We present the first demonstration of oral transmission of Trypanosoma cruzi to raccoons (Procyon lotor), a natural reservoir host in the United States, by ingestion of trypomastigotes and infected bugs, but not infected tissue. To investigate an alternative, non-vector-based transmission method, we tested the hypothesis that raccoons scavenging on infected hosts results in patent infection. Macerated tissue from selected organs infected with amastigote stages of T. cruzi was orally administered to experimental groups of raccoons (n = 2/group) at 2, 12, or 24 hr after collection of the tissue samples. Additionally, raccoons (n=1) in control groups were inoculated intravenously or per os with trypomastigotes. To further elucidate transmission routes of T. cruzi to raccoons, infected Rhodnius prolixus were fed to raccoons (n=2). Raccoons did not become infected after ingestion of amastigote-infected tissues as evidenced by negative polymerase chain reaction results from blood and tissue, lack of seroconversion, and negative parasitemias. However, per os transmission can occur by ingestion of the infective trypomastigote stage or infected reduviid bugs. We conclude from these findings that oral transmission of T. cruzi may be a route of infection for wildlife in sylvatic cycles, but the scavenging behavior of animals is not likely a significant transmission route.  相似文献

The indirect immunofluorescent antibody test (IFAT) and enzyme-linked immunosorbent assay (ELISA) were compared with blood culture for the detection of Trypanosoma cruzi infection in 83 raccoons (Procyon lotor) trapped in 4 counties of southeast Georgia. Both IFAT and ELISA detected 24 of 25 culture-positive samples (96% sensitivity). Cultures from 25 raccoons (30%) were positive for epimastigotes, whereas a total of 50 raccoons (60%) was seropositive by either the IFAT or ELISA. Forty-five of 83 serum samples (54%) were positive for anti-T. cruzi antibodies with the ELISA, and 47 were IFAT positive (57%). Forty-two of the 50 seropositive raccoons (84%) were seropositive by both tests. Endpoint titers of IFAT-positive samples were determined by testing doubling dilutions from 1:40 to 1:1280. High titers of 640 and 320 were observed for 4 raccoons trapped in 1 county (St. Catherines Island, Liberty County) and titers of 160 for 1-2 raccoons from each of the 4 counties sampled. IFAT titers and ELISA optical density values were positively correlated. Both serological tests have a high sensitivity and should be excellent tools for studying the prevalence of T. cruzi in wildlife populations.  相似文献

Long-distance bird migration is fueled by energy gathered at stopover sites along the migration route. The refueling rate at stopover sites is a determinant of time spent at stopovers and impacts the overall speed of migration. Refueling rate during spring migration may influence the fitness of individuals via changes in the probability of successful migration and reproduction during the subsequent breeding season. We evaluated four plasma lipid metabolites (triglycerides, phospholipids, β-OH-butyrate, and glycerol) as measures of refueling rate in free-living semipalmated sandpipers (Calidris pusilla) captured at non-breeding areas. We described the spatial and temporal variation in metabolite concentrations among one winter site in the Dominican Republic and four stopover sites in the South Atlantic and Mid-Atlantic Coastal Plain regions of North America. Triglycerides and β-OH-butyrate clearly identified spatial variation in refueling rate and stopover habitat quality. Metabolite profiles indicated that birds had higher refueling rates at one site in the Mid-Atlantic Coastal Plain than at three sites on the South Atlantic Coastal Plain and one site in the Dominican Republic. Temporal variation in lipid metabolites during the migration season suggested that male semipalmated sandpipers gained more weight at stopovers on the South Atlantic Coastal Plain than did females, evidence of differential migration strategies for the sexes. Plasma lipid metabolites provide information on migration physiology that may help determine stopover habitat quality and reveal how migratory populations use stopover sites to refuel and successfully complete long-distance migrations.  相似文献

Improving biological indicators to better assess the condition of streams   总被引:3,自引:0,他引:3  
Biological indicators of stream condition are in use by water resource managers worldwide. The State of Maryland and many other organizations that use Indices of Biotic Integrity (IBIs) must determine when and how to refine their IBIs so that better stream condition information is provided. With completion of the second statewide round in 2004, the Maryland Biological Stream Survey (MBSS) had collected data from 2500 stream sites, more than doubling the number of sites that were available for the original IBI development. This larger dataset provided an opportunity for the MBSS to address the following shortcomings in the original IBIs: (1) substantial disturbance apparent in some reference sites, (2) fish IBIs could not be applied to very small streams, (3) natural variability within IBIs (based on regions) resulted in some stream types (e.g., coldwater and blackwater streams) receiving lower IBI scores and (4) one IBI was not able to discriminate degradation as desired (i.e., Coastal Plain fish IBI). Therefore, development of new fish and benthic macroinvertebrate IBIs was undertaken to achieve the goals of: (1) increased confidence that the reference conditions are minimally disturbed, (2) including more natural variation (such as stream size) across the geographic regions and stream types of Maryland and (3) increased sensitivity of IBIs by using more classes (strata) and different metric combinations. New fish IBIs were developed for four geographical and stream type strata: Coastal Plain, Eastern Piedmont, warmwater Highlands and coldwater Highlands streams; new benthic macroinvertebrate IBIs were developed for three geographical strata: Coastal Plain, Eastern Piedmont and Highlands streams. The addition of one new fish IBI and one new benthic macroinvertebrate IBI partitioned natural variability into more homogeneous strata. At the same time, smaller streams (i.e., those draining catchments <300 ac), which constituted a greater proportion of streams (25%) sampled in Round Two (2000–2004) than Round One (1995–1997), because of the finer map scale, were included in the reference conditions used to develop the new IBIs. The resulting new IBIs have high classification efficiencies of 83–96% and are well balanced between Type I and Type II errors. By scoring coldwater streams, smaller streams and to some extent blackwater streams higher, the new IBIs improve on the original IBIs. Overall, the new IBIs provide better assessments of stream condition to support sound management decisions, without requiring substantial changes by cooperating stream assessment programs.  相似文献

Raccoons (Procyon lotor) are reservoir hosts for Trypanosoma cruzi. A 3-yr-long serological survey was conducted to determine the prevalence of antibodies to this zoonotic parasite in raccoons collected from Fairfax County, Virginia, a suburban/urban area outside Washington, D.C. Serum samples from 464 raccoons were examined for T. cruzi antibodies at a 1:40 dilution with an indirect fluorescent antibody test (IFAT) and Brazil strain T. cruzi amastigotes and trypomastigotes as antigen. A positive IFAT test was found in 154 (33%) of the 464 samples. The yearly prevalence was 49 of 132 (37%) in 2000; 19 of 120 (16%) in 2001; and 86 of 212 (41%) in 2002. Our study indicates that raccoons in this area of Virginia are frequently exposed to T. cruzi.  相似文献

ABSTRACT Mid-rotation management practices for pine (Pinus spp.) plantations enrolled in cost-share programs have not been widely evaluated for wildlife. Mid-rotation pine plantations often have a substantial hardwood mid-story that limits growth of desirable understory forage species important to white-tailed deer (Odocoileus virginianus; deer). We treated with imazapyr herbicide and prescribed burning (HB) 11 thinned, 13–22-year-old loblolly pine (P. taeda) plantations in the Upper Coastal Plain (UCP; n = 5) and the Lower Coastal Plain (LCP; n = 6) of Mississippi, USA, enrolled in cost-share programs. We then sampled these plantations for production of important deer forages during July of 2003 and 2004, years 1 and 2 posttreatment. Deer foraging habitat was clearly improved by the HB treatment in both regions by year 2. Forb species of annual importance to deer increased in percent cover and biomass in the UCP and in biomass in the LCP. We estimated nutritional carrying capacity using a target diet quality of 14% crude protein; estimates in HB plots were 3 times greater than controls in the UCP and 19 times greater in the LCP. Although UCP sites had baseline carrying capacities nearly 8 times greater than LCP sites, the greater relative response to HB in the LCP eliminated the regional difference. Our results indicate that imazapyr herbicide treatment followed by prescribed fire is a beneficial tool for deer management in mid-rotation pine plantations.  相似文献

Platanthera ciliaris is a butterfly-pollinated, terrestrial orchid with a loose terminal raceme of 10–50 orange flowers, characterized by a long nectariferous spur. In the southeastern United States, P. ciliaris occurs in the Appalachian Mountains and Coastal Plain physiographic provinces, but rarely in the intervening Piedmont. Of 153 plants that flowered in a Mountain population in 1983, 54 (35.2%) flowered again in 1984; in a Coastal Plain population, only 51 of 306 (16.7%) flowered in both years. Experimental pollinations showed that P. ciliaris is self-compatible but not autogamous. Nectar volume increased over the 6- to 7-day life span of a flower, reaching a maximum of 4 μl and 1.0 mg of sugar (Mountains) or 6 μl and 1.2 mg of sugar (Coastal Plain). Nectar concentrations were relatively constant in flowers of all ages and averaged 23% in the Mountains and 19% in the Coastal Plain. Diurnal patterns of nectar secretion were not well-defined, but most nectar appeared to be produced overnight. In the Mountains Papilio troilus (spicebush swallowtail) was the most frequent and effective pollinator, whereas in the Coastal Plain P. palamedes (palamedes swallowtail) was the predominant pollinator. These large butterflies were most active in the late morning (0930–1030) and early afternoon (1230–1400). Levels of pollinator service, as assessed by rates of removal and insertion of pollinia, were higher in the Mountains (0.81, 0.86) than in the Coastal Plain (0.63, 0.67). These values were reflected in higher levels of fruit-set in Mountain (83.9% in 1983, 86.5% in 1984) than in Coastal Plain (63.8, 65.5%) populations. Combining these observations with the fact that hand pollination increased fruit-set of Coastal Plain flowers 6.5–13.5% above open-pollinated controls, it appears that fruit-set in P. ciliaris is pollen-limited.  相似文献

The determination of serologic titers to infectious organisms is a valuable tool for quantitating exposure to disease organisms. Raccoons (Procyon lotor) were live-trapped from September 1989 to October 1993 and samples collected from two distinct locations in west-central Illinois (USA); a state recreational facility (Park) and privately owned farming property (Farm). Sera were submitted for testing Leptospira interrogans (serovars bratislava, canicola, grippotyphosa, hardjo, icterohemmorhagiae, and pomona), canine distemper virus (CDV), pseudorabies virus (PV), and Toxoplasma gondii. Two-hundred and twenty-two (48%) of 459 raccoons were seropositive for L. interrogans. Eighty-five (23%) out of 368 raccoons were seropositive for canine distemper virus. Eighty-two (17%) of 479 raccoons raccoons were seropositive for pseudorabies virus. One hundred and eight-four (49%) of 379 raccoons were seropositive for T. gondii. A significant difference (P < 0.05) in seroprevalence for L. interrogans between the park (43%) and farm (52%) areas was found. A correlation between increasing age and seroprevalence was found for L. interrogans, CDV, PV, and T. gondii. Furthermore, there was a significant difference in seroprevalence for T. gondii during the spring trapping seasons (73%), when compared with the fall (33%). This type of information on exposure to infectious agents is important for developing control programs to manage raccoon-human and raccoon-domestic animals interactions.  相似文献

An ecological study was conducted in May and June of 1995 and 1996 in South Carolina to determine the factors associated with distributions of aquatic Lepidoptera (Crambidae: Nymphulinae). Larvae were found at 65 lotic and lentic sites in three ecoregions (Piedmont, Sandhills, Coastal Plain). Nine species of aquatic Lepidoptera were collected from 12 species of aquatic vascular macrophytes. One to six plant species were used as hosts, depending on the species of lepidopteran; however, the number of host plants used by a lepidopteran was significantly correlated with the lepidopteran's frequency of occurrence. Significant habitat associations were found for five species. Langessa nomophilalis (Dyar) was found under the widest range of temperature and width and occurred in both lotic and lentic habitats. Munroessa icciusalis (Walker) was found in lotic and lentic habitats and had the widest range of recorded depths. Parapoynx maculalis (Clemens) occurred at stream sites with lentic-like conditions. Parapoynx obscuralis (Grote) occupied the widest range of pH and was restricted to lotic habitats, and P. seminealis (Walker) was found in both lotic and lentic habitats. Additional species, collected at fewer than 8% of sites, included M. gyralis, P. allionealis, Synclita obliteralis, and S. tinealis. Overall, the distributions of aquatic Lepidoptera in South Carolina were nonrandom and predictable on the basis of habitat characteristics. Antibodies to Toxoplasma gondii were found in 92 (9.9%) of 929 feral raccoons ( Procyon lotor ) in Japan with the use of the latex agglutination test (LAT, 1∶64 or higher). Seropositivity varied by geographic location, season, and weight of raccoons trapped. Seroprevalences in the northern, central, and western areas of Japan were found to be 7.9% (39/492), 16.5% (47/285), and 3.9% (6/152), respectively. The seroprevalence by season varied from 8.5% (13/153) in spring to 18.9% (14/74) in winter, which was significantly higher than those in other seasons (P < 0.05). Seroprevalence of T. gondii was elevated in accordance with the increase in body weight of raccoons ( r(s) = 0.9), suggesting that the animals acquired the infection postnatally. The results suggest that the raccoon may serve as a useful indicator for the distribution of T. gondii in peridomestic environments in Japan.  相似文献

Summary The phytoplankton of four ponds in the lower Piedmont and upper Coastal Plain of North Carolina was studied to find a possible correlation between bound phosphorus plankton numbers. No correlation was apparent therefore this paper reports a detailed study in relation to other habitat factors.The ponds are all shallow (2 m.). The Piedmont ponds are high in colloidal clay while the Coastal Plain ponds are much lower. The pH ranged around 7.0 except for one Coastal Plain pond which remained close to pH 6.4. Water temperature ranged from 30° C in June to 23° C near the end of August.Phytoplankton numbers were determined by the standard Sedgwick-Rafter method. Populations were low except for brief pulses. One Coastal Plain pond was typical of a fertilized pond and had several tremendous blooms of blue-green algae. The others supported a seemingly typical Caledonian flora, rich in species but low in number of individual organisms.There was no clear correlation between phytoplankton populations and any habitat factor except pH. During 2005-2006, sera and tissues from raccoons (Procyon lotor), coyotes (Canis latrans), and skunks (Mephitis mephitis) from the state of Wisconsin were tested for Toxoplasma gondii infection. Antibodies to T. gondii were found in 32 of 54 (59.2%) raccoons, 18 of 35 (51.4%) coyotes, and 5 of 7 (71.4%) skunks using the modified agglutination test and a cut-off titer of 1:20. Pooled tissues (brains, hearts, and tongues) from 30 raccoons, 15 coyotes, and 1 skunk were bioassayed for T. gondii infection in mice or cats. Viable T. gondii was isolated from 5 of 30 (16.7%) raccoons, 6 of 15 (40.0%) coyotes, and the skunk. Genetic characterization of the 12 parasite isolates by multilocus PCR-RFLP markers revealed 6 different genotypes including 5 atypical and I archetypal II lineages. The results indicate the prevalence of T. gondii in wildlife mammals is high and that these animals may serve as an important reservoir for transmission of T. gondii.  相似文献

The protozoon Toxoplasma gondii has a worldwide distribution and affects many species of warm-blooded animals. In the Canadian prairies, mesocarnivores such as striped skunks (Mephitis mephitis) and raccoons (Procyon lotor) have experienced an increase in density and distribution, and they are in close contact with human dwellings. However, there has been no systematic study on the seroprevalence of T. gondii in these mesocarnivore populations. The objectives of the current project were to determine the serum antibody prevalence of T. gondii in Canadian prairie mesocarnivores and to study the relationship between antibody prevalence and species, sex, age, location, and year of collection. Antibodies to T. gondii were found in 5 of 24 (20.8%) skunks from Saskatchewan trapped in 1999 and 5 of 40 (12.5%) in 2000. Seroprevalences for T. gondii in raccoons and skunks trapped in Manitoba were 2 of 10 (20%) raccoons trapped in 2002, 7 of 44 (15.9%) trapped in 2003, and 16 of 37 (43.2%) trapped in 2004; and in 13 of 99 (13.1%) skunks trapped in 2003, 29 of 131 (22.1%) trapped in 2004, 53 of 165 (32.1%) trapped in 2005, and 30 of 51 (58.8%) trapped in 2006. Age, location, and year, but not the host species, were important variables in the determining the seroprevalence of T. gondii in skunks and raccoons. Results confirm that T. gondii is endemic in the skunk and raccoon populations in the Canadian prairies.  相似文献

Conversion of land from natural to urban or agricultural cover degrades stream ecosystems and results in loss of biodiversity. We compared cumulative frequency distributions to measure responses to land use gradients for aquatic invertebrate taxa to agricultural, urban, and impervious surface cover gradients across the state of Maryland, USA. The technique identifies the upper limit threshold above which taxa cease to occur as well as a lower limit of detection of effect for negatively affected taxa. Urban development and impervious surface cover negatively affected the distributions of 44–56% of the 180 taxa tested, depending on region. Across similar taxa, negative responses occurred at lower levels of urban land covers in the Piedmont compared to the Coastal Plain physiographic province, which suggests that Piedmont aquatic biodiversity may be more vulnerable to urbanization. Most taxa were capable of tolerating high levels of agricultural development, although a number of common taxa in the Coastal Plain and Highlands regions were found to be agriculture-sensitive. Some taxa traditionally used as indicators were tolerant of very high levels of human-altered land uses, suggesting that such taxa require examination prior to use as indicators of landscape stressors. Our analysis method appears to be sufficiently flexible and sensitive to be used for a variety of taxa and systems for stressor detection, ecosystem monitoring, and spatially explicit forecasts of taxa loss as watershed land cover changes.  相似文献

We sought to determine the serological test that could be used for Trypanosoma cruzi seroprevalence studies in Mexico, where lineage I predominates. In a previous study among pregnant women and their newborns in the states of Yucatan and Guanajuato, we reported a 0.8-0.9% of prevalence for T. cruzi -specific antibodies by Stat-Pak and Wiener ELISA. We have expanded this study here by performing an additional non-commercial ELISA and confirming the seropositives with Western blot, using whole antigens of a local parasite strain. We found a seroprevalence of 0.6% (3/500) in Merida and 0.4% in Guanajuato (2/488). The 5 seropositive umbilical cord samples reacted to both non-commercial ELISA and Western blot tests, and only 1 of the maternal samples was not reactive to non-commercial ELISA. A follow-up of the newborns at 10 mo was performed in Yucatan to determine the presence of T. cruzi antibodies in children as evidence of congenital infection. None of the children was seropositive. One newborn from an infected mother died at 2 wk of age of cardiac arrest, but T. cruzi infection was not confirmed. The T. cruzi seroprevalence data obtained with both commercial tests (Stat-Pak and ELISA Wiener) are similar to those from non-commercial tests using a local Mexican strain of T. cruzi.  相似文献

Crimean-Congo haemorrhagic fever (CCHF) is a severe tick-borne viral zoonosis endemic to parts of Africa, Europe, the Middle East and Central Asia. Human cases are reported annually in South Africa, with a 25% case fatality rate since the first case was recognized in 1981. We investigated CCHF virus (CCHFV) seroprevalence and risk factors associated with infection in cattle and humans, and the presence of CCHFV in Hyalomma spp. ticks in central South Africa in 2017–18. CCHFV IgG seroprevalence was 74.2% (95%CI: 64.2–82.1%) in 700 cattle and 3.9% (95%CI: 2.6–5.8%) in 541 farm and wildlife workers. No veterinary personnel (117) or abattoir workers (382) were seropositive. The prevalence of CCHFV RNA was significantly higher in Hyalomma truncatum (1.6%) than in H. rufipes (0.2%) (P = 0.002). Seroprevalence in cattle increased with age and was greater in animals on which ticks were found. Seroprevalence in cattle also showed significant geographic variation. Seroprevalence in humans increased with age and was greater in workers who handled livestock for injection and collection of samples. Our findings support previous evidence of widespread high CCHFV seroprevalence in cattle and show significant occupational exposure amongst farm and wildlife workers. Our seroprevalence estimate suggests that CCHFV infections are five times more frequent than the 215 confirmed CCHF cases diagnosed in South Africa in the last four decades (1981–2019). With many cases undiagnosed, the potential seriousness of CCHF in people, and the lack of an effective vaccine or treatment, there is a need to improve public health awareness, prevention and disease control.  相似文献

Lemurs on St. Catherines Island, Georgia were tested for Trypanosoma cruzi infection to develop a better understanding of the epizootiology of the parasite in nonhuman primates in the southeastern United States. Fifty-six ring-tailed (Lemur catta), blue-eyed black (Eulemur macaco flavifrons), and black-and-white ruffed (Varecia variegata variegata) lemurs were tested by hemoculture and serology to determine the prevalence of T. cruzi in the population. Of those tested 3 (5%) were identified as culture positive and 25 (44.6%) as seropositive. When hemoculture results were compared with those from a similar study performed in 1997, prevalence remained unchanged. Genetic characterization of the 3 culture isolates indicated they belong to the T. cruzi IIa group, which is identical to strains previously isolated from raccoons on the island. Despite the occurrence of T. cruzi in the population, there was no evidence that the health of the lemurs was compromised as a result of infection. Based upon prevalence and available breeding records we speculate that both vertical and vector-mediated transmission play significant roles in the epidemiology of T. cruzi on the island. This also represents the first report of autochthonous infection in blue-eyed black and black-and-white ruffed lemurs.  相似文献

The course of experimental infection of Swiss mice with 95 sylvatic Trypanosoma cruzi isolates included in TCI or TCII genotype was characterized. The purpose was to verify biological properties and its eventual correspondence with original host species, genotype or zymodeme. The isolates of T. cruzi were 100% infective, 55% resulted in patent parasitemia with 69% (36/52) of mortality. A meaningful biological heterogeneity was observed in both, TCI and TCII isolates. TCII isolates resulted in higher patent parasitemia 64% (38/59), in contrast to the 41% TCI infected Swiss mice (14/34). Parasitemia was not always associated to mortality. Higher biological heterogeneity was observed in T. cruzi II isolates derived from L. rosalia from the Atlantic Coastal Rain forest. TCII isolates derived from marsupials resulted in very similar infection profile in Swiss mice.  相似文献

Coastal Fynbos on the Agulhas Plain of the Western Cape, South Africa, has been transformed extensively by crop and pastoral agriculture. Many bird species, e.g. sunbirds, nightjars and mousebirds have consequently been affected negatively by the reduction of suitable habitat for daily requirements. However, populations of certain bird species, especially granivores and omnivores, such as sparrows, larks and pipits, as well as Egyptian Geese and Helmeted Guineafowl have proliferated on the Agulhas Plain. This study investigates the effects of agricultural development on overall bird diversity on the Agulhas Plain. In 1998, twice monthly transect counts at 13 farmland sites recorded 106 bird species. Fifteen species occurred at 75% or more of the sites. Highest avian diversity was recorded at the sites with a mixture of crops and Coastal Fynbos (Mixed regime). A small number of species, e.g. sunbirds, are reliant on the natural Coastal Fynbos for their survival. With the exception of the Mixed regime that gained 50 species, all the other agricultural regimes reflected a loss of diversity when compared with Fynbos. To provide more suitable habitat for a greater diversity of birds, mixed landscapes should be maintained, and further destruction of natural Fynbos habitat should be prevented.  相似文献

A hierarchial population genetic study was conducted on 703 individual Amblyomma americanum from nine populations in Georgia, U.S.A. Populations were sampled from the Coastal Plain, midland Piedmont region, and the upper Piedmont region. Twenty-nine distinct haplotypes were found. A minimum spanning tree was constructed that indicated these haplotypes comprised two lineages, the root of which was distinctly star-like. The majority of the variation found was among ticks within each population, indicating high amounts of gene flow and little genetic differentiation between the three regions. An overall F(ST) value of 0.006 supported the lack of genetic structuring between collection sites in Georgia. Mantel regression analysis revealed no isolation by distance. Signatures of population expansion were detected in the shapes of the mismatch distribution and tests of neutrality. The absence of genetic differentiation combined with the rejection of the null model of isolation by distance may indicate recent range expansion in Georgia or insufficient time to reach an equilibrium where genetic drift may have affected allele frequencies.
