用吸附法固定化培养紫草细胞   总被引:4,自引:0,他引:4  
采用生物活性载体 ,通过吸附固定化方式 ,结合液体石蜡原位萃取技术 ,培养紫草细胞。测定了细胞生长、底物消耗和产物合成的动力学 ,紫草宁产率为 0 .916 g/g干重细胞和 0 .95 3g/g干重接种细胞 ,分别为悬浮培养的 12 .7倍和 6 .3倍。同时 ,对吸附与包埋固定化方法进行了综合比较 ,探讨了吸附固定化方法的应用前景。  相似文献

利用大孔吸附树脂提取蜀葵花色素的研究   总被引:15,自引:0,他引:15  
研究利用大孔树脂吸附和分离蜀葵(Althaearosea(L.)Cavan)花红色素,比较了D-072、D-401、D-301-G、D-101、NKA-9、D-290、D-110七种树脂对该色素的静态吸附情况以及不同极性解吸剂对吸附色素的树脂洗脱的效果,从中选择出吸附和解吸效果最佳的树脂以及较适的解吸剂。结果表明:用D-401大孔吸附树脂作吸附剂,色素吸附率达91%;解吸剂用含0.1%HCl的60%酸化乙醇,色素可被充分洗脱下来,解吸效果较好;树脂通过回收再生后可重复利用。大孔吸附树脂法精制蜀葵花色素工艺相对简单,原料、试剂利用率较高。  相似文献

通过静态吸附解析实验,确定了利用大孔吸附树脂初步纯化唐古特白刺红色素的条件,并根据所得条件做了扩大实验。结果表明.X-5树脂的吸附能力最好,吸附率为76.85%,pH在1—3范围内对吸附效果无影响;较高的乙醇浓度和pH值有利于色素的解析,解析液以80%酸化乙醇(含1%HCI)效果最佳,解析时间为40min;扩大实验得到紫红色色素粉末,得率为1.24%,水溶性好,不含糖类、蛋白质等水溶性杂质;树脂通过回收再生可重复利用。本实验为色素产品的开发及白刺果实的综合利用提供了理论依据。  相似文献

本文分析测定了羽粉水解液在颗粒活性炭柱上层析脱色时柱前柱后液中的氨基酸含量。结果表明,颗粒活性炭在层析吸附羽粉水解液色素时对各种氨基酸都有不同程序的吸附,吸附的平均量为26.50%,尤其是对芳香族氨基酸,吸附量几乎为100%。  相似文献

通过考察真菌诱导子与吸附树脂对新疆紫草毛状根中萘醌积累的影响,获得真菌诱导子与吸附树脂对萘醌类物质积累的最佳处理,为规模化生产提供依据.以新疆紫草毛状根为试验材料,将黑曲霉、米曲霉诱导子及其混合诱导子、大孔吸附树脂添加到M-9培养基中,采用分光光度法测定毛状根总萘醌含量.试验结果表明:在毛状根培养10d时以2.5∶50的比例添加混合诱导子,总萘醌含量是对照的2.28倍;在此结果基础上,在培养第0天添加大孔吸附树脂NKA-9,总萘醌含量最高是对照的3.71倍;黑曲霉诱导子与米曲霉诱导子有协同效应;在生物反应器中添加混合诱导子及大孔吸附树脂NKA-9,其总萘醌含量是对照的4.17倍.米曲霉诱导子、混合诱导子对毛状根增殖有促进作用;同时添加大孔吸附树脂NKA-9及混合诱导子能提高毛状根总萘醌含量.生物反应器培养毛状根为今后利用新疆紫草毛状根规模化生产总萘醌提供了理论参考.  相似文献

研究报道了X-5树脂吸附和分离萝卜红色素的方法。试验结果表明:在pH3.0-5.0时,树脂X-5时萝卜红色素具有较强的吸附能力和吸附选择性,其吸附容量为82.6mg/g树脂,吸附平衡时间30min,X-5树脂对产生萝卜异味的萝卜苷几乎不吸附,同时用50%的乙醇溶液洗脱,可获得90.8%以上的萝卜红色素。用该工艺生产的色素产品为深红色粉状,色价比国家标准提高了13倍。  相似文献

目的:研究非活性深红酵母(Rhodotorula rubra)对重金属离子Pb^2+的生物吸附热力学和动力学特性。方法:采用恒温摇床振荡吸附的实验方法,研究Pb^2+生物吸附的动力学和热力学,并以适当的数学模型对实验数据进行拟合;对吸附前后的酵母进行红外光谱及X射线光电子能谱分析。结果:在20℃~45℃温度范围内,吸附5min时即达到了饱和吸附量的80%以上,2h左右达到平衡;深红酵母对Pb^2+的生物吸附过程适宜用Elovich方程来描述;由二级动力学方程计算的生物吸附活化能为21.56kJ/mol;生物吸附平衡可用Langmuir等温式、Freundlich等温式及Dubinin-Radushkevich等温式来描述,拟合相关系数均接近0.99;Langmuir方程计算所得ΔH^0为13.93kJ/mol。结论:深红酵母对Pb^2+的生物吸附是非均相的扩散过程,由快速吸附和慢速吸附两个阶段组成,以物理吸附为主,并伴随有化学吸附。  相似文献

桑椹红色素是一种安伞、无毒的食用天然色素。本实验埘8种大孔吸附树脂进行筛选,然后通过单因素试验、正交试验和方差分析确定了吸附桑椹红色素的最佳操作条件。结果表明,LSA-21树脂对桑椹红色素的吸附和解吸性能较好;确定的最佳吸附条件为:料液浓度(以吸光度计)为0.425Abs,上柱速度3.5BV/h,料液pH1.57。  相似文献

本文研究了螺旋藻、栅列藻和织线藻的水提物对新疆紫草和硬紫草细胞生长和色素形成的作用。结果表明不同种类的藻的提取物对不同紫草细胞作用呈现差异。在生长阶段,对新疆紫草,上述3种藻的低浓度提取物促进生长但作用不大,高浓度的螺旋藻和栅列藻提取物强烈抑制生长;对硬紫草,各种藻提取液的所有浓度处理均有促生长作用。在色素形成阶段,连续用高浓度织线藻提取物处理可以加速新疆紫草色素形成,同时提高色素含量。低浓度的织线藻提取液处理能提高硬紫草色素含量。栅列藻的水提物对两种紫草的色素形成均起抑制作用。螺旋藻水提物的适当浓度可加速两种紫草的色素形成。B_5培养基中加高浓度织线藻水提物,可抑制新疆紫草生长阶段的色素形成。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

传染性法氏囊病(Infection bursal disease, IBD)是由鸡传染性法氏囊病毒(Infectious bursal disease virus, IBDV)引起的鸡和火鸡的一种高度接触性传染病,给世界各国的禽养殖业带来了巨大损失.自IBDV发现至今新的变异株不断出现,分子结构的改变导致病毒致病力的改变及宿主对疫苗应答的改变,使得传统的疫苗已不能控制其流行,因此各国学者对其基因组结构和功能进行了广泛深入的研究,并积极研制新型有效的疫苗以达到防治的目的.  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献
