潜伏膜蛋白1(LMP1)是由EB病毒编码的致瘤蛋白,众多研究表明LMP1蛋白可通过NF-κB、p38 MAPK、c-JNK等多条重要信号通路引起鼻咽癌细胞的生物学行为改变。我们从EB病毒阳性的B95-8狨猴淋巴瘤细胞中克隆EB病毒LMP1 c DNA,构建携带绿色荧光基因的真核表达质粒p IRES2-Zs-Green1-LMP1,通过脂质体转染的方法将质粒导入鼻咽癌细胞株CNE1、CNE2中,利用质粒所携带的绿色荧光蛋白表达粗略计算转染的效率,通过免疫细胞化学(ICC)、RT-PCR、Western-Blot检测该质粒的表达。本实验室所构建的p IRES2-Zs-Green1-LMP1表达质粒能在鼻咽癌细胞内表达LMP1蛋白,为后续的实验研究奠定基础。  相似文献

探讨EB病毒潜伏膜蛋白1(LMP1)激活激活蛋白1(AP1),和核转录因子(NF-κB)在鼻咽癌细胞SUNE-1及亚细胞株恶性演进中的作用.运用报告基因法和凝胶电泳迁移率法(EMSA)分析AP1和NF-κB反式激活活性和DNA结合活性,蛋白质印迹检测蛋白质表达;裸鼠致瘤实验结合组织制片研究瘤细胞的成瘤和转移能力. 结果显示恶性程度不同的SUNE-1亚细胞株的反式激活活性、DNA结合活性、LMP1蛋白表达及c-Jun氨基端激酶(JNK)活性均存在明显差异,且与细胞恶性程度正相关.这些结果提示LMP1活化AP1和NF-κB的信号通路参与了鼻咽癌细胞SUNE-1的恶性演进过程.  相似文献

EB病毒编码的病毒潜伏膜蛋白1(LMP1)是重要的致瘤蛋白, 一直以来是EB病毒致瘤机制的研究核心. 传统的受体学说认为, 细胞膜受体作为第一信使, 在细胞膜上与其配体结合发挥生物学效应. 但近年来, 对EGFR家族成员受体核移位在基因表达调控中意义的研究, 拓宽了人们对细胞膜受体生物学功能的认识. 利用我们建立的Tet-on系统调控LMP1表达的细胞系, 采用Western blot和激光共聚焦显微镜等技术证实, LMP1可调控EGFR的核移位, 并呈一定的剂量效应. 通过GFP与EGFR及不同突变体的融合蛋白在细胞内的表达发现, EGFR的核定位信号在其核移位过程中起着一定的作用, 并初步发现LMP1调控EGFR核移位为配体非依赖性.  相似文献

 EB病毒潜伏膜蛋白 1 (latentmembraneprotein 1 ,LMP 1 )活化激活蛋白 1 (activatorprotein 1 ,AP 1 )信号传导途径与其致瘤作用密切相关 .为了探讨LMP 1活化AP 1信号传导的分子机制 ,在可诱导调控LMP 1表达的鼻咽癌细胞系L7中 ,首先通过荧光酶双报道系统确定了LMP 1表达能激活AP 1 ;在此基础上 ,用c JunPathDetect系统确定LMP 1表达活化AP 1是通过c Jun的磷酸化 (活化 )介导 .虽然LMP 1不能上调c Jun上游主要调节激酶c JunN端激酶 (c JunN terminalkinase ,JNK)的蛋白表达 ,但能显著促进JNK的磷酸化 (活化 ) ;在L7细胞中导入JNK相互作用蛋白 (JNK interactingprotein ,JIP)基因 ,抑制JNK的核移位能显著抑制LMP 1诱导的AP 1活化 ,同时对NFкВ活化也有部分抑制作用 .结果表明 ,EB病毒LMP 1在鼻咽癌细胞中通过JNK介导AP 1活化  相似文献

建立含有EB病毒的T细胞淋巴瘤细胞系,为探讨EB病毒的致瘤机理,研究EB病毒在T细胞淋巴瘤发生过程中的作用提供手段.在TPA协同EB病毒诱导胸腺恶性T细胞淋巴瘤动物模型的基础上,联合应用IL-2,将诱导的肿瘤组织进行体外细胞培养,成功地分离获得一株在体外长期存活的淋巴细胞TET.T细胞亚群分类实验证实TET细胞为CD4阳性的T淋巴细胞,PCR和原位杂交可检测到EB病毒的EBERs、LMP1和BARF1,并有LMP1蛋白的表达.TET细胞的获得,有望在体外建立转化细胞系,为体外研究EB病毒的致瘤机理及防治提供理想的实验材料.  相似文献

EB病毒LMP1 CTAR1、CTAR2的表达促使人鼻咽癌细胞HNE2增殖   总被引:1,自引:0,他引:1  
探讨EB病毒LMP1不同结构域在鼻咽癌中的致瘤作用,为阐明鼻咽癌分子发病机理,寻找治疗鼻咽癌的分子靶提供实验依据。以转染空白载体为对照,利用电穿孔转染方法,建立稳定表达LMP1不同突变体的鼻咽癌细胞系HNE2-LMP1(1~815)、HNE2-LMP1(1~231)、HNE2-LMP1△187~351,并以这些细胞系为材料,用MTT法检测增殖期活细胞,BrdU掺入法检测细胞增殖状况,比较各组细胞的软琼脂集落形成率和裸鼠成瘤能力,以观察LMP1不同的结构域对鼻咽癌细胞生长的影响。LMP1(1~231)和LMP1△187~351在体外明显促进HNE2细胞增殖,HNE2-LMP1(1~231)、HNE2-LMP1△187~351平均吸光度(A)比值、BrdU掺入率、软琼脂集落形成率均高于HNE2-pSG5与HNE2(P<0 01),而HNE2-LMP1(1~187)与HNE2-pSG5、HNE2相比,这些指标无明显差别。HNE2-LMP1△187~351和HNE2-LMP1(1~231)的裸鼠成瘤潜伏期、倍增时间与平均瘤重明显高于HNE2-pSG5鼻咽癌细胞系,其差异有显著的统计学意义(P<0 05)。而HNE2-LMP1(1~187)、HNE2-pSG5和HNE2鼻咽癌细胞系在潜伏期、倍增时间与平均瘤重方面两两比较,差异无显著的统计学意义(P>0 05)。EB病毒LMP1CTAR1和CTAR2对HNE2细胞生长有明显促进作用,提示EB病毒LMP1可能在鼻咽癌的发生发展中起着重要的作用。  相似文献

EB病毒广泛存在于人群中,它的潜伏感染与鼻咽癌的发生密切相关。EBV在潜伏感染的过程中表达一种潜伏膜蛋白1(LMP1),具有致瘤性,因此编码LMP1的基因BNLF-1被认为是EBV的瘤基因。BNLF-1基因具有广泛的生物学功能,在鼻咽癌的发生发展过程中起重要作用。近年的研究表明,鼻咽癌来源的EBV与标准株EBV比较存在相当的变异,而鼻咽癌来源的LMP1在致瘤性上也明显强于标准株LMP1。本文将综述BNLF-1基因生物学功能研究方面的新进展,并简要介绍变异型LMP1研究的新成果。  相似文献

在EB病毒潜伏膜蛋白LMP1介导的信号传导通路中,TRAFs作为LMP1活化的第一位信号分子,可能扮演着重要的分子开关角色。令人关注的是,在上皮性肿瘤NPC的发生中,EB病毒LMP1能否激活重要的TRAFs信号分子?究竟激活何种TRAFs信号分子,激活的机制何在?将LMP1cDNA导入LMP1表达阴性的HNE2中,建立稳定表达LMP1的鼻咽癌细胞系HNE2-LMP1。以此为材料,应用差异RT-PCR和Western blotting法证实,无论在RNA水平,还是蛋白水平上,TRAF1在HNE2-LMP1中表达较HNE2强,而TRAF2及TRAF3在HNE2-LMP1与HNE2细胞中表达无明显差异;进一步用免疫共沉淀-Western blotting证实LMP1可使TRAF1、TRAF2、TRAF3磷酸化而被活化。这些结果提示在鼻咽癌中,LMP1可能诱导TRAF1表达,而对TRAF2及TRAF3并不影响,但LMP1可磷酸化TRAF1、TRAF2、TRAF3而使其功能性活化。q  相似文献

Control of translation initiation is one means by which cells regulate growth and proliferation, with components of the protein-synthesizing machinery having oncogenic potential. Expression of latency protein LMP2A by the human tumor virus Epstein-Barr virus (EBV) activates phosphatidylinositol 3-kinase/Akt located upstream of an essential mediator of growth signals, mTOR (mammalian target of rapamycin). We show that mTOR is activated by expression of LMP2A in carcinoma cells, leading to wortmannin- and rapamycin-sensitive inhibition of the negative regulator of translation, eukaryotic initiation factor 4E-binding protein 1, and increased c-Myc protein translation. Intervention by this DNA tumor virus in cellular translational controls is likely to be an integral component of EBV tumorigenesis.  相似文献

Epstein-Barr virus (EBV), an oncogenic herpesvirus that causes human malignancies, infects and immortalizes primary human B cells in?vitro into indefinitely proliferating lymphoblastoid cell lines, which represent a model for EBV-induced tumorigenesis. The immortalization efficiency is very low, suggesting that an innate tumor suppressor mechanism is operative. We identify the DNA damage response (DDR) as?a major component of the underlying tumor suppressor mechanism. EBV-induced DDR activation was not due to lytic viral replication, nor did the DDR marks colocalize with latent episomes. Rather, a transient period of EBV-induced hyperproliferation correlated with DDR activation. Inhibition of the DDR kinases ATM and Chk2 markedly increased transformation efficiency of primary B cells. Further, the viral latent oncoprotein EBNA3C was required to attenuate the EBV-induced DDR. We propose that heightened oncogenic activity in early cell divisions activates a growth-suppressive DDR that is attenuated by viral latency products to induce cell immortalization.  相似文献

It has been recently reported that a side population of cells in nasopharyngeal carcinoma (NPC) displayed characteristics of stem-like cancer cells. However, the molecular mechanisms underlying the modulation of such stem-like cell populations in NPC remain unclear. Epstein-Barr virus was the first identified human tumor virus to be associated with various malignancies, most notably NPC. LMP2A, the Epstein-Barr virus encoded latent protein, has been reported to play roles in oncogenic processes. We report by immunostaining in our current study that LMP2A is overexpressed in 57.6% of the nasopharyngeal carcinoma tumors sampled and is mainly localized at the tumor invasive front. We found also in NPC cells that the exogenous expression of LMP2A greatly increases their invasive/migratory ability, induces epithelial–mesenchymal transition (EMT)-like cellular marker alterations, and stimulates stem cell side populations and the expression of stem cell markers. In addition, LMP2A enhances the transforming ability of cancer cells in both colony formation and soft agar assays, as well as the self-renewal ability of stem-like cancer cells in a spherical culture assay. Additionally, LMP2A increases the number of cancer initiating cells in a xenograft tumor formation assay. More importantly, the endogenous expression of LMP2A positively correlates with the expression of ABCG2 in NPC samples. Finally, we demonstrate that Akt inhibitor (V) greatly decreases the size of the stem cell side populations in LMP2A-expressing cells. Taken together, our data indicate that LMP2A induces EMT and stem-like cell self-renewal in NPC, suggesting a novel mechanism by which Epstein-Barr virus induces the initiation, metastasis and recurrence of NPC.  相似文献

Evidence over the last two decades from a number of disciplines has solidified some fundamental concepts in metastasis, a major contributor to cancer associated deaths. However, significant advances have been made in controlling this critical cellular process by focusing on targeted therapy. A key set of factors associated with this invasive phenotype is the nm23 family of over twenty metastasis-associated genes. Among the eight known isoforms, Nm23-H1 is the most studied potential anti-metastatic factor associated with human cancers. Importantly, a growing body of work has clearly suggested a critical role for Nm23-H1 in limiting tumor cell motility and progression induced by several tumor viruses, including Epstein-Barr virus (EBV), Kaposi's sarcoma associated herpes virus (KSHV) and human papilloma virus (HPV). A more in depth understanding of the interactions between tumor viruses encoded antigens and Nm23-H1 will facilitate the elucidation of underlying mechanism(s) which contribute to virus-associated cancers. Here, we review recent studies to explore the molecular links between human oncogenic viruses and progression of metastasis, in particular the deregulation of Nm23-H1 mediated suppression.  相似文献

The association of Helicobacter pylori (H. pylori) with gastric cancer is thus far the best understood model to comprehend the causal relationship between a microbial pathogen and cancer in the human gastrointestinal tract. Besides H. pylori, a variety of other pathogens are now being recognized as potential carcinogens in different settings of human cancer. In this context, viral causes of human cancers are central to the issue since these account for 10-20% of cancers worldwide. In the case of H. pylori and gastric cancer, as well as the human papillomavirus and anal cancer, the causal relationship between the infectious agent and the related cancer in the gastrointestinal tract has been clearly confirmed by epidemiological and experimental studies. Similarly, Epstein-Barr virus and the oncogenic JC virus are being suggested as possible causative agents for cancers in the upper and lower gastrointestinal tract. This review discusses various viral and microbial pathogens and their oncogenic properties in the evolution of gastrointestinal carcinogenesis and summarizes the available experimental data make a convincing agreement favoring the associations between infectious agents and specific human cancers.  相似文献

About 15-20% of human cancers worldwide have viral etiology. Emerging data clearly indicate that several human DNA and RNA viruses, such as human papillomavirus, Epstein-Barr virus, Kaposi's sarcoma-associated herpesvirus, hepatitis B virus, hepatitis C virus, and human T-cell lymphotropic virus, contribute to cancer development. Human tumor-associated viruses have evolved multiple molecular mechanisms to disrupt specific cellular pathways to facilitate aberrant replication. Although oncogenic viruses belong to different families, their strategies in human cancer development show many similarities and involve viral-encoded oncoproteins targeting the key cellular proteins that regulate cell growth. Recent studies show that virus and host interactions also occur at the epigenetic level. In this review, we summarize the published information related to the interactions between viral proteins and epigenetic machinery which lead to alterations in the epigenetic landscape of the cell contributing to carcinogenesis.  相似文献

Cancer remains a major burden for HIV-infected individuals. The majority of cancers associated with HIV infection are driven by oncogenic viruses like Epstein-Barr virus, Kaposi's sarcoma-associated herpesvirus and human papillomavirus. Kaposi's sarcoma is a tumour of endothelium and is the most common malignancy in HIV infected individuals.
