为比较猫的脂肪、骨髓、羊膜、脐带等器官组织4种不同器官来源间充质干细胞(Mesenchymal stem cells,MSC)的生物学特性,本研究通过全血培养法分离培养骨髓来源的间充质干细胞,并采用消化法分离培养脂肪、脐带和胎盘来源的间充质干细胞,比较它们的细胞形态、生长曲线、成脂成骨分化能力以及细胞表面标志物等生物学特性。结果显示,分离得到的4种不同来源的MSC均呈贴壁生长,细胞形态呈梭形、多棱形;均具有成脂成骨分化能力;脂肪源和骨髓源MSC的细胞增殖速度较快,细胞倍增时间较短,而羊膜与脐带源的MSC增殖能力较差,细胞倍增时间较长,4种细胞中脂肪来源MSC的增长活性最好,细胞传至第9代的时候依然保持良好的增殖活性,提示AD-MSC的临床应用可能比较好;4种MSC细胞表面均高表达CD105、CD90和CD44等标志物,低表达CD34。  相似文献

目的:研究再生障碍性贫血(aplasticanemia,从)患者骨髓间充质干细胞(mesenchymalstemcells,MSCs)的生物学特性和初步探讨其异常和AA发生的可能关系。方法:取AA患者骨髓间充质干细胞,测定其生长曲线和倍增时间;流式细胞仪检测其细胞周期和免疫表型;体外定向诱导其向脂肪、成骨、内皮和神经细胞分化;用real-timePCR及油红O染色法比较AA和正常对照组MSCs的成脂分化的不同。结果:AA患者和正常成人的MSCs均呈梭形贴壁生长;AA组细胞倍增时间长于对照组;CD105、CD44、CD29、CD106、FlK-1均阳性;96.51%的细胞处在G0/G1期;AA患者的MSCs保持了多向分化潜能,体外诱导形成脂滴较对照组早,诱导早期的脂蛋白脂酶表达增高。结论:再生障碍性贫血患者的骨髓间充质干细胞增殖能力较正常成人弱,骨髓间充质干细胞的易成脂性可能参与了再障的发病环节。  相似文献

目的:探讨内皮间充质转化(endothelial-to-mesenchymal transition,EMT)是否促进自发性高血压大鼠主动脉纤维化及降压治疗对其干预作用。方法:将24只雄性自发性高血压大鼠(spontaneously hypertensive rats,SHR)随机分为SHR组(S组,蒸馏水灌胃)、哌唑嗪组(P组,哌唑嗪5 mg/(Kg×d)灌胃)和氯沙坦组(L组,氯沙坦10 mg/(Kg×d)灌胃),8只雄性WKY大鼠(W组,蒸馏水灌胃)作为对照组,各组分别干预8周后,通过Masson染色检测各组大鼠主动脉纤维化程度,免疫荧光染色及Western Blot检测各组大鼠主动脉I型胶原、CD31及FSP1蛋白表达变化。结果:Western blot及Masson染色显示从L组、P组到S组大鼠主动脉I型胶原含量依次增多,且主动脉壁厚度均显著大于W组(P0.05),但P组与L组大鼠主动脉管壁厚度无统计学差异(P=0.818);免疫荧光染色表明,各组高血压大鼠主动脉均存在FSP1及CD31共表达(FSP+CD31+)细胞,且从L组、P组到S组FSP+CD31+细胞依次增多;Western Blot检测表明,从W组、L组、P组到S组,CD31蛋白表达量逐渐降少,FSP1蛋白表达量依次增加,差异有统计学意义(P0.05)。结论:EMT可能参与了自发性高血压大鼠的主动脉纤维化,氯沙坦和哌唑嗪可能通过抑制EMT减轻主动脉纤维化。  相似文献

脂肪来源细胞体外增殖规律及定向诱导分化研究   总被引:8,自引:0,他引:8  
脂肪组织由整形外科吸脂术获得(19例,31.5±5.8岁)。酶消化法分离抽吸物中细胞,体外扩增至第10代.测定细胞生长曲线、累计倍增数目,明确其体外生长规律和增殖能力;通过对表面抗原CD29、CD105、CD106、CD166、CD49d、CD34、CD31、3G5等的检测分析脂肪来源细胞的群体组成:分别向软骨、骨、脂肪定向诱导,进一步明确该细胞群体定向分化能力。实验表明,每300ml脂肪抽吸物平均可获得5×10~7个有核细胞,体外扩增10代,平均每代倍增数目为1.59±0.224.累计倍增数目为15.53。流式细胞学及免疫细胞化学检测显示,干细胞相关抗原CD29、CD105、CD106、CD166等表达率均>60%,但与造血系相关的CD34、CD31表达率也分别达到7.3%、29.2%。ADC向软骨诱导可检测到Ⅱ型胶原表达;向成骨诱导可见矿化结节形成,并可检测到AKP、Osteonectin基因表达;向脂肪诱导可检测到PPARr2、GLU-4、Leptin基因表达,细胞内有脂滴形成。脂肪来源的细胞获得量大,体外增殖能力强,并含有具有多向分化潜能细胞,有可能作为组织构建的种子细胞。  相似文献

目的 观察成年 (16周龄 )自发性高血压大鼠 (spontaneouslyhypertensiverat,SHR)与同龄对照组 (WKY)大鼠之间细胞外基质成分的差异及血管紧张素Ⅱ (AngiotensinⅡ ,AngⅡ )在SHR大鼠左室肥厚形成过程的作用。方法 用尾袖法间接测定大鼠血压 ;检测左心室组织及血浆中的血管紧张素转化酶 (angiotensinconvertingenzyme ,ACE)活性 (紫外分光光度法 ) ;放免法测定左室心肌AngⅡ含量。免疫组化测定左室心肌胶原含量 ,用3H -Proline掺入量测定体外培养心肌成纤维细胞 (cardiacfibroblast,CFB)胶原的合成率。结果  (1) 16周龄SHR大鼠血压明显高于对照组 (WKY)大鼠 ,分别为 (2 7.6 3± 2 .6 7)kPa和 (16 39± 0 54)kPa ,P <0 .0 5;(2 )SHR大鼠左室心肌AngⅡ含量明显高于WKY组 ,分别为 (2 6 6± 75)pg/ 10 0mg和 (134± 4 1)pg/ 10 0mg ,P <0 .0 5;(3)左室重量 (Leftventricalarmass,LVM)SHR明显高于WKY组 ,分别为 (10 14.3± 6 2 .1)mg和 (895.7± 86 .4 )mg ,P <0 .0 5;(4 )心体比 (Letventricrlarmass/bodyeight,LVM/BW )SHR明显高于WKY组 ,分别为 (3.4 4± 0 .15)mg/g和 (2 .17± 0 .11)mg/g ,P <0 .0 5;(5)体外细胞培养的心肌成纤维细胞3H -Proline掺入量随着AngⅡ浓度升高而增加 ,1μmol/L的AngⅡ使SH  相似文献

为了探索血小板源生长因子 (platelet derivedgrowthfactor,PDGF)受体介导的信号转导在自发性高血压大鼠(spontaneouslyhypertensiverats,SHR)心肌肥大中的作用 ,实验采用Westernblot法检测SHR及其对照WKY大鼠心肌PDGF受体β和细胞外信号调节激酶 (extracellularsignal regulatedkinase1/ 2 ,ERK 1/ 2 )的蛋白表达和ERK 1/ 2磷酸化水平的变化。结果显示 :4周龄SHR的收缩压、舒张压、±dp/dtmax和心肌肥大指数与同龄WKY大鼠相比均无明显差异 ,而 12周龄SHR上述指标与同龄WKY大鼠相比均明显升高 ,表明 12周SHR已发生高血压 ,心脏收缩功能代偿性增强 ,并出现心肌肥大。 4周龄SHR心肌PDGF受体 β和ERK1/ 2的磷酸化水平以及ERK 1/ 2的蛋白表达水平与同龄对照相比均无明显变化 ,12周时SHR心肌PDGF受体 β的蛋白表达较同龄WKY增加 32 77% (P <          0 0 5 ) ,PDGF受体介导的信号转导通路的下游信号分子ERK 1/ 2的磷酸化水平较同龄WKY升高 19 6 % (P =0 0 1) ,表明ERK 1/ 2的活化增加 ,但ERK 1/ 2的蛋白表达水平尚无变化。为进一步明确PDGF受体 β在心肌细胞生长中的作用及其与ERK 1/ 2活性的关系 ,采用PDGF BB刺激培养的乳鼠心肌细胞 ,发现 [3 H]亮氨酸掺入量明显增加 ,ERK 1/ 2的磷酸化水平明  相似文献

目的 :研究运动对高血压肥大心脏心肌初级和次级应答基因 (immediateearlygeneandlateresponsegene)表达的影响。方法 :采用Northern分子杂交方法对游泳运动 10周后自发性高血压大鼠 (spontaneouslyhypertensiverats ,SHR)心肌初级应答基因c fosmRNA和次级应答基因心钠素 (atrialnatriureticfactor ,ANF)mRNA的表达进行比较研究。结果 :游泳SHR收缩压和舒张压分别比安静SHR降低 2 2 %和 2 5 % (P <0 .0 1) ,但左心室重 /体重比值两组间无明显差异 (P >0 .0 5 )。SHR最后一次游泳 2 4h后 ,心肌c fosmRNA表达与安静SHR相比无明显差异 ,但两组大鼠比SHR的正常血压对照鼠WistarKyoto(WKY)分别提高 83 %和 80 %。游泳SHR心肌ANFmRNA表达比安静SHR降低 3 2 % ,但仍比WKY大鼠高 2 9%。结论 :SHR经过游泳运动后 ,出现心室肌ANF基因表达降低与c fos基因表达增强的不一致现象可能是运动改善高血压肥大心脏的分子机制之一。  相似文献

目的建立小鼠骨片间充质干细胞(MSC)分离培养及扩增的方法。方法取小鼠胫骨和股骨,洗去骨髓后,用胶原酶I消化疏松骨密质,利用MSC具有迁徙和贴壁生长的能力进行分离。并对获取的细胞进行流式鉴定和诱导分化。结果培养2d小鼠骨片边缘爬出成纤维样细胞,呈克隆和鱼群样生长,并可以进行持续传代培养。流式鉴定结果显示这群细胞表达MSC标志Scall(92.7%),CD29(98.4%),CD90(91.6%),不表达造血细胞标志CD34(1.57%),CD45(3.99%),CD11b(0.63%),并可成功诱导分化成骨细胞和脂肪细胞。结论成功建立从小鼠骨片中获得MSC的方法,为实验研究提供可靠的细晌实源.  相似文献

本文旨在探讨血管紧张素转换酶抑制剂雷米普利(Ramipril)是否通过调节脑动脉血管细胞缝隙连接蛋白43 (connexin43, Cx43)的表达发挥其降压及保护脑动脉的作用。Wistar-Kyoto (WKY)和自发性高血压大鼠(spontaneously hypertensive rat,SHR)随机分为4组:WKY组、WKY+Ramipril组、SHR组、SHR+Ramipril组(n=8)。运用无创尾动脉测压仪测量收缩压;采用苏木素-伊红染色观察脑动脉病理学改变;应用压力肌动图技术检测各组脑动脉血管收缩率;应用免疫荧光和免疫组织化学技术分析脑动脉上Cx43的分布及表达;采用real-time PCR和Western blot技术分别检测脑动脉上Cx43 mRNA及蛋白表达。结果显示:(1) SHR组收缩压显著高于WKY组(P 0.01, n=8);SHR+Ramipril组收缩压明显低于SHR组(P 0.01, n=8)。(2)相比于WKY组,SHR组脑动脉管壁增厚明显(P 0.01, n=8),而SHR+Ramipril组相比于SHR组,动脉管壁厚度明显减少。(3) SHR组脑动脉收缩率高于WKY组(P 0.05, n=8);SHR+Ramipril组脑动脉收缩率低于SHR组(P 0.05, n=8)。应用2-APB (Cx43非特异性阻断剂)或Gap26 (Cx43特异性阻断剂)预孵育后,SHR+Ramipril组动脉收缩率显著下降(P 0.05, n=8);给予Cx43非特异性激动剂AAP10预孵育后,SHR+Ramipril组动脉收缩率显著升高(P 0.05, n=8)。(4) SHR组脑动脉上Cx43 mRNA及蛋白表达水平高于WKY组(P 0.05, n=8);SHR+Ramipril组脑动脉上Cx43 mRNA及蛋白表达水平明显低于SHR组(P 0.05, n=8)。以上结果提示,雷米普利能够下调SHR脑动脉血管细胞间Cx43 mRNA和蛋白的表达,降低血压,改善高血压诱发的脑动脉重塑以及血管功能障碍。  相似文献

目的:探讨高血压合并动脉粥样硬化与血管紧张素原的相关性。方法:30只雄性16周龄SHR大鼠随机均分为SHR组和SHR合并AS组,另设15只同龄雄性WKY大鼠作为正常血压对照组即WKY组。SHR合并AS组饲以高脂饲料并辅以大剂量VitD3灌胃建立高血压动脉粥样硬化大鼠模型,SHR组和15只同龄雄性WKY大鼠均饲以标准饲料。各组大鼠分别于0、6、12周时光镜、电镜下评估血管病变,全自动生化分析仪检测血脂水平,并采用ELISA法检测血清AGT、AngII浓度。结果:SHR血清AGT、AnglI浓度显著高于WKY大鼠(P〈0.05)。存在AS病变的SHR合并AS组,血清AGT、AngII浓度明显高于无AS病变的SHR组,且随着AS病变严重性的增加,血清AGT、AngII浓度亦增加(P〈0.05)。结论:抑制AGT的表达可能为高血压患者中AS的防治提供一种新的方法。  相似文献

Total and regional skeletal muscle flows (radiolabeled microspheres) were determined in isolated maximally vasodilated hindquarters of spontaneously hypertensive rats (SHR) and age-matched (11-12 mo) normotensive Wistar-Kyoto rats (WKY) to assess the vascular flow capacity of the skeletal muscle vascular beds. Vascular flow capacity was estimated by measuring total hindquarters and regional muscle blood flows (under conditions of maximal vasodilation with papaverine or papaverine plus isoproterenol) over a wide range of perfusion pressures in WKY and SHR. Capillary exchange capacity was estimated by determining the capillary filtration coefficient. Isogravimetric capillary pressures and segmental vascular resistances were determined in each hindquarter. Isogravimetric flows and capillary pressures were not different between WKY and SHR. However, total and precapillary vascular resistances were significantly elevated in SHR, and postcapillary resistances were not different compared with WKY. Maximal capillary filtration coefficient values for the SHR group averaged 20% lower than WKY values, suggesting that hypertension was associated with a reduction in the microvascular surface area available for fluid exchange and, therefore, the capillary exchange capacity. Over the perfusion pressures studied, total hindquarters flows averaged 60% lower in SHR than in WKY. Flows to individual skeletal muscles averaged 76% lower in SHR than in WKY regardless of the muscle fiber type. Thus, modifications exist in the hindlimb skeletal muscle vasculature of SHR that reduces the capillary exchange capacity and limit the capacity of deliver flow at a given perfusion pressure gradient.  相似文献

目的 :探讨分离培养脐血CD-3 4 细胞来源间充质干细胞 (MSC)及研究其生物学特征。方法 :取足月妊娠健康产妇胎儿脐血 ,分离其中单个核细胞 (MNC) ,去除CD 3 4 细胞 ,体外用低糖型DMEM培养基培养。观察细胞形态、测定生长曲线、利用流式细胞仪对培养细胞进行表型测定、细胞周期分析、体外诱导分化实验以及检测造血因子的表达情况。结果 :脐血CD-3 4 细胞中可培养出间充质干细胞 ,可诱导向成骨和脂肪细胞分化并表达IL 6、SCF和SDF 1等造血生长因子。结论 :从足月妊娠健康产妇脐血CD-3 4 细胞可分离培养出间充质干细胞 ,具有与其它来源MSC类似的表型及分化潜能 ,在体外传代可保持其低分化状态并表达造血因子 ,可作为组织工程的种子细胞和具有促进造血作用  相似文献

Oxidative stress plays an important role in arterial hypertension and propionyl-L-carnitine (PLC) has been found to protect cells from toxic reactive oxygen species. In this work, we have evaluated the antioxidant capacity of chronic PLC treatment in spontaneously hypertensive rats (SHR) by measuring the activity of antioxidant enzymes and the lipid peroxidation in liver and cardiac tissues. The activity of glutathione peroxidase was decreased in liver and cardiac tissues of SHR when compared with their normotensive controls, Wistar- Kyoto (WKY) rats, this alteration being prevented by PLC treatment. Glutathione reductase activity was increased in hypertensive rats and no effect was observed after the treatment. No significant changes in superoxide dismutase activity were observed among all experimental groups. Liver of hypertensive rats showed higher catalase activity than that of normotensive rats, and PLC enhanced this activity in both rat strains. Thiobarbituric acid reactive substances, determined as a measure of lipid peroxidation, were increased in SHR compared with WKY rats, and PLC treatment decreased these values not only in hypertensive rats but also in normotensive ones. The content of carnitine in serum, liver and heart was higher in PLC-treated rats, but PLC did not prevent the hypertension development in young SHR. In addition, triglyceride levels, which were lower in SHR than WKY rats, were reduced by chronic PLC treatment in both rat strains. These results demonstrate: i) the hypotriglyceridemic effect of PLC and ii) the antioxidant capacity of PLC in SHR and its beneficial use protecting tissues from hypertension-accompanying oxidative damage.  相似文献

BACKGROUND: The unique potential of mesenchymal stromal cells (MSC) has generated much research interest recently, particularly in exploring the regenerative nature of these cells. Previously, MSC were thought to be found only in the BM. However, further studies have shown that MSC can also be isolated from umbilical cord blood, adipose tissue and amniotic fluid. In this study, we explored the possibility of MSC residing in the cornea. METHODS: Human cornea tissues were chopped to fine pieces and cultured in DMEM supplemented with 10% FBS. After a few days, the crude pieces of cornea were removed. Isolated keratocytes that were adherent to tissue culture flasks were grown until confluency before being passaged further. The immunophenotype was evaluated by flow cytometry. Assays were performed to differentiate cultured cells into adipocytes and osteocytes. RESULTS: Isolated corneal keratocytes exhibited a fibroblastoid morphology and expressed CD13, CD29, CD44, CD56, CD73, CD90, CD105 and CD133, but were negative for HLA-DR, CD34, CD117 and CD45. These properties are similar to those of BM-MSC (BM-MSC). In addition, corneal keratocytes were able to differentiate into adipocytes and osteocytes. DISCUSSION: Our results indicate that corneal keratocytes have MSC-like properties similar to those of BM-MSC. This study opens up the possibility of using BM-MSC in corneal tissue engineering and regeneration. Furthermore, discarded corneal tissue can also be used to generate MSC for tissue engineering purposes.  相似文献

Periodontal ligament (PDL) is a specialized connective tissue that connects cementum and alveolar bone to maintain and support the teeth in situ and preserve tissue homeostasis. Recent studies have revealed the existence of stem cells in human dental tissues including periodontal ligament that play an important role, not only in the maintenance of the periodontium but also in promoting periodontal regeneration. In this study, human periodontal ligament cells (hPDLCs) were isolated by outgrowth and enzymatic dissociation methods. Expression of surface markers on PDLCs as human mesenchymal stem cells (MSCs) was identified by flow cytometry. In addition, proliferation and differentiation capacity of cultured cells to osteoblasts, adipocytes were evaluated. As a result, we successfully cultured cells from the human periodontal ligament tissues. PDLCs express mesenchymal stem cell (MSC) markers such as CD44, CD73, and CD90 and do not express CD34, CD45, and HLA-DR. PDLCs also possess the multipotential to differentiate into various types of cells, such as osteoblast and adipocytes, in vitro. Therefore, these cells have high potential to serve as materials for tissue engineering, especially dental tissue engineering.  相似文献

Chronic treatment of spontaneously hypertensive rats (SHR) and Kyoto-Wistar normotensive rats (WKY) with nadolol was carried out from gestation until 28 weeks of age. Nadolol treatment caused some lowering of blood pressure but did not prevent the development of hypertension or cardiac hypertrophy in the SHR, in spite of significant beta-blockade. The lumen of large mesenteric arteries from control SHR was smaller than from WKY, and nadolol treatment increased the lumen size in the SHR. An increased number of smooth muscle cell layers present in the control SHR as compared with WKY was reduced slightly by nadolol treatment. However, the changes produced by nadolol did not reach the levels of control and treated WKY. In the aorta, the incidence of polyploid smooth muscle cells was higher in the SHR than the WKY in the control group. Nadolol treatment reduced the percentage of polyploid cells in both SHR and WKY, so that the difference between these two groups of animals was eliminated in the treated groups. The tissue level of norepinephrine in the plasma, heart, mesenteric arteries, and adrenal glands in the SHR and WKY was not affected by the treatment. We suggest that the ineffectiveness of nadolol in preventing hypertension development may be due to its lack of effect in preventing primary changes in the resistance arteries, and that the development of polyploidy of smooth muscle cells may be mediated by beta-receptors.  相似文献

The locus ceruleus (LC) contains a high density of angiotensin II (All) receptors. The role of All receptors at the LC in genetic hypertension and organ function is unclear. Spontaneously hypertensive (SHR) rats and Wistar-Kyoto (WKY) rats were studied, and blood pressure of animals was measured using the tail-cuff method. Animals were decapitated and the heart weight (HW) and testicular weight (TW) of animals measured. All receptor binding was carried out by incubating the LC tissue sections with 200 pM [125I]-All receptor ligand, and measured using quantitative autoradiography. Results showed that the HW/BW ratio was significantly higher in SHR rats than WKY rats. However, the TW/BW ratio was higher in SHR rats than WKY rats only at two hypertensive stages, whereas All receptor binding capacity in the LC was also statistically higher in SHR rats than WKY rats. Results indicated that cardiac and testicular hypertrophies were related to higher All receptor binding in the LC of SHR rats, when compared with WKY rats. Interestingly, the literature shows that there is an LC-testes axis. In conclusion, this study indicated that All receptors in the LC are associated with genetic hypertension, and testicular weight could be a reasonable index for essential hypertension.  相似文献

We investigated the catecholamine-stimulated lipolytic response of perirenal adipocytes isolated from spontaneously hypertensive (SHR) and normotensive (C) rats of the Sprague-Dawley (SD) and Wistar-Kyoto (WKY) strain. Younger rats (10-17 weeks) were matched with respect to age and body weight. Age-matched SHR rats were smaller than their C counterparts, had equal-size adipocytes, and demonstrated lipolytic responses equal to C cells. Weight-matched SHR rats were older than normotensive controls, had larger adipocytes, and showed depressed norepinephrine (NE)-stimulated responses with a rightward shift in the dose-response curve. Rates of lipolysis of SHR and C cells were not different in the simultaneous presence of norepinephrine and theophylline. Nine- to ten-month-old rats were of comparable body weight and adipocyte size regardless of blood pressure status; however, SHR cells still showed a significantly blunted response to catecholamine stimulation. We conclude that (1) the NE-stimulated lipolytic response of adipocytes of SHR rats is significantly less than that elicited from C cells; (2) this function difference seems unrelated to a size difference between cells of younger, SHR and C rats, thus implicating the adrenergic system; and (3) whole body growth (as reflected by body weight) and perirenal adipocyte growth do not proceed in parallel in actively growing SHR rats.  相似文献

In hypertension arterial wall properties do not necessarily depend on increased blood pressure alone. The present study investigates the relationship between the development of hypertension and thoracic aortic wall properties in 1.5-, 3-, and 6-mo-old spontaneously hypertensive rats (SHR); Wistar-Kyoto rats (WKY) served as controls. During ketamine-xylazine anesthesia, compliance and distensibility were assessed by means of a noninvasive ultrasound technique combined with invasive blood pressure measurements. Morphometric measurements provided in vivo media cross-sectional area and thickness, allowing the calculation of the incremental elastic modulus. Extracellular matrix protein contents were determined as well. Blood pressure was not significantly different in 1.5-mo-old SHR and WKY, but compliance and distensibility were significantly lower in SHR. Incremental elastic modulus was not significantly different between SHR and WKY at this age. Media thickness and media cross-sectional area were significantly larger in SHR than in WKY, but there was no consistent difference in collagen density and content between the strains. Blood pressure was significantly higher in 3- and 6-mo-old SHR than in WKY, and compliance was significantly lower in SHR. The findings in this study show that in SHR, in which hypertension develops over weeks, alterations in functional aortic wall properties precede the development of hypertension. The decrease in compliance and distensibility at a young age most likely results from media hypertrophy rather than a change in intrinsic elastic properties.  相似文献
