抗菌和细胞毒活性海洋细菌的筛选及其次生代谢基因证据   总被引:1,自引:0,他引:1  
从不同海域的海水、海泥和海洋生物中分离海洋细菌,利用琼脂扩散法和MTT法对细菌培养液的乙酸乙酯提取物进行了抗菌和细胞毒活性筛选,并对具有细胞毒活性的细菌菌株进行了16SrRNA系统发生学分析和多聚酮合酶(PKSⅠ型)、非核糖体肽合成酶(NRPS)的筛选。结果显示,在分离到的346株海洋细菌中,42株细菌具有抗菌活性,12株具有细胞毒活性。对12株具有细胞毒活性的细菌菌株进行了16SrRNA系统发生学分析,它们分别属于Agrobacterium,Pseudoalteromons,Bacillus,Paracoccus,Rheinheimera,Aerococcus,Exiguobacterium和Alteromonas8个属。对这12株具有细胞毒活性的细菌菌株进行进一步的PKS和NRPS筛选,得到了4株含有PKSⅠ型的KS结构域或NPRS的A结构域的海洋细菌,为从聚酮和非核糖体肽等生物合成途径去深入研究其次生代谢产物提供了基因学的证据。  相似文献

从硇洲岛和徐闻珊瑚礁自然保护区潮间带采集海水和沉积物标本,采用纯培养的方法分离其中的海洋细菌;以金黄色葡萄球菌、枯草芽抱杆菌和大肠埃希菌等为指示菌,以氨苄青霉素、青霉素-链霉素为阳性对照,采用琼脂扩散法筛选抗菌活性菌株;采用基于16S rDNA序列比对及其系统发育分析对分离培养的阳性海洋细菌进行分类鉴定和生物多样性分析;为进一步从海洋细菌资源中发掘新型抗菌药物奠定基础。结果从106株海洋细菌中筛选出了44株抗细菌活性菌株,阳性率为41.5%。其中,11株具有抗金黄色葡萄球菌作用,31株有抗枯草芽孢杆菌作用,13株有抗大肠埃希菌作用。抗菌活性菌株分布于31属,优势属为芽孢杆菌属(Bacillus)和弧菌属(Vibrio)。这表明分离自硇洲岛和徐闻珊瑚礁自然保护区潮间带的海洋细菌中的抗菌活性菌株具有丰富的生物多样性。  相似文献

【目的】本文对微小卡罗藻共附生微生物进行分离并对其抗菌和细胞毒活性进行初步研究,以期望获得既具有抗菌又具有细胞毒的高活性菌株,为从共附生微生物的角度去研究微小卡罗藻毒素的合成途径以及真正来源提供研究材料。【方法】利用琼脂扩散法和MTT法对细菌培养液的乙酸乙酯提取物进行抗菌和细胞毒活性筛选,并对具有细胞毒活性的细菌菌株进行了16S rRNA系统发生学分析。【结果】在分离到的38株海洋细菌中,25株细菌具有抗菌活性,5株细菌(W-14-2、W-2-2、W-12、E-8-2和W-4)具有细胞毒活性。对这5株具有细胞毒活性的细菌菌株进行16S rRNA系统发生学分析显示它们分别与Alteromonas alvinellae、Stappia aggregata、Pelagibaca bermudensis、Marinobacter kribbensis和Maribacter dokdonensis的16S rRNA基因序列具有较高的相似性。【结论】在分离到的微小卡罗藻共附生微生物中含有较为丰富的活性菌株,且获得5株具有抗菌活性又具有细胞毒的高活性菌株。  相似文献

海带藻际微生物抗菌活性及其多样性的初步研究   总被引:1,自引:0,他引:1  
从浙江台州海域健康海带、病烂海带(Laminaria japonica)及其周围海水中分离纯化海带藻际微生物, 利用双层琼脂扩散法对获得的菌株进行抗菌活性筛选, 比较活性菌株与来源的相关性, 并对具有抗菌活性的菌株进行16S rRNA系统发生学分析。结果显示, 在分离的143株海带藻际微生物中, 有42株细菌具有抗菌活性, 其中来源于健康海带和海水的活性菌株比例(35%和29%)大于来源于病烂海带的细菌(23%)。对其中16株具有广谱抗菌活性的菌株进行的16S rRNA系统发生学分析显示, 分别属于Pseudoalteromonas、Rahnella、Donghaeana、Bacillus和Exiguobacterium 5个属。为从藻际微环境的微生物多样性入手了解海带病烂机制以及寻找新的抗菌药物提供研究材料。    相似文献

抗肿瘤活性海洋放线菌的筛选及菌株HGF26的初步鉴定   总被引:1,自引:0,他引:1  
对采自连云港海域的海泥样品进行放线菌选择性分离,用其发酵液进行抗肿瘤活性筛选,并对活性较好的菌株HGF26进行了初步鉴定。从海泥样品中共分离得到放线菌78株,以人肝癌细胞HepG2为靶标,活性筛选得到细胞毒活性达60%以上的阳性菌株3株,以其他5株肿瘤细胞为靶标的复筛表明,菌株HGF26的发酵液对多种肿瘤细胞具有显著的细胞毒活性,其中对胃癌细胞BGC823的细胞毒活性为79%,活性产物具有较好的酸碱和热稳定性。通过对菌株的培养特征、形态特征、生理生化特征和细胞壁成分分析,将菌株HGF26初步鉴定为微白黄链霉菌的海洋变种。  相似文献

该文设计9种分离培养基,采用稀释涂布法从14份真红树植物的46份组织样品中分离纯化内生细菌。并基于菌株形态学特征和16S rRNA基因序列确定分离菌株的种属及分析其物种多样性,采用秀丽隐杆线虫模型筛选菌株延缓衰老活性。结果表明:(1)通过基因序列去重复后从46份真红树植物组织样品中获得32株海洋细菌,基于菌株16S rRNA基因序列信息分析,覆盖12科17属,其中芽孢杆菌属(Bacillus)为优势菌属,并获得1株疑似橙单胞菌属(Aurantimonas)新种,16S rRNA基因序列相似性低于97%;(2)经过秀丽隐杆线虫粗筛发现3株海洋细菌具有显著延缓线虫衰老的活性(P0.05)。以上结果表明海南西海岸真红树内生细菌具有物种多样性,部分菌株具有延缓线虫衰老活性。  相似文献

从海参、海胆、海葵、海兔、石莼、羊栖菜、裙带菜分离得到125种共附生海洋微生物,以6种敏感菌为指示菌,从中获得具有抑菌活性的细菌21株,放线菌8株,真菌2株。21株抑菌海洋细菌中芽孢杆菌属为7株,占33.3%,弧菌属为11种,占52.2%,其余3株为假单孢杆菌属,占14.5%。8株抑菌海洋放线菌中链霉菌属为5株,占62.5%,小单孢菌属为3株,占36.5%。2株抑菌海洋真菌均为青霉属。  相似文献

从海泥、海水及海洋动物中分离到数十株细菌 ,对其产生活性物质的菌株进一步筛选 ,得到产蛋白酶、淀粉酶和抑菌活性等生物活性物质的几株海洋细菌 ,将其中的活性高的四株菌纯化后进行菌种鉴定。通过对革兰氏染色、个体及群体形态观察、糖类发酵、硝酸盐还原等生理特性的研究 ,依据《伯杰氏细菌鉴定手册》确定它们的分类地位 ,它们分别应归属为欧文氏菌属 (Erwiniasp .)、气单孢菌属 (Aeromonassp .)、微球菌属 ( (Micrococoussp .)、和假单孢菌属 ( (Pseudomonassp .)。  相似文献

细菌在海洋生物地球化学循环中发挥着重要作用。为更好地了解海洋细菌的特征及其在海洋环境中的潜在作用, 本文利用纯培养与16S rRNA基因高通量测序技术对中印度洋与南海西部海域表层海水细菌多样性进行研究。纯培养结果表明, 自中印度洋与南海西部表层海水中共分离275株可培养海洋细菌, 隶属于4门49属75种。变形菌门是绝对优势类群(占总株数的68.7%), 其次是放线菌门(21.5%)、拟杆菌门(9.1%)和厚壁菌门(0.7%)。在属水平, 微杆菌属(Microbacterium)与弧菌属(Vibrio)是主要的优势属, 共占总株数的30.0%。在3种分离培养基中, 自1/10 × 2216E培养基中分离细菌的数目与种类最多(89株, 30属); 分离菌株中的细菌菌株有7、9与3个属分别仅在2216E、1/10 × 2216E及葡萄糖甘露糖(glucose-mannose, GM)培养基中生长。此外, 共分离培养出50株细菌(26种)可能代表潜在新分类单元。高通量测序结果显示, 中印度洋和南海西部表层海水中共有23个门531个属。优势门类为变形菌门(72.2%)和拟杆菌门(15.3%), 优势属为嗜冷杆菌属(Psychrobacter, 24.4%)、盐单胞菌属(Halomonas, 16.3%)和亚硫酸杆菌属(Sulfitobacter, 13.9%)。此外, 中印度洋表层海水细菌Shannon-Wiener指数与Pielou均匀度指数显著高于南海西部(P < 0.05), 且细菌群落结构显著不同(P < 0.05)。综合纯培养与原位细菌数据得出, 中印度洋与南海西部海洋细菌具有丰富的多样性, 具有进一步开发研究的价值。  相似文献

白木香内生真菌的分离鉴定及其抑菌活性   总被引:2,自引:0,他引:2  
从白木香Aquilaria sinensis(Lour.)Gilg木质部的树脂形成部位和健康部位中共分离获得42株内生真菌,经初步鉴定,产孢的33株分属于3目4科7属,其余未产孢的9株暂归为无孢菌群。采用杯碟法和MTT法分别测定了各菌株的发酵上清液对3种病原菌的体外抑菌活性和2种肿瘤细胞的体外细胞毒活性。结果表明,白木香木质部健康部位内生真菌以枝顶孢霉属为优势属,而树脂形成部位的内生真菌种类比健康部位要多,且以青霉属为优势属。其中26株至少能抑制一种指示菌,占总数的61.9%;7株对指示瘤株具有细胞毒活性,占总数的16.7%。抑菌活性菌株主要分布在枝顶孢霉属和青霉属。枝顶孢霉属菌株抑菌活性较强,其抗菌活性成分值得进一步研究。  相似文献

从上海郊区海域采集的海水、海泥样中分离出193株海洋细菌,对其进行抑菌活性菌株的筛选。对分离出的菌株分别利用琼脂块法和管碟法进行初筛和复筛,结果显示有29株海洋细菌具有抑菌活性,占总分离菌株的15%,其中抑菌能力较强的G4抑菌谱较广,同时能抑制革兰氏阳性和阴性指示菌。  相似文献

分离并筛选具有抑菌活性的海洋细菌对于开发和利用海洋微生物具有重要意义,该研究从6份海泥样品中共分离到78株海洋细菌,以6种细菌作为敏感指示菌,采用覆盖技术对分离菌株进行拮抗试验,17株海洋细菌具有抑菌活性,对其中2株具有较强抑菌活性的海洋细菌进行革兰氏染色,耐盐试验,运动性观察,过氧化氢酶测定,明胶液化试验,硫化氢产生试验,石蕊牛奶试验,糖类发酵,硝酸盐还原等特性分析,依据《伯杰氏细菌鉴定手册》进行分类鉴定,它们分别应归属为气单孢菌属(Aeromonas sp.)和假单孢菌属(Pseudomonas sp.)。  相似文献

Bacilli of the species Bacillus subtilis, B. pumilus, B. mycoides, B. marinus and B. licheniformis (a total of 53 strains) were isolated from 15 invertebrate species and the water of the Vostok Bay, Peter the Great Bay, Sea of Japan. Bacilli were most often isolated from bivalves (22.7%) and sea cucumbers (18.9%); they occurred less frequently in sea urchins and starfish (13.2 and 7.5%, respectively). Most of bacilli strains were isolated from invertebrates inhabiting silted sediments. No Bacillus spp. strains were isolated from invertebrates inhabiting stony and sandy environments. The species diversity of bacilli isolated from marine objects under study was low. Almost all bacterial isolates were resistant to lincomycin. Unlike B. pumilus, B. subtilis isolates were mostly resistant to benzylpenicillin and ampicillin. Antibiotic sensitivity of B. licheniformis strains was variable (two strains were resistant to benzylpenicillin and oxacillin, while one was sensitive). A significant fraction of isolated bacilli contained pigments. Pigmented strains were more often isolated from seawater samples, while colorless ones predominated within hydrobionts. B. subtilis colonies had the broadest range of colors. In the Bacillus strains obtained, DNase, RNase, phosphatase, elastolytic, chitinase, and agarolytic activity was detected. Bacilli strains with hydrolytic activity occurred in invertebrates more often than in seawater.  相似文献

【目的】认识智利海洋沉积物中放线菌的多样性。【方法】分别采用选择性分离培养和非培养的基于16SrRNA基因序列系统发育分析方法,对来自智利南部海域海洋沉积物中放线菌多样性进行研究。采用6种选择性分离培养基分离放线菌;利用放线菌特异性引物对样品总DNA进行16SrRNA基因序列扩增并构建了16SrRNA基因克隆文库。分别挑选不同培养特征的22株放线菌和59个基因克隆进行16SrRNA基因序列的系统进化分析;测定分离的放线菌对海水的依赖性及产生抗菌活性化合物的能力。【结果】共分离到328株放线菌;挑选的22株放线菌分别属于小单孢菌属、多形孢菌属、链霉菌属、迪茨氏菌属、气微菌属和短状杆菌属;挑取的59个克隆属于40个分类单元,其中60%分类单元属于放线菌门放线菌亚纲、酸微菌亚纲和红色杆菌亚纲,另外40%的分类单元在放线菌内形成几个独立的进化分支,有可能代表放线菌新类群。22株放线菌有19株具有抗菌活性,50%的生长依赖海水的放线菌也具有抗菌活性。【结论】智利海域沉积物存在丰富的放线菌系统发育多样性并能产生活性次级代谢产物,而且还蕴藏丰富的新类型的放线菌资源。  相似文献

从中国北部湾海域采集的海蜇样品中分离到577株海洋细菌,以条件致病真菌白色念珠菌(Candida albicans)作为敏感测试菌株,采用琼脂块法测定了577株海洋细菌的抗真菌活性,结果表明有119株具有抗白色念珠菌活性,占总数的20.6%。对其中的40株高活性菌株进行抗真菌和抗细菌活性研究,结果表明,有21株海洋细菌对3种或3种以上的真菌有抑制活性,23株对细菌有不同程度的抑制作用,其中1株海洋细菌I0s4-18具有广谱的抗菌作用,对革兰氏阴性细菌、革兰氏阳性细菌、酵母菌及丝状真菌均具有抑制作用。海蜇共附生的微生物中能产生抗真菌活性的微生物的比例是很高的,I0s4-18菌株具有良好的开发前景。  相似文献

A total of 319 bacterial strains isolated from the surfaces of seaweeds and invertebrates were tested for their effects on settlement of Ulvalactuca spores and Hydroidesezoensis larvae in laboratory bioassays. Of the 192 bacterial strains isolated from the surfaces of seaweeds 63 isolates were shown to be inhibitory against the settlement of algal spores and 62 isolates were inhibitory against larval settlement. Thirty-seven percent of the 127 bacterial strains isolated from the surfaces of marine invertebrates were shown to be inhibitory against algal spores and larval settlement. The strain CI4 isolated from an adult Cionaintestinalis showed the strongest inhibitory effect and was identified as Pseudoalteromonas sp. via 16S rRNA gene sequencing. The high proportions of host associated bacteria producing antifouling compounds suggest that these bacteria may help the host organism in the defense against fouling.  相似文献

The bacteria associated with marine invertebrates are a rich source of bioactive metabolites. In the present study bacteria associated with the sponge Suberites domuncula and its primmorphs (3-dimensional aggregates containing proliferating cells) were isolated and cultured. These bacteria were extracted, and the extracts were assayed for antiangiogenic, hemolytic, antimicrobial, and cytotoxic activities. Our studies revealed that extract obtained from the bacterium (PB2) isolated from sponge primmorphs is a potent angiogenesis inhibitor. In the chick chorio-allantoic membrane (CAM) assay, it showed 50% activity at 5 μg ml−1 and 100% activity at 10 and 20 μg ml−1 concentrations. Extracts obtained from 5 bacterial strains isolated from sponge and its primmorphs showed hemolytic activity. The sponge-associated bacteria belonging to the α subdivision of Proteobacteria and the primmorph-associated bacterium identified as a possible novel Pseudomonas sp. displayed remarkable antimicrobial activity. It is important to note that these bacterial extracts were strongly active against multidrug-resistant clinical strains such as Staphylococcus aureus and Staphylococcus epidermidis, isolated from hospital patients. The bacterial extracts having antimicrobial activity also showed cytotoxicity against HeLa and PC12 cells. In summary, this investigation explores the importance of sponge-associated bacteria as a valuable resource for the discovery of novel bioactive molecules.  相似文献
