株高是影响植物株型建成的重要农艺性状之一,直接决定作物的倒伏性和生物产量,但目前关于苜蓿等豆科牧草株高性状形成的分子调控机制尚不清楚。通过定向筛选豆科模式植物蒺藜苜蓿Tnt1逆转座子插入突变体库,分离鉴定了一个蒺藜苜蓿矮化突变体compact stalk internodes(costin),该突变体的矮化表型是由于茎节伸长受到抑制所致。通过基因表型连锁分析成功克隆了COSTIN基因,该基因编码一个钙离子交换蛋白,与拟南芥的CALCIUM EXCHANGER 7(CAX7) 基因高度同源。qRT-PCR检测发现COSTIN基因在茎、叶和果荚等组织中有较高的表达。进一步研究发现在costin突变体中赤霉素合成途径关键基因MtCPS、MtKAO1、MtGA20ox4、MtGA20ox7和MtGA3ox1表达下调;外施赤霉素GA3可以恢复costin突变体的矮化表型。上述研究表明COSTIN基因通过影响植物激素赤霉素的生物合成来调控蒺藜苜蓿的茎节伸长。  相似文献

蒺藜苜蓿(Medicago truncatula G)花器官特异表达基因是参与其花器官形成与发育的重要基因。筛选蒺藜苜蓿的花器官特异表达基因,寻找这类基因在其他模式植物中的直系同源基因,并将其表达模式在不同植物间进行比较,有利于深入的理解这类基因在蒺藜苜蓿花器官发育中的功能。根据蒺藜苜蓿表达谱,并以其PISTILLAZA(PI)基因为模板,文章筛选了97个蒺藜苜蓿花器官特异表达基因(Ratio≥10,且Z≥7.9).通过同源比对,确定了这类基因在拟南芥(Arabidopsis thaliana L.)、大豆(Glycinemax L.)、百脉根(Lotusjaponicus L.)和水稻(Oryzasativa L.)中的直系同源基因。对这类基因在5种植物中的表达量、表达部位和功能进行比较,发现进化关系较近的植物,直系同源基因的表达变异较小,而进化关系较远的植物,直系同源基因的表达变异较大。进一步对表达分化较大的直系同源基因进行启动子分析,发现不同植物中直系同源基因表达模式的变化与启动子中调控元件的特性有关。  相似文献

赤霉素受体(GID)是赤霉素信号转导途径的重要成员,直接影响着赤霉素对植物体效应的发挥。该研究利用同源克隆的方法,首次从紫花苜蓿中克隆得到1个赤霉素受体基因,命名为MsGID1b。序列分析发现,MsGID1b基因开放阅读框长度为1 053bp,编码350个氨基酸,推测其蛋白质分子量为39.839kD,是一个无信号肽和跨膜结构的亲水性蛋白。序列比对结果表明,MsGID1b基因与蒺藜苜蓿MtGID1b基因的核苷酸序列相似性为98%,氨基酸序列相似性为99%,且具有HSL家族典型的HGG和GXSXG保守结构域及GA、DELLA蛋白结合位点。荧光定量PCR分析表明,MsGID1b基因在紫花苜蓿各组织中的表达丰度依次为:根盛花初花茎叶荚果;经GA3、ABA、NaCl、PEG和黑暗诱导后该基因表达上调,尤其是在GA3诱导下,MsGID1b基因的表达量一直维持在较高水平,表明MsGID1b基因可能参与紫花苜蓿的抗逆调控。  相似文献

为了解叶绿素合成关键酶基因在大豆不同生育期的表达情况,本研究以拟南芥叶绿素合成关键酶基因序列设计引物,大豆科丰14和它的持绿突变体BN108为试材,利用半定量RT-PCR分析了9种叶绿素合成关键酶基因在开花期和成熟前期表达量的变化。结果表明,HEMA、GSA、HEME、POR、CHLG基因在成熟前期表达量显著下降(p0.05),尤其是HEMA和GSA基因的表达量下降幅度较大,说明上述基因在叶绿素生物合成过程中起到主要调控作用;而突变体BN108有别于科丰14,GSA、HEME、CHLM和CAO基因表达量在成熟前期不降反升,推测持绿突变体在成熟前期叶绿素能够维持较大的合成量,是植物持绿现象的重要原因之一。  相似文献

类黄酮代谢对于植物生长发育和植物-环境互作至关重要,其中糖基转移酶介导的糖基化修饰在类黄酮代谢中发挥着重要作用。为了研究蒺藜苜蓿中糖基转移酶的生物学功能,通过定向筛选蒺藜苜蓿Tnt1逆转座子插入突变体库,获得了一类植株矮小、叶片深绿的突变体small and emerald1 (se1)。通过基因表型连锁性分析成功克隆了SE1基因,该基因编码1个糖基转移酶,与拟南芥中调控类黄酮生物合成的AtUGT84A1氨基酸同源性为52.8%。对野生型和se1突变体叶片的类黄酮含量进行测定发现类黄酮总量在se1突变体中显著降低(P<0.01)。进一步研究发现在se1突变体中类黄酮合成途径关键基因CHS、F3H和F3’H表达水平下降。亚细胞定位显示SE1可能在细胞质和细胞核中发挥生物学功能。研究表明糖基转移酶基因SE1可能参与蒺藜苜蓿类黄酮合成代谢调控,进而影响其生长发育。此外,研究还发现SE1基因对于叶绿素合成可能具有负向调控作用。  相似文献

多药和有毒化合物外排转运蛋白MATE能转运金属、激素、次生代谢物等多种底物,因而在植物的生长发育中发挥重要作用。本研究基于紫花苜蓿(Medicago sativa L.)基因组数据和缺铁胁迫的转录组数据,克隆获得紫花苜蓿MsMATE1及pMsMATE1启动子(GenBank登录号分别为MN547958和MT505313)。MsMATE1基因长1470 bp,编码489个氨基酸。系统进化分析表明,紫花苜蓿MsMATE1蛋白属于mate家族,与蒺藜苜蓿MtMATE(XP013453190.1)亲缘关系最近。生物信息学分析表明,MsMATE1具有典型的MATE_like超家族结构域,属于H+势驱动的真核亚类;MsMATE1为跨膜蛋白,二级结构的主要构成元件是α螺旋。pMsMATE1启动子长1598 bp,序列分析显示内含多个植物激素和逆境胁迫响应元件。MsMATE1在紫花苜蓿幼苗的根茎叶中都有表达,茎中的表达量最高。高铁和缺铁胁迫下,MsMATE1基因在紫花苜蓿幼苗各部位的表达显著上调,茎中上调最明显。高铁和缺铁胁迫,转MsMATE1基因烟草的3种抗氧化酶活性、叶绿素和可溶性蛋白含量显著增加,丙二醛含量显著降低。以上结果表明,MsMATE1在烟草中的异源表达提高了植物抵御高铁和缺铁胁迫的能力。MsMATE1可以作为应用基因工程方法改良植物铁胁迫耐受的重要候选基因,本研究为深入了解MsMATE1蛋白在植物响应铁胁迫中的分子机制奠定基础,MsMATE1基因应对其他金属胁迫或环境胁迫的功能有待于进一步鉴定。  相似文献

CIPK是植物中一类丝氨酸/苏氨酸蛋白激酶,在植物响应逆境胁迫中发挥着重要的作用。本研究根据盐碱胁迫下紫花苜蓿(Medicago sativa L.)转录组数据设计引物,通过RT-PCR克隆获得紫花苜蓿MsCIPK8基因,该基因CDS全长1341bp,编码446个氨基酸,编码蛋白相对分子质量50.73 kD,等电点6.72,具有CIPKS家族蛋白所特有的N端激酶域和C端NAF/FISL结构域。生物信息学分析结果显示,MsCIPK8为可溶性蛋白,二级结构多为无规矩卷曲;系统进化分析表明,紫花苜蓿MsCIPK8与蒺藜苜蓿(Medicago truncatula Gaertn.)MtCIPK8亲缘关系最近。两个蛋白序列比对发现存在4个差异位点,其中3个在保守结构域内。MsCIPK8在低温、干旱、盐和盐碱胁迫下表达量均受到诱导上调表达。低温胁迫下,MsCIPK8在根和叶中的表达量分别在12 h和3 h达到峰值;盐胁迫下,MsCIPK8在根中的表达量12 h达到峰值;盐碱胁迫下,根和叶中MsCIPK8的表达量在12 h后持续高表达;干旱胁迫下,MsCIPK8在根和叶中的表达量在12 h均达到峰值。上述结果表明MsCIPK8参与紫花苜蓿对干旱、低温、盐和盐碱等非生物胁迫的应答。  相似文献

Scirtothrips perseae Nakahara was discovered attacking avocados in California, USA, in 1996. Host plant surveys in California indicated that S. perseae has a highly restricted host range with larvae being found only on avocados, while adults were collected from 11 different plant species. As part of a management program for this pest, a “classical” biological control program was initiated and foreign exploration was conducted to delineate the home range of S. perseae, to survey for associated natural enemies and inventory other species of phytophagous thrips on avocados grown in Mexico, Guatemala, Costa Rica, the Dominican Republic, Trinidad, and Brazil. Foreign exploration efforts indicate that S. perseae occurs on avocados grown at high altitudes (>1500 m) from Uruapan in Mexico south to areas around Guatemala City in Guatemala. In Costa Rica, S. perseae is replaced by an undescribed congener as the dominant phytophagous thrips on avocados grown at high altitudes (>1300 m). No species of Scirtothrips were found on avocados in the Dominican Republic, Trinidad, or Brazil. In total, 2136 phytophagous thrips were collected and identified, representing over 47 identified species from at least 19 genera. The significance of these species records is discussed. Of collected material 4% were potential thrips biological control agents. Natural enemies were dominated by six genera of predatory thrips (Aeolothrips, Aleurodothrips, Franklinothrips, Leptothrips, Scolothrips, and Karnyothrips). One genus each of parasitoid (Ceranisus) and predatory mite (Balaustium) were found. Based on the results of our sampling techniques, prospects for the importation of thrips natural enemies for use in a “classical” biological control program in California against S. perseae are not promising.  相似文献

Parsimony analyses of the internal transcribed spacer regions of nuclear ribosomal DNA (ITS 1 & ITS 2) for 38 taxa sampled from the Phebalium group (Rutaceae: Boronieae) and two outgroups confirm that, with the exception of Phebalium sensu stricto and Rhadinothamnus, six of the currently recognised genera within the group are monophyletic. The data indicate that Phebaliums. str. is paraphyletic with respect to Microcybe, and Rhadinothamnus is paraphyletic with respect to Chorilaena. Rhadinothamnus and Chorilaena together are the sister group to Nematolepis. Drummondita, included as an outgroup taxon, clustered within the ingroup as sister to Muiriantha and related to Asterolasia.The phylogeny suggests that the evolution of major clades within a number of these genera (e.g. Phebalium) relates to vicariance events between eastern and south-western Australia. Leionema is an eastern genus, with the most basal taxon being the morphologically distinct Leionema ellipticum from northern Queensland. Leionema also includes one species from New Zealand, but this species (as with some others) proved difficult to sequence and its phylogenetic position remains unknown. Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.

Komárek has recently reviewed the various species assigned to the green algal genusNeochloris Starr (Chlorococcales, Chlorococcaceae) and removed those with uninucleate vegetative cells to a new genus,Ettlia. Watanabe & Floyd, unaware ofKomárek's work, also reviewed the species ofNeochloris and distributed them among three genera—Neochloris, Chlorococcopsis gen. nov., andParietochloris gen. nov.—on the basis of details of the covering of the zoospore and the arrangement of the basal bodies of the flagellar apparatus. Little is known about how tropical land-use systems contribute to the conservation of functionally important insect groups, including dung beetles. In a study at the margin of Lore Lindu National Park (a biodiversity hotspot in Central Sulawesi, Indonesia) dung-beetle communities were sampled in natural forest, young secondary forest, agroforestry systems (cacao plantations with shade trees) and annual cultures (maize fields), each with four replicates (n = 16 sites). At each site we used 10 pitfall traps, baited with cattle dung, along a 100 m transect for six 3-day periods. The number of trapped specimens and species richness at the natural forest sites was higher than in all land-use systems, which did not significantly differ. Each land-use system contained, on average, 75% of the species richness of the natural forest, thereby indicating their importance for conservation. However, a two-dimensional scaling plot based on NESS indices (m = 6) indicated distinct dung beetle communities for both forest types, while agroforestry systems and annual cultures exhibited a pronounced overlap. Mean body size of dung beetles was not significantly influenced by land-use intensity. Five of the six most abundant dung beetle species were recorded in all habitats, whereas the abundance of five other species was significantly related to habitat type. Mean local abundance and number of occupied sites were closely correlated, further indicating little habitat specialisation. The low dung beetle diversity (total of 18 recorded species) may be due to the absence of larger mammals in Sulawesi during historical times, even though Sulawesi is the largest island of Wallacea. In conclusion, the dung beetle fauna of the lower montane forest zone in Central Sulawesi appears to be relatively robust to man-made habitat changes and the majority of species did not exhibit strong habitat preferences.  相似文献

The genusKarschia, in the earlier sense, including saprophytes and parasites on lichens, has been thought to be a non-lichenized parallel genus of the lichen genusBuellia. Modern workers included it on the one hand inBuellia, on the other hand combined it with bitunicate ascomycetes. It is now proved thatKarschia is heterogeneous and contains but superficially similar members both of the genusBuellia of theLecanorales and of typical or masked bitunicateAscomycetes. Therefore, it can not be regarded as a link betweenLecanorales andDothideales. The type species ofKarschia belongs to theDothideales.
