关。  相似文献

目的:探讨Notch信号通路对肝癌细胞迁移能力及钙粘附蛋白E(E-cadherin)、环氧化酶-2(COX-2)表达的影响。方法:体外培养肝癌细胞系(SMMC-7721、MHCC97H)、正常非肿瘤肝细胞系(HL-7702),Transwell小室用于测定细胞的迁移侵袭能力,Western blot蛋白印迹法用于测定Notch1、E-cadherin、COX-2蛋白的表达水平,并采用DAPT阻断Notch信号通路,比较肝癌细胞系与正常非肿瘤肝细胞系的迁移侵袭能力及肝癌细胞中E-cadherin、COX-2蛋白的表达水平的改变。结果:SMMC-7721细胞、MHCC97H细胞的迁移能力强于HL-7702细胞,差异有统计学意义(P0.05);相比于HL-7702细胞,MHCC97H细胞、SMMC-7721细胞中的Notch1、COX-2表达水平均显著升高,E-cadherin的表达水平明显降低(P0.05);DAPT处理后,SMMC-7721细胞、MHCC97H细胞发生迁移的能力均弱于对照组,差异有统计意义(P0.05);DAPT处理后,SMMC-7721细胞、MHCC97H细胞内COX-2、Notch1的表达量明显降低,而E-cadherin的表达水平升高(P0.05)。结论:Notch信号通路参与肝癌细胞迁移过程,其机制可能与E-cadherin、COX-2的表达相关。  相似文献

目的:探究二十碳五烯酸(Eicosa Pentaenoic Acid.EPA)对SMMC-7721人肝癌细胞的凋亡、端粒逆转录酶h TERT的调控作用及端粒酶表达活性的影响。方法:体外培养SMMC-7721人肝癌细胞,用不同浓度的EPA(0μM、25μM、50μM、100μM、200μM)作用于SMMC-7721肝癌细胞(24 h、48 h、72 h)后,显微镜下观察其形态学变化;应用MTT法检测SMMC-7721肝癌细胞细胞增殖变化情况;Western-blot法检测h TERT、Bax、Bcl-2蛋白表达水平变化;Real Time-PCR检测h TERTm RNA的表达变化;ELISA法检测SMMC-7721肝癌细胞端粒酶活性的表达水平。结果:EPA可诱导肝癌细胞SMMC-7721发生细胞凋亡,具有明显的时间计量依赖关系。在此过程中Bcl-2蛋白表达的降低和Bax蛋白表达上调,同时端粒酶逆转录酶h TERT蛋白及其m RNA的表达水平和端粒酶活性均明显降低。结论:抑制端粒酶逆转录酶基因(h TERTm RNA)表达而抑制端粒酶的活性、诱导癌细胞凋亡,可能是EPA的抗癌作用机制之一。  相似文献

目的研究枸杞多糖(lycium barbarum polysaccharide,LBP)对SMMC-7721肝癌细胞迁移、侵袭影响的机制。方法运用MTT法检肝癌细胞SMMC-7721的增殖率,Transwell检测细胞的迁移力和侵袭力,应用血管内皮生长因子(vascular endothelial growth factor,VEGF)小分子干扰RNA沉默VEGF表达,转染pcDNA 3.1-VEGF过表达VEGF,Western blot和qRT-PCR检测VEGF、MMP-2和MMP-9表达。结果 LBP(100、200、400μg/ml)处理可抑制SMMC-7721细胞的增殖、迁移和侵袭,并抑制MMP-2、MMP-9和VEGF表达;沉默VEGF可降低SMMC-7721细胞迁移、侵袭相关蛋白MMP-2和MMP-9水平,过表达VEGF可逆转LBP对SMMC-7721细胞迁移和侵袭相关蛋白MMP-2和MMP-9水平的抑制作用。结论枸杞多糖可抑制肝癌细胞的迁移和侵袭,其机制可能与直接抑制VEGF有关。  相似文献

为探讨连香树精油的体外抗肿瘤活性。以人肝癌SMMC-7721细胞为受试细胞株,利用MTT法、流式细胞术、JC-1法和Western blot法评价连香树精油的体外抗肿瘤活性。结果表明:连香树精油处理24 h后可降低SMMC-7721细胞存活率;细胞中G0/G1期的比例下降,G2/M期的比例升高;细胞线粒体膜电位降低;自噬相关蛋白LC3-Ⅱ、Beclin-1表达上升,加入自噬抑制剂氯喹后LC3-Ⅱ蛋白与Bcl-2抗凋亡蛋白表达下降。综上说明连香树精油能抑制SMMC-7721细胞的增殖活性,通过阻滞G2/M期抑制细胞分裂,通过降低线粒体膜电位诱导SMMC-7721细胞凋亡,而自噬在SMMC-7721细胞凋亡过程中作为一种保护机制而存在。  相似文献

【目的】探讨斑蝥素酸镁对人肝癌细胞SMMC-7721及其裸鼠皮下移植瘤的影响。【方法】1、采用磺酰罗丹明染色法(SRB法)检测不同浓度斑蝥素酸镁在体外对人肝癌细胞SMMC-7721增殖的抑制作用;2、流式细胞术检测斑蝥素酸镁对人肝癌细胞SMMC-7721细胞周期和细胞凋亡的影响;3、Hoechst33342染色观察斑蝥素酸镁对人肝癌细胞SMMC-7721细胞形态的影响;4、透射电镜观察斑蝥素酸镁作用后人肝癌细胞SMMC-7721超微结构的变化;5、建立人肝癌细胞SMMC-7721裸鼠皮下移植瘤模型,实验组每只裸鼠瘤周注射斑蝥素酸镁6.26×10?5 mmol,对照组给予相同容积的无菌生理盐水瘤周注射,计算抑瘤率;6、原位末端标记染色(TUNEL)法检测人肝癌裸鼠皮下移植瘤组织细胞凋亡情况。【结果】1、斑蝥素酸镁对人肝癌细胞SMMC-7721有比较明显的抑制作用,抑制率随药物浓度的增加而升高,呈剂量效应关系,其半数抑制浓度(IC50)为1.79?mol/L;2、流式细胞检测结果显示:人肝癌细胞SMMC-7721在斑蝥素酸镁的作用下,G0/G1期细胞减少,G2/M期细胞增加,细胞出现G2/M期阻滞;细胞凋亡率随斑蝥素酸镁浓度加大而逐渐增加;3、Hoechst33342染色镜下显示:斑蝥素酸镁作用后人肝癌细胞SMMC-7721出现凋亡细胞形态特征;4、透射电镜观察:斑蝥素酸镁作用后人肝癌细胞SMMC-7721出现细胞核异形、染色质聚集成团、边集,见凋亡小体;5、斑蝥素酸镁组肿瘤体积、重量显著小于生理盐水组(P﹤0.05),抑瘤率为49%;6、TUNEL法提示斑蝥素酸镁组移植瘤组织细胞凋亡率显著高于生理盐水组(χ2=92.609,P﹦0.000)。【结论】斑蝥素酸镁对人肝癌细胞SMMC-7721在体内外均有抑制增殖作用,并可以诱导肿瘤细胞凋亡,其凋亡的发生与细胞分裂期阻滞有关。  相似文献

目的:瞬转以及筛选出能够稳定表达URI(RPB5-mediating protein)基因的SMMC-7721细胞株,以其为模型研究URI基因对人肝癌SMMC-7721细胞增殖的影响.方法:首先,抽提URI的重组质粒并转染到SMMC-7721细胞中,在G418药物的筛选下选出能够稳定表达URI基因的细胞株,RT-PCR法和酶切检测该稳定细胞中URI基因的表达效率以确认URI是否稳定表达,MTT法和克隆形成实验检测URI基因对SMMC-7721细胞增殖的影响.结果:成功建立URI基因过表达的稳定细胞株.与SMMC-7721细胞对照组比较,其URI mRNA表达水平显著上调,能稳定表达URI细胞株的增殖,克隆形成率明显升高.结论:pFLAG-CMV-4-URI重组质粒能使URI在肝癌SMMC-7721细胞内稳定表达,过表达URI基因有可能帮助细胞通过G2/M期检验点来提高肝癌细胞SMMC-7721的增殖能力.  相似文献

目的:探讨棕榈酸(Palmiticacid,PA)对人肝癌细胞系SMMC-7721侵袭转移能力的影响,并通过检测肝癌细胞系中CD147-MMPs信号通路在PA影响下的变化,初探PA影响肝癌细胞侵袭转移的分子机制。方法:PA(0、20、50、100μM)作用SMMC-7721细胞后(8、16、24h),MTT法检测细胞增殖,划痕及Transwell实验评价细胞迁移侵袭能力,Western-blot及real-time PCR检测CD147蛋白及其mRNA的水平,ELISA检测基质金属蛋白酶(MMP-2,MMP-9)的水平。结果:与对照组相比,PA作用SMMC-7721细胞后,细胞存活率无显著差异(P0.05);细胞迁移和侵袭能力显著增高(P0.05);CD147蛋白及其mRNA的表达显著增高(P0.05);培养上清中MMP-9的浓度显著增高(P0.05),MMP-2的水平则无变化。不同的梯度组之间相比较,细胞迁移和侵袭能力、CD147的表达水平(蛋白及其mRNA)以及培养上清中MMP-9的浓度均随PA作用时间和作用剂量的增大而产生更显著的增高。结论:PA通过活化CD147-MMPs信号通路促进SMMC-7721细胞的迁移侵袭。  相似文献

芪类化合物在癌的起始、促进和发展阶段均有化学抗癌活性,对其化学结构进行改造并进行作用机制研究,提高其抗肿瘤活性,有可能发现新的抗肿瘤药物.在合成23种芪类化合物的基础上,以MTT法测定其对9种肿瘤细胞系的抑制作用,发现多种化合物对SMMC-7721、BGC-823、HepG2、Bel-7402、MCF-7、SGC-7901细胞系具有较明显的抑制活性.以细胞周期分析、Hoechst 33258荧光染色及线粒体膜电位变化进行检测,发现经抑制作用较强的化合物HCQ-10处理后的 Bel-7402细胞发生G2/M期阻滞,并继发细胞凋亡.实验结果说明,合成芪类化合物对SMMC-7721、BGC-823、HepG2、Bel-7402、MCF-7、SGC-7901细胞具有较为明显的增殖抑制作用,其抑制Bel 7402细胞增殖的作用机制为阻滞细胞于G2/M期并诱导细胞继发凋亡.  相似文献

Phage display technology has been used as a powerful tool in the discovery of ligands specific to receptor(s) on the surface of a cancer cell and could also impact clinical issues including functional diagnosis and cell-specific drug delivery. After three rounds of in vitro panning and two rounds of reverse absorption, a group of phages capable of addressing BEL-7402 enormously were obtained for further analysis. Through a cell-based ELISA, immunofluorescence, FACS, and in vivo binding study, WP05 (sequence TACHQHVRMVRP) was demonstrated to be the most effective peptide in targeting four kinds of liver cancer cell lines (BEL-7402, BEL-7404, SMMC-7721, and HepG2), but not the normal liver cell line HL-7702. In conclusion, the peptide WP05 which was screened by in vitro phage display technology was proved to be a targeting peptide to several common hepatocellular carcinoma cell lines.  相似文献

目的:肝癌的转移与复发是肝癌治疗的一大难题,盐霉素是近年来新发现的具有抗肿瘤作用的抗生素,本文研究了盐霉素在体外及体内对人肝细胞癌转移与侵袭能力的作用及机制。方法:在体外对肝癌细胞株HepG2,SMMC-7721,BEL-7402给予盐霉素处理,体内建立裸鼠肝脏原位肿瘤模型,并给予腹腔注射盐霉素治疗。观察肿瘤细胞的转移侵袭能力以及肝内肿瘤转移灶的情况,进一步测定E.cadherin,Vimentin的表达,来研究盐霉素对肝癌转移及侵袭能力的影响及机制。结果:经盐霉素处理后,肝癌细胞株HepG2,SMMC.7721,BEL.7402的转移及侵袭能力明显下降,肝内转移灶的数目也减少。分子机制检测发现盐霉素处理后E.cadherin表达增高,Vimentin表达下降。结论:盐霉素在体内与体外都抑制了肝癌的转移与侵袭,其机制可能抑制了肿瘤细胞的上皮间质化(EMT)过程。这为控制肝癌的转移和复发提供了新的治疗思路。  相似文献

An immortal human hepatic cell line HL-7702 and human hepatoma cell line SMMC-7721 were treated with 3–30 μM SeO2. SeO2 at 30 μM markedly inhibited cell proliferation and viability, and prompted apoptosis of both normal hepatic and hepatoma cells after 48 h treatment. SeO2 could also down-regulate the Bcl-2 level, greatly in HL-7702 and slightly in SMMC-7721 cells, but up-regulate wild type P53 level a little in HL-7702 and significantly in SMMC-7721 cells. The Bcl-2/P53 value was closely correlated with the apoptotic rate as well as SeO2 concentrations.  相似文献

This study aimed to evaluate the correlation of integrin alpha 7 (ITGA7) with clinical outcomes and its effect on cell activities as well as stemness in hepatocellular carcinoma (HCC). HCC tumor tissues and paired adjacent tissues from 90 HCC patients were obtained and ITGA7 expression was detected using immunohistochemistry assay. Cellular experiments were conducted to examine the effect of ITGA7 on cell activities, astemness via ITGA7 ShRNA transfection, and compensation experiments were further performed to test whether ITGA7 functioned via regulating PTK2-PI3K-AKT signaling pathway. ITGA7 was overexpressed in tumor tissues compared with paired adjacent tissues and its high expression was correlated with larger tumor size, vein invasion and advanced Barcelona Clinic Liver Cancer stage, and it also independently predicted worse overall survival in HCC patients. In cellular experiments, ITGA7 was upregulated in SMMC-7721, Hep G2, HuH-7 and BEL-7404 cell lines compared with normal human liver cells HL-7702. ITGA7 knockdown suppressed cell proliferation but promoted apoptosis, and it also downregulated CSCs markers (CD44, CD133 and OCT-4) as well as PTK2, PI3K and AKT expressions in SMMC-7721 and Hep G2 cell lines. ITGA7 overexpression promoted cell proliferation but inhibited apoptosis, and it also upregulated CSCs markers in HL-7702 cells. Further compensation experiments verified that ITGA7 regulated cell proliferation, apoptosis and CSCs markers via PTK2-PI3K-Akt signaling pathway. ITGA7 negatively associates with clinical outcomes in HCC patients, and it regulates cell proliferation, apoptosis and CSCs markers via PTK2-PI3K-Akt signaling pathway.  相似文献

Zuojinwan (ZJW), a famous Chinese medicinal formula, contains two medicinal herbs Coptis chinese Frach and Evodia rutaecarpa (Juss.) Benth in the ratio of 6: 1. The inhibitory effects of ZJW on eight kinds of human cancer cell lines including SMMC-7721, BEL-7402, BEL-7404, HepG2, A549, NCI-H446, NCI-H460 and HCT- 116 cells were evaluated, and the possible mechanism was investigated. The growths of the eight kinds of cancer cells were inhibited by ZJW assessed through MTT assay. Flow cytometry assay revealed a sub-G1 peak with reduced DNA content was formed. The cell cycle was arrested in the G0/G1 phase in ZJW-treated SMMC-7721 and HepG2 cells, and in the S phase for NCI-H460 cells. Significant DNA damage was produced by ZJW assessed with single-cell gel electrophoresis assay. Morphological changes were also observed. Caspase-3 and -9 activities were increased following ZJW treatment. Western blot analysis showed that Bax and Bak protein levels were increased after ZJW treatment, while Bcl-2 and Bcl-xl protein levels were decreased. Our results suggest that ZJW has significant anti-cancer activities due to induction of mitochondria- dependent apoptosis pathway. Therefore, ZJW has the potential to be a novel chemotherapy drug to treat hepatoma, lung cancer and colon cancer by suppressing tumor growth.  相似文献
