 应用四室隔网系统研究了菌丝桥在日本落叶松(Larix kaempferi)幼苗间传递磷的作用。结果表明,供体接种卷缘桩菇(Paxillus involutus)和彩色豆马勃(Pisolithus tinctorius)后,其外延菌丝可以穿过隔离层侵染受体落叶松,在供体和受体落叶松间形成了菌丝桥。供体植株接种菌根真菌后生物量明显增加,但是对受体植株没有显著的影响。菌根真菌侵染的供体和受体植株的根、地上部吸磷量均分别显著高于对照,而且供体植株根、地上部吸磷量增加的程度明显高于受体。被卷缘桩菇和彩色豆马勃侵染的受体植株体内32P的放射性强度分别是对照的10倍和6倍,两者形成菌丝桥后传递到受体植株的32P分别为供体植株体内32P的1.10%和0.22%。供体植株吸收的32P可以通过菌丝桥传递给受体,但是绝对数量十分有限,对受体植株磷营养没有产生显著的影响,但P. involutus和P. tinctorius侵染受体植株后,促进了受体落叶松对磷的吸收,这是菌丝桥形成后,真菌帮助受体植株吸收磷引起的。  相似文献

应用5室分隔法研究了供体三叶草体内的32P通过菌丝桥向受体黑麦草的传递作用。结果表明,菌根侵染供体三叶草根系之后,根外菌丝可穿过中室到达受体植株根室而再度侵染受体黑麦草的根系,从而形成三叶草-黑麦草根系之间的菌丝桥;供体三叶草体内的32P可通过根间菌丝桥传递给受体黑麦草,32P的传递量随受体植株施磷水平的提高而降低.  相似文献

为探究外生菌根真菌菌丝对马尾松Pinus massoniana幼苗氮素的吸收、传递及分配规律的影响,本研究应用15N示踪技术,采用四室隔网系统进行盆栽试验。研究表明:供体接种外生菌根真菌后,根外菌丝能够穿过粘土层和隔离层到达受体室,在供、受体植株之间形成菌丝桥。接种不同外生菌根真菌均显著增加供体植株苗高、地径及生物量(P<0.05),但对受体植株的影响有差异,其中接种橙黄硬皮马勃Scleroderma citrinum (Sc)对植株苗高、地径及生物量具有显著的促进作用(P<0.05)。接种对供、受体植株全氮含量无显著性的规律(P>0.05),只有接种东方豆马勃Pisolithus orientalis (Po)的植株全氮含量显著高于其他处理。接种能够显著提高供体植株根、茎、叶的吸氮量,且叶>根>茎,受体植株中,接种Po、Sc和褐环乳牛肝菌Suillus luteus (Sl)的植株总吸氮量较不接种(NE)显著增加(P<0.05),而接种混合菌(Sm)的植株与NE无显著差异(P>0.05)。接种Sl的植株15N传递率最高(75.86%),受体植株对15N吸收量最大(91.68 μg/pot),Po次之。供体植株中,除接种Sl的植株15N主要分配在叶外,其他处理的15N分配中心均在根部,在受体植株中,接种Po、Sl、Sm的植株15N主要分配在根部,而NE和接种Sc的植株15N主要分配在茎。综上,在马尾松供、受体苗木间能形成菌丝桥,接种外生菌根真菌能够促进马尾松供体植株苗高、地径的生长及生物量的积累,提高植株吸氮量,对于受体植株不同菌种的作用有差异,其中接种Sc的苗木表现最好。此外,不同的菌种菌丝对植株的全氮含量、15N吸收量、传递率及分配率的影响也不同,其中,以接种Po和Sl的苗木表现较好。  相似文献

应用五室方法研究了三叶草根间菌线桥传递衰亡根系中磷的作用。三叶草生长至10周切除供体地上部让根系衰亡,11周收收获样品进行分析测定。结果表明:菌线桥可以在植株间传递^32P,从而使受体三叶草地上部磷营养状况得到改善;供体植株地上部切除后有利于^32P通过菌丝桥从衰亡根系向受体植株的转移,表现为受体植株含磷量有所增加,但对植株的生长影响不大。  相似文献

菌根菌丝桥是植物间在地下进行物质交流的通道, 但它能否作为植物间地下化学通讯的通道来传递抗病信号则缺乏研究. 本文利用丛枝菌根真菌(AMF)摩西球囊霉在供体与受体番茄植株间建立菌丝桥, 对供体植株接种早疫病病原菌茄链格孢菌, 研究供体与受体番茄植株根系间是否存在抗病信号的传递. 荧光定量PCR检测表明, AMF侵染后的供体番茄植株再接种病原菌, 其根系中苯丙氨酸解氨酶基因(PAL)、脂氧合酶基因(LOX)和几丁质酶基因(PR3)的转录水平显著高于仅接种病原菌、未接种病原菌和AMF, 以及只接种AMF的番茄植株. 更重要的是, 与供体有菌丝桥连接的受体番茄根系中PAL、LOX和PR3的基因的表达量也显著高于无菌丝桥连接、菌丝桥连接被阻断以及有菌丝桥连接但供体植物未接种病原菌的处理,3个基因最高转录水平达到无菌丝桥连接对照受体植物的4.2、4.5和3.5倍. 此外, 供体植株根系启动防御反应的时间(18和65 h)比受体(100和140 h)早. 表明病原菌诱导番茄供体根系产生的抗病信号可以通过菌丝桥传递到受体根系.  相似文献

研究了不同外源磷水平条件下,接种丛枝菌根真菌根内球囊霉(Glomus intraradices)对寄主植物甜玉米菌根侵染率、地上部和地下部鲜重、氮磷含量、精氨酸含量影响。结果表明:丛枝菌根真菌能够很好的侵染于玉米植株根系。且不同磷水平条件下,菌根侵染率差异较显著。在低磷水平下,菌根侵染率较高。孢子数量随着磷水平提高而增加。菌丝室根外菌丝鲜重在P40时最高。菌根化的甜玉米生物量及氮磷含量显著高于对照组。此外,低磷水平促使甜玉米地上部和地下部鲜重显著提高。甜玉米地上部总氮和地下部总氮含量分别在P40、P80和P20、P40时最高。地上部总磷和地下部总磷含量分别在P80和P160时最高。菌根精氨酸含量在低磷(P20)时最高。研究表明接种丛枝菌根真菌可促进甜玉米幼苗生长并与外源磷水平有关。  相似文献

以豆科植物紫花苜蓿为试验材料,应用三室(供体室-间隔室-受体室)培养系统,研究在供体和受体紫花苜蓿根系之间菌丝网络形成的时间效应以及间隔室中不同植物对菌丝网络建成的介导作用.第一个试验在供体和受体植物生长8、10、12、14周之后进行收获以检验菌丝网络形成的时间效应;第二个试验则在间隔室分别种植紫花苜蓿、羊草和独行菜,以考察菌根依赖性不同的植物对菌丝网络形成的介导作用.试验结果显示:(1)接种丛枝菌根真菌的供体紫花苜蓿根系能够形成良好的菌根共生,其外延菌丝可穿过尼龙网和间隔室侵染受体植物根系;植物生长8周后,在受体植物根系检测到菌根侵染,证实供体和受体植物间形成了根间菌丝网络;10周后,尽管供体室和受体室植物的侵染率已无差异,但二者的生物量和地上部磷浓度差异却加大,表现出菌丝网络对植物种内竞争影响的不对称性.(2)试验条件下,不同介导植物对受体植物的菌根侵染及生物量均无明显影响,但显著降低了供体植物生物量和地上部磷浓度;间隔室无介导植物或种植独行菜时,受体植物地上部和根系生物量显著低于供体植物,而当介导植物为紫花苜蓿和羊草时,受体和供体植物生物量无显著差异.研究表明,植物根间菌丝网络的形成受时间和介导植物的影响,同时也具有调节植物间资源分配和植物相互作用的功能.  相似文献

【背景】西南桦是兼具内生、外生菌根的典型菌根营养型树种,菌根化育苗是其壮苗培育的有效措施。【目的】揭示外生菌根真菌对西南桦无性系幼苗生长和养分含量的影响,为其菌根化育苗筛选优良外生菌根真菌提供科学依据。【方法】以BY1、FB4、FB4+和A5等4个西南桦优良无性系为研究对象,选用土生空团菌(Cenococcumgeophilum)、松乳菇(Lactariusdeliciosus)、黄硬皮马勃(Scleroderma flavidum)、多根硬皮马勃(S. polyrhizum)、褐环乳牛肝菌(Suillus luteus)和红绒盖牛肝菌(Xerocomuschrysenteron)6个外生菌根真菌进行盆栽接种试验,分析接种处理间及无性系间苗高、地径、生物量以及养分含量差异。【结果】6个菌种均能与西南桦无性系幼苗形成外生菌根共生体,接种多根硬皮马勃与黄硬皮马勃显著促进了幼苗生长和养分吸收(P0.05),说明其与幼苗的亲和力明显优于其它菌种。尽管菌根侵染率在4个无性系之间无显著差异(P≥0.05),但各菌种对FB4、BY1幼苗生长的促进作用显著强于其它2个无性系。【结论】多根硬皮马勃和黄硬皮马勃可作为西南桦菌根化育苗的优选菌种。  相似文献

应用^32P同位素示踪法,人工接种VA菌根菌(Glomus mosseae),对种植在根箱中的杨树(Populus euramericana cv.‘I-214')和刺槐(Robinia pseudoacia)苗木进行了树种间P素养分转移途径的研究.结果表明,5室根箱中,接种VA菌根菌的杨树一例,菌根侵染率为34%,根箱隔网另一侧的刺槐根系侵染率为26%,而对照的杨树和刺槐根系侵染率均为零.在杨树一例施放射性同位累^32P,施用后第14—27天,刺槐一例的放射性同位素值,处理显著高于对照(P<0.05)、在根箱中的杨树和刺槐根系间观察到菌丝连接,表明人工接种VA菌根菌能在杨树和刺槐根系间产生菌丝桥,菌丝桥可以在杨树和刺槐根系间传递P素.养分转移定量分析表明,根系接触和根系分泌物是树种间P素转移的主要途径,其转移P素量占转移总量的62%;菌根菌等微生物活动及其与根系接触和根系分泌物两种途径的交互作用占38%;菌丝桥通过隔网发挥的作用仅表现为一种趋势.  相似文献

Ectomycorrhizal fungi constitute an important component of forest ecosystems that enhances plant nutrition and resistance against stresses. Diversity of ectomycorrhizal (EcM) fungi is, however, affected by host plant diversity and soil heterogeneity. This study provides information about the influence of host plants and soil resources on the diversity of ectomycorrhizal fungal fruiting bodies from rainforests of the Democratic Republic of the Congo. Based on the presence of fungal fruiting bodies, significant differences in the number of ectomycorrhizal fungi species existed between forest stand types (p < 0.001). The most ectomycorrhizal species‐rich forest was the Gilbertiodendron dewevrei‐dominated forest (61 species). Of all 93 species of ectomycorrhizal fungi, 19 demonstrated a significant indicator value for particular forest stand types. Of all analysed edaphic factors, the percentage of silt particles was the most important parameter influencing EcM fungi host plant tree distribution. Both host trees and edaphic factors strongly affected the distribution and diversity of EcM fungi. EcM fungi may have developed differently their ability to successfully colonise root systems in relation to the availability of nutrients.  相似文献

Eucalypt plantations in China have largely been established on soils that are low in phosphorus (P) and have few eucalypt-compatible ectomycorrhizal fungi. Effects of P application and ectomycorrhizal fungal inoculation on early tree growth in plantations of Eucalyptus urophylla Blake in Guangdong (Gaoyao) and E. globulus Labill. in Yunnan (Chuxiong) in southern China were investigated as part of a larger study. Application of superphosphate at establishment, in the presence of a basal fertiliser, increased early growth of E. urophylla and E. globulus. The optimum treatments for maximum stand volume at year 3 were 200 kg P ha–1 which increased stand volume by 750% on the strongly acidic, P-deficient lateritic red oxisol at Gaoyao, and 40 kg P ha–1 which increased stand volume by 55% on the mildly P-deficient red ultisol at Chuxiong, at 3 years. Superphosphate increased tree survival at Gaoyao as well as at Chuxiong. Nursery inoculation of eucalypt seedlings with ectomycorrhizal fungi significantly affected tree height and stand volume of the E. urophylla plantation, but the effect (positive or negative) was isolate-dependent and related to tree survival rate. A Laccaria isolate (CSIRO E4728) significantly increased stand volume by 27% at Gaoyao and a Scleroderma (MURU LH041) increased growth by 15% at Chuxiong at age 3 years. All isolates increased tree growth under P-limited soil conditions and only one isolate increased tree growth at marginal soil P. The results suggest that tree growth should be able to be optimised in plantations by the use of effective ectomycorrhizal fungi combined with a judicious fertilisation program at establishment.  相似文献

The trajectory of forests establishing on reclaimed oil sands mines in the Canadian boreal forest is uncertain. Soil microbes, namely mycorrhizal fungi, partly underlie successional trajectories of plant communities, yet their role in restoration is often overlooked. Here, we tested the relative importance of common management tools used in restoration—species planted and soil placement—on the recovery of ectomycorrhizal fungal communities over 4 years. Importantly, we further compared the community assembly of fungi on reclaimed landscapes to that in reference ecosystems disturbed to different degrees. This latter test addresses whether disturbance intensity is more important than common management interventions to restore fungal communities in these ecosystems. Three main findings emerged. (1) The effect of tree species planted and soil placement on ectomycorrhizal fungal communities establishing on reclaimed landscapes was dynamic through time. (2) Disturbances that remove or disrupt the organic layer of soils substantially affect the composition of ectomycorrhizal fungal communities. (3) Shifts in the community composition of ectomycorrhizal fungi were driven to a greater extent by disturbance severity than either tree species planted or soil placement.  相似文献

Fertilization typically reduces ectomycorrhizal diversity shortly after its application but less is known about its longer-term influence on fungal species. Long-term effects are important in forests where fertilizer is rarely applied. We compared fungal species composition in western hemlock control plots with plots last fertilized 7 years ago with nitrogen (N) or nitrogen plus phosphorus (N + P). The N + P fertilization had a significant lingering effect, increasing the tree size and foliar P content of the western hemlocks. From ectomycorrhizal roots of 24-year-old trees from northern Vancouver Island, Canada, we identified fungi from 12 samples per treatment, by amplifying, cloning, and sequencing fungal ribosomal DNA fragments, placing sequences with 97% or more identity in the same operational taxonomic unit (OTU). Diversity was high across treatments; we detected 77 fungal OTUs, 52 from ectomycorrhizal genera, among 922 clone sequences. The five most frequent OTUs were similar in abundance across treatments. Only 19 OTUs matched any of the 197 previously reported ectomycorrhizal species of western hemlock. Species composition but not diversity in nitrogen plus phosphorus plots differed significantly from control or nitrogen plots. Two Cortinarius OTUs were indicator species for nitrogen plus phosphorus plots and presence of Cortinarius cinnamomeus was correlated with control or nitrogen plots. After 7 years, fertilization history had made no detectable difference in ectomycorrhizal fungal diversity, but long-lasting changes in environment resulting from fertilization had a lingering effect on fungal ectomycorrhizal species composition. 晚生型外生菌根真菌通常出现在森林演替的后期,是成熟林中的优势外生菌根真菌类群。对四川都江堰一片亚热带针阔混交林中的菌根真菌地上群落进行调查,并应用二元逻辑回归分析对晚生型外生菌根真菌的主要类群,即鹅膏菌科、牛肝菌科和红菇科,与周围(5m×5m样方)树种组成的关系进行研究。还应用次级变量分析方法对主要外生菌根真菌类群的空间格局进行了分析。结果表明,非外生菌根树种及某些外生菌根树种对特定类群菌根真菌子实体的出现有抑制作用,而不同类群外生菌根真菌在克隆生长上的差异并不是子实体空间分布的决定因素。我们认为,当研究自然林中外生菌根子实体的空间分布时,除了宿主植物的分布,也应考虑非宿主植物的分布以及菌根真菌相互作用的影响。  相似文献

简要回顾了一百多年来外生菌根研究史;综述了外生菌根资源调查。分类鉴定,生态,生理,分子生物学以及生物技术研究与应用诸方面的阶段性成果和进展,通过对外生菌根研究与应用现状的归纳与分析,就我国菌根研究与应用等方面存在的主要问题提出了以下4点建议;1,加强菌根资源的系统调查;确认对应植物属种,进行编目与建立数据库以及菌根DNA文库。2,采集与收集菌根真菌,进行分离,培养,繁育与保藏,建立菌根真菌菌种库和DNA文库,3,加强菌根技术研究,侧重研究造林树种菌根化育苗和造林的新技术,新工艺,4,改革传统的育苗与造林的方式,方法,选育对应的优良菌根真菌菌株应用于育苗造林;制定相关的政策,法规以规范和推广菌根技术。  相似文献

Partitioning of soil phosphorus (P) pools has been proposed as a key mechanism maintaining plant diversity, but experimental support is lacking. Here, we provided different chemical forms of P to 15 tree species with contrasting root symbiotic relationships to investigate plant P acquisition in both tropical and subtropical forests. Both ectomycorrhizal (ECM) and arbuscular mycorrhizal (AM) trees responded positively to addition of inorganic P, but strikingly, ECM trees acquired more P from a complex organic form (phytic acid). Most ECM tree species and all AM tree species also showed some capacity to take up simple organic P (monophosphate). Mycorrhizal colonisation was negatively correlated with soil extractable P concentration, suggesting that mycorrhizal fungi may regulate organic P acquisition among tree species. Our results support the hypothesis that ECM and AM plants partition soil P sources, which may play an ecologically important role in promoting species coexistence in tropical and subtropical forests.  相似文献
