[目的]西花蓟马Frankliniella occidentalis (Pergande)是番茄斑萎病毒(Tomato spotted wilt orthotospovirus,TSWV)的主要传播媒介,以循环增殖方式传播TSWV,但在传播过程中参与的互作蛋白尚不清楚。本研究以TSWV的外鞘糖蛋白GN (GlycoproteinN)为诱饵筛选与西花蓟马互作的蛋白,为阐明TSWV与西花蓟马的互作机制提供依据。[方法]构建以pPR3-N为载体的西花蓟马泛素分裂酵母双杂交膜系统cDNA文库,利用Gene Ontology (GO)通路注释互作蛋白,分析其生物学功能。[结果]西花蓟马cDNA文库库容为3.2×10~6,除去重复序列、载体序列和移码序列,筛选到74个与G_N互作的蛋白,参与了细胞过程、代谢过程、生物调节过程等12种生物过程。[结论]成功构建了西花蓟马酵母双杂交膜体系cDNA文库,筛选到与TSWV GN互作的蛋白,为进一步研究TSWV与西花蓟马的互作机制奠定了基础。  相似文献

入侵害虫西花蓟马及其他8种常见蓟马的分子鉴定   总被引:12,自引:0,他引:12  
用PCR产物直接测序法对入侵害虫西花蓟马和其他8种蓟马的线粒体 COⅠ基因433 bp片段测序,获得62个个体的序列。分子数据分析显示: 种内个体间平均遗传距离在0~0.005之间,2003年在北京发现的西花蓟马与欧洲等地区报导的西花蓟马不存在明显的遗传差异; 9种蓟马种间平均遗传距离为0.213。构建的NJ树可以很好的显示蓟马的聚类,物种各单元型最初分支自展值均达到100%。结果表明,基于PCR及直接测序技术的分子鉴定可以达到准确鉴定蓟马物种之目的。  相似文献

为明确榕管蓟马Gynairothrips uzeli Zimmerman最嗜颜色及粘虫色板田间诱集效果,以垂叶榕种植基地为试验点,比较榕管蓟马对10种颜色的嗜好差异,测定粘虫板颜色与悬挂高度、颜色与悬挂密度对榕管蓟马的互作效应,查验3种偏嗜颜色-悬挂高度-悬挂密度组合粘虫板的田间诱集效果。结果表明:榕管蓟马最嗜颜色为黄色,其次为紫色和青色,对其他7种颜色的嗜好较弱;粘虫板颜色、悬挂高度和悬挂密度均极显著影响榕管蓟马诱集,颜色与悬挂高度、颜色与悬挂密度对榕管蓟马诱集存在显著的互作效应;黄、青、紫等3种偏嗜颜色粘虫板的悬挂高度均以色板底端高出垂叶榕顶部10 cm为佳,适宜悬挂密度则分别为4片/100 m2、5片/100 m2和5片/100 m2;3种偏嗜颜色-悬挂高度-悬挂密度组合中,黄色组合粘虫色板对榕管蓟马的田间诱集效果最好。  相似文献

西花蓟马Frankliniella occidentalis(Pargande)是世界性害虫,2003年在我国首次发生危害。针对西花蓟马与其他种类蓟马形态相似、难以快速区分的问题,本文在SCAR标记基础上,采用TaqMan实时荧光定量PCR技术,设计1对特异性引物和1条MGB探针,扩增出大小为138bp的特异片段。以质粒DNA为标准品建立了标准曲线(R2=0.9965),种特异性检验结果显示,该引物和探针只能检测到西花蓟马的荧光信号,而对其他种类的蓟马不具有检测能力。并且可以定量检测西花蓟马不同虫态靶标DNA片段的拷贝数。该检测体系重复性强、稳定性高,在口岸检疫以及植物种苗及其产品调运中的有害生物检测和监测中具有重要意义。  相似文献

【背景】外来入侵害虫西花蓟马于2003年首次在北京郊区被发现,并逐渐扩散蔓延,目前已在我国局部地区暴发成灾。【方法】于2008~2009年通过定点调查的方法对山东省115个地区258个样点的主要花卉上的蓟马种类进行了分析。【结果】山东省主要花卉上发生的蓟马有11种,包括花蓟马、黄胸蓟马、黄蓟马、西花蓟马、棕榈蓟马、烟蓟马、禾花蓟马、横纹蓟马、腹小头蓟马、黑白纹蓟马、油加律带蓟马等,其优势种为花蓟马。外来入侵害虫西花蓟马在山东省18个地区的花卉上被发现。其中,荣成市区发生最重,占蓟马采集总量的51.86%;德州市区和青岛市区发生次之,分别占蓟马采集总量的34.21%和33.88%;其他地区西花蓟马发生量占采集总量的1.39%~18.75%。【结论与意义】西花蓟马已在山东省定殖并广泛分布,相关部门应做好西花蓟马的预防与控制工作。  相似文献

2013年对广东地区榕树虫瘿中的蓟马种类进行了调查,共发现榕母管蓟马Gynaikothrips ficorum(Marchal)、榕管蓟马Gynaikothrips uzeli(Zimmerman)、榕端宽管蓟马Mesothrips jordani Zimmermann、拉马棘腿管蓟马Androthrips ramachandrai Karny和黄胫棘腿管蓟马Androthrips monsterae(Moulton)5种管蓟马科种类,其中黄胫棘腿管蓟马为中国新纪录种。榕母管蓟马和榕管蓟马为营瘿蓟马,诱导初始虫瘿具有寄主专一性,榕母管蓟马是细叶榕Ficus microcarpa初始虫瘿的诱导者,而榕管蓟马仅在垂叶榕Ficus benjamina Linn.上诱导形成虫瘿;但当两种榕树种植在一起时,榕母管蓟马和榕管蓟马偶尔会入侵到对方成熟虫瘿中并与之生活在一起。榕端宽管蓟马属于盗寄生种类,而拉马棘腿管蓟马和黄胫棘腿管蓟马主要捕食营瘿蓟马的幼体(包括卵、若虫和"蛹"),三者均为寄居性蓟马,无寄主专一性。  相似文献

西花蓟马是近年来在我国局部地区暴发成灾的重要外来入侵害虫,有关西花蓟马入侵对本地蓟马种群动态、空间分布及优势种影响的报道较少。对云南省昆明市近郊蔬菜花期的蓟马种群动态和空间分布研究表明,蔬菜上的蓟马种类主要是西花蓟马Frankliniella occidentalis(Pergande)、花蓟马F.intonsa(Trybom)、棕榈蓟马T.palmi(Karny)和端大蓟马Megalurothrips distalis(Karny);不同蔬菜上的蓟马优势种存在一定差异,其中辣椒和茼蒿上的蓟马优势种为西花蓟马;韭菜、茄子和四季豆上的蓟马优势种分别为花蓟马、棕榈蓟马和端大蓟马。各蔬菜上的蓟马种群数量以花期为多,盛花期达最大值,其中茄子花上的蓟马成虫平均虫口密度最高,为14.93头/朵。利用聚集度指标进行空间分布检测表明,不同蔬菜上蓟马成虫的空间分布型均为聚集分布,且聚集程度随密度的增加而增大。本研究可为深入探讨西花蓟马对本地蓟马的竞争取代机制积累资料,同时为西花蓟马的综合治理奠定理论基础。  相似文献

对广西省所有地市的30多个县植食性蓟马种类及其分布进行了调查。调查结果表明:花蓟马Frankliniella intonsa Trybom、华简管蓟马Haplothrips chinensis Priesner、棕榈蓟马Thrips palmi Karny、黄胸蓟马Thrips hawaiiensis Morgan的分布最广且数量也最丰富,是广西省最为常见的蓟马种类。调查中发现花蓟马属Frankliniella Karny的1个国内新纪录种:梳缺花蓟马Frankliniella schultzei Trybom。编制了广西省常见植食性蓟马成虫种类检索表,为从事广西省蓟马种类研究者提供参考。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
