用雌、孕激素处理去卵巢家兔后观察子宫内膜鞘糖脂的含量和组成。结果表明、雌激素处理后、子宫内膜Gls含量是孕激素处理的十余倍,而孕激素增加NGSL含量的作用却比雌激素更强。两种激素对鞘糖脂组分的影响也明显不同。雌激素给药组子宫内膜鞘糖脂以多糖基组分GD_3 GT_(1b) CPH为主,孕激素组则以短糖链组分GM_3 CMH CDH为主。雌激素预先作用后再用孕激素或雌、孕激素同时给药,与单用雌激素后比较其短糖链组分明显增加,多糖基组分则明显减少,表现出两者的共同作用。上述结果提示:雌、孕激素对鞘糖脂代谢影响明显不同,雌激素使糖链复杂化、孕激素则使糖链趋向于简单。  相似文献

 妊娠期家兎子宫内膜的神经节苷脂(Gls)的含量明显低于动情期的,而中性鞘糖脂(NGSL)的含量则以妊娠中、晚期的最高,动情期最低。鞘糖脂组成变化最显著的是妊娠早期,由动情期到早孕GM_3从28.0%增加到52.7%,CMH.CDH由未测出分别增加到29.2%和21.9%,而糖链复杂的组分GD_3,GTlb和CPH的百分含量则明显减少,到妊娠中、晚期、短糖链组分逐渐减少,而复杂糖链组分渐增。中期妊娠内膜的(GIs)以GD_3为主要组分,占45%,明显高于其它各期。NGSL在妊娠中、晚期CPH增高达70%,与动情期水平相当。结果提示,妊娠期间子宫内膜的鞘糖脂含量与组成均发生明显变化,这些变化可能与子宫功能密切相关。特别是早孕对的变化,推测与子宫内膜和胚泡的识别,粘连特性的获得有关。  相似文献

本文测定了新生儿、生育期、更年期和足月妊娠人子宫肌肉的神经节苷脂(Gg)与中性鞘糖脂(N-GSL)的含量,比较了两种鞘糖脂的HPTLC谱。新生儿期Gg的总含量(以脂结合唾液酸LBSA量表示)最高,每克湿重组织约45.2μg,足月妊娠子宫肌肉中的含量最低,为10.4μg,生育期为32.8μg、更年期为39.5μg。N—GSL的含量却以足月妊娠子宫肌肉中最多,达99.4μg。按HPTLC谱分析子宫肌肉中Gg的主要组分为GD_3和GM_3,在子宫发育成熟与妊娠时,肌肉组织中这两种组分的含量变化明显:生育期样品的GD_3由新生儿的25.4%增加到56.6%(按占LBSA总量的百分比计算),GM_3则由33.2%降至16.9%。此外,GM_1和GD_(1a)也明显减少。N—GSL在生育期CMH、CDH和CTH的含量(按占含糖基量的百分比计算)成倍增加,而含五糖基以上的组分则仅为新生儿子宫的1/5。足月妊娠与新生儿子宫肌肉的两类鞘糖脂的HPTLC谱类似,但前者GT1b占19.4%,明显高于新生儿样品(6.1%)。  相似文献

采用高效薄层层析(HPTLC)对两株具有不同淋巴道转移潜能的小鼠腹水型肝癌瘤株细胞膜鞘糖脂组分进行了比较分析.低转移的CL-A_2瘤株神经节苷脂以GM_3为主,高转移的CL-16A_3瘤株则以GM_2为主.两细胞株中性鞘糖脂各组分相对百分含量无较显著差异.脂结合唾液酸含量测定表明,CL-16A_3瘤株脂结合唾液酸含量约为CL-A_2瘤株的三倍.提示,具有不同淋巴道转移潜能的瘤细胞,其质膜鞘糖脂的组成也不同.  相似文献

 利用气相色谱、气相色谱-质谱联用等方法测定了狗小肠鞘氨醇糖脂中的长链碱组成。其主要的长链碱为鞘氨醇(Sphingosine)、异鞘氨醇(isosphingosine)、二氢鞘氨醇(Sphinganine)和植物鞘氨酸(Phyto-sphingosine)。一共分离出十三个鞘氨醇糖脂。在唯一的五糖基神经酰胺中异鞘氨醇是主要成份。在一个一糖基神经酰胺中植物鞘氨醇是主要成份。植植物鞘氨酸也是两个二糖基神经酰胺和一个三糖基神酰胺的主要长链碱。说明它不仅存于植物体内。  相似文献

从月经周期各期、早妊(6—8周)及足月妊娠子宫内膜中提取、纯化了神经节苷脂(Gls)和中性鞘糖脂(N-GSL),分别测定了其含量。对两类鞘糖脂的组成进行了HPTLC图谱分析。初步观察了不同时期子宫内膜中CMP—NeuAc:LacCer唾液酰转移酶(ST_1)和CMP-NeuAc:CM_3唾液酰转移酶(ST_2)的活性。结果表明:分泌期的GLs总含量低于增生期(P<0.02);分泌期GD_3含量较生长期增多(P<0.01);妊娠后,GM_3含量增加,而GD_3减少;相应地,ST_1活性增高,ST_2活性降低。分泌期CMH含量为增生期的4.7倍。结果提示子宫内膜鞘糖脂含量和组成的变化可能与子宫的功能有关,而且受女性激素水平的影响。  相似文献

丝状真菌作为一类重要的微生物,被广泛应用于发酵食品、工业酶和次生代谢物等工业生产中。真菌鞘糖脂主要由鞘氨醇、脂肪酸链和特殊的极性基团组成,根据极性基团的不同,分为中性鞘糖脂和酸性鞘糖脂两大类。鞘糖脂不仅参与真菌生长、细胞分化、增殖、细胞凋亡、逆境胁迫等重要生理活动,中性鞘糖脂还可作为功能性医药用品、化妆品和保健食品的重要活性组分。本文论述了真菌鞘糖脂的主要种类、结构、生物合成途径和及其参与丝状真菌生长、分化和响应逆境胁迫的生物学功能;探讨了真菌中性鞘糖脂作为抗菌肽的靶点和酸性鞘糖脂在开发抗真菌药物中的应用;同时还综述了中性鞘糖脂作为化妆品的保湿成分或保健食品的功能成分,在改善皮肤屏障功能和预防特应性皮炎中的重要作用的相关研究进展,尤其是来源于曲霉的中性鞘糖脂,可显著增强皮肤屏障功能,并可作为益生元预防肠道损伤;另外还探讨了曲霉尤其是米曲霉作为开发中性鞘糖脂生物资源的优势。  相似文献

GM3对J6-2细胞的促分化作用及对鞘糖脂与磷脂组分的影响   总被引:4,自引:2,他引:2  
本文在用形态学与组化手段证实神经节苷脂GM3促进国人白血病细胞系J6-2分化的基础上,研究了分化前后细胞鞘糖脂及磷脂组分的变化。作者用自行提纯的GM3表明,J6-2细胞在分化前后没有显著的神经节苷脂组分的改变,中性鞘糖脂组分CMH在分化后明显增多。GM3使LPC、SPM及PI+PS等磷脂组分减少,但使PC组分增多。此项结果提示了神经节苷脂的一个新的功能——调节磷脂代谢,但其具体机制尚待阐明。  相似文献

糖鞘脂是一类广泛分布在动物细胞膜表面的糖脂类物质,它在调控细胞识别、黏附、增殖以及凋亡等方面均有重要的生物学作用.本综述主要讨论了在现代分析技术范畴中,糖鞘脂的鉴定及其糖链结构的分离与解析方面的研究进展和糖鞘脂在癌症等疾病发生发展中所起的生物学功能,以及糖鞘脂作为疾病治疗靶标的可能性.随着现代仪器技术,尤其是质谱技术和色谱-质谱联用技术的发展,糖鞘脂的分离与检测也进入了高速发展的时代.目前,使用质谱技术在肝癌、结直肠癌、乳腺癌等恶性肿瘤的组织样本中均发现了不同种类糖鞘脂不同程度的异常表达.其中,岩藻糖基化的糖鞘脂上调表达在众多癌症糖鞘脂检测中尤为突出,故岩藻糖基化的糖鞘脂可能会成为一类癌症的早期诊断标志物.近年来,随着对糖鞘脂理解的不断深入,糖鞘脂在诸多疾病,如癌症血管生成过程中的功能研究成为了热点之一.例如,从肿瘤细胞表面脱落的大多数糖鞘脂在肿瘤微环境中主要起到了促进血管生成的作用,而与此相反的是,另一种结构简单的神经节苷脂GM3却起到了抑制血管生成的作用.本综述汇集了对上述现象在分子水平上的不同解读以及利用此现象对癌症靶向治疗的研究与探索,并对基于抑制糖鞘脂合成的靶向治疗的发展前景进行了分析展望.  相似文献

Lymphocytes from spontaneous thymoma in AKR mice and from X-ray induced thymoma in C57B1/6 mice showed elevated levels (by 50% and 100%, respectively) of lipid-bound sialic acid as compared with lymphocytes from normal thymuses used as controls. Some ganglioside fractions in thymomas were elevated 4-6-fold over those in normal thymuses while other fractions decreased or disappeared. Neutral glycosphingolipid (NGSL) content in lymphocytes from thymomas was also changed. Thin-layer chromatography of NGSLs showed that the fractions migrating as ceramide monohexoside (CMH), dihexoside (CDH) and below globoside standards were increased, respectively, 2-3-fold, 3-6-fold and 2-fold in both types of thymomas. Methylation and gas-liquid chromatography analysis confirmed the presence of CMH, CDH and globoside in NGSLs isolated from X-ray induced thymoma.  相似文献

Density dependent chain elongation of neutral glycosphingolipids (NGSL) is associated with contact inhibition of mitosis in several normal cultured cell lines. Transformed non-neural cell lines which have impaired contact inhibition frequently lose this biochemical response. To determine if either of these phenomena occur in human neural cells we determined NGSL compositions of cultured glioblastoma multiforme and normal fetal brain cells. Fetal cells generally had more total NGSL than the tumor cells. As a percentage of total NGSL, both cell lines at higher cell densities had larger proportions of ceramide trihexoside and globoside, but smaller proportions of cerebroside. This decrease was mainly in nonhydroxy fatty acid cerebroside of glioma cells, but in hydroxy fatty acid cerebroside of normal fetal brain cells. These results demonstrate that although glioblastoma multiforme cells have markedly impaired growth control, they still preserve density dependent chain elongation of NGSL. A role for this phenomenon in normal cellular growth control has yet to be established.  相似文献

Perinatal (1-2 days of age) and one-month-old (24-32 days of age) male goats were used to investigate the effect of age and long-term culture (24 h) of perirenal and omental adipose explants in the presence of insulin, cortisol and bovine somatotropin (alone or in different combinations) on net glucose-stimulated lipogenesis (NGSL, i.e. the rate of lipogenesis in the presence of glucose minus the rate of lipogenesis in the absence of glucose) in the absence and in the presence of catecholamines in acute incubations (2 h). Mean values of NGSL in both freshly prepared and cultured explants were consistently lower in perinatal than in one-month-old goats. Cortisol alone decreased and combinations of insulin plus cortisol increased NGSL in perirenal explants of one-month-old animals. When perirenal explants from these one-month-old goats were cultured in the presence of insulin plus cortisol plus bovine somatotropin, the rates of lipogenesis were lower than those in cultures with insulin plus cortisol. No such effects of these hormones were noted in omental explants of both perinatal and one-month-old animals. In freshly prepared perirenal and omental explants, the rates of NGSL were inhibited by isoprenaline in tissues of both groups of animals and by noradrenaline in omental tissues of animals of the older group only. The mean values of NGSL in cultured explants of perinatal animals were not affected by noradrenaline. Isoprenaline inhibited NGSL in omental but not in perirenal tissue. In older animals the rates of NGSL were decreased by both noradrenaline and isoprenaline in perirenal and omental adipose tissues. Isoprenaline was more effective than noradrenaline in perirenal adipose tissue.  相似文献

Net glucose-stimulated lipogenesis (NGSL: the rate of lipogenesis in the presence of glucose minus the rate of lipogenesis in the absence of glucose) in omental adipose tissue explants from young castrated male goats was evaluated in control animals (n = 3; placebo-treated) and in animals treated with the sustained release of recombinant bovine somatotropin (n = 4; bST; 100 mg at 7-day intervals in a 147 days lasting experiment). The rate of fatty acid synthesis was determined in acute incubations in both freshly prepared and chronically cultured explants. Adipose explants remained metabolically active and retained their ability to respond to hormones when maintained in a tissue culture medium. NGSL in explants cultured for 24 h in the presence of insulin alone or bST alone, was non-significantly increased (more in the controls) and decreased (more in bST-treated animals), respectively. However, cortisol alone decreased (P<0.05) NGSL in explants from both control and bST-treated animals. In tissues from bST-treated animals, cortisol acted synergistically with insulin to produce a higher rate of NGSL than that observed in cultures with insulin alone. bST inhibited insulin plus cortisol-stimulated lipogenesis significantly (P<0.05) in explants from bST-treated animals but non-significantly in control animals. The rates of NGSL were decreased (P<0.05) by catecholamines in explants from both control and bST-treated animals. Norepinephrine (NE) and isoprenaline (ISO) were equally effective in the controls, whereas isoprenaline was more effective than norepinephrine in bST-treated animals.  相似文献

Gangliosides of human gastric and mammary tumours and of homologous normal tissues were studied by using biochemical methods and specific antisera. It was found that in most cases GM3, GD3 and GM1 are predominant gangliosides, whereas several polar components are minor ones. A comparison of the relative amount of ganglioside fractions revealed that in gastric tumours the per cent content of polar compounds is higher than in intact tissue; however, the absolute content of all gangliosides is markedly increased. A comparative study of the composition of mammary tumour and normal tissue gangliosides demonstrated two types of changes: i) the absolute content of all gangliosides in tumour tissue was increased and, ii) the increase in the content of total gangliosides was paralleled with the appearance of a new fraction (presumably GM4), the decrease of the GD3 content and the disappearance of polar gangliosides. A possible mechanism of this effect is discussed.  相似文献

We recently identified ganglioside GM3 as a modulator of glomerular hypertrophy in streptozotocin-induced diabetic rats (Life Sci., 72: 1997-2006, 2003). This study examined whether alteration of ganglioside GM3 expression could modulate the high glucose-induced proliferation of glomerular mesangial cells (GMCs). GMCs isolated from rat kidneys were cultured under normal (5.6 mM) or high (25 mM) glucose condition for 24-72 hrs. Cell proliferation was predominantly stimulated when GMCs were cultured with high glucose as well as 20 microM of d-threo-PDMP, an inhibitor of ganglioside biosynthesis, for 24 hrs, whereas raising ambient glucose significantly reduced the mesangial sialic acid contents. Based upon mobility on high-performance thin-layer chromatography (HPTLC), GMCs showed a complex pattern of ganglioside expression that consisted of three major components of gangliosides, mainly GM3. High glucose induced a significant reduction of ganglioside expression with apparent changes in the composition of major ganglioside GM3, and semi-quantitative analysis by HPTLC showed that ganglioside GM3 was reduced to 62% of GMCs cultured under normal glucose condition. A prominent immunofluorescence microscopy using anti-GM3 monoclonal antibody also showed a dramatic disappearance of immunoreactivity in high glucose-treated GMCs. Moreover, high glucose significantly lowered the Km values of GM3 synthase (16 microM vs. 49 microM), but did not change the Vmax. These results provide the pathophysiological relationship between the high glucose-induced proliferation of GMCs and the decreased expression of ganglioside GM3, indicating a mechanism for the negative regulation of mesangial proliferation by ganglioside GM3. This mechanism may play an important role in the development of diabetic glomerulopathy.  相似文献
