The singlet excited state lifetime of the chlorophyll a (Chi a) in cytochrome b6f (Cyt b6f) complex was reported to be shorter than that of free Chl a in methanol, but the value was different for Cyt b6f complexes from different sources (~200 and ~600 ps are the two measured results). The present study demonstrated that the singiet excited state lifetime is associated with the detergents n-dodecyl-β-D-maltoside (DDM) and n-octyl-β-D-glucopyranoside (β-OG), but has nothing to do with the different sources of Cyt b6f complexes. Compared with the Cyt b6f dissolved in β-OG, the Cyt b6f in DDM had a lower fluorescence yield, a lower photodegradation rate of Chl a, and a shorter lifetime of Chl a excited state. In short, the singlet excited state lifetime, ~200 ps, of the Chl a in Cyt b6f complex in DDM is closer to the true in vivo.  相似文献

A study on energy transfer among chlorophylls(Chls)in the trimeric unit of the major light-harvesting complex Ⅱ(LHC Ⅱ)from Bryopsis corriculan,was carried out using time-correlated singlephoton counting.In the chlorophyll Q region of LHC Ⅱ,six molecules characterized as Chlb_(628),Chlb_(646),Chlb_(652)~(654,657),Chla_(664)~(666),Chla_(674)~(677.680)and Chla_(682)~(683) were discriminated according to their absorption spectrumand fluorescence emission spectrum.Then,excited by pulsed light of 628 nm,fluorescence kinetics spectrain the chlorophyll Q region were measured.In accordance with the principles of fluorescence kinetics,thesekinetics data were analyzed with a multi-exponential model.Time constants on energy transfer were obtained.An overwhelming percentage of energy transfer among chlorophylls undergoes a process longer than 97picoseconds(ps),which shows that,before transferring energy to another Chl,the excited Chl might convertenergy to vibrations of a lower state with different multiplicity(intersystem crossing).Energy transfer at thelevel of approximately 10 ps was also obtained,which was interpreted as the excited Chls may go throughinternal conversion before transferring energy to another Chl.Although with a higher standard deviation,timeconstants at the femtosecond level can not be entirely excluded,which can be attributed to the ultrafastprocess of direct energy transfer.Owing to the arrangement and direction of the dipole moment of Chls inLHC Ⅱ,the probability of these processes is different.The fluorescence lifetimes of Chlb_(652)~(654,657),Chla_(664)~(666),Chla_(674)~(677.680)and Chla_(682)~(683)were determined to be 1.44ns,1.43 ns,636 ps and 713 ps,respectively.Thepercentages of energy dissipation in the pathway of fluorescence emission were no more than 40% in thetrimeric unit of LHC Ⅱ.These results are important for a better understanding of the relationship between thestructure and function of LHC Ⅱ.  相似文献

据报道, 细胞色素b6f蛋白复合体(cytochrome b6f, Cyt b6f)中叶绿素a (chlorophyll a, Chl a)分子的单线激发态寿命(或荧光寿命)短于甲醇中游离Chl a的单线激发态寿命(~4 ns), 但是文献报道不同来源的Cyt b6f中Chl a单线激发态寿命并不一致(一种为200 ps左右, 一种结果为600 ps左右). 本研究证明, 不同来源Cyt b6f中Chl a的单线激发态寿命测定结果的不同, 与Cyt b6f来源无关, 而与溶解膜蛋白的去垢剂—八烷基葡萄糖苷 (n-Octyl β-D-glucopyranoside, β-OG)和十二烷基麦芽糖苷(n-Dodecylβ-D-maltoside, DDM)有关. 溶解在DDM中的样品和溶解在b-OG中的样品相比, 其Chl a的荧光产率低, 强光照射下抗光破坏的能力强, 单线激发态的寿命更短(~200 ps). 显然, 溶解在DDM中样品的Chl a单线激发态的寿命更接近体内真实的情况.  相似文献

An Isolated photosystem (PS) II reaction center (RC) with altered pigment content was obtained by chemical exchange of native chlorophyll a (Chl) with externally added Cu-Chl a (Cu-Chl). Pigment composition and spectroscopic properties of the RC exchanged with Cu-Chl were compared with native RC and RC treated with Chl In the same way. High-performance liquid chromatography analysis showed approximately 0.5 Cu-Chl per two pheophytln in the Cu-Chl-reconstltuted RC preparation. Insertion of Cu-Chl resulted in a decrease In absorption at 670 nm and an Increase at 660 nm, suggesting that the peripheral Chl may have been displaced. Fluorescence emission spectra of the Cu-Chl-reconstituted RC displayed a marked decrease In fluorescence yield and a blue shift of the band maximum, accompanied by the appearance of a broad peak at a shorter wavelength, Indicating that energy transfer In the modified RC was disturbed by Cu-Chl, a quencher of the excited state. However, there were few differences in the circular dichrolsm (CD) spectra, suggesting that the arrangement of pigments and proteins responsible for the CD signal was not significantly affected. In addition, no obvious change In peptlde components was found after the exchange procedure.  相似文献

The main light-harvesting chlorophyll a/b -protein complex (LHC Ⅱ) has been isolated directly from thylakoid membranes of shiphonous 8Teen alga, Bryopsis corticulans Setch. by using two consecutive runs of anion exchange and gel-filtration chromatography. Monomeric and trimeric subcomplexes of LHC Ⅱ were obtained by using sucrose gradient ultracentrifugation. Pigment analysis by reversed-phase high performance liquid chromatography showed that chlorophyll a (Chl a), chlorophyll b (Chl b), neoxanthin, violaxanthin and siphonaxanthin were involved in LHC Ⅱ from B. corticulans. The properties of electronictransition of monomeric LHC Ⅱ showed similarities to those of trimeric LHC Ⅱ. Circular dichroism spectroscopy showed that strong intramolecular interaction of excitonic dipoles between Chl a and between Chl b exist in one LHC Ⅱ apoprotein, while the intermolecular interaction of these dipoles can be intensified in the trimeric structure. The monomer has high efficient energy transfer from Chl b and siphonaxanthin to Chl a similarly to that of the trimer. Our results suggest that in B. corticulans, LHC Ⅱ monomer has high ordered pigment organization that play effective physiological function as the trimer, and thus it might be also a functional organization existing in thylakoid membrane of B.corticulans.  相似文献

Terahertz time-domain spectroscopy (THz-TDS) is a new technique in studying the conformational state of a molecule in recent years. In this work, we reported the first use of THz-TDS to examine the dena- turation of two photosynthesis membrane proteins: CP43 and CP47. THz-TDS was proven to be useful in discriminating the different conformational states of given proteins with similar structure and in monitoring the denaturation process of proteins. Upon treatment with guanidine hydrochloride (GuHCl), a 1.8 THz peak appeared for CP47 and free chlorophyll a (Chl a). This peak was deemed to originate from the interaction between Chl a and GuHCl molecules. The Chl a molecules in CP47 interacted with GuHCl more easily than those in CP43.  相似文献

The response of Prorocentrum donghaiense and Thalassiosira weissfiogii pigments under nitrate (N) and phosphate (P) limitation were studied using HPLC and in vivo fluorescence protocols in batch cultures. For P. donghaiense, the pigment ratio was kept stable under different nutrient conditions from the results of HPLC. For T. weissflogii, there was a lower ratio of chlorophyllide to Chl a during the exponential phase, but the reverse during the stationary phase. Different members of the phytoplankton had different pigments response mechanisms under nutrient limitation. From the results of in vivo fluorescence, the ratio of peridinin to Chl a for P. donghaiense increased in nutrient-free culture, while it was kept stable for nutrient-limited cultures during the exponential phase. For T. weissflogii, the ratio of fucoxanthin to Chl a for each culture increased during the exponential phase, but the ratio under N limitation was apparently lower than that for P limitation during the stationary phase. The results indicate that both pigment ratios from HPLC and in vivo fluorescence of To weissflogii were changed greatly under different nutrient conditions, which suggests that both ratios could be used as indicators of algal physiological status in different nutrient conditions.  相似文献

We previously demonstrated using noninvasive technologies that the interferon-gamma (IFN-γ) receptor complex is preassembled [ 1 ]. In this report we determined how the receptor complex is preassembled and how the ligand-mediated conformational changes occur. The interaction of Statl with IFN-γR1 results in a conformational change localized to IFN- γR1. Jakl but not Jak2 is required for the two chains of the IFN-γ receptor complex (IFN-γR1 and IFN-γR2) to interact; however, the presence of both Jakl and Jak2 is required to see any ligand-dependant conformational change. Two IFN- γR2 chains interact through species-specific determinants in their extracellular domains. Finally, these determinants also participate in the interaction of IFN-γR2 with IFN-γR1. These results agree with a detailed model of the IFN-γ receptor that requires the receptor chains to be pre-associated constitutively for the receptor to be active.  相似文献

Various abnormal phenomena have been observed during PCR so far. The present study performed a series of consecutive PCRs (including many rounds of re-amplification continuously) and found that the abortion of re-amplification was inevitable as long as a variety of complicated product appeared. The aborting stages varied, according to the lengths of targets. Longer targets reached the abortion earlier than the shorter ones, marked by appearance of the complex that was immobile in electropho-resis. Denatured gel-electrophoresis revealed that the complex was mainly made up of shorter or partially synthesized strands, together with small amounts of full-length ones. Able to be digested by S1 nuclease but unable by restriction endonucleases (REs), the complex was proved to consist of both single regions and double-helix regions that kept the complex stable thermodynamically. Simulations gave evidence that partial strands, even at lower concentration, could disturb re-amplification effec- tively and lead to the abortion of re-amplifications finally. It was pointed out that the partial strands formed chiefly via polymerase’s infidelity, and hence the solution to lighten the abnormality was also proposed.  相似文献

Carabids(Coleoptera:Carabidae)seem to be suitable bioindicalors of the environmental impacts of novel agrotechnologies,including deployment of the genetically engineered(GE)crops.In this article,we describe our effort to employ carabids in the environmental risk assessment(ERA).GE maize MON88017,its near-isogenic hybrid nontreated or treated with the soil insccticide chlorpyrifos,and two reference hybrids were used to compare three different ways how to utilize carabids in ERA.The analysis of abundance of all captured carabids or of the most abundant carabid specics did not disclose any differences between the treatments.The analysis based on the categories of functional traits revealed distinct fecatures of some treatments and proved suitable for ERA because it permitted field data transportability in spite of different species compositions.Our results indicate that GE maize has no detrimental environmental effecl,On the other hand,we found significant trends toward lower abundance and lower species number(including analysis of all carabid species together)in plots treated with the insecticide,and some tendencies to higher abundance and higher species number in plots sown with the reference hybrid PR38N86.Using functional group indicators alows identification of unintended changes in ecological functions of agroecosystem and comparability across geographics.We recommend data evaluation at the level of the categories of functional traits in ERA of GE crops and other agricultural practices.  相似文献

The first record of Ptyas nigromarginata(Blyth,1854) from Thailand and a new record from China are reported.Literature and internet sources were searched for previous records of this species to establish its geographic and altitudinal distributions in Asia.The distribution in India was adapted to the new state division of the northern part of the country.The results show that the species almost exclusively occurs in hill evergreen and montane forests of the Himalayan foothills and mountain ranges continuous with it,at altitudes of 1000–2300 m above sea level.  相似文献

Aims The increased atmospheric nitrogen (N) deposition due to human activity and climate change greatly causes grassland ecosystems shifting from being naturally N-limited to N-eutrophic or N-saturated, and further affecting the growth of grass species. The aims of this study are: 1) to evaluate the effects of different N addition levels on morphology and photosynthetic characteristics of Leymus chinensis; 2) to determine the critical N level to facilitate L. chinensis growth. Methods We conducted a different N addition levels experiment in dominant species in the temperate steppe of Nei Mongol. The aboveground biomass, morphological and leaf physiological traits, pigment contents, chlorophyll a fluorescence parameters and biochemical parameters of L. chinensis were investigated. Important findings Our results showed that aboveground biomass first increased and then decreased with the increased N, having the highest values at the 10 g N·m-2·a?1 treatment, but the 25 g N·m-2·a?1 still significantly increased the aboveground biomass relative to 0 g N·m-2·a?1. Leymus chinensis accommodate low N situation through allocating less N to carboxylation system and decreasing leaf mass per area (LMA) in order to get more light energy. Moderate N addition captured more light energy through increasing total chlorophyll (Chl) contents and decreasing the ratio of Chl a/b. Moderate N addition increased LMA, carboxylation efficiency, maximum car boxylation rate (Vcmax), maximum electron transport rate (Jmax) and decreased Jmax/Vcmax, thus allocating more N to carboxylation system to enhance carboxylation capability. Moreover, the photochemical activity of PSII was increased through higher effective quantum yield of PSII photochemistry, electron transport rate and photochemical quenching coefficient. Excessive N addition had negative effects on physiological variables of L. chinensis due to lower carboxylation capability and photochemical activity of PSII, further leading to decreased net photosynthetic rate, whereas increased non-photochemical quenching coefficient and carotenoids played the role in the dissipation of excess excitation energy. Overall, moderate N addition facilitated the photosynthetic characteristics of dominant species, but excessive N addition inhibited photosynthetic characteristics. The most appropriate N addition for the growth of L. chinensis was 5-10 g N·m-2·a?1 in the temperate steppe of Nei Mongol, China.  相似文献

Cas1 is a key component of the CRISPR adaptation complex, which captures and integrates foreign DNA into the CRISPR array,resulting in the generation of new spacers. We have determined crystal structures of Thermus thermophilus Cas1 involved in new spacer acquisition both in complex with branched DNA and in the free state. Cas1 forms an asymmetric dimer without DNA.Conversely, two asymmetrical dimers bound to two branched DNAs result in the formation of a DNA-mediated tetramer, dimer of structurally asymmetrical dimers, in which the two subunits markedly present different conformations. In the DNA binding complex, the N-terminal domain adopts different orientations with respect to the C-terminal domain in the two monomers that form the dimer. Substrate binding triggers a conformational change in the loop 164–177 segment. This loop is also involved in the 3′ fork arm and 5′ fork arm strand recognition in monomer A and B, respectively. This study provides important insights into the molecular mechanism of new spacer adaptation.  相似文献

Field trials were carried out from June to August in 2004 at Wuyishan (Wuyi Mountains), Fujian province, China, to determine the effects of habitat diversification on arthropod communities. Two Chinese cabbage, Brassica chinensis, field 1 (F1) and field 2 (F2) surrounded by diverse vegetable cultivars were selected, while a monoculture of Chinese cabbage served as the control field (CK). The results showed that: (i) when comparing insect abundance of each order between different habitats, significantly higher numbers of lepidopterous insects (39.76% from the each order) and lower densities of Hymenoptera (19.82%) were found in CK than in F1 and F2; (ii) compared with CK, F1 and F2 had a lower percentage of species richness and an abundance of herbivorous insects, but increased richness, abundance and biodiversity of predatory insects; (iii) no differences were observed in neutral insects' guild between different fields; and (iv) the dominant species for each guild depends on the habitat types and sampling dates. This study suggests that intercropping could conceivably be used in these habitats to increase the population of natural predators, thus achieving desirable and ecologically friendly results in vegetable fields.  相似文献

A lower extremity exoskeleton, SJTU-EX, is proposed, which mainly aims to help soldiers and workers to support a payload in motions. To solve the issues of the exoskeleton-environment and exoskeleton-human interactions, four types of foot contact are proposed based on their different kinematic characteristics. By the combination of single leg states, sixteen exo- skeleton states are presented using a series of meaningful notations from the topological point of view. The generalized function set (GF set) theory is employed to achieve the kinematic characteristics of the end effectors in different states. Moreover six mathematical formulations of the dynamics of the exoskeleton and its interactions with the human wearer are developed for different exoskeleton states. The applicability and potential of the proposed classification method are demonstrated by analyzing common lower limb motions, which are described concisely by a sequence of notations. Finally, a new concept of characteristic state is put forward to uniquely indicate the type of motion.  相似文献

Only β-NGF, the subunit of the 7S NGF complex, exhibits NGF activity, but the function ofthe zinc ion in native β-NGF has received little attention. Flameless atomic absorption spectroscopy (FAAS)measurements reveal that native β-NGF contains Zn(Ⅱ) with a Zn(Ⅱ)/β-NGF stoichiometry of 1 : 14.6.The presence of Zn(Ⅱ) in the native molecule results in significant changes of the secondary structure andlocal tertiary structure around Trp(s) with respect to those of apo β-NGF, as suggested by spectra offluorescence and circular dichrosim. Stopped-flow studies show that there are at least two steps during theinteraction of Zn(Ⅱ) with the apo form. In comparison with its apo form, the native β-NGF shows a higherability to trigger the proliferation of TF1 cells and mediate the survival of PC 12. Thus it is most likely that thestructural changes caused by the presence of Zn(II) directly lead to the increase in the biological activity of β-NGE All results indicate that Zn(Ⅱ) in native β-NGF plays an important role in the structure and thebiological activity of the protein.  相似文献

To investigate the biochemical response of freshwater green algae to elevated CO2 concentrations, Chlorella pyrenoidosa Chick and Chlamydomonas reinhardtii Dang cells were cultured at different CO2 concentrations within the range 3-186μmol/L and the biochemical composition, carbonic anhydrase (CA),and nitrate reductase activities of the cells were investigated. Chlorophylls (Chl), carotenoids, carbonhydrate,and protein contents were enhanced to varying extents with increasing CO2 concentration from 3-186μmol/L. The CO2 enrichment significantly increased the Chl a/Chl b ratio in ChloreUa pyrenoidosa, but not in Chlamydomonas reinhardtii. The CO2 concentration had significant effects on CA and nitrate reductase activity. Elevating CO2 concentration to 186μmol/L caused a decline in intracellular and extracellullar CA activity. Nitrate reductase activity, under either light or dark conditions, in C. reinhardtii and C. pyrenoidosa was also significantly decreased with CO2 enrichment. From this study, it can be concluded that CO2 enrichment can affect biochemical composition, CA, and nitrate reductase activity, and that the biochemical response was species dependent.  相似文献

In recent times,systemic coagulation,fibrinolysis,and cardio-pulmonary injury has been recognized in patients with COVID-19,the clinical disease state caused by infection of the novel coronavirus,SARS-CoV-2.While originally believed to be a primary lower respiratory infection,as more cases are identified,treated,and examined,hematologic complications are being identified as a significant driver of morbidity and mortality associated with the disease.Elevated D-dimer levels(>1μg/ml)on hospital admission have been identified as being associated with increased mortality[1],and levels greater than 2μg/ml predict fatal outcomes in patients[2].Alongside these results,there is also a greater risk of thrombotic events in COVID-19 patients,with risk increasing to as high as 31%[3],and pulmonary embolism risk increases proportionately alongside this[4].Whilst understanding of the pathophysiology is incomplete,there is clearly a component of coagulative disorder in these patients.D-dimer could prove a valuable tool to identify patients who are likely to have poorer outcomes and allow for prophylactic treatment and monitoring of secondary complications(Fig.1).  相似文献

Two-thirds of the coastal rivers and bays in the United States are degraded from nutrient pollution, and nitrogen inputs these waters continue to increase. The nitrogen comes from a variety of sources, including runoff from agricultural fields, concentrated animal feeding operations, atmospheric deposition from fossil fuel combustion, and sewage and septic wastes. Technical solutions for nitrogen pollution exist at reasonable cost. That most of these solutions have not yet been implemented to any significant extent across the United States suggests that new policy approaches are necessary. The best solution may involve a combination of voluntary and mandatory approaches, applying different approaches to different sources of nitrogen pollution. A watershed-based approach that relies heavily on voluntary mechanisms (such as crop-yield insurance to reduce over-fertilization) is likely to be the most effective for some sources of nitrogen (such as runoff from agricultural fields), while a uniform national regulatory approach may be better for others (such as NOx emissions from fossil fuel combustion). Implementation of management strategies should be carefully coupled to monitoring programs to assess the effectiveness of these strategies. While both nitrogen and phosphorus are important to control, the focus should be on nitrogen management, in part because nitrogen is more generally the causal agent of coastal eutrophication. Also, while nitrogen-control practices tend to also reduce phosphorus pollution, phosphorus-control practices often have little effect on nitrogen. Although current scientific and technical knowledge is sufficient to begin to make substantial progress toward solving coastal nitrogen pollution, progress will be made more quickly and more cost effectively with increased investment in appropriate scientific research.  相似文献
