UV-B胁迫小麦上麦长管蚜的生命表参数和取食行为   总被引:1,自引:0,他引:1  
【目的】为明确UV-B胁迫小麦对麦长管蚜Sitobion avenae (Fabricius)种群生命表参数和取食行为的影响。【方法】采用特定年龄生命表方法和刺探电位图谱(EPG)技术, 研究了不同强度UV-B胁迫处理小麦对麦长管蚜生命表参数和取食行为的影响。【结果】生命表结果表明, 取食高强度(0.75 mW/cm2) UV-B辐射小麦后, 麦长管蚜种群内禀增长率rm、 净增殖率R0、 繁殖力F下降, 平均世代周期T缩短, 且差异显著(P<0.05); 取食低强度(0.2 mW/cm2)UV-B辐射小麦后, 麦长管蚜种群生命表参数与对照组(正常光照处理小麦饲喂的蚜虫种群)无显著差异(P>0.05)。刺探电位图谱结果显示, 高强度UV-B处理组np波(即非取食)数量显著增加, pd波(刺探波)首次出现的时间延迟, 表明紫外处理小麦干扰了蚜虫的在叶表面的正常刺探活动, 延长了搜索和刺探时间; C波持续时间显著延长, 反映取食难度加大, 有效取食时间缩短, 从而影响种群生命表参数; 而低强度UV-B处理组小麦对蚜虫取食行为影响与对照组差异不显著, 但pd波首次出现时间显著延长。【结论】高强度UV-B胁迫小麦会严重影响麦长管蚜的生长发育、 繁殖与取食行为。  相似文献

在加倍CO2浓度(0.7 mL/L)的人工气候箱中培养小麦感蚜品种(铭贤169)和抗蚜品种(KOK1679)幼苗30d,正常CO2浓度(0.35 mL/L)处理为对照.应用刺探电位图谱(EPG)技术对麦长管蚜Sitobion avenae (Fab.),在苗期小麦上的取食行为进行了比较研究.结果表明:蚜虫在加倍CO2浓度下生长的2个小麦品种上的取食过程中,电势降落次数(pd波个数)显著增加,刺探总时间(C波总时间)显著延长,首次出现在小麦韧皮部取食的时间(E2)显著滞后,被动取食持续时间(E2波的总时间)显著拉长.因此,加倍CO2浓度可能通过影响小麦而对麦长管蚜的取食行为产生了明显的间接影响.  相似文献

选用经田间鉴定抗蚜性表现不同的10个小麦品种,在室内外较系统地研究了其对禾谷缢管蚜和麦长管蚜存活、生长发育和生殖的影响,分析了室内外试验结果的相关性.结果表明,小麦品种的抗生性并不能造成若蚜个体的直接死亡,但对若蚜发育历期、成蚜寿命和产仔数影响显著,差异均达极显著水平或显著水平.其中对两种蚜虫若蚜发育历期的影响室内外表现了较好的一致性,可作为小麦品种抗蚜性鉴定的主要指标;其次是对成蚜产仔数的影响,禾谷缢管蚜室内外试验结果比较的吻合性也较好  相似文献

【目的】明确苜蓿斑蚜Therioaphis trifolii对不同品种苜蓿Medicago sativa的取食行为,探明抗蚜因子和抗虫位点,筛选抗蚜苜蓿品种。【方法】利用刺吸电位技术(electrical penetration graph, EPG)记录苜蓿斑蚜成蚜在10个苜蓿品种上的取食行为,并基于聚类分析方法筛选合适的EPG参数进行抗蚜性评价。【结果】苜蓿斑蚜成蚜在取食苜蓿过程中呈现np波、pd波、A波、B波、C波、E波、F波和G波8种波形,其中E波、F波和G波的总时间在不同品种间表现出显著差异。在5 h测试期间,E波持续时间在敖汉上最长,金皇后和中苜3号上次之,草原2号和阿尔冈金上最短;F波总时间以准格尔、阿尔冈金和金皇后上最长,敖汉、德宝和草原2号上最短,说明前者具有较强的机械抗性而后者机械抗性较弱。以第1次刺探持续时间、总刺探时间、F波总时间、C波总时间、E波总时间作为聚类分析的指标,10个苜蓿品种被聚成3类:阿尔冈金、草原2号、WL168HQ、德宝、中苜2号和新牧2号为第Ⅰ类,金皇后、中苜3号和准格尔为第Ⅱ类,敖汉为第Ⅲ类。【结论】苜蓿斑蚜成蚜在不同苜蓿品种上的取食行为存在差异,草原2号、阿尔冈金和WL168HQ在叶表皮、叶肉和韧皮部层次对苜蓿斑蚜成蚜存在抗性,金皇后在叶表皮和叶肉部位表现出抗性;苜蓿斑蚜成蚜在敖汉上能够长时间刺吸取食,因此敖汉抗性最弱。本研究为深入探讨苜蓿抗虫机理和蚜虫综合治理提供了理论参考。  相似文献

应用50个小麦品种对麦长管蚜抗性鉴定指标值进行模糊统计回归分析,提出了品种自然感蚜量、蚜虫在品种上的累积存活率、平均发育历期、产仔率、内禀增长率(rm)及品种耐害性等抗性指标的模糊隶属函数,建立了品种抗性的模糊综合决策模型。对10个品种的抗蚜性综合决策结果表明,陕167的综合抗蚜性最强,83(37)65、(87)113、82246次之,小偃6号最差。同时,综合型决策模型的决策结果最优,精度高,保留较多信息。  相似文献

基于EPG的麦长管蚜、麦二叉蚜和禾谷缢管蚜取食行为比较   总被引:6,自引:0,他引:6  
麦长管蚜Sitobion avenae (Fabricius),麦二叉蚜Schizaphis graminum (Rondani)和禾谷缢管蚜Rhopalosiphum padi (Linnaeus)虽然都取食小麦,但其取食部位、传毒能力均有差异。利用EPG研究了3种麦蚜在同一寄主植物上的取食行为,结果显示:(1)禾谷缢管蚜在取食过程中G波出现的时间最早,G波总的持续时间和每次刺探G波的平均持续时间都明显长于麦长管蚜和麦二叉蚜。麦长管蚜第1次E1波的时间、E1波总的持续时间和每次刺探E1波的平均持续时间都明显比麦二叉蚜和禾谷缢管蚜要长。麦二叉蚜E2波总的持续时间和每次刺探E2波的平均持续时间都明显长于麦长管蚜和禾谷缢管蚜。表明3种蚜虫各自拥有不同取食策略。禾谷缢管蚜比其它两种蚜虫需要更多的水分和无机盐,麦长管蚜通过分泌大量唾液增大对食物的利用率,麦二叉蚜通过增加韧皮部取食时间以满足对营养的需要。(2)刺探过程中麦二叉蚜pd波出现的次数为(51.97±7.82)次,高于麦长管蚜的(44.73±4.52)次和禾谷缢管蚜的(32.99±4.22)次。麦二叉蚜pd波Ⅱ-2和Ⅱ-3亚波的时间最长,禾谷缢管蚜最短,三者之间差异达显著水平。表明与其它两种麦蚜相比,麦二叉蚜在取食过程中口针与细胞间的接触频率更高,与细胞内获毒和传毒相关的Ⅱ-3波和Ⅱ-2波持续时间更长,增加了获毒和传毒的几率,是其传毒能力最强的机理。  相似文献

应用刺探电位图谱(EPG)技术对禾谷缢管蚜Rhopalosiphum padi在不同小麦品种(Ww2730、小偃22和Batis)苗期的取食行为进行了比较研究。结果表明:在小偃22上蚜虫开始取食的第1次刺探时间最晚,且持续时间最短;在Ww2730上取食受到机械阻力的个体最多,且出现F波的几率和持续时间最长;两品种上蚜虫在木质部主动摄取汁液(G波)花费的时间最长。 在Batis上,蚜虫口针第1次到达韧皮部时需要分泌较多水溶性唾液(E1波),但随后只需分泌较少的水溶性唾液就可以成功取食,而且被动吸食韧皮部汁液的时间(E2波)最长。蚜虫口针在到达小偃22韧皮部取食之前,出现多次的口针试探、回撤,并且多次、多量分泌水溶性唾液(E1波);虽然蚜虫在小偃22上口针最先到达韧皮部,但被动吸食韧皮部汁液的时间(E2波)最短。由此得出结论:小偃22表皮部、韧皮部存在阻碍禾谷缢管蚜取食的物理和生化因素; 禾谷缢管蚜在Ww2730取食遇到更多的是细胞间机械阻力; Batis是较感蚜的品种。  相似文献

根据小麦苗期1~6级蚜害分级标准,1996年~1997年利用国际通用的生物型鉴别寄主(品种)对北京地区麦二叉蚜种群进行了生物型鉴定。各鉴别品种的蚜害反应型分级是:小麦S28A(gb1)为6,Amigo(Gb2)为1,Largo(Gb3)为2,C117959(Cb4)为6,C117882(Cb5)为3,GRSl201(Gb6)为3.5,燕麦C11580为1,黑麦Insave为1。结果表明,北京麦二叉蚜的致害性显著地不同于10个已知的生物型(从A到J),是一新的生物型,命名为中国1型(BiotypeCHN-1)。此外,针对麦二叉蚜生物型分化问题,对小麦抗蚜育种中的品种抗性持久化与累积抗性品种的培育,以及分子标记辅助育种技术的应用等策略进行了探 讨。  相似文献

利用田间抗蚜性鉴定模糊识别技术,结合室内刺探电位(EPG)植物抗性鉴定技术,比较分析了不同遗传背景的小麦种质资源的抗蚜性,为筛选新型小麦抗蚜种质材料提供依据。2年田间抗蚜性鉴定结果表明: 小偃麦多表现为中抗或低抗,而小黑麦多为中感或低感。选取抗性性状稳定且抗性级别不同的小偃麦21(中抗)、小偃麦22(低抗)、小黑麦31(中感)和小黑麦32(低感)进行麦长管蚜取食行为分析。对非刺探波(Np)、刺探波(P)、电势落差(Pd)、水溶性唾液分泌波(E1)、韧皮部取食波(E2)、细胞机械阻碍波(F)和木质部取食波(G)等基本波形的分析显示,麦长管蚜在小偃麦上首次开始刺探的时间显著长于小黑麦,且在小偃麦上的E1波的持续时间显著大于小黑麦;麦长管蚜在小偃麦21上的F波和小偃麦22上Np波的持续时间最长,在小黑麦31上的P波和小黑麦32上的G波的持续时间最长。以E1、F和Np波的持续时间为指标,基于刺探电位的小麦种质资源抗性水平鉴定结果与田间鉴定结果基本一致。因此,使用EPG技术筛选抗蚜小麦材料时,建议采用E1、F和Np波作为评价小麦抗性水平的指标。小偃麦21、22对麦长管蚜的抗性水平较高,可作为小麦抗蚜育种的种质材料。  相似文献

本研究采用室内、外结合的方法,分苗期和拔节期,对大田抗蚜性鉴定中表现不同的10个小麦品种的抗性对麦长管蚜种群增长的影响进行了研究。研究结果显示,品种抗性对麦长管蚜的存活率、发育进度、繁殖力有一致的抑制作用;品种间抗蚜性存在显著差异。同时通过组建不同品种上麦长管蚜繁殖特征生命表,从量的角度揭示了小麦品种抗性与麦长管蚜种群增长之间的关系。  相似文献

主要抗蚜小麦品种(系)的抗性类型及其生化抗性机制   总被引:6,自引:0,他引:6  
通过对10个抗麦蚜品种(系)室内苗期生命参数、抗性类型和抗蚜次生物质的研究,明确了不同抗性级别的品种对麦蚜种群控制力及部分生化抗蚜机制。实验结果表明,参试的抗蚜品种(系)中30%左右为不选择性:表现为爬行频繁,定殖率低,但是定殖个体生长发育良好;70%为抗生性;表现为使麦长管蚜Sitobion avenae(F.)和禾谷缢管蚜只Rhopalosiphum padi(L.)的发育历期分别延长2.1%~28.2%和3.7%~13.9%,若蚜死亡率增加1.0~3.6倍和1.0~2.25倍,平均寿命缩短10.2%~96.5%和37.5~97,1%,繁殖力下降3.4%~72.8%和25%~97.2%。苗期生化测定结果表明:不同抗源的单宁和总酚含量明显高于感蚜品种,其总酚含量与抗麦长管蚜级别呈显著负相关,以抗生性为主的品种其总酚含量亦与麦长管蚜的内禀增长力(rm)呈显著负相关(P<0.05),表明总酚是小麦抗长管蚜的关键因子之一,而与禾谷缢管蚜抗性水平无关;单宁含量与麦蚜抗性关系不密切。  相似文献

Acyrthosiphon pisum (Harris) is a major pest of alfalfa worldwide. Here, we evaluated the resistance of eight alfalfa cultivars to A. pisum to identify resistant cultivars. The Giga-8 DC-EPG technique was used to record differences in electrical waveform patterns of probing behavior to identify cultivars with higher resistance. The frequency and average duration of EPG waveforms significantly differed among the cultivars, with eight documented waveforms (np, C, pd, E1, E2, sE2, F, and G). The longer duration of np, C, F and E1 waveforms reflected stronger resistance, whereas longer sE2 waveforms reflected weaker resistance. The resistance-related waveforms, np and C, occurred most frequently with Sardi 7, the time of the first E waveforms were the latest, duration of G waveforms and F waveforms were the longest, the feeding waveforms E2 and sE2 were the shortest, the time of reaching phloem for feeding was the latest, with notable resistance in leaf epidermis, mesophyll and phloem. The number of C waveforms occurring with Gibraltar were the least, duration of np waveforms was short, feeding waveforms were long, time to reach the phloem for feeding was early, and the resistance was not pronounced in various tissues. After comprehensive evaluation, the resistance levels of 8 alfalfa cultivars were as follows: High resistance, Sardi 7; Resistance, Zhongcao No. 3, Derby, WL319HQ; Low resistance, Golden Empress, Algonquin, Zhaodong; Susceptible, Gibraltar. In conclusion, evaluation of cultivar resistance to aphids can provide baseline data for selecting and planting cultivars with high local resistance.  相似文献

通过福林法、葸酮法、滴定法等分别测定了不同抗蚜性小麦品种植株中酚、糖含量及酸度。结果表明:小麦对禾缢管蚜尺Rhopalosiphum padi(Linnaeus)的抗性与小麦体内酚含量有关,抗性强的小麦其体内含酚量相对高,反之低。同时小麦抗性的强弱与体内糖含量,酸度亦有相关性。另外还测走了在不同抗性小麦品种上生长的蚜虫体内淀粉酶及羧酸酯酶的活性,结果表明在抗性强的小麦品种上生长的蚜虫体内淀粉酶,羧酸酯酶活性强,反之弱。  相似文献

European pear psylla Cacopsylla pyri L. (Hemiptera Psyllidae) is one of the worst pests of pear (Pyrus communis L.) in Europe. We investigated probing behaviour in adults and nymphs of C. pyri by full EPG on a psylla‐resistant pear selection, NY 10353. Concerning stylet probing behaviour on the plant surface, the results showed no significant differences between the resistant selection and the susceptible cultivar Bartlett, and no differences were also detected for epidermis and mesophyll resistance in the same conditions. For mesophyll/phloem, no differences were found in adults. However, in nymphs, weak resistance factors (longer stylet penetration and mesophyll salivation) were detected in the resistant selection. In phloem, EPG data indicate strong resistance factors in NY 10353, especially for nymphs and summer‐form adults (longer time before the first phloem ingestion and a lower duration of each phloem ingestion event). No prolonged (>10 min) phloem ingestion was performed by nymphs and adults in the resistant selection. The results support the hypothesis that NY 10353 resistance factors are located in the phloem sap and cause high C. pyri nymph mortality: this could be useful as a basis for further investigations of resistance mechanisms at the metabolic, chemical and genetic levels.  相似文献

The electrical penetration graph (EPG) method was used to analyse the feeding behaviour of apterous, adultMyzus persicae (Sulzer) (Homoptera: Aphididae) onNicotiana clevelandii (Gray) seedlings, treated systemically with azadirachtin. A preliminary experiment showed that the effects of tethering aphids for EPG recording were minimal. The percentage of the 9 h recording period devoted to non-penetration activities, and to stylet pathway patterns increased as the azadirachtin concentration in the root treatment increased. The number of probes initiated, and the numbers of sieve tube penetrations also increased with increased azadirachtin concentration. The mean time elapsing between the initiation of the first probe to reach a sieve element and contact with this tissue was not significantly altered by azadirachtin treatment. However, azadirachtin treatment significantly reduced the percentage of probes that reached sieve elements and increased non-penetration activity before and after the first perid of ingestion from the sieve elements. The percentage of the recording period spent in the EPG pattern associated with sieve tube penetration was significantly reduced by an azadirachtin concentration of 300 ppm, and the duration of each individual penetration was significantly reduced by an azadirachtin concentration of 100 ppm. When the total EPG was split into 3 h periods, significant interactions were seen between time period and azadirachtin concentration for the duration of non-penetration, pathway, and sieve tube penetration patterns.  相似文献

Analysis of electrically recorded feeding behaviour of aphids was combined with colony‐development tests to search for sources of resistance to Myzus persicae (Sulzer) (Homoptera: Aphididae) in tuber‐bearing Solanum species (Solanaceae), aiming at a reduction of potato leaf roll virus (PLRV) transmission. Twenty genotypes, originating from 14 gene bank accessions, representing 13 wild tuber‐bearing Solanum spp., three Solanum tuberosum L. (potato) cultivars, and one S. tuberosum breeding line, were selected. Colony‐development tests were carried out in no‐choice experiments by placing adult aphids on plants of each genotype and counting numbers of nymphs and adults on young plants after 8 and 15 days, and on flowering plants after 14 and 30 days. Large differences were observed among genotypes: some developed small colonies and others developed large ones. Also, in a few genotypes, resistance in mature plants was different for leaves of different ages; young leaves were resistant to aphids whereas old senescent leaves were susceptible. The electrical penetration graph (DC‐EPG system) technique was used to study aphid feeding behaviour on each Solanum genotype for 6 h. Electrical penetration graph (EPG) results also showed large differences among the genotypes, indicating resistance at the leaf surface and at three different levels of plant tissue (epidermis, mesophyll, and phloem). Therefore, it was concluded that different mechanisms of resistance to M. persicae exist among the genotypes analysed. EPGs recorded from aphids on Solanum berthaultii Hawkes and Solanum tarijense Hawkes with and without glandular trichomes showed that strong surface resistance can bias EPG parameters associated with resistance located in deeper tissues. Experimental evidence is presented that the resistance to aphids in the genotypes with glandular trichomes strongly depends on these morphological structures.  相似文献

棉铃虫对Bt生物农药早期抗性及与转Bt基因棉抗虫性的关系   总被引:19,自引:0,他引:19  
用饲料感染法建立了棉铃虫Helicoverpa rmigera(Hubmer)敏感品系(SUS1)对Bt生物农药的敏感毒力基线和区分剂量,1995年测定了五省六县棉铃虫初孵幼虫对Bt生物农药的敏感性,结果表明:山东阳谷、河北邯郸、河南新乡、安徽萧县及江苏丰县棉铃虫已产生早期抗性,抗性个体百分率为5%~10%,与敏感品系相比,LC50值稍有增加,但斜率b值明显变小;而江苏东台棉铃虫仍属敏感。这是国内外首次诊测到棉铃虫对Bt生物农药抗性。用棉叶喂饲法测定比较了转Bt基因棉花品系对不同种群棉铃虫的抗虫性效果,结果表明:用早期抗性的阳谷和新乡棉铃虫初孵幼虫接虫5d后平均死亡率较敏感品系下降16%~29%,说明棉铃虫对Bt农药与转Bt生物基因棉花品系间存在交互抗性。还讨论了Bt农药的抗性治理对策。  相似文献

The role of morphological characteristics of tomato leaves inthe infection process of the pathogen Pseudomonas syringae pv.tomato was studied in 15 cultivars of varying susceptibility.Natural openings such as stomata, broken trichomes and cuticularcracks were counted. 棉蚜获毒后禁食对其保持并传播黄瓜花叶病毒的影响   总被引:5,自引:0,他引:5  
采用棉蚜Aphis gossypii 甜瓜Cumumis melo 黄瓜花叶病毒(cucumber mosaic virus, CMV)体系,研究棉蚜获毒后在空气中禁食对其保持并传播黄瓜花叶病毒的影响。结果表明获毒后的禁食时间与棉蚜传毒效率呈负相关。运用EPG (electrical penetration graph)及其即时显示、即时中断技术研究分析棉蚜禁食后的早期传毒行为细节。结果显示:禁食处理没有显著影响电势落差(potential drop,pd)数目及穿刺过程中出现的第一个pd波形前穿刺时间这两个重要指标,但禁食处理能引起pd波的两个亚波形pdⅡ-1和pdⅡ-2持续时间的显著减短。进一步分析未禁食棉蚜传毒作用与pd亚波形的关系,显示传毒可能与pdⅡ-2的持续时间相关(P=0.06)。因此,pdⅡ-2的持续时间可能是与棉蚜传毒相关的一个行为指标。该研究还建立了新的高效而稳定的获毒方法---5pd获毒法,与传统的5min获毒法相比,获毒效率显著提高。  相似文献
