不同木质纤维素基质上白腐菌降解特性的研究   总被引:14,自引:0,他引:14  
通过测定木质素、纤维素、半纤维素和漆酶分泌的变化,研究白腐菌在稻草、木屑、粗纤维素、滤纸、黑液木素基质上的降解特性。结果表明,除黑液木素上白腐菌不能生长外,在前25d,各基质中纤维素、半纤维素和木质素含量呈持续下降趋势,之后,降解速率减少,其中木质素的降解速率大于纤维素和半纤维素的降解速率。漆酶分泌在生长初期呈快速上升趋势,第10d酶活达到最大,第10~20d快速下降,其后基本不变,基质中酶活大小顺序为稻草基质、木屑基质、粗纤维和滤纸基质,显示了木质素存在对漆酶分泌的诱导作用。  相似文献

漆酶是一种绿色高效的多酚氧化酶,在降解双酚A方面具有巨大潜力。为降低发酵产漆酶的成本及考察漆酶在双酚A降解中的能力,本研究以麸皮和柚皮为主要基质,优化了栓菌固态发酵产漆酶条件,对优化后获得的漆酶在双酚A降解中的应用进行了研究。结果表明,在培养基组分为:麸皮和柚皮粉比例为6:4(W/W)、固液比1:2.5(W/V)、铜离子2%(W/W)、蔗糖3%(W/W)、硝酸钾2%(W/W)、稻壳20%(W/W)的条件下,栓菌发酵产漆酶的酶活最高,发酵11d后,酶活可达到38.4U/gds。当双酚A初始浓度为10μg/mL时,在55℃条件下酶解140min后,双酚A基本降解完全。  相似文献

松软毡被孔菌是一种可产漆酶的多孔菌,漆酶可用于废水处理、降解有毒物质等,因此对采自黑龙江胜山国家级自然保护区的松软毡被孔菌进行生物学特性的研究,可为漆酶的开发、生产提供基础数据。根据野外采集的标本和出菇子实体形态学特性的分析和分子生物学(ITS,rDNA)比较研究,通过形态学观察和分子工具辅助鉴定,确定为松软毡被孔菌。同时对其适宜碳源、氮源、pH和温度进行初步研究,研究表明适合松软毡被孔菌生长适宜条件:碳源为葡萄糖、唯一氮源为酵母浸粉、温度为30℃、pH值为8.0。松软毡被孔菌可在人工培养基上(培养料配方:78%阔叶树木屑,20%麸皮,1%石膏,1%蔗糖)形成子实体。本研究结果为漆酶的开发研究、药理活性研究提供理论依据。  相似文献

毛栓菌胞外漆酶经盐析、透析、sephadexG75和G2 5四步纯化 ,粗酶液被纯化了 39 1倍 ,比活力 12 152 ,回收率 4 5 3%。漆酶最适pH值为 4 0 ,最适反应温度为 30℃。K Cu 2 、Zn2 离子可激活漆酶 ,而Ag 、Fe3 离子可抑制漆酶的活性。漆酶的Km值为 1 81× 10 3mol/L。  相似文献

本文旨在研究阿魏蘑菌体形态与漆酶产量之间的关系。结果显示,玻璃珠的添加可改变发酵过程中菌体形态,漆酶产量在球状菌体条件下高于丝状、絮状菌丝:直径分布在0.2~0.4 mm范围的菌球对漆酶的合成具有明显的促进作用;适合的葡萄糖、玉米粉和麸皮添加量,对直径在0.2~0.4 mm范围的菌球形成具有重要影响。此外,添加惰性载体同样可以控制菌球的直径分布,但对漆酶的合成无促进作用。  相似文献

【目的】为提高漆酶产量,降低生产成本,以山核桃蒲壳作为基质,对粗毛栓菌D2固态发酵产漆酶的营养条件进行研究。【方法】对不同碳源、氮源、碳氮比、蒲壳含量对漆酶产量的影响进行分析。【结果】山核桃蒲壳是粗毛栓菌生长的良好载体,能够促进漆酶的合成。粗毛栓菌D2漆酶固态发酵培养基干物质组成为:山核桃蒲壳40%(质量比),玉米粉24%(质量比),菜籽饼粉36%(质量比)。发酵6 d时,漆酶活性为126.8 U/g干基。【结论】粗毛栓菌固态发酵山核桃蒲壳产漆酶具有效率高,生产成本低的优点,具有潜在的工业化应用前景。  相似文献

以稻草粉和麸皮为主要原料,对白腐菌(White-rot fungi) NS75、黑曲霉(Aspergillus niger)NS83和絮凝酵母(Saccharomyces cerevisiae)SP5混合菌固态发酵产纤维素酶进行研究.实验结果显示,在白腐菌和黑曲霉双菌混合培养2d后接入絮凝酵母,培养到第7d产酶达到峰值;三菌混合发酵产纤维素酶酶活明显高于白腐菌和黑曲霉双菌混合培养,其β-葡萄糖苷酶(β-G)和羧甲基纤维素酶(CMCase)酶活比白腐菌(White-rot fungi) NS75和黑曲霉(Aspergillus niger)NS83双菌发酵产酶分别提高了143.3%和68.2%.单因素实验和正交实验结果表明,当稻草粉麸皮质量比为8∶2,料水比为1∶2,白腐菌NS75、黑曲霉NS83和絮凝酵母SP5的接种比例为1:2∶1.5 (v/v/v)时,于30℃培养7d,固态发酵基中β-G和CMCase酶活分别达到62305 U/g和30241 U/g.  相似文献

利用筛选的绿色木霉ZY-1(Trichoderma viride ZY-1)固态发酵产纤维素酶,采用稻草和麸皮为底物,考察稻草与麸皮比例随发酵时间对产酶的影响。结果表明:底物中,在m(稻草):m(麸皮)为0:5和1:4时,发酵48h,pH保持4.5左右,还原糖量急剧上升,胞外蛋白产量最低;仅以稻草作底物时,整个发酵过程中pH约为7,还原糖量最低,胞外蛋白产量较高而滤纸酶活、羧甲基纤维素酶(CMCase)和β-葡萄糖苷酶(β-Gase)酶活均较低;在m(稻草):m(麸皮)为3:2时,发酵96h,滤纸酶活达最大值5.01U/g干曲;m(稻草):m(麸皮)为1:4时,发酵96h,β-Gase酶活达最大值4.6U/g干曲;m(稻草):m(麸皮)为4:1时,发酵72h,CMCase酶活达最大值6.01U/g干曲。因此,底物中存在适量的稻草和麸皮有利于Trichoderma viride ZY—1产纤维素酶。  相似文献

以稻草秸秆加20%棉籽壳为培养基质,接种侧耳Z17、921、1024菌株,在不同生长阶段。测定培养物的主要化学成分和有关酶活的变化。结果表明,从接种到子实体形成,所试菌株培养物的纤维素、木质素等呈持续不断的下降趋势,水份、粗蛋白含量却逐渐升高,基质中漆酶酶活性在菌丝生长初期呈迅速上升趋势,后稍降低,而愈创木酚酶活性在菌丝上长初期及子实体形成时达到高峰,后有所降低或消失。  相似文献

本研究经过选择性培养基分离及特异性底物筛选从干旱区荒漠植物松叶猪毛菜和红砂灌丛土壤中分离出两株产漆酶的真菌Z45和H53,扩增其ITS序列进行分子鉴定,并分析温度、碳源、氮源、碳氮比和pH对菌株Z45生长的影响,在此基础上利用正交试验优化其培养条件.结果 表明,产漆酶真菌Z45和H53均隶属于端梗霉属(Acrophialophora sp.),Z45温度适应范围广且耐高温,最适生长温度为40℃.最优培养基组合为:分别以麦芽糖和硝酸铵为碳、氮源,按15∶1的碳源/氮源比配制,pH值6.0.本研究从干旱区荒漠土壤中获得了1株耐高温、产漆酶、应用价值高的子囊菌端梗霉,丰富了产漆酶微生物的种质资源库.  相似文献

Reactive dyes are one of the major sources of waste-water pollution. Efficient degradation of these dyes with enzymes produced from agricultural waste has attracted tremendous recent interests in both the scientific community and the general public. In this study, we took advantage of solid-state fermentation of four agro-byproducts (rape stem, wheat bran, peanut shell and rice hull) for producing laccase from the fungus Trametes sp. AH28-2. Higher laccase activities were obtained in multiple-substrates media than in single substrate media. The maximum yield of laccase (2.10 × 106 U/kg) was obtained in the medium containing 60% rape stem, 20% peanut shell and 20% wheat bran, without the supplement of any toxic inducers. Our results further demonstrated that the textile reactive dyes Levafix Blue CA and Cibacron Blue FN-R (1.0 g/l) were completely decolorized by the crude laccase (5.0 U/l) obtained within 15 h in the absence of any mediator. Therefore, the agro-byproducts could be re-utilized to produce laccase for the decolorization of textile reactive dyes. Q.Y. Sun and Y.Z. Hong contributed equally to the study.  相似文献

粗糙脉孢菌为子囊菌中的高效纤维素降解菌,可以直接以纤维素为营养源进行生长。本研究以粗糙脉孢菌为实验对象,利用基因工程技术构建甾醇还原酶基因erg24的高表达菌株,分别以蔗糖、麦麸、玉米秸秆、小麦秸秆、杨树木屑、水稻秸秆6种物质的粉末为碳源培养野生型粗糙脉孢菌和erg24高表达菌株,利用半定量RT-PCR测定在不同培养条件下erg2erg24erg6 3个麦角甾醇合成相关基因的表达水平,采用HPLC方法测定不同培养条件下麦角甾醇的积累量。研究结果表明,分别以玉米秸秆、杨树木屑、水稻秸秆这3种粉末为碳源时,培养物中的erg2erg24erg6 3个基因表达量较高。在不同培养条件下erg24高表达菌株合成麦角甾醇量显著高于野生型粗糙脉孢菌的合成量,且以杨树木屑粉末为碳源培养时,所获得的麦角甾醇产量最高,为30.53μg/mg。结果表明erg24基因是粗糙脉孢菌合成麦角甾醇的关键基因之一,利用玉米秸秆、小麦秸秆、杨树木屑或水稻秸秆粉末为碳源培养粗糙脉孢菌时,可获得较高产量的麦角甾醇。研究结果为以农业废弃物为营养源,利用真菌生产麦角甾醇奠定了基础。  相似文献

本文旨在探明家蝇Musca domestic幼虫对高粱秸秆和小麦秸秆营养成分利用和木质纤维素的降解情况,以及不同配比秸秆饲料对家蝇生物学指标的影响,为今后产业化利用家蝇来生物降解秸秆类有机废弃物奠定理论基础.本研究采用70%高粱秸秆粉、70%麦秸秆粉和纯麦麸作为饲料,每种饲料各设置对照组、发酵组和家蝇幼虫取食组3种处理,检测并分析3种饲料各处理组中一般营养成分和木质纤维素的含量及变化;同时采用7种配比饲料喂养家蝇,观察各组家蝇不同发育阶段的体长、体重、存活率和发育历期等.结果表明:(1)3种饲料的各处理组中,一般营养成分和木质纤维素的含量均呈现:对照组>发酵组>家蝇取食组的趋势,此外除高粱秸秆组粗蛋白外,其余各组成份均差异显著(P<0.05);(2)小麦秸秆的木质纤维素含量>高粱秸秆>麦麸,且各组间均差异显著(P<0.05);(3)在相同秸秆配比条件下,高粱秸秆饲喂的家蝇其各虫态体长、体重、总存活率和发育历期均要优于麦秸秆喂养,且总存活率和发育历期与麦麸喂养相比差异不显著(P>0.05);(4)纯高粱秸秆喂养下家蝇仍能保持正常的生长发育,而在90%小麦秸秆喂养下,家蝇则无法存活.综上所述,家蝇适合作为生物转化器用于秸秆、尤其是高粱秸秆的降解,且降解效果优于单纯的微生物发酵降解,研究结果为今后规模化利用家蝇降解秸秆类有机废弃物提供了理论依据和科学基础.  相似文献

Aim:  To produce high laccase activities from the white-rot fungus Fomes fomentarius .
Methods and Results:  Different culturing methods, viz, cell immobilization on stainless steel sponges and plastic material and solid-state fermentation (SSF) using wheat bran as substrate were used for laccase production by the white-rot fungus F. fomentarius . The SSF study expresses the highest laccase activities, nearly to 6400 U l−1 after 13 days of laboratory flasks cultivation. When the wheat bran medium was supplemented with 2 mmol l−1 copper sulfate, laccase activity increased by threefold in comparison to control cultures, reaching 27 864 U l−1. With the medium thus optimized, further experiments were performed in a 3 l fixed-bed bioreactor (working volume 1·5 l) leading to a laccase activity of about 6230 U l−1 on day 13.
Conclusions:  The results obtained clearly showed the superiority of wheat bran for laccase production over stainless steel sponges and plastic material. Supplementing the wheat bran solid medium with 2 mmol l−1 copper sulfate allowed obtaining high activities at flask scale. The system was scaled to fixed-bed laboratory reactor.
Significance and Impact of the Study:  The high enzyme production along with the low-cost of the substrate, showed the suitability of the system F. fomentarius – SSF for industrial purposes.  相似文献

Trametes versicolor was shown to produce extracellular laccase during surface cultivation on glucose, wheat straw and beech wood. Growth on both wheat straw and beech wood led to an increase as high as 3.5-fold in extracellular laccase activity, in comparison with growth on glucose. The corresponding yields in fungal biomass reached only about 20% of the value obtained on glucose. Manganese peroxidase activity␣appeared during growth on wheat straw and beech wood. Mycelia grown on glucose, wheat straw and beech wood also showed intracellular laccase activities, monitored with 2,6-dimethoxyphenol, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid), 4-hydroxy-3,5-dimethoxybenzaldehyde azine (syringaldazine) and 3,4-dihydroxyphenylalanine (l-DOPA). Assaying intracellular laccase with 2,6-dimethoxyphenol, syringaldazine and l-DOPA showed the maximum oxidation rates to be at pH values different from those producing maximum oxidation rates with extracellular laccase. In each case most of the total laccase activity was recovered from the culture filtrates. Growth on wheat straw and beech wood led to increased values for both extra- and intracellular laccase activities, based on fungal dry weight, in comparison with growth on glucose. 用愈创木酚平板法对14株白腐真菌进行初筛,通过测定漆酶活力进行复筛,筛选出1株生活力较强,产漆酶活力高的菌株MZ-1,经ITS-5.8S rDNA序列分析,初步鉴定为Trametes versicolor。在固态发酵培养基的基础上,对该菌株产漆酶的培养基组成进行正交优化,得到最优发酵培养基:麸皮∶秸秆粉∶豆粕∶玉米粉为3∶3∶2∶1,可溶性淀粉2%,(NH4)2SO4+蛋白胨1%,KH2PO40.1%,料水比1∶2。接种4个菌塞,温度为30℃,发酵8 d后酶活可达到1 555.57 U/g。  相似文献

Cultivation of the oyster mushroom, Pleurotus sajor-caju, on rice and wheat straw without nutrient supplementation was investigated. The effects of straw size reduction method and particle size, spawn inoculation level, and type of substrate (rice straw versus wheat straw) on mushroom yield, biological efficiency, bioconversion efficiency, and substrate degradation were determined. Two size reduction methods, grinding and chopping, were compared. The ground straw yielded higher mushroom growth rate and yield than the chopped straw. The growth cycles of mushrooms with the ground substrate were five days shorter than with the chopped straw for a similar particle size. However, it was found that when the straw was ground into particles that were too small, the mushroom yield decreased. With the three spawn levels tested (12%, 16% and 18%), the 12% level resulted in significantly lower mushroom yield than the other two levels. Comparing rice straw with wheat straw, rice straw yielded about 10% more mushrooms than wheat straw under the same cultivation conditions. The dry matter loss of the substrate after mushroom growth varied from 30.1% to 44.3%. The straw fiber remaining after fungal utilization was not as degradable as the original straw fiber, indicating that the fungal fermentation did not improve the feed value of the straw.  相似文献

In this study, Bacillus sphaericus NRC 69 was grown in culture media, in which 12 agricultural wastes were tested as the main carbon, nitrogen and energy sources under solid state fermentation. Of the 12 tested agricultural by-products, wheat bran was the most efficient substrate for the production of B. sphaericus mosquitocidal toxins against larvae of Culex pipiens (LC50 1.2 ppm). Mixtures of tested agricultural wastes separately with wheat bran enhanced the produced toxicity several folds and decreased LC50 between 3.7- and 50-fold in comparison with that of agricultural wastes without mixing. The toxicity of B. sphaericus grown in wheat bran/rice hull at 8/2 (g/g) and wheat bran/barley straw at 1/4 (g/g) showed the same toxicity as that in wheat bran medium (LC50 decreased 17- and 16-fold, in comparison with that in rice hull or barely straw media, respectively). In wheat bran medium, the maximum toxicity of the tested organism obtained at 50% moisture content, inoculum size 84 × 106 CFU/g wheat bran and incubation for 6 days at 30°C. Addition of cheese whey permeate at 10% to wheat bran medium enhanced the toxicity of B. sphaericus NRC 69 about 46%.  相似文献
