脂肪干细胞(ADSCs)来源于脂肪,是一种多能性成体干细胞,它具有许多其他间充质干细胞所不具备的优点如取材丰富、增殖活性高和免疫原性低等。在不同的信号刺激下,ADSCs能分化成多种不同的体细胞,其多向分化的潜能已被广泛应用于各种组织再生的研究中,已成为再生医学中优秀的种子细胞。本文就近年来ADSCs在伤口愈合、骨骼重建、肝脏再生、心肌修复及中枢神经系统修复与再生中应用的研究进展做一综述,并阐述了应用ADSCs治疗疾病时潜在的风险和安全问题。  相似文献

干细胞是目前生命科学研究的热点方向。干细胞具有自我更新及定向分化的潜在能力。近年来,干细胞移植治疗在治疗压力性尿失禁和膀胱损伤方面已成为研究重点,不同来源的干细胞在治疗膀胱损伤已取得瞩目的研究成果。干细胞对阴茎勃起神经和海绵体血管内皮细胞起着修复保护作用。干细胞具有向多种谱系细胞转化的能力来治疗压力性尿失禁。干细胞移植为泌尿系统的神经肌肉疾病的修复重建提供了一条新途径,使认为不可修复的的神经肌肉疾病的结构修复和组织重建成为可能。干细胞包括脂肪干细胞(adipose-derived stem cells,ADSCs)、骨髓间充质干细胞(bone narrow mesenchymal stem,BMSCs)和肌源性干细胞(muscle-derived stem cell,MDSCs)等。组织工程学是一类交叉学科,主要包括综合细胞培养、材料构建和细胞种植等。组织工程技术为泌尿外科临床医师提供了一条修复乃至重建受损脏器的新思路。本文就利用干细胞作为种子细胞,对膀胱缺损、压力性尿失禁、勃起功能障碍泌尿系疾病的组织工程修复进行综述。  相似文献

再生医学近年来受到越来越多的重视。它开启了治疗由于老化,损伤及一些先天性缺陷所造成的缺损畸形的新途径。其临床应用已涉及到各种组织的修复,包括血液,皮肤,角膜,软骨和骨等。在口腔领域,目前治疗牙缺失主要依靠修复体,种植体和牙移植。然而这些方法都存在一定的缺陷。而通过再生医学的原理和方法实现牙再生治疗可以为机体提供有生命的,有功能的,相容性好的组织结构。种子细胞是牙再生的基础与关键。在牙再生研究中,牙髓间充质干细胞,牙乳头细胞,牙周膜间充质细胞,牙囊细胞及牙源性上皮细胞等牙源性干细胞常通过诱导分化为成釉细胞或成牙本质细胞来作为种子细胞应用,在临床上却难以获取,近来研究也有用骨髓间充质干细胞或脂肪间充质干细胞细胞等非牙源性干细胞者,但其牙向分化能力及分化调控机制还不明确。跻带间充质干细胞在新近的研究中较其它非牙源性干细胞表现出更大的优势,脐带间充质干细胞更原始、具有更高可塑性、更大扩增分化潜能。在此,本文就脐带间充质干细胞向牙细胞系分化的可能性做一论述,并对其可能实现的牙向分化给出可能的方法和策略,为牙再生种子细胞的选取提供新的思路。  相似文献

组织干细胞是成体组织中存在的一类尚未分化、能自我更新和增殖的特殊细胞群,具有分化为多种组织细胞的潜能.一般处于休眠状态,在组织损伤修复和维持组织的动态平衡中发挥重要的作用.体育运动、激素、生长因子和药物能激活内源性的组织干细胞,促进组织再生或伤口修复.利用内源性修复机制刺激组织再生,一直是生物医学领域的梦想.最新的研究表明,这个梦想现在可能成为现实.本文简要介绍了组织干细胞的内源性修复、内源性修复的生理学机制、招募内源性干细胞的主要方法及目前采用招募内源性干细胞修复和再生组织这一策略尚需克服的困难和临床应用前景.  相似文献

再生医学的目的是为了减轻组织损伤所致的不可逆性功能损害,多种类型的干细胞可促进神经再生,发挥治疗作用。近来,在骨髓和其他组织中发现了一种数量极少的极小胚胎样干细胞(VSELs),其分子标志为Oct-4+CXCR4+SSEA-1+Sca-1+lin-CD45-,它们可动员到外周血中。在给予动员剂或组织损伤等应激情况下可向损伤区迁移,现在认为它是高度迁移的外胚层/生殖系源性的干细胞群,具有多能干细胞特征,能分化为三个胚层细胞,综上所述,极小胚胎样干细胞可能通过促进神经再生修复中枢神经系统损伤。  相似文献

间充质干细胞因具有易于分离培养获得、低免疫源性、多向分化潜能及旁分泌功能等特征,而被越来越多地用于组织再生与创伤修复研究,并显现了广阔的应用前景。间充质干细胞治疗皮肤创伤的研究已开展了十余年,无论动物模型或是临床病人,均显示了不错的疗效。然而,该治疗仍面临很多问题和挑战,离转化为临床常规治疗手段尚有距离。该文拟对间充质干细胞在皮肤创伤修复中的作用及其机制和提高疗效的手段等方面作一综述,并探讨面临的问题和未来的研究方向。  相似文献

Adipose‐derived stem cells (ASCs) have been discovered for more than a decade. Due to the large numbers of cells that can be harvested with relatively little donor morbidity, they are considered to be an attractive alternative to bone marrow derived mesenchymal stem cells. Consequently, isolation and differentiation of ASCs draw great attention in the research of tissue engineering and regenerative medicine. Cartilage defects cause big therapeutic problems because of their low self‐repair capacity. Application of ASCs in cartilage regeneration gives hope to treat cartilage defects with autologous stem cells. In recent years, a lot of studies have been performed to test the possibility of using ASCs to re‐construct damaged cartilage tissue. In this article, we have reviewed the most up‐to‐date articles utilizing ASCs for cartilage regeneration in basic and translational research. Our topic covers differentiation of adipose tissue derived mesenchymal stem cells into chondrocytes, increased cartilage formation by co‐culture of ASCs with chondrocytes and enhancing chondrogenic differentiation of ASCs by gene manipulation. Bone tissue engineering represents one of the most challenging emergent fields for scientists and clinicians.Current failures of autografts and allografts in many pathological conditions have prompted researchers to find new biomaterials able to promote bone repair or regeneration with specific characteristics of biocompatibility,biodegradability and osteoinductivity.Recent advancements for tissue regeneration in bone defects have occurred by following the diamond concept and combining the use of growth factors and mesenchymal stem cells(MSCs).In particular,a more abundant and easily accessible source of MSCs was recently discovered in adipose tissue.These adipose stem cells(ASCs)can be obtained in large quantities with little donor site morbidity or patient discomfort,in contrast to the invasive and painful isolation of bone marrow MSCs.The osteogenic potential of ASCs on scaffolds has been examined in cell cultures and animal models,with only a few cases reporting the use of ASCs for successful reconstruction or accelerated healing of defects of the skull and jaw in patients.Although these reports extend our limited knowledge concerning the use of ASCs for osseous tissue repair and regeneration,the lack of standardization in applied techniques makes the comparison between studies difficult.Additional clinical trials are needed to assess ASC therapy and address potential ethical and safety concerns,which must be resolved to permit application in regenerative medicine.  相似文献

The recent identification of a mesenchymal stem cell population in adipose tissue has led to an abundance of research focused on the regenerative properties of these cells. As such, adipose‐derived stem cells (ASCs) and potential therapies in craniofacial regeneration have been widely studied. This review will discuss the identification and potential of ASCs, and specifically, preclinical and clinical studies using ASCs in craniofacial repair. Studies involving ASCs in the repair of defects caused by craniosynostosis and Treacher Collins syndrome will be discussed. A comprehensive review of the literature will be presented, focusing on fat grafting and biomaterials‐based approaches that include ASCs for craniofacial regeneration. This article presents the stem and progenitor cells from subcutaneous adipose tissue,briefly comparing them with their bone marrow counterparts,and discussing their potential for use in regenerative medicine.Subcutaneous adipose tissue differs from other mesenchymal stromal/stem cells(MSCs) sources in that it contains a pre-adipocyte population that dwells in the adventitia of robust blood vessels.Pre-adipocytes are present both in the stromal-vascular fraction(SVF;freshly isolated cells) and in the adherent fraction of adipose stromal/stem cells(ASCs;in vitro expanded cells),and have an active role on the chronic inflammation environment established in obesity,likely due their monocyticmacrophage lineage identity.The SVF and ASCs have been explored in cell therapy protocols with relative success,given their paracrine and immunomodulatory effects.Importantly,the widely explored multipotentiality of ASCs has direct application in bone,cartilage and adipose tissue engineering.The aim of this editorial is to reinforce the peculiarities of the stem and progenitor cells from subcutaneous adipose tissue,revealing the spheroids as a recently described biotechnological tool for cell therapy and tissue engineering.Innovative cell culture techniques,in particular 3 D scaffold-free cultures such as spheroids,are now available to increase the potential for regeneration and differentiation of mesenchymal lineages.Spheroids are being explored not only as a model for cell differentiation,but also as powerful 3 D cell culture tools to maintain the stemness and expand the regenerative and differentiation capacities of mesenchymal cell lineages.  相似文献

In 2001, researchers at the University of California, Los Angeles, described the isolation of a new population of adult stem cells from liposuctioned adipose tissue that they initially termed Processed Lipoaspirate Cells or PLA cells. Since then, these stem cells have been renamed as Adipose-derived Stem Cells or ASCs and have gone on to become one of the most popular adult stem cells populations in the fields of stem cell research and regenerative medicine. Thousands of articles now describe the use of ASCs in a variety of regenerative animal models, including bone regeneration, peripheral nerve repair and cardiovascular engineering. Recent articles have begun to describe the myriad of uses for ASCs in the clinic. The protocol shown in this article outlines the basic procedure for manually and enzymatically isolating ASCs from large amounts of lipoaspirates obtained from cosmetic procedures. This protocol can easily be scaled up or down to accommodate the volume of lipoaspirate and can be adapted to isolate ASCs from fat tissue obtained through abdominoplasties and other similar procedures.  相似文献

One of the most important issues in orthopaedic surgery is the loss of bone resulting from trauma, infections, tumours or congenital deficiency. In view of the hypothetical future application of mesenchymal stem cells isolated from human adipose tissue in regenerative medicine, we have analysed and characterized adipose-derived stem cells (ASCs) isolated from adipose tissue of rat, rabbit and pig. We have compared their in vitro osteogenic differentiation abilities for exploitation in the repair of critical osteochondral defects in autologous pre-clinical models. The number of pluripotent cells per millilitre of adipose tissue is variable and the yield of rabbit ASCs is lower than that in rat and pig. However, all ASCs populations show both a stable doubling time during culture and a marked clonogenic ability. After exposure to osteogenic stimuli, ASCs from rat, rabbit and pig exhibit a significant increase in the expression of osteogenic markers such as alkaline phosphatase, extracellular calcium deposition, osteocalcin and osteonectin. However, differences have been observed depending on the animal species and/or differentiation period. Rabbit and porcine ASCs have been differentiated on granules of clinical grade hydroxyapatite (HA) towards osteoblast-like cells. These cells grow and adhere to the scaffold, with no inhibitory effect of HA during osteo-differentiation. Such in vitro studies are necessary in order to select suitable pre-clinical models to validate the use of autologous ASCs, alone or in association with proper biomaterials, for the repair of critical bone defects.  相似文献

Tissue resident mesenchymal stem cells (MSCs) are known to participate in tissue regeneration that follows cell turnover, apoptosis, or necrosis. It has been long known that aging impedes an organism's repair/regeneration capabilities. In order to study the age associated changes, the molecular characteristics of adipose tissue derived MSCs (ASCs) from three age groups of healthy volunteers i.e., young, middle aged, and aged were investigated. The number and multilineage differentiation potential of ASCs declined with age. Aging reduces the proliferative capacity along with increases in cellular senescence. A significant increase in quiescence of G2 and S phase was observed in ASCs from aged donors. The expression of genes related to senescence such as CHEK1 and cyclin-dependent kinase inhibitor p16ink4a was increased with age, however genes of apoptosis were downregulated. Further, an age-dependent abnormality in the expression of DNA break repair genes was observed. Global microRNA analysis revealed an abnormal expression of mir-27b, mir-106a, mir-199a, and let-7. In ubiquitously distributed adipose tissue (and ASCs), aging brings about important alterations, which might be critical for tissue regeneration and homeostasis. Our findings therefore provide a better understanding of the mechanism(s) involved in stem cell aging and regenerative potential, and this in turn may affect tissue repair that declines with aging.  相似文献

近年来,间充质干细胞(mesenchymal stem cells,MSCs)衍生的外泌体在组织再生领域引发许多关注。MSCs衍生外泌体作为细胞间通讯的信号分子,具有天然靶向性强、免疫原性低等特点,其通过MSCs旁分泌途径被细胞吸收,参与调控发挥促进细胞或组织再生功能。水凝胶作为再生医学领域的支架材料,具有良好的生物相容性、降解性等特点。将二者制成复合物联合使用后不仅可以提高外泌体在病变位置的滞留时间,且可通过原位注射等方法提高外泌体到达病变位置的剂量,在病变区域治疗效果显著且持续性改善。文中总结了现阶段外泌体与水凝胶复合物材料共同作用促进组织修复、再生的研究结果,以期为未来组织再生领域中的相关研究工作提供借鉴。  相似文献
