Anthropogenic nitrogen (N) deposition is an expanding problem that affects the functioning and composition of forest ecosystems, particularly the decomposition of forest litters. Legumes play an important role in the nitrogen cycle of forest ecosystems. Two litter types were chosen from Zijin Mountain in China: Robinia pseudoacacia leaves from a leguminous forest (LF) and Liquidambar formosana leaves from a non-leguminous forest (NF). The litter samples were mixed into original forest soils and incubated in microcosms. Then, they were treated by five forms of N addition: NH4 +, NO3 ?, urea, glycine, and a mixture of all four. During a 6-month incubation period, litter mass losses, soil microbial biomass, soil pH, and enzyme activities were investigated. Results showed that mixed N and NO3 ?-N addition significantly accelerated the litter decomposition rates of LF leaves, while mixed N, glycine-N, and urea-N addition significantly accelerated the litter decomposition rates of NF leaves. Litter decomposition rates and soil enzyme activities under mixed N addition were higher than those under single form of N additions in the two forest types. Nitrogen addition had no significant effects on soil pH and soil microbial biomass. The results indicate that nitrogen addition may alter microbial allocation to extracellular enzyme production without affecting soil microbial biomass, and then affected litter decomposition process. The results further reveal that mixed N is a more important factor in controlling litter decomposition process than single form of N, and may seriously affect soil N cycle and the release of carbon stored belowground.

We compared soil moisture content, pH, total organic carbon (C org), total nitrogen (TN), total phosphorus (TP) and inorganic N (NH4 +–N, NO3 ?–N) concentrations, soil potential C and N mineralization rates, soil microbial biomass C (C mic), soil metabolic quotient (qCO2), soil microbial quotient (C mic/C org) and soil enzyme (urease and invertase) activities in semiarid sandy soils under three types of land cover: grassland, Mongolian pine (Pinus sylvestris var. mongolica) plantation, and elm (Ulmus punila)–grass savanna in southeastern Keerqin, in northeast China. Soil C org, TN and TP concentrations (0–10, 10–20, 20–40 and 40–60 cm) were lower while soil C/N and C/P ratios were higher in the plantation than in grassland and savanna. The effects of land cover change on NH4 +–N and NO3 ?–N concentrations, soil potential nitrification and C mineralization rates in the surface soil (0–10 cm) were dependent on sampling season; but soil potential N mineralization rates were not affected by land cover type and sampling season. The effects of land cover change on C mic and qCO2 of surface soil were not significant; but C mic/C org were significantly affected by land cover change and sampling season. We also found that land cover change, sampling season and land cover type?×?sampling season interaction significantly influenced soil enzyme (urease and invertase) activities. Usually soil enzyme activities were lower in the pine plantations than in grassland and savanna. Our results suggest that land cover change markedly influenced soil chemical and biological properties in sandy soils in the semiarid region, and these effects vary with sampling season.

Many vital ecosystem processes take place in the soils and are greatly affected by the increasing active nitrogen (N) deposition observed globally. Nitrogen deposition generally affects ecosystem processes through the changes in soil biochemical properties such as soil nutrient availability, microbial properties and enzyme activities. In order to evaluate the soil biochemical responses to elevated atmospheric N deposition in bamboo forest ecosystems, a two-year field N addition experiment in a hybrid bamboo (Bambusa pervariabilis × Dendrocalamopsis daii) plantation was conducted. Four levels of N treatment were applied: (1) control (CK, without N added), (2) low-nitrogen (LN, 50 kg N ha−1 year−1), (3) medium-nitrogen (MN, 150 kg N ha−1 year−1), and (4) high-nitrogen (HN, 300 kg N ha−1 year−1). Results indicated that N addition significantly increased the concentrations of NH4 +, NO3 , microbial biomass carbon, microbial biomass N, the rates of nitrification and denitrification; significantly decreased soil pH and the concentration of available phosphorus, and had no effect on the total organic carbon and total N concentration in the 0–20 cm soil depth. Nitrogen addition significantly stimulated activities of hydrolytic enzyme that acquiring N (urease) and phosphorus (acid phosphatase) and depressed the oxidative enzymes (phenol oxidase, peroxidase and catalase) activities. Results suggest that (1) this bamboo forest ecosystem is moving towards being limited by P or co-limited by P under elevated N deposition, (2) the expected progressive increases in N deposition may have a potential important effect on forest litter decomposition due to the interaction of inorganic N and oxidative enzyme activities, in such bamboo forests under high levels of ambient N deposition.

The effects of temperature on N mineralization were studied in two organic surface horizons (LF and H) of soil from a boreal forest. The soil was incubated at 5 °C and 15 °C after adding 15 N and gross N fluxes were calculated using a numerical simulation model. The model was calibrated on microbial C and N, basal respiration, and KCl-extractable NH4 +, NO3 , 15NH4 + and 15 NO3 . In the LF layer, increased temperature resulted in a faster turnover of all N pools. In both layers net N mineralization did not increase at elevated temperature because both gross NH4 + mineralization and NH4 + immobilization increased. In the H layer, however, both gross NH4 + mineralization and NH4 + immobilization were lower at 15 °C than at 5 °C and the model predicted a decrease in microbial turnover rate at higher temperature although measured microbial activity was higher. The decrease in gross N fluxes in spite of increased microbial activity in the H layer at elevated temperature may have been caused by uptake of organic N. The model predicted a decrease in pool size of labile organic matter and microbial biomass at elevated temperature whereas the amount of refractory organic matter increased. Temperature averaged microbial C/N ratio was 14.7 in the LF layer suggesting a fungi-dominated decomposer community whereas it was 7.3 in the H layer, probably due to predominance of bacteria. Respiration and microbial C were difficult to fit using the model if the microbial C/N ratio was kept constant with time. A separate 15N-enrichment study with the addition of glucose showed that glucose was metabolized faster in the LF than in the H layer. In both layers, decomposition of organic matter appeared to be limited by C availability. This revised version was published online in June 2006 with corrections to the Cover Date.

Soil microbial properties play a key role in belowground ecosystem functioning, but are not well understood in forest ecosystems under nitrogen (N) enrichment. In this study, soil samples from 0–10 cm and 10–20 cm layers were collected from a Dahurian larch (Larix gmelinii Rupr.) plantation in Northeast China after six consecutive years of N addition to examine changes in soil pH, nutrient concentrations, and microbial biomass and activities. Nitrogen addition significantly decreased soil pH and total phosphorus, but had little effect on soil total organic carbon (TOC) and total N (TN) concentrations. The NO 3 ? -N concentrations in the two soil layers under N addition were significantly higher than that in the control, while NH 4 + -N concentrations were not different. After six years of N addition, potential net N mineralization and nitrification rates were dramatically increased. Nitrogen addition decreased microbial biomass C (MBC) and N (MBN), and MBC/TOC and MBN/TN in the 0–10 cm soil layer, but MBC/MBN was increased by 67% in the 0–10 cm soil layer. Soil basal respiration, microbial metabolic quotient (qCO2), and β-glucosidase, urease, acid phosphomonoesterase and nitrate reductase activities in the two soil layers showed little change after six years of N addition. However, soil protease and dehydrogenase activities in the 0–10 cm layer were 41% and 54% lower in the N addition treatment than in the control, respectively. Collectively, our results suggest that in the mid-term N addition leads to a decline in soil quality in larch plantations, and that different soil enzymes show differentiated responses to N addition.

Wildfire in California annual grasslands is an important ecological disturbance and ecosystem control. Regional and global climate changes that affect aboveground biomass will alter fire-related nutrient loading and promote increased frequency and severity of fire in these systems. This can have long-term impacts on soil microbial dynamics and nutrient cycling, particularly in N-limited systems such as annual grasslands. We examined the effects of a low-severity fire on microbial biomass and specific microbial lipid biomarkers over 3?years following a fire at the Jasper Ridge Global Change Experiment. We also examined the impact of fire on the abundance of ammonia-oxidizing bacteria (AOB), specifically Nitrosospira Cluster 3a ammonia-oxidizers, and nitrification rates 9?months post-fire. Finally, we examined the interactive effects of fire and three other global change factors (N-deposition, precipitation and CO2) on plant biomass and soil microbial communities for three growing seasons after fire. Our results indicate that a low-severity fire is associated with earlier season primary productivity and higher soil-NH4 + concentrations in the first growing season following fire. Belowground productivity and total microbial biomass were not influenced by fire. Diagnostic microbial lipid biomarkers, including those for Gram-positive bacteria and Gram-negative bacteria, were reduced by fire 9- and 21-months post-fire, respectively. All effects of fire were indiscernible by 33-months post-fire, suggesting that above and belowground responses to fire do not persist in the long-term and that these grassland communities are resilient to fire disturbance. While N-deposition increased soil NH4 +, and thus available NH3, AOB abundance, nitrification rates and Cluster 3a AOB, similar increases in NH3 in the fire plots did not affect AOB or nitrification. We hypothesize that this difference in response to N-addition involves a mediation of P-limitation as a result of fire, possibly enhanced by increased plant competition and arbuscular mycorrhizal fungi–plant associations after fire.

As a major driving element of the structure and function of arid and semiarid ecosystems, rainfall is the essential factor limiting plant biological processes. To clarify the characteristics of transpiration and responses to summer rainfall, sap flow density (Fd) of Pinus tabulaeformis and Hippophae rhamnoides was monitored using thermal dissipation probes. In addition, midday leaf water potential (ψm) and leaf stomatal conductance (Gs) were also analyzed to determine water use strategies. The results indicated that the diurnal variation in the normalized Fd values exhibited a single‐peak curve for P. tabulaeformis, while H. rhamnoides showed multiple peaks. The normalized Fd for P. tabulaeformis remained relatively stable regardless of rainfall events. However, there was also a significant increase in the normalized Fd for H. rhamnoides in response to rainfall in June and August (< .05), although no significant differences were observed in July. The normalized Fd values for P. tabulaeformis and H. rhamnoides fitted well with the derived variable of transpiration, an integrated index calculated from the vapor pressure deficit and solar radiation (Rs), using an exponential saturation function. The differences in fitting coefficients suggested that H. rhamnoides showed more sensitivity to summer rainfall (< .01) than P. tabulaeformis. Furthermore, during the study period, P. tabulaeformis reduced Gs as soil water decreased, maintaining a relatively constant ψm; while H. rhamnoides allowed large fluctuations in ψm to maintain Gs. Therefore, P. tabulaeformis and H. rhamnoides should be considered isohydric and anisohydric species, respectively. And more consideration should be taken for H. rhamnoides in the afforestation activities and the local plantation management under the context of the frequently seasonal drought in the loess hilly region.

A 5-month laboratory incubation experiment was conducted to study the immobilization-mineralization of N in soil to which dried or composted 15N labelled ryegrass (Lolium italicum L.) had been added. Cellulose was added to dried ryegrass to give a C/N ratio similar to that of composted ryegrass. Exchangeable NH4 + and NO3 , HCl-hydrolyzable N forms, microbial biomass N, NaOH-soluble and insoluble N were monitored during incubation. Dried ryegrass brought about a significant increase in total and labelled exchangeable NH4 +, while a rapid immobilization and a subsequent slow release of exchangeable NH4 + was observed in soil with composted ryegrass, together with a resistance to degradation of the labelled humic substances. Compounds synthesized during the composting process and resistant to microbial decomposition probably caused an increase in the amino-acid fraction of soil. These findings suggest that composting can reduce the risk of N losses.

The pattern of natural restoration in soil components and processes was documented in five landslide-damaged (1–58-year-old) sites in the moist tropical sal (Shorea robusta) forest ecosystem of Nepal Himalaya. Comparisons were made with an undisturbed forest site in the same region. Concentrations of soil organic C, total N, total P and extractable nutrients (Ca, Mg and K) increased with the age of sites. The 58-year-old site showed concentrations of soil organic C, total N and total P that were 75–89% of concentrations in the undisturbed sal forest. The soil microbial biomass, the active fraction of soil organic matter, showed similar seasonal variations at all sites. The amount of mean microbial biomass (expressed as C, N and P contents) increased 4–5 times at the 58-year-old site relative to the 1-year-old site, and the bulk increase occurred within the initial 15 year. The increase in the C/N ratio of soil microbial biomass with age (9.4–11.6 years) reflected change in its composition. Although the net N-mineralization rate increased consistently until 58 years of age, the proportion of nitrification rate relative to ammonification rate distinctly decreased beyond 40 years. On the other hand, the soil available-N (both NO3 and NH4+) concentrations increased from 1 to 40 year and then declined; with age the proportion of NH4+ increased, however. Rates of restoration in soil properties were faster in the early successional stages (1–15 year) than late stages. Among different soil properties the restoration of soil microbial biomass (C and N) was faster than soil organic C and total N. Best fit power function models showed that the estimated times for the 58-year-old site to reach the level of the undisturbed, mature sal forest would be about 30–35 year for microbial biomass (C and N) and about 100–150 year for organic C and total N. Higher accumulation of soil microbial biomass and high N-mineralization rate at late successional stages indicated the re-establishment of enriched soil and restitution of nutrient cycling during the course of ecosystem restoration.

A field experiment was carried out at the Shenyang Experimental Station of Ecology (CAS) in order to study the effects of slow-release urea fertilizers high polymer-coated urea (SRU1), SRU1 mixed with dicyandiamide DCD (SRU2), and SRU1 mixed with calcium carbide CaC2 (SRU3) on urease activity, microbial biomass C and N, and nematode communities in an aquic brown soil during the maize growth period. The results demonstrated that the application of slow-release urea fertilizers inhibits soil urease activity and increases the soil NH4 +-N content. Soil available N increment could promote its immobilization by microorganisms. Determination of soil microbial biomass N indicated that a combined application of coated urea and nitrification inhibitors increased the soil active N pool. The population of predators/omnivores indicated that treatment with SRU2 could provide enough soil NH4 +-N to promote maize growth and increased the food resource for the soil fauna compared with the other treatments.

Nitrogen (N) is a crucial nutrient for soil biota, and its cycling is determined by the organic carbon decomposing process. Some endophytic fungi are latent saprotrophs that trigger their saprotrophic metabolism to promote litter organic matter cycling as soon as the host tissue senesces or dies. However, the effects of endophytic fungi on litter and soil N dynamics in vitro have rarely been investigated. In this study, we investigated N dynamics (total and mineral N) in both litter and soil in incubations of a pure culture of an endophytic fungus Phomopsis liquidambari with litter and following soil burial of the litter. Soil enzymes and microbial communities participating in the N transformations were also investigated. A pure culture of P. liquidambari released litter NH 4 + –N in the initial stages (10 days) of the incubation. However, following soil burial, the presence of both P. liquidambari and soil ammonia-oxidizing bacteria (AOB) resulted in an increase in soil NO 3 ? –N. These results indicate that the endophytic fungus P. liquidambari in vitro stimulates organic mineralization and promote NH 4 + –N release. Such effects triggered soil AOB-driven nitrification process.

Identifying soil microbial responses to anthropogenically driven environmental changes is critically important as concerns intensify over the potential degradation of ecosystem function. We assessed the effects of elevated atmospheric CO2 on microbial carbon (C) and nitrogen (N) cycling in Mojave Desert soils using extracellular enzyme activities (EEAs), community‐level physiological profiles (CLPPs), and gross N transformation rates. Soils were collected from unvegetated interspaces between plants and under the dominant shrub (Larrea tridentata) during the 2004–2005 growing season, an above‐average rainfall year. Because most measured variables responded strongly to soil water availability, all significant effects of soil water content were used as covariates to remove potential confounding effects of water availability on microbial responses to experimental treatment effects of cover type, CO2, and sampling date. Microbial C and N activities were lower in interspace soils compared with soils under Larrea, and responses to date and CO2 treatments were cover specific. Over the growing season, EEAs involved in cellulose (cellobiohydrolase) and orthophosphate (alkaline phosphatase) degradation decreased under ambient CO2, but increased under elevated CO2. Microbial C use and substrate use diversity in CLPPs decreased over time, and elevated CO2 positively affected both. Elevated CO2 also altered microbial C use patterns, suggesting changes in the quantity and/or quality of soil C inputs. In contrast, microbial biomass N was higher in interspace soils than soils under Larrea, and was lower in soils exposed to elevated CO2. Gross rates of NH4+ transformations increased over the growing season, and late‐season NH4+ fluxes were negatively affected by elevated CO2. Gross NO3 fluxes decreased over time, with early season interspace soils positively affected by elevated CO2. General increases in microbial activities under elevated CO2 are likely attributable to greater microbial biomass in interspace soils, and to increased microbial turnover rates and/or metabolic levels rather than pool size in soils under Larrea. Because soil water content and plant cover type dominates microbial C and N responses to CO2, the ability of desert landscapes to mitigate or intensify the impacts of global change will ultimately depend on how changes in precipitation and increasing atmospheric CO2 shift the spatial distribution of Mojave Desert plant communities.

A silt loam soil from Pakistan was incubated at 30°C with increasing levels (67, 133, 200, 267 and 333 μg N g?1 soil) of15N-labelled (NH4)2SO4 and glucose (C/N ratio of 30 for all additions). At a stage when all of the applied15N was immobilized (transformed into microbial biomass), moist soil samples were subjected to the chloroform fumigation-incubation method (CFIM) for determination of KN and microbial biomass. Mineralization of biomass derived from the applied15N and the native soil N was studied under anaerobic conditions. In situ values of KN varied from 0.19 to 0.42 and increased with increasing levels of amendment (N + glucose). From 10 to 18% of the native soil N was found as microbial biomass. Anaerobic incubation of the soils resulted in the mineralization (determined as NH 4 + ) of 15.08 to 29.23% of the biomass15N at different levels of amendment; 2.90 to 4.43% of the native soil N was mineralized. From 70 to 90% of the N mineralized from native soil N was derived from microbial biomass; the rest was attributed to non-biomass N.

We obtained soil samples from geographically diverse switchgrass (Panicum virgatum L.) and sorghum (Sorghum bicolor L.) crop sites and from nearby reference grasslands and compared their edaphic properties, microbial gene diversity and abundance, and active microbial biomass content. We hypothesized that soils under switchgrass, a perennial, would be more similar to reference grassland soils than sorghum, an annual crop. Sorghum crop soils had significantly higher NO3 ? -N, NH4 + -N, SO4 2? -S, and Cu levels than grassland soils. In contrast, few significant differences in soil chemistry were observed between switchgrass crop and grassland soils. Active bacterial biomass was significantly lower in sorghum soils than switchgrass soils. Using GeoChip 4.0 functional gene arrays, we observed that microbial gene diversity was significantly lower in sorghum soils than grassland soils. Gene diversity at sorghum locations was negatively correlated with NO3 ? -N, NH4 + -N, and SO4 2? -S in C and N cycling microbial gene categories. Microbial gene diversity at switchgrass sites varied among geographic locations, but crop and grassland sites tended to be similar. Microbial gene abundance did not differ between sorghum crop and grassland soils, but was generally lower in switchgrass crop soils compared to grassland soils. Our results suggest that switchgrass has fewer adverse impacts on microbial soil ecosystem services than cultivation of an annual biofuel crop such as sorghum. Multi-year, multi-disciplinary regional studies comparing these and additional annual and perennial biofuel crop and grassland soils are recommended to help define sustainable crop production and soil ecosystem service practices.

This study examines the effect of elevated CO2 on short-term partitioning of inorganic N between a grass and soil micro-organisms. 15N-labelled NH4+ was injected in the soil of mesocosms of Holcus lanatus (L.) that had been grown for more than 15 months at ambient or elevated CO2 in reconstituted grassland soil. After 48 h, the percentage recovery of added 15N was increased in soil microbial biomass N at elevated CO2, was unchanged in total plant N and was decreased in soil extractable N. However, plant N content and microbial biomass N were not significantly affected by elevated CO2. These results and literature data from plant–microbial 15N partitioning experiments at elevated CO2 suggest that the mechanisms controlling the effects of CO2 on short- vs. long-term N uptake and turnover differ. In particular, short-term immobilisation of added N by soil micro-organisms at elevated CO2 does not appear to lead to long-term increases in N in soil microbial biomass. In addition, the increased soil microbial C:N ratios that we observed at elevated CO2 suggest that long-term exposure to CO2 alters either the functioning or structure of these microbial communities.

Several boreal and alpine forests are depleted in nutrients due to acidification. Fertilization may be a remedy, but rapidly-soluble salts (N, P, K, Mg) may pose nitrate problems for the groundwater or decrease microbial activity. With the aim to investigate potential nitrogen leaching after fertilization we set up an experiment employing intact soil cores (11 cm diameter, 20–40 cm long) from a mixed forest and a Picea abies stand (soil type Rendsina) in the Northern Calcareous Alps of Austria. The cores were fertilized with a commercial NPK fertilizer or a methylene-urea-apatite-biotite (MuAB) fertilizer at a rate corresponding to 300 kg N ha-1 and incubated for 28 weeks together with unfertilized controls. Both soil water (retrieved 5 cm below the soil surface) and leachate were analyzed for nitrate and ammonium in regular intervals. After the incubation, soil microbial biomass and basal repiration were determined and a nitrogen mineralization assay was performed. For the control, in the soil water and leachate maximum NH4 + and NO3 - concentrations of 5 and 11 mg N L-1, respectively, were found. Compared to that, MuAB fertilizer resulted in a slow increase of NH4 + and NO3 - in the soil water (up to 11 and 35 mg N L-1 respectively) and in the leachate (4 mg NH4 +-N L-1 and 44 mg NO3 --N L-1). Highest nitrogen loads were found for the fast release NPK fertilizer, with NH4 + and NO3 - concentrations up to 170 and 270 mg N L-1, respectively, in the soil water. NH4 +-N levels in the leachate remained below 5, while NO3-N levels were up to 190 mg L-1. Fast- release NPK caused a significant decrease of microbial biomass and basal respiration. These parameters were not affected by MuAB fertilizer. The results suggest that the MuAB fertilizer may be an ecologically appropriate alternative to fast-release mineral fertilizers for improving forest soils.

Microbes drive global soil nitrogen mineralization and availability
Soil net nitrogen mineralization rate (Nmin), which is critical for soil nitrogen availability and plant growth, is thought to be primarily controlled by climate and soil physical and/or chemical properties. However, the role of microbes on regulating soil Nmin has not been evaluated on the global scale. By compiling 1565 observational data points of potential net Nmin from 198 published studies across terrestrial ecosystems, we found that Nmin significantly increased with soil microbial biomass, total nitrogen, and mean annual precipitation, but decreased with soil pH. The variation of Nmin was ascribed predominantly to soil microbial biomass on global and biome scales. Mean annual precipitation, soil pH, and total soil nitrogen significantly influenced Nmin through soil microbes. The structural equation models (SEM) showed that soil substrates were the main factors controlling Nmin when microbial biomass was excluded. Microbe became the primary driver when it was included in SEM analysis. SEM with soil microbial biomass improved the Nmin prediction by 19% in comparison with that devoid of soil microbial biomass. The changes in Nmin contributed the most to global soil NH4+‐N variations in contrast to climate and soil properties. This study reveals the complex interactions of climate, soil properties, and microbes on Nmin and highlights the importance of soil microbial biomass in determining Nmin and nitrogen availability across the globe. The findings necessitate accurate representation of microbes in Earth system models to better predict nitrogen cycle under global change.
