A genetic linkage map for the duck (Anas platyrhynchos) was developed within a cross between two extreme Peking duck lines by linkage analysis of 155 polymorphic microsatellite markers, including 84 novel markers reported in this study. A total of 115 microsatellite markers were placed into 19 linkage groups. The sex-averaged map spans 1353.3 cM, with an average interval distance of 15.04 cM. The male map covers 1415 cM, whereas the female map covers only 1387.6 cM. All of the flanking sequences of the 155 polymorphic loci--44 monomorphic loci and a further 41 reported microsatellite loci for duck--were blasted against the chicken genomic sequence, and corresponding orthologs were found for 49. To integrate the genetic and cytogenetic map of the duck genome, 28 BAC clones were screened from a chicken BAC library using the specific PCR primers and localized to duck chromosomes by FISH, respectively. Of 28 BAC clones, 24 were detected definitely on duck chromosomes. Thus, 11 of 19 linkage groups were localized to 10 duck chromosomes. This genetic and cytogenetic map will be helpful for the mapping QTL in duck for breeding applications and for conducting genomic comparisons between chicken and duck.  相似文献

以384只北京鸭 (Z2系、Z4系、Z2×Z4杂交系)和樱桃谷鸭为材料, 利用PCR-SSCP结合测序技术, 对前胰岛素原基因外显子2与部分内含子的多态性进行了研究, 并分析对屠体性状的遗传效应。结果发现存在2个单核苷酸突变位点, 即在第179位和第195位分别发生了T→C和C→T的突变。适合性χ2检验结果表明, 北京鸭各品系和樱桃谷鸭均处于Hardy-Weinberg平衡状态(P>0.05)。最小二乘分析SNPs与屠体性状的关系表明, 在北京鸭3个品系中, 基因型 BB 在胴体重、全净膛重和胸肌重上极显著(P<0.01)高于基因型AA和AB, 在腿肌重和皮脂重上极显著(P<0.01)高于基因型AB; 基因型AA在皮脂率和全净膛重上极显著(P<0.01)和显著(P<0.05)高于基因型AB。而对于樱桃谷鸭, 只有AB型在皮脂重和腹脂重上显著(P<0.05)高于基因型AA。研究结果表明, 鸭前胰岛素原基因多态性与鸭的部分屠体性状存在显著相关性, 且B等位基因有利于增加鸭的胴体重和胸肌重。  相似文献

RFLP and RAPD markers were evaluated and compared for their ability to determine genetic relationships in a set of three B. napus breeding lines. Using a total of 50 RFLP and 92 RAPD markers, the relatedness between the lines was determined. In total, the RFLP and the RAPD analysis revealed more than 500 and 400 bands, respectively. The relative frequencies of loci with allele differences were estimated from the band data. The RFLP and RAPD marker sets detected very similar relationships among the three lines, consistent with known pedigree data. Bootstrap analyses showed that the use of approximately 30 probes or primers would have been sufficient to achieve these relationships. This indicates that RAPD markers have the same resolving power as RFLP markers when used on exactly the same set of B. napus genotypes. Since RAPD markers are easier and quicker to use, these markers may be preferred in applications where the relationships between closely-related breeding lines are of interest. The use of RAPD markers in fingerprinting applications may, however, not be warranted, and this is discussed in relation to the reliability of RAPD markers.  相似文献

This study presents an attempt to supply breeders of hybrid rye with more genetic information on inbred lines, using molecular markers. Eighteen polymorphic loci detected by means of the RAPD (Random Amplified Polymorphic DNA) technique and mapped on 2R-7R rye chromosomes, were applied to study genetic similarities among forty inbred lines of rye. The lines were grouped in four main clusters revealed on dendrogram, which was generally consistent with the pedigree data. Mapped RAPD markers were shown to be a useful tool for phenetic studies in rye. Additionally, a system of 20 polymorphic fragments, detected by three primers, was developed for fingerprinting of rye lines. The system of RAPD markers, which was developed in this study, should be helpful in characterisation of rye genetic stocks used for breeding.  相似文献

Contrary to chicken and livestock mammals, duck genome has not been explored much. Nowadays a relatively small number of reports on molecular variability and mapping of loci in Peking ducks has been published. Therefore, the objective of this study was to detect single loci affecting body weight, carcass and meat traits in Peking ducks (Anas platyrhynchos). The study was based on an F2 cross between two parental lines A-55 and GL-30. Phenotypes of 387 birds from generation F2 including carcass and meat quality traits were collected. We have isolated and characterized 18 microsatellite loci in the Peking duck (Anas platyrhynchos). The average number of alleles per locus was 3.5, ranging from one to six in domestic Peking ducks (n = 40). All of the markers were polymorphic in a sample of five mallards (A. platyrhynchos; two to eight alleles). Seventeen of the 18 markers amplified in Muscovy ducks (Cairina moschata) with 11 being polymorphic in our sample (n = 14). Amplification of the markers in different species comprising the subfamilies Anatinae and Anserinae indicates their potential value for population genetic applications in a wide range of waterfowl species.  相似文献

Random amplified polymorphic DNA (RAPD) markers have been used to characterize the genetic diversity among 35 spring wheat cultivars and lines with different levels of Fusarium resistance. The objectives of this study were to determine RAPD-based genetic similarity between accessions and to derive associations between Fusarium head blight (FHB) and RAPD markers. Two bulked DNA from either highly resistant lines or susceptible lines were used to screen polymorphic primers. Out of 160 screened primers, 17 primers generated reproducible and polymorphic fragments. Genetic similarity calculated from the RAPD data ranged from 0.64 to 0.98. A dendrogram was prepared on the basis of a similarity matrix using the UPGMA algorithm, which corresponded well with the results of principal component analysis and separated the 35 genotypes into two groups. Association analysis between RAPD markers and the FHB index detected three RAPD markers, H19(1000), F2(500) and B1(2400), significantly associated with FHB-resistant genotypes. These results suggest that a collection of unrelated genotypes can be used to identify markers linked to an agronomically important quantitative trait like FHB. These markers will be useful for marker-assistant breeding and can be used as candidate markers for further gene mapping and cloning.  相似文献

 RAPD markers and agronomic traits were used to determine the genetic relationships among 32 breeding lines of melon belonging to seven varietal types. Most of the breeding lines were Galia and Piel de Sapo genotypes, which are currently being used in breeding programmes to develop new hybrid combinations. A total of 115 polymorphic reliable bands from 43 primers and 24 agronomic traits were scored for genetic distance calculations and cluster analysis. A high concordance between RAPDs and agronomic traits was observed when genetic relationships among lines were assessed. In addition, RAPD data were highly correlated with the pedigree information already known for the lines and revealed the existence of two clusters for each varietal type that comprised the lines sharing similar agronomic features. These groupings were consistent with the development of breeding programmes trying to generate two separate sets of parental lines for hybrid production. Nevertheless, the performance of certain hybrids indicated that RAPDs were more suitable markers than agronomic traits in predicting genetic distance among the breeding lines analysed. The employment of RAPDs as molecular markers both in germplasm management and improvement, as well as in the selection of parental lines for the development of new hybrid combinations, is discussed. Association between the RAPD markers and the resistance to race 4 of the black rot causative agent was studied in Brassica rapa L. Experiments were carried out using doubled haploid lines, obtained via crosses between the race 4-susceptible fodder turnip and resistant pak-choi, and the F2 progeny of the crosses between the doubled haploid lines with contrasting resistance. The WE(22)980 RAPD marker inherited from the pak-choi and associated with the clubroot susceptibility was also linked to the locus responsible for the resistance to race 4 of Xanthomonas campestris pv. campestris. The two other RAPD markers were linked to susceptibility to black rot. Simultaneous association of the same DNA markers with the resistance/susceptibility to two different obligate pathogens favored the hypothesis on cluster organization of the resistance genes in plants. The markers described can be used in plant breeding and in further investigation of the genetic bases of resistance in plants.  相似文献

In our previous work (J. Palyga, Genetic polymorphisms of histone H1. b in duck erythrocytes. Hereditas 114, 85-89, 1991) we reported a genetic polymorphism of duck erythrocyte histone H1.b. Here, we screened H1 preparations in a two-dimensional polyacrylamide gel to refine the distribution of allelic forms of H1.b in fifteen duck populations. We have revealed that the frequency of H1.b allelic variants was significantly different among many conservative and breeding duck groups. While b(1) and b(3) were common in all populations screened, the allele b(2), with a slightly lower apparent molecular weight, was confined mainly to brown-feathered ducks (Khaki Campbell and Orpington) and descendent lines.The C- and N-terminal peptides released upon cleavage with N-bromosuccinimide and Staphylococcus aureus protease V8 from duck allelic histones H1. b2 and H1.b3, respectively, migrated differently in the gel, probably as a result of potential amino acid variation in a C-terminal domain.  相似文献

In order to study duck microsatellites, we constructed a library enriched for (CA)n, (CAG)n, (GCC)n and (TTTC)n. A total of 35 pairs of primers from these microsatellites were developed and used to detect polymorphisms in 31 unrelated Peking ducks. Twenty-eight loci were polymorphic and seven loci were monomorphic. A total of 117 alleles were observed from these polymorphic microsatellite markers, which ranged from 2 to 14 with an average of 4.18 per locus. The frequencies of the 117 alleles ranged from 0.02 to 0.98. The highest heterozygosity (0.97) was observed at the CAUD019 microsatellite locus and the lowest heterozygosity (0.04) at the CAUD008 locus, and 11 loci had heterozygosities greater than 0.50 (46.43%). The polymorphism information content (PIC) of 28 loci ranged from 0.04 to 0.88 with an average of 0.42. All the above markers were used to screen the polymorphism in other bird species. Two markers produced specific monomorphic products with the chicken DNA. Fourteen markers generated specific fragments with the goose DNA: 5 were polymorphic and 9 were monomorphic. But no specific product was detected with the peacock DNA. Based on sequence comparisons of the flanking sequence and repeat, we conclude that 2 chicken loci and 14 goose loci were true homologous loci of the duck loci. The microsatellite markers identified and characterized in the present study will contribute to the genetic map, quantitative traits mapping, and phylogenetic analysis in the duck and goose.  相似文献

As a special species of avian, Peking duck is often used as a model for exploring effective factors against cardio-cerebrovascular diseases, and therefore investigations of antioxidant enzymes including superoxide dismutase are intriguing. By using 3(')-RACE with a gene-specific primer, a cDNA encoding duck Cu,Zn SOD was amplified from the total RNA extracted from Peking duck liver. Three free cysteine residues are found in the deduced amino acid sequence of duck SOD, among which Cys153 at the carbonyl-terminal is a distinctive feature. Production with a high yield of recombinant duck Cu,Zn SOD was achieved in Escherichia coli after the reconstituted expression vector pET-3a-dSOD was transformed into the bacterial strain BL21(DE3)pLysS. After two steps of anion exchange chromatography, a great quantity of the purified enzyme (100mg/L fermented culture) with an enzymatic activity comparable to that of native duck and bovine SOD was finally obtained. Duck SOD is a homodimer with 153 residues for each subunit. The molecular mass of the recombinant enzyme is 15,540.0Da measured by mass spectrum, which well coincides with the estimated size of the sequence but significantly differs from that of the native counterpart. Five charge isomers were observed on isoelectricfocusing (IEF). The most interesting observation is that the thermal stability of duck SOD is much lower than that of the bovine enzyme as revealed by irreversible heat inactivation at 70 degrees C. These properties are discussed in relation to the distinctive free Cys residues in duck Cu,Zn SOD.  相似文献

In order to elucidate the domestication history of Peking ducks, 190 blood samples from six Chinese indigenous duck breeds were collected with186 individualsgenotyped by 15 microsatellite markers. Both the FST and Nei’s standard genetic distances (Ds) from the microsatellite data indicated high genetic differentiation between Peking duck and other Chinese indigenous breeds. The haplotype network with mtDNA data showed that most of the Peking duck haplotypes were distinctly different from those of other domestic breeds. Although the H01 haplotype was shared by all domesticated duck breeds, Peking ducks displayed 12 specific domestic duck haplotypes, including four similar haplotypes H02, H04, H08 and H22, that formed a single haplogroup (A). Both H02 and H22 haplotypes were also shared by mallard and Peking ducks, indicating that Peking ducks originated from wild mallard ducks.  相似文献

Genetic diversity among 83 lentil genotypes including 23 wild types, 19 indigenous varieties, 5 exotic lines and 36 advanced breeding lines was studied using molecular markers. A total of 112 amplicons were produced using 15 RAPD and 8 SSR markers. Dendrogram based on Jaccard similarity coefficient and UPGMA analysis revealed two major clusters and one minor cluster. Cluster I comprised 21 wild accessions of L. orientalis and 1 L. ervoides subspecies. Nineteen Indian varieties grouped together in subcluster IIA indicating their narrow genetic base. Subcluster IIB consisted of 41 genotypes including 5 exotic and 36 advanced breeding lines mainly derived from exotic genotypes. The narrow genetic base of released cultivars and germplasm lines emphasized the need for broadening of genetic base of breeding material using exotic collections and wild species to ascertain genetic improvement upon existing cultivars.  相似文献

用40个10碱基随机引物对福建2个生态类型的7个地方鸭种的基因组池DNA进行了RAPD分析,从扩增产物表现为多态的引物筛选出9个特异性强的引物进行个体DNA的RAPD分析,并利用Shannon指数估算了2个生态区域鸭群的遗传多样性,探讨了它们与生态环境之间的关系及其变化规律.结果表明,福建境内的地方鸭种具有较高的遗传多样性,闽东沿海地区鸭群有67.97%的遗传变异来自群体内,32.03%来自于群体间;闽西山地丘陵地区鸭群有59.05%的遗传变异来自群体内,40.95%来自于群体间;闽东地区鸭群的遗传变异明显高于闽西地区鸭群.同时,用非加权组平均法(UPGMA)对各种群的聚类分析表明,闽西地区的连城白鸭、泰宁麻鸭、龙岩山麻鸭和三明麻鸭归为一类;闽东地区的龙海金定鸭和莆田黑鸭聚为一类.不同生态区域内鸭群的遗传变异与地理位置存在一定的相关性.  相似文献

利用32个微卫星标记研究了5个福建省地方鸭品种(金定鸭、连城白鸭、莆田黑鸭、山麻鸭和番鸭)和2个对照品种(北京鸭和卡基康贝尔鸭)的遗传多样性和种质特性.结果表明:32个微卫星标记在7个鸭品种中检测到371个等位基因,平均每个基因座有11.719个等位基因,平均多态信息含量为0.522,有效等位基因数在5.141~6961,基因杂合度变化范围在0.512~0.700,杂合度较高;基于Nei氏标准遗传距离(DS)和DA遗传距离,采用UPGMA方法构建了系统发生树,将金定鸭、莆田黑鸭、连城白鸭、山麻鸭和卡基康贝尔鸭聚为一类,而北京鸭和番鸭各自聚为一类.所得聚类图与种群的真实关系较为符合.  相似文献

RAPD markers generated by mixtures of two different 10-mer primers were developed for eight different olive cultivars used as parental lines in olive-breeding programs. Two RAPD bands were converted into dominant SCAR markers by direct sequencing of the RAPD products, avoiding the costly and time-consuming cloning step. The SCARs generated have maintained the original RAPD polymorphism among the cultivars and segregated according to Mendelian inheritance. Preliminary results suggest the use of the SCAR SCOeMS-2 for the marker-assisted selection of the high flesh/stone ratio. This strategy provides a rapid method for the characterization of RAPD markers and for the development of PCR-based markers with applications in olive mapping, paternal testing and germplasm characterization. The use of these markers in multiplexed PCRs, and the direct ethidium bromide detection of the PCR products in the test tube, facilitate their efficient and reliable breeding applications. Up to now a single cytoplasmic male sterility (CMS) source, PET1, is used worldwide for hybrid breeding in sunflower. Introgression of the restorer gene Rf1, responsible for fertility restoration, into new breeding material requires tightly linked markers to perform an efficient marker-assisted selection. A survey of 520 decamer primers by bulked segregant analyses identified five RAPD markers linked to the restorer gene Rf1. In a F(2) population of 183 individuals one of the RAPD markers, OPK13_454, mapped 0.8 cM from Rf1, followed by OPY10_740 with 2 cM. Bulked segregant analyses using 48 AFLP primer combinations identified 17 polymorphisms, which could be mapped in the same linkage group as Rf1. E33M61_136, and E41M48_113 were mapped 0.3 cM and 1.6 cM from the gene, respectively. Conversion of E41M48_113 into a sequence-specific marker resulted in a monomorphic pattern. However, two of the RAPD markers, OPK13_454 and OPY10_740, were successfully converted into SCAR markers, HRG01 and HRG02, which are now available for marker-assisted selection. To investigate the utility of these SCAR markers in other cross-combinations they were tested in a set of 20 lines. Comparison of the patterns of 11 restorer and nine maintainer lines of PET1 demonstrated that the markers OPK13_454/HRG01 and HRG02 were absent in all maintainer lines but present in all restorer lines, apart from the high oleic line RHA348 and the dwarf line Gio55. In addition, restorer lines developed from the interspecific hybrids Helianthus annuus x Helianthus mollis and H. annuus x Helianthus rigidus gave the same characteristic amplification products.  相似文献
