探讨大蒜素预处理对大鼠大负荷运动后即刻和12、24 h血液抗氧化应激能力的影响,并分析其可能机制。将成年雄性SD大鼠64只,随机分为安慰剂组(C组)和大蒜素组(D组),D组每日定时经口腔灌胃给予大蒜素30 mg/kg体重,C组给予相同体重比例的安慰剂(蒸馏水)。两周后测定运动前及大负荷运动后即刻、运动后12 h、运动后24 h各4个时相血清谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)及总抗氧化能力(TAC)的变化。结果发现D组GSH-Px在运动后即刻、12 h非常显著高于C组(P0.01);GSH、SOD在运动后即刻、12 h显著高于C组(P0.05),MDA变化则与SOD呈相反趋势;TAC在运动后即刻、12 h、24 h三个时相均显著高于C组。提示大蒜素预处理有助于大鼠大负荷运动后血液抗氧化应激能力的恢复。  相似文献

为了考察食品级槲皮素粉对机体最大摄氧量和耐力的影响,本研究纳入20名健康的大学生志愿者作为本研究的研究对象。将受试者随机分为A组和B组,每组10名,A组饮用剂量为1 mg/mL的槲皮素饮料,B组饮用安慰剂饮料,每天饮用1 000 m L。饮用7 d后,通过自行车分级运动测试最大摄氧量(VO2max),通过骑行疲劳时间测试耐力,同时检测血清丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)含量。然后进行交叉实验并测试VO2max、骑行疲劳时间及抗氧化指标。研究显示,与基线VO2max相比,饮用槲皮素饮料后VO2max显著升高13.21%,而饮用安慰剂饮料后的VO2max与基线无显著差异。与基线骑行疲劳时间相比,饮用槲皮素饮料后骑行疲劳时间显著升高25.15%,而饮用安慰剂饮料后的骑行疲劳时间与基线无显著差异。与基线血清MDA相比,饮用安慰剂饮料后受试者血清MDA显著升高27.15%,而饮用槲皮素饮料可抑制血清MDA的升高。与基线血清SOD和GSH-Px相比,饮用安慰剂饮料后受试者血清SOD和GSH-Px分别降低了20.49%和21.08%,而饮用槲皮素饮料可抑制血清SOD和GSH-Px的降低,表明槲皮素可显著提高骑行运动过程中受试者的VO2max和耐力。本研究初步表明,补充槲皮素可通过降低运动过程中MDA水平来减少脂质过氧化损伤。另外,槲皮素通过抑制运动过程中SOD和GSH-Px的降低来提高机体的抗氧化能力,从而延缓疲劳。  相似文献

目的:探讨竹节参对大强度耐力训练大鼠心肌线粒体抗氧化能力的影响,为该药运用于抗运动疲劳提供理论依据。方法:将大鼠随机分为安静对照组,大强度耐力训练组(训练组),大强度耐力训练+竹节人参组(训练加药组),测定心肌线粒体脂质过氧化产物丙二醛(MDA)和过氧化氢(H2O2)的含量以及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)的活性,研究竹节参对大强度耐力训练大鼠心肌线粒体的保护作用。结果:力竭运动引起大鼠心肌线粒体MDA、H2O2含量显著升高(P0.01),心肌线粒体抗氧化酶CAT、GSH-Px、SOD活性显著下降(P0.01);训练加药组大鼠心肌线粒体MDA、H2O2含量明显低于训练组(P0.01),CAT、GSH-Px、SOD活性明显高于训练组。结论:竹节参可明显提高大强度耐力训练大鼠心肌线粒体的抗氧化能力,保护心肌线粒体的氧化损伤。  相似文献

目的探讨麝香保心丸对一次性力竭运动大鼠心肌损伤标记物和C反应蛋白的影响。方法选择雄性Wistar大鼠52只,随机分成实验组和对照组,每组26只。两组大鼠每天自由进食饮水相同,实验组大鼠每天麝香保心丸2粒(每粒22.5 mg)分2次灌胃,共两周。两组均接受力竭游泳运动,制成力竭运动动物模型,分别测定和比较两组大鼠血清和心肌组织肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、肌钙蛋白T(Tn T)、B型脑钠肽(BNP)和C反应蛋白(CRP)水平。结果大鼠平均力竭运动游泳时间实验组较对照组明显延长(P0.01)。实验组血清和心肌组织CK、CK-MB、Tn T、BNP和CRP水平均明显低于对照组(P0.05)。结论麝香保心丸可显著降低力竭运动大鼠血清和心肌中CK、CK-MB、Tn T、BNP和CRP水平,能够减轻力竭运动后心肌损伤。  相似文献

氦氖激光对大鼠心肌超微结构、SOD和MDA影响的观察   总被引:2,自引:1,他引:1  
为了解低强度氦氖激光对心肌细胞超微结构、超氧化物歧化酶(SOD)和丙二醛(MDA)的影响,采用2.87J/cm^2,7.17J/cm^2和28.66J/cm^2三种剂量氦氖激光照射大鼠心前区,电镜观察心肌超微结构,测定心肌组织SOD和MDA的含量。结果发现心肌线粒体有轻微损伤,心肌组织SOD含量在7.17J/cm^2剂量显著下降(P<0.05),而在28.66J/cm^2剂量显著升高(P<0.05),心肌组织MDA含量在7.17J/cm^2和28.66J/cm^2两种剂量下降有显著升高(P<0.01),在2.87J/cm^2剂量,心肌组织SOD和MDA含量均无显著变化(P<0.05)。结果提示心肌SOD含量和脂质过氧化随不同的激光剂量而变化。  相似文献

目的:研究黄连素对高脂血症大鼠的降脂作用和抑制肝脏脂质过氧化的作用。方法:随机抽取10只大鼠作为对照组,其余大鼠制作大鼠高血脂模型。成模大鼠随机分为模型组,黄连素低、中、高剂量组和血脂康组,每组10只。各给药组大鼠每天一次给予相应药物灌胃,连续30 d。观察黄连素对高脂血症大鼠血清中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)的影响和肝组织二脂酰甘油酰基转移酶(DGAT),羟甲基戊二酸单酰辅酶A(HMG-Co A),胆固醇7α-羟化酶(CYP7A),肝脏甘油三脂脂肪酶(HTGL)以及脂质过氧化产物丙二醛(MDA)含量、超氧化物歧化酶(SOD)与谷胱甘肽过氧化物酶(GSH-Px)活性的影响。结果:与正常组比较,高脂血症组大鼠的肝脏系数、TC、TG、LDL-C、DGAT、HMG-Co A和MDA显著升高(P0.01),HDL-C、CYP7A、HTGL、SOD和GSH-Px显著降低(P0.01)。与高脂血症组比较,黄连素中、高剂量组和血脂康组大鼠肝脏系数、TC、TG、LDL-C、DGAT、HMG-Co A和MDA明显减低(P0.01),HDL-C、CYP7A、HTGL、SOD和GSH-Px显著升高(P0.01);黄连素低剂量组TC、TG、LDL-C、DGAT、HMG-Co A和MDA含量显著降低(P0.05;P0.01),CYP7A和HTGL显著升高(P0.01)。结论:黄连素可降低高血脂大鼠的血脂水平,抑制肝脏脂质过氧化过程,减少肝脏损伤。  相似文献

目的:探讨还原型谷胱甘肽(GSH)对阿霉素所致大鼠心脏毒性的保护作用及其机制。方法:选取40只健康SD大鼠作为实验动物,将其随机分为4组,即GSH(小剂量)组+阿霉素组(小剂量组)、GSH(大剂量)+阿霉素组(大剂量组)、阿霉素组及生理盐水组,每组各10只。上述前3组均给予阿霉素;小剂量组给予GSH 250 mg/kg;大剂量组给予GSH 500 mg/kg;生理盐水组给予相同体积的生理盐水。末次给药24 h后,应用免疫组化学SP法检测大鼠心肌组织中BAX和BCL-2蛋白的表达情况,应用ELISA法测定大鼠血清中CK(肌酸激酶)、CK-MB(肌酸激酶同工酶)、LDH(乳酸脱氢酶)的含量以及心肌组织中超氧化物歧化酶(SOD)、丙二醛(MDA)的活性,并对实验数据进行统计学分析。结果:1与阿霉素组相比,应用GSH干预的大鼠心肌组织中BCL-2的表达显著升高,BAX的表达显著降低,差异均有统计学意义(P0.05);大、小剂量组间BAX和BCL-2蛋白的表达水平比较无统计学差异(P0.05);2与阿霉素组相比,GSH干预的大鼠血清CK、CK-MB、LDH水平均显著下降,但大、小剂量组间比较无显著性差异(P0.05);3与阿霉素组相比,应用GSH干预的大鼠心肌组织MDA水平降低,SOD活力升高(P0.05),但大、小剂量组间比较无显著性差异(P0.05)。结论:还原型谷胱甘肽能够抑制阿霉素导致的心脏毒性作用,其作用机制可能与提高心肌组织BCL-2蛋白的表达与SOD水平、降低MDA水平以及BAX蛋白的表达有关。  相似文献

目的:观察大鼠二次脑损伤后脑组织SOD及MDA的变化,进一步探讨过氧化反应在二次脑损伤机制中的作用。方法:90只雄性SD大鼠随机分为正常对照组(A组)、弥漫性脑损伤组(B组)和二次脑损伤组(C组),建模成功后,分别于伤后1 h、3 h、6h、12 h、24 h、48 h处死动物,取额叶组织匀浆,分别测定大鼠脑内SOD及MDA的含量。结果:伤后1h开始,B、C组中SOD含量呈现先上升后下降的趋势,并随时间呈持续下降趋势直至伤后24 h,C组较B组下降程度更加明显(P0.05);伤后3 h,B、C组中SOD含量均增高,B组增高程度和速率均高于C组(P0.05)。伤后1 h开始,B、C组MDA含量呈升高趋势,并于24小时达峰值,C组升高趋势较B组更显著,伤后6 h、24 h、48 h,C组与B组之间相比统计学差异显著(P0.05)。结论:创伤性脑损伤及二次脑损伤后,脑组织内过氧化反应明显加重,SOD及MDA均出现明显变化,二次脑损伤的过氧化反应较脑损伤组更加严重且持续时间更长,且MDA较SOD的变化具有滞后性。  相似文献

目的:通过美洲大蠊提取物(PAE)对力竭运动大鼠心肌自由基代谢的影响,探讨其对心肌氧化损伤的保护作用。方法:雄性SPF级健康SD大鼠40只,随机分为安静组、运动组、美洲大蠊提取物安静组、美洲大蠊提取物运动组(n=10)。服药组每天灌服2 ml美洲大蠊提取物(美洲大蠊提取物按50 mg/kg配制,溶于2 ml蒸馏水中灌胃给药),对照组每次灌蒸馏水2 ml。每天灌胃1次,连续灌胃14 d后,美洲大蠊提取物运动组与运动组大鼠进行一次性力竭游泳运动建立力竭模型,记录大鼠力竭运动时间。力竭运动结束时即刻取样,检测血清中丙二醛(MDA)含量、谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活性,并检测观察心肌组织中一氧化氮合酶(NOS)基因的表达情况。结果:与安静组相比,一次力竭游泳后,运动组心肌SOD、GSH-Px的活性明显降低(P0.01),而MDA含量显著升高(P0.01);而美洲大蠊提取物能够显著提高力竭SD大鼠的心肌SOD、GSH-Px的活性(P0.01),降低MDA含量(P0.01),e Nos基因表达增高。结论:大鼠力竭运动后心肌会发生氧化损伤,美洲大蠊提取物干预后能够增加力竭运动后大鼠心肌的抗氧化能力,对力竭运动所致心肌损伤具有一定的保护作用,进而增强大鼠运动能力。  相似文献

研究人参茎叶皂甙(GSL)对高胆固醇饮食大鼠心肌再灌注性心律失常(RPA_r)和脂质过氧化的影响。方法:将胆固醇乳剂用灌胃法饲养大鼠14d,建立高脂血症模型,各组大鼠进行心肌缺血再灌注实验,观察高脂血症和GSL对大鼠心肌缺血再灌注2h后血丙二醇(MDA),超氧化物歧化酶(SOD)和一氧化氮(NO)水平的影响和对再灌注性心律失常发生率的影响。结果显示:(1)用胆固醇乳剂饲养大鼠14d,成功建立高脂血症模型。同时给予GSL14d有明显降脂作用。(2)高脂血症状态下,心肌缺血再灌注2h后,血MDA升高(p<0.01),SOD降低(p<0.01)和NO(p<0.05)降低,再灌注10min内RPAr的发生率增高。(3)GSL组再灌注后2h的血MDA降低,而SOD和NO水平显著升高;使RPAr发生率大为降低,无VF发生。实验显示高脂血症加重心肌缺血再灌注损伤和提高RPAr发生率及动物死亡率,GSL可减少高脂饮食大鼠脂质过氧化和诱导体内NO生成而减轻缺血再灌注心肌损伤,降低缺血再灌注性心律失常发生率。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献
