摘要:【目的】从耐碱性木聚糖酶高产短小芽孢杆菌中克隆得到带有自身启动子的木聚糖酶基因,将其在巨大芽孢杆菌中进行表达,并对表达产物进行性质分析。【方法】将克隆得到的木聚糖酶基因xynA以及带有自身启动子序列的结构基因, 构建在芽孢杆菌表达载体pWH1520和改造后的载体pWG03中,得到重组质粒pWTEJX和pWGXYN,分别转化到巨大芽孢杆菌BM70中,获得重组巨大芽孢杆菌BMJXH9和BMGpp12;经过诱导产酶培养,均得到分泌表达。【结论】重组巨大芽孢杆菌BMGpp12比BMJXH9产酶活力提高了三倍  相似文献

【目的】凝结芽孢杆菌(Bacillus coagulans)是一种非常有工业应用前景的微生物,但遗传转化的困难是限制对其进行代谢工程改造的关键因素。本研究主要考察生长培养基、电击缓冲液、复苏培养基中添加高渗剂如山梨醇、甘露醇等对凝结芽孢杆菌转化效率稳定性的影响,并对凝结芽孢杆菌电击转化条件进行优化。【方法】用穿梭质粒p NW33N电转化凝结芽孢杆菌P4-102B,系统地考察高渗条件下细胞生长阶段、感受态菌体浓度、电击缓冲液组分和复苏培养基组成等因素对转化效率的影响。【结果】同一电场压力下高渗体系转化效率较低渗体系明显提高且稳定性较好,菌体在含有0.5 mol/L山梨醇的LB培养基中生长到OD600为0.8时收集,用SMG[0.5 mol/L山梨醇,0.5 mol/L甘露醇,10%(质量体积比)甘油]电击缓冲液洗涤菌体4次制备感受态,在固定电场强度14 k V/cm、脉冲时间5 ms、1 mm规格的电转杯进行电击转化,电转化后立即加入含有0.5 mol/L山梨醇和0.38 mol/L甘露醇的LB复苏培养基培养,能够获得最佳的转化效率2.7×102 CFU/μg DNA。【结论】使用高渗电击转化法能够提高电击转化的稳定性和重复性,并且可以获得较高的转化效率。  相似文献

【背景】感染产气荚膜梭菌会引起动物坏死性肠炎,通常使用抗生素进行预防和治疗。随着我国饲料禁抗、养殖减抗的实施,寻找绿色微生态制剂及其代谢产物成为当前研究的热点。【目的】旨在研究前期筛选的一株抑制产气荚膜梭菌的枯草芽孢杆菌BS-2特性。【方法】检测了菌株生长曲线、代谢物质的抑菌特性及细菌素基因簇mRNA表达。【结果】枯草芽孢杆菌BS-2代谢物质对革兰氏阴性菌无抑制作用,而对革兰氏阳性菌具有较强的抑菌性能,并且对产气荚膜梭菌的抑菌性能在2-12 h内迅速增长,在12-24 h内抑菌性能较稳定;该抑菌性能不受胃蛋白酶、胰蛋白酶、蛋白酶K的影响,具有良好的热稳定性;进一步分析抑菌物质基因簇mRNA表达,发现枯草芽孢杆菌BS-2抑制产气荚膜梭菌的活性可能与表面活性素(surfactin)和美杀菌素(mersacidin)表达有关。【结论】枯草芽孢杆菌BS-2对产气荚膜梭菌具有较强的抑制作用,可能通过抑菌物质surfactin和mersacidin表达发挥作用。  相似文献

【目的】实现地衣芽孢杆菌麦芽糖淀粉酶在枯草芽孢杆菌中的高效异源表达,并研究该重组酶的酶学性质。【方法】克隆巨大芽孢杆菌木糖异构酶基因的启动子区域及其调控蛋白,构建一个大肠杆菌/芽孢杆菌穿梭型诱导表达质粒,使用该诱导型启动子介导麦芽糖淀粉酶编码基因,实现其在枯草芽孢杆菌中的功能表达。对重组枯草芽孢杆菌的诱导条件进行优化,提高麦芽糖淀粉酶的产量。【结果】获得了诱导表达麦芽糖淀粉酶基因的重组枯草芽孢杆菌菌株。最适诱导温度为45°C,最适诱导剂添加浓度为1%,最适添加诱导剂时间为接种培养9 h后。重组酶蛋白分子量大小为67 k D,对该酶的酶学性质研究发现,以可溶性淀粉为底物,反应生成麦芽糖和葡萄糖,其中麦芽糖含量为60.42%。重组酶最适作用温度为45°C,最适作用p H为6.5,Ca2+、Co2+、EDTA对该重组麦芽糖淀粉酶具有激活作用。【结论】通过木糖诱导表达系统可以实现麦芽糖淀粉酶在枯草芽孢杆菌中的高效诱导型表达,酶活最高可达296.64 U/m L发酵液,在工业上有着较好的应用前景。  相似文献

【背景】微生物蛋白酶已经成为工业用蛋白酶的主要来源,筛选具有特殊环境适应性的微生物成为生物酶资源的开发热点。【目的】通过对青藏高原土壤微生物产蛋白酶菌株的筛选、优化及相关特性研究,寻找新的蛋白酶资源,为高原菌种资源利用提供科学依据。【方法】采用形态学和分子生物学对筛选菌株进行菌种鉴定,利用单因素试验和正交试验对菌株进行发酵条件优化及酶学性质的探究。【结果】筛选出一株高产蛋白酶菌株XC2,经鉴定菌株XC2为枯草芽孢杆菌(Bacillus subtilis)。XC2最优产酶条件:可溶性淀粉4.0%,牛肉膏1.0%,K~+0.6%,培养温度34°C、初始pH 7.0、接种量2.0%的条件下200 r/min振荡培养13 h,所产蛋白酶活力最高为638.5 U/mL。XC2所产蛋白酶最适反应温度60°C,最适pH9.0;40-50°C、pH8.0-10.0条件下酶活稳定性较高;Mn~(2+)对酶活力有明显激活作用,而Zn~(2+)、Cu~(2+)、Fe~(2+)、Fe3+对酶活力有明显抑制作用。【结论】枯草芽孢杆菌XC2有较强的产碱性蛋白酶的能力,具有较好的应用前景。  相似文献

【目的】利用基因敲除技术构建突变菌株BS-AP-K来研究枯草芽孢杆菌的分泌型氨肽酶对菌体生长的作用。【方法】基于Xer/dif重组系统敲除Bacillus subtilis 168基因组中ywaD基因,研究比较野生型与BS-AP-K菌株在不同培养基中的生长情况。【结果】通过比较两菌株的生长情况,发现敲除分泌型氨肽酶会对菌体生长带来不利影响,而这种影响可以通过在培养基中添加多种游离氨基酸来弥补。【结论】研究结果表明胞外氨肽酶通过酶切外源蛋白质以及多肽来为细胞生长提供营养所需。  相似文献

【目的】通过对一株地衣芽孢杆菌来源的角蛋白酶N端进行分子改造,研究其对角蛋白酶活力和热稳定性的影响,进而提高角蛋白酶的热稳定性。【方法】将角蛋白酶N端前5个氨基酸进行分段缺失,并通过序列比对将N端的前5个氨基酸替换为来源于Thermoactinomyces vulgaris的嗜热蛋白酶的N端,将野生型和突变体角蛋白酶基因在枯草芽孢杆菌WB600中进行表达,并对重组酶进行纯化与酶学性质研究。【结果】角蛋白酶N端不同长度的缺失大幅度地降低了角蛋白酶的活力,其中缺失前5个氨基酸完全丧失了酶活力。将角蛋白酶N端前5个氨基酸替换为嗜热蛋白酶N端前12个氨基酸,虽然降低了近70%的活力,但是却增加了角蛋白酶的热稳定性,60°C条件下的半衰期t1/2由原来的9 min提高到20 min。【结论】角蛋白酶的N端对其酶活力具有较大的影响,与嗜热蛋白酶来源的N端进行替换可以有效提高角蛋白酶的热稳定性。  相似文献

【目的】克隆嗜热脱氮土壤芽孢杆菌中的β-葡萄糖苷酶基因bglB,在E.coli中异源表达,纯化并研究其酶学性质。【方法】利用PCR技术从嗜热脱氮土壤芽孢杆菌的基因组DNA中克隆得到bglB基因,将该基因克隆到表达载体pGEX-2TL上并在大肠杆菌BL21(DE3)中表达,对纯化后的β-葡萄糖苷酶的酶学性质及寡聚状态进行分析。【结果】重组表达的β-葡萄糖苷酶最适温度为65°C,最适pH为7.0,能在pH 5-10、60°C下稳定存在4 h,并能在较高的离子强度(880 mmol/L K+)下发挥其功能。Al3+离子对其有强烈的激活作用,Co2+有一定的抑制作用。最适反应条件下该酶比活力为0.043 IU/mg。该酶具有多种寡聚体形式,这些寡聚体均有β-葡萄糖苷酶活性。【结论】获得一个耐热耐盐的中性β-葡萄糖苷酶,为进一步研究β-葡萄糖苷酶的催化作用机理,提高其热稳定性提供一定的帮助。  相似文献

【目的】采用响应面法(RSM)优化枯草芽孢杆菌5 L发酵罐产β-1,3-1,4-葡聚糖酶的发酵条件。【方法】利用Box-Behnken设计和方差分析。【结果】获得最佳发酵条件为:转速、通气量和培养基pH分别为500 r/min、1.05 vvm和5.08,发酵时间仅为22 h产β-1,3-1,4-葡聚糖酶活力达2 294.4 U/mL。【结论】实验结果表明响应面法优化5 L发酵罐发酵产β-1,3-1,4-葡聚糖酶的条件合理可行。  相似文献

摘要:【目的】系统研究棉秆制浆废液用于培养具有生物防治功能的枯草芽孢杆菌的适宜条件和转化液的安全性。【方法】以初始pH值、温度、曝气量和接种量作为单因素,通过实验优化培养条件,并以大鼠和大耳兔作为测试动物,对转化液的安全性进行分析。【结果】结果表明,APMP棉秆制浆废液培养枯草芽孢杆菌的最适条件为:初始pH7.0,温度30℃,曝气量16 L/h,接种量0.8 g/L,反应器内加入填料,装填率为30%,在此培养条件下菌株的活芽孢数为6.27×109 CFU/mL,废液COD 转化率达70.4%。生物转化后,转化液的成分含量与转化前相比均有不同程度的下降。经4种毒性试验检测表明,未发现转化液的生物致病性。【结论】APMP 棉秆制浆废液可为枯草芽孢杆菌的生长和代谢提供必要的营养,可用于生物转化,且转化液为无刺激性、无毒性物质。  相似文献

The process of biocalcification, recognised as precipitation of calcium carbonate, has been described as a widespread phenomenon associated with a wide range of different bacterial species. This biocalcifying activity, and factors that affect it, have been widely studied in moderately halophilic bacteria but there is a lack of information on factors that affect biocalcification by freshwater bacteria. In this paper, we study how temperature can affect biocalcification by freshwater bacteria that potentially could be used for the process of bioconsolidation during conservation. Ten isolates were characterised by standard biochemical and API 20NE tests. Their biocalcifying activity was studied at temperatures between 10 and 40°C in B4 liquid medium. Mineralogical and quantitative analyses of the crystals were carried out by XRD, and morphological studies by SEM. Biocalcification only occurred when bacteria were present and were able to grow. Carbonate precipitation by bacteria increased with time and temperature of incubation. Temperature affected not only the amount of precipitation but also crystal quality and morphology. As bioconsolidant agents, these organisms could be applied to stone when the temperature does not exceed 40°C depending on the type of isolate.  相似文献

The influence of mineral substrate composition and structure on bacterial calcium carbonate productivity and polymorph selection was studied. Bacterial calcium carbonate precipitation occurred on calcitic (Iceland spar single crystals, marble, and porous limestone) and silicate (glass coverslips, porous sintered glass, and quartz sandstone) substrates following culturing in liquid medium (M-3P) inoculated with different types of bacteria (Myxococcus xanthus, Brevundimonas diminuta, and a carbonatogenic bacterial community isolated from porous calcarenite stone in a historical building) and direct application of sterile M-3P medium to limestone and sandstone with their own bacterial communities. Field emission scanning electron microscopy (FESEM), atomic force microscopy (AFM), powder X-ray diffraction (XRD), and 2-dimensional XRD (2D-XRD) analyses revealed that abundant highly oriented calcite crystals formed homoepitaxially on the calcitic substrates, irrespective of the bacterial type. Conversely, scattered spheroidal vaterite entombing bacterial cells formed on the silicate substrates. These results show that carbonate phase selection is not strain specific and that under equal culture conditions, the substrate type is the overruling factor for calcium carbonate polymorph selection. Furthermore, carbonate productivity is strongly dependent on the mineralogy of the substrate. Calcitic substrates offer a higher affinity for bacterial attachment than silicate substrates, thereby fostering bacterial growth and metabolic activity, resulting in higher production of calcium carbonate cement. Bacterial calcite grows coherently over the calcitic substrate and is therefore more chemically and mechanically stable than metastable vaterite, which formed incoherently on the silicate substrates. The implications of these results for technological applications of bacterial carbonatogenesis, including building stone conservation, are discussed.  相似文献

【目的】为了探讨细菌对碳酸盐矿物种类和形态的影响。【方法】本文利用丛毛单胞菌HJ-1菌株进行了持续50 d的培养实验。在实验过程中,对细菌数量、沉淀物重量、培养液中Ca2+和Mg2+浓度等进行了动态监测。利用扫描电子显微镜对矿物形态进行了观察,并利用X-射线衍射仪对矿物成分进行测定。【结果】丛毛单胞菌HJ-1菌株具有显著的诱导碳酸盐矿物沉淀的能力,碳酸盐矿物的重量随着培养时间的延长而逐渐增加。X-射线衍射结果表明,形成的碳酸盐沉淀主要由文石和高镁方解石组成,其中文石的最高含量达86%。上述矿物在形态上复杂多样,主要有杆状、柱状、哑铃形、球状和板状以及不规则状和鳞片状集合体。【结论】通常,在Mg/Ca≤2并且有微生物参与的条件下极少形成文石。本文在Mg/Ca为2,不含碳酸根离子的培养基中培养HJ-1菌株的过程中发现了文石。作者认为,低Mg/Ca条件下文石的形成主要与HJ-1菌株分泌较多的胞外多糖有关。  相似文献

Abstract

Microbially induced calcite precipitation (MICP) is currently appraised for the improvement of sandy soils, but only few studies use it to improve sand-clay mixtures. The effect of contents of kaolin clay and the effect of ions in kaolin clay on bacterial urease activity and productive rates for calcium carbonate were studied. Moreover, sand solidification tests were conducted and the solidifying effects of MICP for sand-clay mixtures were evaluated. The results show that adding kaolin clay has an inhibitory effect on the urease activity of bacteria, and adding too many kaolin clays also decrease the productive rates for calcium carbonate. With adding Al2O3 or FeCl3, urease activity both decreases and it becomes lower with adding more Al2O3 or FeCl3. The permeability of sand columns all decreased gradually with MICP curing. With more kaolin clay, the increasing range of bacterial utilization rates of those with larger particle sizes is bigger. The maximum productive rate for calcium carbonate of samples with smaller particle sizes exists in sample with 5% of kaolin clay while other samples with 7.5% of clay have more calcium carbonate. Sand columns with different sand particle sizes have different suitable amounts of added kaolin clays for MICP solidification.  相似文献

Summary The otoliths of embryos and young animals of the lizard Podarcis s. sicula were studied by X-ray diffraction and scanning electron microscopy. Two types of crystal that give different X-ray diffraction patterns were found in the membranous labyrinth of Podarcis. The crystals consist of calcite or aragonite and are easily distinguished by scanning electron microscopy because of their different morphology. The two calcium carbonate crystal forms are not mixed at random but are present in the embryo from the very beginning in specific sites. The endolymphatic sac contains aragonite crystals while the saccule contains calcite crystals adjacent to the wall, in addition to a preponderance of aragonite crystals. The utricle and lagena contain only calcite crystals. The presence of two crystal forms of calcium carbonate in the membranous labyrinth are discussed in terms of differing genetic and functional significance.  相似文献

[目的]研究天然群体感应抑制剂(Quorum sensing inhibitors,QSI)分子对海洋生态功能菌生物膜形成的影响.[方法]以对污损生物幼虫附着具有诱导作用的海洋细菌为目标菌,通过在其生物膜的形成过程中添加天然群体感应抑制剂,研究其对目标菌成膜细菌数和浮游细菌数、生物膜形态以及生物膜表面胞外多糖含量的影响.[结果]呋喃酮和吡啶在50 mg/L时,对8株目标菌的成膜有显著的抑制作用,抑制率在80%左右,吲哚、青霉烷酸和香豆素在较高浓度800 mg/L才有比较好的抑制活性.生长抑制实验结果显示,同等浓度下,QSI分子对目标菌成膜的抑制活性明显高于其对浮游细菌生长的抑制活性.结果表明,QSI分子主要通过干扰目标菌群体感应系统以抑制生物膜的形成.[结论]研究证实QSI分子在海洋菌生物膜形成过程中具有一定的调控作用.通过添加QSI可能能够间接抑制由生物膜诱导的污损生物附着,从而以新的角度研制新型抗污损物质.  相似文献

Scanning electron microscopy and X-ray dispersive energy microanalysis were used to investigate the formation of carbonate crystals by Deleya halophila. The formation of calcium carbonate crystals (polymorphous aragonite) by D. halophila is a sequential process that commences with a nucleus formed by the aggregation of a few calcified bacterial cells and the subsequent accumulation of more calcified cells and carbonate, which acts to weld the bacteria together. The process leads to the formation of spherical bioliths measuring approximately 50 μm in diameter. The mechanism of carbonate precipitation by D. halophila under our working conditions represents a process of induced biomineralization.  相似文献

【目的】研究添加泥浸汁与否对太湖沉积物中可培养细菌的影响。【方法】采用R2A培养基和添加泥浸汁R2A培养基对沉积物中细菌进行分离培养,16S r RNA基因系统发育分析比较种群结构。【结果】培养基中添加泥浸汁,可使可培养细菌的种类数量增加到1.6倍。16S r RNA基因序列分析表明,培养的优势细菌类群存在明显差别。R2A培养基上生长的细菌主要为厚壁菌门(52%)、放线菌门(24%)、变形菌门(20%)和拟杆菌门(4%),其中大部分细菌与芽孢杆菌属、假单胞菌属、节杆菌属等关系密切;而添加泥浸汁的R2A培养基上生长的细菌则主要为变形菌门(40%)、放线菌门(35%)、厚壁菌门(22.5%)和拟杆菌门(2.5%),与鞘脂单胞菌属、芽孢杆菌属、副球菌属、节杆菌属等关系密切。【结论】添加泥浸汁原始营养因子可提高沉积物中可培养细菌的多样性,提高菌种可培养效率。  相似文献

To better understand the formation mechanism of carbonate minerals by microbes, culture experiments with a duration of 70 days were performed under the mediation of strain GW-M isolated from soil using modified Lagoa Vermelha (LV, a hypersaline coastal Lagoon, Rio de Janeiro, Brazil) medium with 6:1 Mg/Ca molar ratio. The results demonstrated that strain GW-M can mediate the formations of both high-Mg calcite and aragonite and that dumbbell-, cauliflower-, rhombohedra-shaped, and irregular minerals coexist in the modified LV medium. The amount of rhombohedra-shaped crystals increased significantly with culture time. A proposed mechanism for these formations is the following. Heterogeneous nucleation on the surface of the extracellular polymeric substances (EPS) always occurred, and carbonates with irregular shape existed in experimental products at any stages. The morphologies evolved from rod to dumbbell and finally to cauliflower. At the initial stage (till day 20), hydrogen ions and EPS secreted by the bacteria only influenced the microenvironment around the cells, and carbonates were precipitated on the surface of bacterial cells. At the middle and late stages (on days 45 and 70), microbes and their secretions influenced the whole medium. Under these conditions, rhombohedra-shaped crystals were formed when homogeneous nucleation occurred. In addition, the results of energy-dispersive spectrometry (EDS) showed that Mg contents in the synthesized carbonate minerals with rhombohedra-shaped were significantly lower than those of carbonates with other shapes, though relationship between morphology and species of mineral cannot be obtained by this phenomenon alone. These results shed further light on the mechanism of carbonate precipitation in the presence of microbes.  相似文献

为探讨洞穴微生物沉积碳酸钙作用对洞穴沉积物的影响,利用传统生物学方法,采集贵州中西部地区石将军洞洞穴沉积物表面的微生物样品,结合洞穴监测数据和理化背景资料,利用B-4培养基和B-4C培养基对洞穴细菌进行筛选和纯化,分离出能沉积碳酸钙的菌种,观察和了解洞穴细菌形成的CaCO3晶体,应用X射线衍射分析仪(XRD)分析细菌形成的CaCO3晶体成分,并利用扫描电子显微镜(SEM)观察晶体结构特征。结果表明:1)在B-4培养基下微生物产生的碳酸钙晶体主要为方解石、球霰石和方解石混合物、球霰石,这种变化与培养基pH值的增幅相关;同时,在添加Mg离子的B-4C培养基下形成的碳酸钙晶体主要为方解石,此外,研究中并未检测到文石晶体。2)通过SEM扫描,发现微生物作用形成的碳酸钙晶体存在不规则六方体、柱状体、四方体层状、半球状等,这些晶体形态在化学作用系统下少见,多见于微生物作用形成的方解石。此外,晶体中微生物作用痕迹明显,微生物作用贯穿于整个沉积过程。  相似文献
