Age-associated changes in different bandwidths of the human electroencephalographic (EEG) spectrum are well documented, but their functional significance is poorly understood. This spectrum seems to represent summation of simultaneous influences of several sleep–wake regulatory processes. Scoring of its orthogonal (uncorrelated) principal components can help in separation of the brain signatures of these processes. In particular, the opposite age-associated changes were documented for scores on the two largest (1st and 2nd) principal components of the sleep EEG spectrum. A decrease of the first score and an increase of the second score can reflect, respectively, the weakening of the sleep drive and disinhibition of the opposing wake drive with age. In order to support the suggestion of age-associated disinhibition of the wake drive from the antagonistic influence of the sleep drive, we analyzed principal component scores of the resting EEG spectra obtained in sleep deprivation experiments with 81 healthy young adults aged between 19 and 26 and 40 healthy older adults aged between 45 and 66 years. At the second day of the sleep deprivation experiments, frontal scores on the 1st principal component of the EEG spectrum demonstrated an age-associated reduction of response to eyes closed relaxation. Scores on the 2nd principal component were either initially increased during wakefulness or less responsive to such sleep-provoking conditions (frontal and occipital scores, respectively). These results are in line with the suggestion of disinhibition of the wake drive with age. They provide an explanation of why older adults are less vulnerable to sleep deprivation than young adults.

Although circadian and sleep research has made extraordinary progress in the recent years, one remaining challenge is the objective quantification of sleepiness in individuals suffering from sleep deprivation, sleep restriction, and excessive somnolence. The major goal of the present study was to apply principal component analysis to the wake electroencephalographic (EEG) spectrum in order to establish an objective measure of sleepiness. The present analysis was led by the hypothesis that in sleep-deprived individuals, the time course of self-rated sleepiness correlates with the time course score on the 2nd principal component of the EEG spectrum. The resting EEG of 15 young subjects was recorded at 2-h intervals for 32-50 h. Principal component analysis was performed on the sets of 16 single-Hz log-transformed EEG powers (1-16 Hz frequency range). The time course of self-perceived sleepiness correlated strongly with the time course of the 2nd principal component score, irrespective of derivation (frontal or occipital) and of analyzed section of the 7-min EEG record (2-min section with eyes open or any of the five 1-min sections with eyes closed). This result indicates the possibility of deriving an objective index of physiological sleepiness by applying principal component analysis to the wake EEG spectrum.

Sleep-wake regulation involves reciprocal interactions between sleep- and wake-promoting processes that inhibit one another. To uncover the signatures of the opponent processes underlying ultradian sleep cycles, principal component analysis was performed on the sets of 16 single-Hz log-transformed electroencephalographic (EEG) power densities (1-16?Hz frequency range). Data were collected during unrestricted night sleep followed by 9 20-min naps (14 women aged 17-55 yrs) and during 12 20-min naps after either restriction or deprivation of sleep (9 males and 9 males, respectively, aged 18-22 yrs). It was found that any subset of power spectra could be reduced to the invariant four-principal component structure. The time courses of scores on these four components might be interpreted as the spectral EEG markers of the kinetics of two pairs of opponent chronoregulatory processes. In a sequence of ultradian sleep cycles, the 1st and 2nd components represent the alternations between competing drives for sleep and wakefulness, respectively, whereas the 3rd and 4th components reflect the alternations between light and deep sleep, respectively. The results suggest that principal component structuring of EEG spectrum can be employed for derivation of the parameters of the quantitative models conceptualizing the three major aspects of sleep-wake regulation—homeostatic, circadian, and ultradian processes.

Sleeping brain activity reflects brain anatomy and physiology. The aim of this study was to use high density (256 channel) electroencephalography (EEG) during sleep to characterize topographic changes in sleep EEG power across normal aging, with high spatial resolution. Sleep was evaluated in 92 healthy adults aged 18–65 years old using full polysomnography and high density EEG. After artifact removal, spectral power density was calculated for standard frequency bands for all channels, averaged across the NREM periods of the first 3 sleep cycles. To quantify topographic changes with age, maps were generated of the Pearson’s coefficient of the correlation between power and age at each electrode. Significant correlations were determined by statistical non-parametric mapping. Absolute slow wave power declined significantly with increasing age across the entire scalp, whereas declines in theta and sigma power were significant only in frontal regions. Power in fast spindle frequencies declined significantly with increasing age frontally, whereas absolute power of slow spindle frequencies showed no significant change with age. When EEG power was normalized across the scalp, a left centro-parietal region showed significantly less age-related decline in power than the rest of the scalp. This partial preservation was particularly significant in the slow wave and sigma bands. The effect of age on sleep EEG varies substantially by region and frequency band. This non-uniformity should inform the design of future investigations of aging and sleep. This study provides normative data on the effect of age on sleep EEG topography, and provides a basis from which to explore the mechanisms of normal aging as well as neurodegenerative disorders for which age is a risk factor.

A generalized subspace approach is proposed for single channel brain evoked potential (EP) extraction from background electroencephalogram (EEG) signal. The method realizes the optimum estimate of EP signal from the observable noisy signal. The underlying principle is to project the signal and noise into signal and noise coefficient subspace respectively by applying projection matrix at first. Secondly, coefficient weighting matrix is achieved based on the autocorrelation matrices of the noise and the noisy signal. With the coefficient weighting matrix, we can remove the noise projection coefficients and estimate the signal ones. EP signal is then obtained by averaging the signals estimated with the reconstruction matrix. Given different signal-to-noise ratio (SNR) conditions, the algorithm can estimate the EP signal with only two sweeps observable noisy signals. Our approach is shown to have excellent capability of estimating EP signal even in poor SNR conditions. The interference of spontaneous EEG has been eliminated with significantly improved SNR. The simulation results have demonstrated the effectiveness and superior performance of the proposed method.

Different biological signals are recorded in sleep labs during sleep for the diagnosis and treatment of human sleep problems. Classification of sleep stages with electroencephalography (EEG) is preferred to other biological signals due to its advantages such as providing clinical information, cost-effectiveness, comfort, and ease of use. The evaluation of EEG signals taken during sleep by clinicians is a tiring, time-consuming, and error-prone method. Therefore, it is clinically mandatory to determine sleep stages by using software-supported systems. Like all classification problems, the accuracy rate is used to compare the performance of studies in this domain, but this metric can be accurate when the number of observations is equal in classes. However, since there is not an equal number of observations in sleep stages, this metric is insufficient in the evaluation of such systems. For this purpose, in recent years, Cohen’s kappa coefficient and even the sensitivity of NREM1 have been used for comparing the performance of these systems. Still, none of them examine the system from all dimensions. Therefore, in this study, two new metrics based on the polygon area metric, called the normalized area of sensitivity polygon and normalized area of the general polygon, are proposed for the performance evaluation of sleep staging systems. In addition, a new sleep staging system is introduced using the applications offered by the MATLAB program. The existing systems discussed in the literature were examined with the proposed metrics, and the best systems were compared with the proposed sleep staging system. According to the results, the proposed system excels in comparison with the most advanced machine learning methods. The single-channel method introduced based on the proposed metrics can be used for robust and reliable sleep stage classification from all dimensions required for real-time applications.Electronic supplementary materialThe online version of this article (10.1007/s11571-020-09641-2) contains supplementary material, which is available to authorized users.
