测定了小叶锦鸡儿(Caragana microphylla Lam.)花粗提物和不同极性组分总黄酮含量和各项抗氧化能力指标。结果表明:小叶锦鸡儿花不同极性的黄酮组分的抗氧化活性有明显差别,其总抗氧化能力与溶剂极性之间有显著正相关性(P<0.05),但其它抗氧化活性与溶剂极性之间均无显著相关性(P>0.05)。同一极性黄酮组分在不同抗氧化体系中的抗氧化活性有较大差别。对DPPH.自由基的清除能力最强的为乙酸乙酯层(EC50=0.11 mg/mL),其次是二氯甲烷层(EC50=0.38 mg/mL)和乙醇粗提物(EC50=0.56 mg/mL)。总抗氧化能力(T-AOC)最强的为水层,在1 mg/mL时为1.482个T-AOC能力单位。清除羟自由基.OH能力最强的为水层,其次为正己烷层和乙醇粗提物,在3.57 mg/mL时清除率分别为86.23%、66.04%和64.63%。清除超氧阴离子自由基O2-.能力最强的为水层,其次为乙醇粗提物,在1 mg/mL时其清除率分别相当于0.15 mg/mL Vc清除率的336.17%和318.20%。可见,小叶锦鸡儿花的粗提物和不同极性黄酮组分具有较好的抗氧化活性,可用于提取、制备抗氧化特性和作用不同的天然抗氧化剂。  相似文献

四溴双酚A(tetrabromobisphenol A,TBBPA)是新型高关注度污染物之一,对鱼类具有强毒性。山核桃叶中的黄酮类化合物具有抗氧化和抗细胞凋亡等作用。通过硅胶柱洗脱、薄层色谱(TLC)检测对山核桃叶进行提取和纯化,得到黄酮类化合物产物B;设置4个实验组(空白对照,0.5 mg/L TBBPA,1.0 mg/L产物B,0.5 mg/L TBBPA和1.0 mg/L产物B),记录斑马鱼胚胎发育过程中的形态学指标;同时,检测不同处理组中活性氧(reactive oxygen species,ROS)水平和凋亡相关基因的表达情况。结果发现,山核桃叶提取的总黄酮产物B中包含50%的查尔酮、27%的黄酮或者黄酮醇、18%的二氢黄酮或者二氢黄酮醇、5%的异黄酮等22种黄酮类化合物;富含黄酮类化合物的产物B对TBBPA诱导斑马鱼胚胎的卵凝结率、畸形率及32 h血流障碍等指标具有显著降低作用;可显著抑制TBBPA诱导的ROS水平升高;对TBBPA诱导的凋亡基因p53上调和Bcl-2下调均有显著改善作用。实验结果表明山核桃叶总黄酮通过降低ROS水平,抑制凋亡基因的异常表达,发挥抗氧化和抑制凋亡作用。实验结果有助于了解山核桃叶有效成分总黄酮对TBBPA诱导的斑马鱼胚胎毒性的保护作用,为水生态系统和人体健康危害的防治提供新思路。  相似文献

本文采用ABTS清除体系、DPPH清除体系、H2O2/Fe2+/水杨酸检测体系、亚硝基清除体系、普鲁士蓝法、邻苯三酚自氧化法研究了国槐叶黄酮的抗氧化活性,并同芦丁、Vc进行了比较。结果表明,国槐叶黄酮对几种自由基均有不同的清除效果,在质量浓度为50μg/mL时,国槐叶黄酮对羟基自由基(·OH)、ABTS、DPPH、亚硝基自由基(NO2-)和超氧阴离子自由基(O2-·)的清除率为分别62.09%、47.70%、43.28%、22.11%、21.83%;与芦丁、Vc相比较,国槐叶黄酮清除ABTS、DPPH自由基的能力低于芦丁和Vc,而清除NO2-、.OH的能力高于芦丁和Vc,对O2-·的清除能力低于Vc而高于芦丁,其次国槐叶黄酮具有很好的还原力。由此说明,国槐叶提取物可以作为天然抗氧化剂,可以应用在医药食品领域。  相似文献

夹竹桃叶乙醇提取物对斑马鱼的毒性评价   总被引:1,自引:0,他引:1  
研究表明, 夹竹桃(Nerium indicum Mill.)及其提取物对多种害虫具有较强的毒杀作用, 已在害虫生物防治中显示出较大作用。为了合理开发夹竹桃植物, 需要进一步评价夹竹桃植物对水生生物的安全性和对动物的毒理学。文章利用索氏提取法提取夹竹桃叶乙醇粗提物, 并进一步用氯仿萃取浓缩、硅胶柱层析方法提取分离乙醇精提物, 经鉴定为强心甙组分。在此基础上, 委托浙江省医学科学院评价了乙醇粗提物对实验动物的毒理学, 并采用静态法评价了这二种提取物对斑马鱼(Brachydanio rerio)的急性毒性和慢性效应。结果表明: 夹竹桃叶乙醇粗提物的提取率为45%, 乙醇精提物(主要成分为强心甙组分)的提取率为0.25%。乙醇粗取物对大鼠经口和经皮毒性为低毒级, 对家兔皮肤和眼无刺激, 对豚鼠的致敏反应属弱致敏性。这说明夹竹桃叶乙醇粗提物对哺乳动物十分安全。在急性毒性试验中, 发现斑马鱼的死亡率与提取物处理浓度和处理时间均呈明显的正相关。处理浓度越大, 斑马鱼的死亡率越高; 同一浓度处理时间越长, 死亡率越高。用24 mg/L 乙醇粗提物处理6、12、24 和48h 的死亡率分别为26.67%、60%、91.11%和95.56%; 用0.5 mg/L乙醇精提物处理12、24、48、72 和96h 的死亡率分别为6.25%、33.33%、52.08%、54.17%和60.42%; 用24 mg/L 乙醇粗提物处理96h 或32 mg/L 处理48h, 或者用1 mg/L 乙醇精提物处理24h, 斑马鱼全部死亡。在慢性毒性试验中, 用3.33-10.0 mg/L 乙醇粗提物处理斑马鱼28d, 其死亡率为12%-20%。乙醇粗提物处理斑马鱼后, 急性毒性LC50=12.52 mg/L(药后96h)10 mg/L, 慢性毒性LC50=199.51 mg/L(药后28d); 乙醇精提物处理斑马鱼后, 急性毒性LC50=0.46 mg/L(药后96h)1 mg/L, 根据《化学农药环境安全评价试验准则》(1990)中农药对鱼类的毒性分级标准, 说明夹竹桃叶乙醇粗提物对斑马鱼的毒性属于低毒, 比较安全; 乙醇精提物对斑马鱼的毒性则为高毒, 十分不安全。    相似文献

翅茎香青中黄酮物质的提取及其抗氧化活性研究   总被引:1,自引:0,他引:1  
目的:以翅茎香青的茎、叶和花为研究对象,测定其总黄酮含量及其黄酮粗提物对羟自由基(OH·)和超氧自由基(02-·)的清除能力.方法:用索氏提取法以乙醇(95%)为溶剂提取翅茎香青各部位的黄酮物质,采用紫外分光光度法对各部位总黄酮含量以及黄酮粗提物对OH·及02-·的清除率进行测定.结果:实验结果表明:翅茎香青花的总黄酮含量较高,对OH·的清除作用也较好,其叶的黄酮粗提物对O2-·的抑制作用较好.结论:翅茎香青茎、叶和花的黄酮粗提物对OH·和O2-·均具有一定的清除活性,这将为我们日后的研究工作起到铺垫及导向作用.  相似文献

uvrA基因对SOS显色法检测遗传毒物的影响   总被引:6,自引:0,他引:6  
用SOS显色法对45种化学物质(包括致癌物质、抗癌药物、赫基衍生物、代谢抑制物、食品添加剂和含铬废水)进行遗传毒性检测。分别用大肠杆菌的uvrA-菌株PQ37和uvrA+菌株GC 4415作为测试菌株以考察uvr A 基因对SOS显色法检测遗传毒性灵敏度的影响。带有uvrA 突变的菌株PQ37比较灵敏,不带uvrA突变的菌株GC 4415测不出消癌芥、吖啶黄和SIPI系列化台物的遗传毒性。但在实验中也观察到某些化合物诸如丝裂霉素c、嘹啶酮酸、甲氨喋呤、5一氟尿嘧啶和5一溴脱氧尿苷等诱导菌株PQ37的SOS应答的能力匣而低。已知某些化学物质诱导SOS应答的能力部分取决于uvrA基因产物的存在,因此应使用带有uvrA突变和不带uvrA突变的菌株来进行SOS的显色测定。  相似文献

通过响应面法对山茱萸叶黄酮的提取工艺进行优化,并对其体外抑菌和抗氧化活性进行研究。经优化后,山茱萸叶黄酮在超声功率290 W、液料比31∶1(mL/g)、乙醇体积分数61%、提取时间23 min的工艺条件下提取得率最高,达到6.31%,与模型预测值相符。抑菌实验结果表明山茱萸叶黄酮对沙门氏菌有较强的抑菌作用,对绿脓杆菌、金黄色葡萄球菌和中华根霉有较弱的抑菌作用,说明山茱萸叶黄酮对细菌有较强的抑菌作用,对真菌有较弱的抑菌作用。山茱萸叶黄酮浓度为1.2 mg/mL时其对DPPH自由基的清除率为77.24%,对羟基自由基的清除率达到83.54%;山茱萸叶黄酮清除DPPH自由基和羟基自由基的IC_(50)分别为0.699和0.631 mg/L,表明山茱萸叶黄酮具有较强的抗氧化活性。  相似文献

以赶黄草茎叶为实验材料,用乙酸乙酯、正丁醇对赶黄草乙醇粗提物(EF)进行分级萃取,依次得到乙酸乙酯相(AF)、正丁醇相(BF)和水相(WF),分别测定各相黄酮含量、体外抗氧化和抑菌活性。结果表明,乙酸乙酯相的黄酮含量最高,为133.36 mg/g,超过赶黄草总黄酮含量的50%;乙酸乙酯相的DPPH自由基、羟自由基、超氧阴离子清除能力和亚铁离子螯合能力最强,其IC_(50)值分别为0.076、0.209、0.038、0.593 mg/mL;乙酸乙酯相的抑菌效果显著,对金黄色葡萄球菌和铜绿假单胞菌的最低抑菌浓度均为0.075 mg/mL。  相似文献

本实验以枸骨叶为研究对象,经75%乙醇浸提,用极性递增的溶剂分离制备获得枸骨叶提取物,测定了枸骨叶提取物对酪氨酸酶的抑制作用,并使用1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基清除的方法检测了枸骨叶提取物抗氧化能力。实验结果表明:枸骨叶三氯甲烷和正丁醇提取物对酪氨酸酶具有较强的抑制作用,IC50值分别达到0.414 mg/m L和0.667 mg/m L,均为混合型可逆抑制类型。同时,二者均具有一定的清除DPPH自由基的能力,IC50值分别为0.132 mg/m L和0.039 mg/m L,表明枸骨叶提取物同时具有抑制酪氨酸酶活性和抗氧化的功效,在美容、药物及食品等行业具有潜在的应用价值。  相似文献

通过还原力、清除超氧阴离子自由基、清除羟自由基和抑制H2O2诱导红细胞氧化溶血实验来评价沙棘茶水溶性多糖(WPHT)体外抗氧化能力,并与Vc进行了比较.结果表明,WPHT具有较强的还原能力,对O-·2和·OH具有较强的清除作用,IC50分别为:394 μg/mL、182 μg/mL;对H2O2诱导红细胞氧化溶血及MDA生成有很强的抑制作用,IC50分别为:221 μg/mL、202 μg/mL.说明WPHT在一定浓度范围内具有较强的抗氧化能力.  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献
