从新疆巩留县伊犁贝母的新鲜鳞茎中分离到一株具有分泌抑菌活性物质的内生尖孢镰孢菌Fusarium oxysporumY1,该菌在7种不同培养基上生长时显示出不同的菌落生长特征,而且只在沙氏培养基中生长时才具有分泌抑菌活性物质的能力。抑菌活性筛选结果表明:由该菌及其发酵液制备的发酵液浸膏、菌体裂解液浸膏以及经进一步纯化后获得的乙酸乙酯浸膏和正丁醇浸膏均具有明显的抑菌活性,其中以发酵液的乙酸乙酯浸膏和菌体裂解液的正丁醇浸膏活性最强,它们对金黄色葡萄球菌Staphylococcus aureus、表皮葡萄球菌Staphylococcus epidermidis、枯草芽孢杆菌Bacillus subtilis、藤黄八叠球菌Sarcina lutea和大肠杆菌Escherichia coli的最低抑菌浓度均小于25μg/mL。  相似文献

目的 探讨东莞地区政标本2的常见病原菌及其对抗生素的敏感性。方法 参照《全国临床检验操作规程》处理下呼吸道痰液标本2,分离培养和鉴定菌株。用Kirby-Bauer(K-B)法进行药数测定。结果 93全怀标本共检出98株菌,其中甘兰阴性杆菌居多共63株,占64%(其中又以非发本报菌居多共41株,占41.8%),球菌仅有31株,占31.6%。革兰阴性杆菌对泰能量敏感,丁胺卡那霉素次之,葡萄球菌只泰能较  相似文献

目的 研究羧甲基壳聚糖对5株口腔颌面部感染细菌的抑菌性能,为在临床应用提供相关资料。方法 采用液体梯度稀释法分别测定羧甲基壳聚糖对牙龈卟啉菌、中间普氏菌、金黄色葡萄球菌、大肠埃希菌和铜绿假单胞菌的最低抑菌浓度(MIC)。结果 羧甲基壳聚糖对5株菌的MIC分别为:20、5、2.5、5、10 g/L。结论 羧甲基壳聚糖对口腔颌面部感染重要相关细菌具有抑菌作用。  相似文献

该文研究36种常用中药材80%乙醇提取物在体外抗临床常见致病菌的抗菌活性。采用药敏纸片法测耐药菌的耐药谱,中药粗粉用80%乙醇浸泡提取,提取液减压浓缩得浸膏,通过琼脂打孔法测定提取物抑菌圈,再通过微量倍比稀释法测定最低抑菌浓度(MIC)和最低杀菌浓度(MBC)。结果表明:36种中药材醇提物中,有15种具有广谱抗菌活性,对实验中各标准菌表现出不同程度的抑制作用,对MRSA抗菌活性也较强。其中,岩陀、卷柏、首乌藤、苏木、乌药、夏枯草6种药材的抗菌活性比较突出,抑菌圈均大于11 mm,细菌对其表现为中高度敏感;它们对7株标准菌的MIC/MBC值除个别为12.5 mg·m L~(-1)以外,均小于1.563 mg·m L~(-1),对16株MRSA的MIC/MBC值均小于1.563 mg·m L~(-1),它们的萃取层活性均小于1 mg·m L~(-1)。所筛选出的15种抗菌活性较强的中药材,可为后续研究其活性单体化合物和作用机制,研发有效的抗多重耐药菌的中药制剂以及解决细菌耐药性问题提供一定的参考。  相似文献

阴道炎、子宫颈炎常见菌培养及药敏结果分析   总被引:3,自引:2,他引:1  
了解阴道炎、子宫颈炎的常见菌及药敏情况,指导治疗。用无菌棉拭子擦取宫颈或阴道分泌物,立即置于血平板及淋球菌分离巧克力培养基内,37 ℃48h 鉴定细菌及药物敏感测定。543 例患者标本培养出441 株细菌,阳性率83.1 % ,其中常见菌葡萄球菌93 株,阴道加特纳菌70 株,念珠菌53 株,淋球菌51 株,大肠埃希菌43 株,不动杆菌31 株,解脲支原体34 株。并对细菌的种类和药敏测定结果进行了讨论,结果显示革兰阳性菌株对头孢菌素类比较敏感,而革兰阴性菌株对头孢氨苄、氧氟沙星、丁胺卡那、氯霉素效果较好。不断监测与总结较好的抗生素对各类细菌的敏感性是非常重要的。  相似文献

广州地区淋球菌抗生素耐药性及质粒图谱研究   总被引:1,自引:0,他引:1  
目的:了解广州地区淋球菌对抗生素的耐药性及质粒图谱。方法:用琼脂稀释法测定5种抗生素对167株菌的最低抑菌浓度(MIC)及用破裂解法分析160株菌的质粒图谱。结果:167株淋球菌检出PPN9株(5.4%),TRNG16株(9.6%),环丙沙星耐药率达78.4%,头孢三嗪,壮观霉素未发现耐药菌株。160株菌其质粒谱型2.6Md 4.5Md(24.4%),2.6Md 4.5Md 24.5Md(20.6%),2.6Md 24.5Md(20.6%),4.5Md检出率(45%)24.5Md检出率(41.3%)。结论:表明了广州地区淋球菌抗生素的耐药性和质粒图谱,有助于淋病的治疗和防治。  相似文献

为筛选一株产黑色素能力强的菌株并优化其培养条件。通过ITS测序鉴定11株供试菌株,以菌丝生长速度、平板L值等指标筛选出一株产黑能力强的香灰菌,并对其生长所需碳源、氮源、pH等培养条件进行优化。研究表明,11株供试菌株均为香灰菌(Hypoxylon sp.),其中Hp.sp0006菌丝生长速度较快、菌球大且均匀、L值最低,并且发酵液黑色素含量最高。该菌株最优培养条件是,以葡萄糖为碳源、牛肉浸膏为氮源、碳氮比20∶1并添加10 mg维生素B1,黑色素含量可达(1.21±0.17)g/L。香灰菌Hp.sp0006是一株产黑色素较高的菌株,优化后的培养基更有利于黑色素的合成,为香灰菌黑色素的开发利用奠定基础。  相似文献

目的:探讨亚抑菌浓度头孢他啶对大肠埃希菌生物膜形成的影响与细菌耐药性、超广谱β-内酰胺酶(ESBLs)产生及ESBLs基因分型的相关性,为临床生物膜感染的治疗和抗生素的合理使用提供理论依据。方法:大肠埃希菌最低抑菌浓度(MIC)检测采用琼脂平板倍比稀释法,超广谱β-内酰胺酶(ESBLS)表型确证实验采用双纸片协同法,大肠埃希菌ESBLs基因检测采用PCR扩增,生物膜形成能力检测采用96孔板结晶紫染色法。结果:50株大肠埃希菌临床株对青霉素类、氟喹诺酮类、头孢哌酮及复方新诺明具有较高的耐药性,而对阿米卡星、哌拉西林/他唑巴坦敏感性较高。所有菌株均对碳青霉烯类抗菌药物敏感。31株大肠埃希菌为ESBLs阳性菌株。CTX-M、TEM、OXA、SHV和VEB基因阳性率分别为93.5%、83.9%、19.4%、16.1%和3.2%。亚-MIC头孢他啶对9株(18.0%)大肠埃希菌生物膜形成具有抑制作用。亚-MIC头孢他啶对大肠埃希菌生物膜形成的影响与细菌耐药性和ESBLs均无相关性(P0.05)。结论:亚-MIC头孢他啶对大肠埃希菌生物膜形成的调控作用与细菌耐药性、产ESBLs及ESBLs基因分型均无相关性。  相似文献

目的 从临床分离的念珠菌中进一步鉴定都柏林念珠菌,并评价3种表型鉴别白念珠菌和都柏林念珠菌的方法.方法 对17株临床分离并初步鉴定的白念珠菌和1株ATCC白念珠菌标准株,采用PHR1同源序列PCR法检测,鉴定出其中的都柏林念珠菌;分别采用45℃生长试验、YEPD(1%酵母浸膏,2%蛋白胨,2%葡萄糖)液基39℃芽管生成试验、Staib琼脂(鸟食琼脂)厚壁孢子形成试验对两种菌的表型特点进行比较.结果 17株临床分离的白念珠菌中有3株鉴定为都柏林念珠菌;45℃时,两种菌在改良沙堡弱琼脂上均无明显生长,YEPD液基中仅有1株白念珠菌生长良好;YEPD液基39℃培养2种菌均无芽管生成;Staib琼脂培养72h,3株都柏林念珠菌中有2株可形成厚壁孢子,而白念珠菌则无,与PHR1同源序列检测结果基本一致.结论 PHR1同源序列检测是鉴别都柏林念珠菌与白念珠菌的可靠方法,Staib琼脂厚壁孢子形成试验有助于鉴别两菌,45℃生长试验和YEPD液基39℃芽管生成试验则不能有效鉴别两菌.  相似文献

动植物共生菌是药物发现的重要来源之一。为了获取结构新颖且具有良好生物活性的微生物次生代谢产物,本文利用划线分离方法从蜈蚣肠道中分离获得一株共生菌WG4(未鉴定),经土豆液体发酵培养,发酵液经乙酸乙酯萃取、浓缩,获得次级代谢产物浸膏。浸膏经硅胶柱层析、高效液相色谱等分离方法反复纯化,获得3个化合物。通过核磁共振谱和低分辨质谱测试,3个化合物分别鉴定为1-羟基吩嗪(1)、吩嗪-1-甲酰胺(2)和吩嗪-1-羧酸(3)。据文献报道,吩嗪化合物具有抗菌、抗虫、抗病毒、抗肿瘤等生物活性,本实验证实了动植物来源的共生菌具有生产药源化合物的潜力。  相似文献

Inhibition of Helicobacter pylori by garlic extract (Allium sativum)   总被引:6,自引:0,他引:6  
Abstract The antibacterial effect of aqueous garlic extract (AGE) was investigated against Helicobacter pylori . Sixteen clinical isolates and three reference strains of H. pylori were studied. Two different varieties of garlic were used. The concentration of AGE required to inhibit the bacterial growth was between 2–5 mg ml−1. The concentration, for both AGE types, to inhibit 90% (MIC90) of isolates was 5 mg ml−1. The minimum bactericidal concentration (MBC) was usually equal to, or two-fold higher than, minimum inhibitory concentration (MIC). Heat treatment of extracts reduced the inhibitory or bactericidal activity against H. pylori ; the boiled garlic extract showed a loss of efficacy from two-to four-fold the values of MIC and the MBC obtained with fresh AGR. The antibacterial activity of garlic was also studied after combination with a proton pump-inhibitor (omeprazole) in a ratio of 250:1. A synergistic effect was found in 47% of strains studied; an antagonistic effect was not observed.  相似文献

Strains of Zymomonas mobilis grown in media containing either glucose or sucrose were assessed for the production of hydrogen sulphide (H2S). In a liquid medium with low glucose concentration (20 g l?1) only a proportion of the strains tested formed H2S, but in medium containing a higher glucose concentration (100 g l?1) all the strains tested produced H2S. Four Z. mobilis strains were assayed quantitatively for H2S production and strain ZM4 was found to produce the most H2S in glucose medium. The amount of yeast extract and glucose, and the type of sugar used in the medium affected the amount of H2S formed by strain ZM4. A mutant, designated ZM4701, of strain ZM4 was isolated which did not produce any detectable H2S in liquid medium containing yeast extract plus either glucose or sucrose. The nutritional requirements of ZM4701 were investigated.  相似文献

Summary Decomposition of chitin by Cytophaga johnsonii was investigated. Unlike other chitinolytic bacteria, some strains of C. johnsonii did not liberate chitinase extracellularly; instead the cells of such strains had need for close contact with the chitin particles in order to hydrolyze them. The cell-free extract of one of these strains, viz., C. johnsonii C35 did show presence of chitinase. The remaining strains liberated an extracellular chitinase and a chitobiase. The partially purified chitinase from the culture filtrates of C. johnsonii C31 was most active at pH 6.3–6.5 and at 40°C.Metabolism of N-acetylglucosamine, a product of chitin hydrolysis, by C. johnsonii C35 and C31 was investigated. The nature of some of the products formed and the steps involved in their transformations by the resting cells and the cell-free extracts suggested that N-acetylglucosamine first gets deacetylated to glucosamine before it is further oxidized to glucosaminic acid by these strains. Both strains were also able to dehydrogenate gluconate to 2-ketogluconate, the former being in all probability the product of deamination of glucosaminic acid.  相似文献

目的:研究新疆特色林果核桃、巴达木果壳乙醇提取物的抑菌特性。方法:用滤纸片法检验果壳乙醇提取物对金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌和变形杆菌的抑菌活性,通过液体培养基稀释法测定最低抑菌浓度。结果:巴达木乙醇提取物(25%乙醇提取10 h)对大肠杆菌有较好的抑菌效果,最低抑菌浓度(MIC)为25%;核桃乙醇提取物(25%乙醇提取24 h)对大肠杆菌有较好的抑菌效果,MIC为6.25%。结论:核桃、巴达木果壳乙醇提取物对受试菌具有较好的抑菌效果,为进行大规模开发利用提供了有效依据。  相似文献

Soluble proteins of the Japanese, Philippine, and Formosan strains of Schistosoma japonicum were separated by disc electrophoresis using polyacrylamide gel columns. Differences between strains and sexes were investigated on the basis of Rf values of the bands, location of prominent peaks, quantitative comparisons of major bands, and overall densitometric profiles. With male extract, 29, 28, and 29, distinct bands were resolved for the Japanese, Philippine, and Formosan strains, respectively. Female extracts gave 31, 26, and 25 distinct bands for the Japanese, Philippine, and Formosan strains, respectively. Both qualitative and quantitative differences were found among the three strains and between sexes. The closest relationship of densitometric profile was between the Japanese and Formosan strains.  相似文献

We found that extract from petals of Rosa canina L. (rose red) strikingly reduced the minimum inhibitory concentration of beta-lactams in methicillin-resistant Staphylococcus aureus. We isolated two compounds that reduced the minimum inhibitory concentrations of beta-lactams from the extract, tellimagrandin I and rugosin B. Tellimagrandin I was very effective regarding the reduction of the minimum inhibitory concentration, and rugosin B showed some effect. Tellimagrandin I showed a weak bactericidal action when added together with oxacillin. Judging from the fractional inhibitory concentration index, the effect of tellimagrandin I and oxacillin was synergistic. Tellimagrandin I also significantly reduced the minimum inhibitory concentration of tetracycline in some strains of methicillin-resistant Staphylococcus aureus.  相似文献

Immunogenicity and protective activity against Brucella ovis of detergent-extracted recombinant Omp31 (rOmp31 extract) from Brucella melitensis produced in Escherichia coli, purified rough lipopolysaccharide from B. ovis (R-LPS) and a mixture of rOmp31 extract and R-LPS (rOmp31 extract + R-LPS) were assessed in BALB/c mice. The experimental vaccines were compared with a hot saline extract (HS extract) from B. ovis mainly composed of outer membrane proteins (OMPs) and R-LPS, and known to be protective in mice against a B. ovis infection. Serum antibodies to Omp31 and R-LPS were detected in the corresponding mice using Western blotting with B. ovis whole-cell lysates and ELISA with purified antigens. Protection was evaluated by comparing the levels of infection in the spleens of vaccinated mice challenged with B. ovis. A significantly lower number of B. ovis colony-forming units in spleens relative to unimmunized (saline injected) controls were considered as protection. Mice immunized with rOmp31 extract or rOmp31 extract mixed with R-LPS developed antibodies that bound to the B. ovis surface with similar titers. Vaccination with rOmp31 extract plus R-LPS provided the best protection level, which was comparable with that given by HS extract. Similar protection was also obtained with rOmp31 extract alone and, to a lesser degree, with R-LPS. Comparisons between groups showed that an extract from E. coli-pUC19 (devoid of Omp31) provided no protection relative to either HS extract, rOmp31 extract or rOmp31 extract mixed with R-LPS. In conclusion, the recombinant Omp31 associated or not with B. ovis R-LPS, could be an interesting candidate for a subcellular vaccine against B. ovis infection.  相似文献

Citrobacter freundii has been implicated in food spoilage and food poisoning outbreaks. This study examines the effects of some compounds (e.g. citric acid, ascorbic acid, lactic acid, sodium chloride, andThymus vulgaris extract) on growth of two strains of Citrobacter freundii at 31 °C and 5 °C. At 31 °C, lactic acid (0.2%) or ascorbic acid (0.2%) alone completely inhibited growth of the tested strains, as there was 100% reduction in growth of the strains after 24 h incubation in nutrient broth containing these compounds.Thymus vulgaris extract (0.3%) reduced the growth rate (p<0.05), the percentages of inhibition after 24 h incubation were about 60% for both strains. NaCl (5%) greatly reduced growth, the percentages of inhibition were about 84% for both strains. Combination ofT. vulgaris extract (0.3%) and NaCl (4%) together completely inhibited growth ofC. freundii species tested. Ascorbic acid (0.1%) or citric acid (0.03%) did not affect growth of the strains (p>0.05), but a lag occurred before increase in number could be observed. In chicken and fish homogenates, combination of NaCl (4%) and ascorbic acid (0.1%) reduced the growth (p < 0.05) (growth inhibition was 40%). At 5 °C, lactic acid (0.1%) alone greatly reduced the growth (p<0.05). The activity of NaCl, or ascorbic acid alone against the tested strains was greatly increased (p<0.05). ForC. freundii 4, the percentage of growth inhibition after 6 days incubation in broth containing 3% NaCl or 0.1% ascorbic acid were 88% and 72%, respectively. ForC. freundii 38, the percentage of growth inhibition after 6 days incubation in broth containing these compounds were 60% and 54%, respectively.  相似文献

用稀释法从腐殖泥中分离菌株,根据它们在羧甲基纤维素钠(CMC-Na)-刚果红培养基上的透明圈直径及对滤纸的崩解能力,获得7株纤维素分解细菌(编号:B-37、B-35、B-31、B-25、B-17、Z-a和Z-b),其中菌株Z-b的纤维素崩解能力最强,分子鉴定结果表明它与Stenotrophomonas maltophilia的16S rDNA序列有99.8%的同源性,初步确定为嗜麦芽窄食单胞菌。适合7个菌株生长的C源为马铃薯浸出液,无机盐组分为:CaCl_2 0.20、MgSO_4 1.25、NaCl 5.00、(NH_4)_2SO_4 1.30、KH_2PO_4 1.35、FeSO_4·7H_2O 0.015、Na-EDTA 0.02g/L。在滤纸为唯一碳源的培养基中,菌株Z-b的滤纸酶(FPase)和CMC酶(CMCase)活性最大,为0.099 U/mL和0.075 U/mL,而在固体PSA上,菌株B-31和B-37的FPase和CMCase活性最高,为0.131 U/mL和0.175 U/mL。7个细菌单独发酵,Z-b对滤纸的崩解能力最强,滤纸块完全崩解成粉未状;与真菌34混合发酵,菌株组合Z-a+34、Z-b+34、B-31+34、B-25+34将滤纸完全水解为水溶性物质。可见,各菌株的纤维素酶活与培养条件密切相关,某些真菌、细菌间存在协同作用,它们混合发酵可大大提高纤维素的水解效率。  相似文献
