To obtain basic information on the endocrine mechanisms underlying sex change in the protandrous anemonefish Amphiprion clarkii, we examined the immunolocalization of the steroidogenic enzyme cytochrome 11beta-hydroxylase (P45011beta), which is involved in 11-ketotestosterone (11-KT) production, and analyzed the ability of gonads to produce steroid hormones throughout the sex differentiation process and at the breeding stage. Immunopositive reactions against P45011beta appeared in sexually undifferentiated gonads at 30 days post hatching (dph). The number of immunopositive cells continued to increase during ovarian differentiation (from 60 to 180 dph) and throughout the formation of ambisexual gonads with both ovarian and testicular tissue until 270 dph. In the male phase, strongly immunopositive cells were observed in the cellular interstices of both testicular and ovarian tissues. P45011beta was localized only in the theca cells enclosing developed oocytes in the female phase. In-vitro 11-KT production in the gonads gradually increased with testicular differentiation (before, during, and after differentiation). Production of 11-KT in the gonads was higher in the male phase than during testicular differentiation or in the female phase. Our results suggest that androgen is involved in testicular differentiation during sex differentiation and spermatogenesis.  相似文献

Mating system and protandrous sex change of the lizard flathead Inegocia japonica were investigated off Nagashima, Kagoshima, Japan. The reproductive season of this species was estimated to be from June to September by a fluctuation of gonadsomatic index from monthly sampling data. The hermaphrodite fish, having testicular and ovarian parts, were collected from February to December, indicating that the sex change occurred in both reproductive and nonreproductive seasons. Females were larger than males and hermaphrodites. Moreover, five cases of the sex change from male to female were directly confirmed during the 1995–1997 field census in the behavioral observation area. Home ranges, where no aggressive interaction took place among any individuals, overlapped within and between sexes. Seven pair spawnings observed in the area were not of the same individual combination, and the size of pair fish was likely to be random. This species may thus have a promiscuous mating system, i.e., random pairing, which favors protandry as predicted in the size-advantage model.  相似文献

The majority of XX ? XY chimeric mice develop into fertile males. The sexual differentiation of the gonads in these animals has been examined on days 12–14 postcoitum to determine if their development parallels that of normal testes. It was found that 50% of chimeric fetuses, the proportion predicted to be XX ? XY, had neither normal testes nor ovaries. Instead, ovotestes were present, with varying proportions of presumptive ovarian and testicular tissue. On day 12 the ovotestes were organized with testicular tissue in the central region and ovarian tissue at the craniad and/or caudad poles. In the more advanced fetuses there was evidence of regression of the ovarian portion, which would account for the testes found in adults. These results are discussed in light of current theories of sex determination and differentiation and what was previously known about gonads of sex mosaics.  相似文献

To clarify the relationship between steroid hormones and sex differentiation of the protandrous anemonefish Amphiprion clarkii, we histologically examined its gonadal differentiation. From hatching to 30 days post hatching (dph), all of the gonads surveyed were sexually undifferentiated. The gonads of all fish first differentiated into ovaries at 60 dph, and the oocytes gradually developed and increased in number as the ovaries grew up until 183 dph. Some cysts of differentiated spermatogenic germ cells appeared in the ovaries at 214 dph, and ambisexual gonads with both ovarian and testicular tissues formed by 273 dph. Using immunohistochemistry, we then investigated the expression of cytochrome P450 cholesterol side-chain cleavage enzyme (P450scc), during gonadal sex differentiation. P450scc-immunopositive reactions first appeared in sexually undifferentiated gonads at 30 dph. Beginning at 60 dph, the number of strongly positive cells increased throughout the differentiation of the ovaries and continued to increase during the testicular differentiation until 210 dph. Immunopositive cells were observed more frequently in ovarian tissue than in testicular tissue in the ambisexual gonads at 270 dph. These results suggest that endogenous steroid hormones are important for the sex differentiation, including the primary sex differentiation and subsequent testicular differentiation, of the anemonefish.  相似文献

Aromatase inhibitors administered before sexual differentiation of the gonads can induce sex reversal in female chickens. To analyze the process of sex reversal, we have followed for several months the changes induced by Fadrozole, a nonsteroidal aromatase inhibitor, in gonadal aromatase activity and in morphology and structure of the female genital system. Fadrozole was injected into eggs on day four of incubation, and its effects were examined during the embryonic development and for eight months after hatching. In control females, aromatase activity in the right and the left gonad was high in the middle third of embryonic development, and then decreased up to hatching. After hatching, aromatase activity increased in the left ovary, in particular during folliculogenesis, whereas in the right regressing gonad, it continued to decrease to reach testicular levels at one month. In treated females, masculinization of the genital system was characterized by the maintenance of the right gonad and its differentiation into a testis, and by the differentiation of the left gonad into an ovotestis or a testis; however, in all individuals, the left Müllerian duct and the posterior part of the right Müllerian duct were maintained. In testes and ovotestes, aromatase activity was lower than in gonads of control females (except in the right gonad as of one month after hatching) but remained higher than in testes of control and treated males. Moreover, in ovotestes, aromatase activity was higher in parts displaying follicles than in parts devoid of follicles. The main structural changes in the gonads during sex reversal were partial (in ovotestes) or complete (in testes) degeneration of the cortex in the left gonad, and formation of an albuginea and differentiation of testicular cords/tubes in the two gonads. Testicular cords/tubes transdifferentiated from ovarian medullary cords and lacunae whose epithelium thickened and became Sertolian. Transdifferentiation occurred all along embryonic and postnatal development; thus, new testicular cords/tubes were continuously formed while others degenerated. The sex reversed gonads were also characterized by an abundant fibrous interstitial tissue and abnormal medullary condensations of lymphoid-like cells; in the persisting testicular cords/tubes, spermatogenesis was delayed and impaired. Related to aromatase activity, persistence of too high levels of estrogens can explain the presence of oviducts, gonadal abnormalities and infertility in sex reversed females.  相似文献

为探明中国大鲵(Andrias davidianus)雌雄幼体的性腺发育特征,确定适合的性别分子鉴定方法,对15尾5月龄和17尾17月龄养殖个体进行形态测量、解剖观察、性腺组织切片及PCR扩增雌性特异DNA片段。结果发现,引物adf225和adf340的扩增效果好,判定5月龄个体8雌7雄;17月龄个体8雌9雄,与依据性腺形态结构区分的结果一致。体视显微镜下5月龄幼体中肾腹侧有两条半透明细条状的原始生殖嵴;组织切片显示生殖细胞形态分化不明显。17月龄卵巢波浪状弯曲,有颗粒感,精巢呈光滑的白条状,形态分化明显;组织切片显示,卵巢分化出体积较大的卵母细胞,同时保留原始卵泡,精巢分化出生精小叶和精原细胞、支持细胞。外形测量显示,5月龄与17月龄性二型不明显,不能根据外形判断性别。本研究确定了大鲵幼体性别分子鉴定的最佳引物,可用于养殖过程中雌雄选配,以节约资源。  相似文献

Handling is a source of stress for farm bred blue foxes. The influence of handling during the late gestational period was investigated in 10-day-old male and female blue foxes. Testosterone and estradiol were measured by RIA in the plasma, gonadal homogenates andin vitro incubates, from blue foxes of both sexes. The gonads were incubatedin vitro without or with human chorionic gonadotropin. In cubs of both sexes, the gonad weights and ovarian estradiol production were decreased by stress. The testicular testosterone and ovarian estradiol contents were increased in prenatally stressed cubs as compared to the controls. The testicular content and baselinein vitro production of testosterone were not affected by prenatal stress, but the testicular response to human chorionic gonadotropin was higher in the stressed group. This study suggests that prenatal stress induced by handling pregnant vixens may influence gonadal steroidogenesis and that this effect was more pronounced in female cubs.  相似文献

The present paper deals with gonadal formation, sex succession and sexual function in the Japanese hawkfishes,Cirrhitichthys aureus (Temminck et Schlegel),C. aprinus (Cuvier),C. falco Randall,Cirrhitops hubbardi (Schultz) andCyprinocirrhites polyactis (Bleeker). Detailed studies were made forC. aureus collected from Suruga Bay, central Japan. In this species the gonad of a young fish of 21.8 mm SL had begun to initially differentiate into an ovary forming an early ovarian cavity of the parovarian type. The gonadal structures of 63 further specimens ofC. aureus, ranging from 23.9 to 114.3 mm SL, could be separated into three categories: viz., ovaries (7), ambisexual gonads (54) and secondary testes (2). Reproduction ofC. aureus in Suruga Bay took place from June to November. Although ovaries which appeared throughout the year presented only in the immature stage and occurred only in small fish ranging from 25.1 to 46.6 mm SL, functional gonads occurred in 32 ambisexual fish ranging from 61.5 to 113.5 mm SL and in one secondary male of 101.5 mm SL, collected during the reproductive season. The smaller ambisexual fish, ranging from 61.5 to 92.0 mm SL, showed active oogenetic processes in the major ovarian zones with many ripened eggs ovulating into the ovarian cavity. These gynogenic gonads undoubtedly functioned as female; however, distinct meiosis proceeded partially in the minor testicular zones with a few spermatozoa occurring in the vas deferens. The larger ambisexual specimens, ranging from 71.5 to 113.5mm SL, had slender androgenic gonads, which showed active spermatogenesis in various testicular zones and which might function as males with reduced ovigerous lamellae. The secondary males had testes occupied by seminal lobules, but which retained the ovarian cavity within. According to aquarium rearing experiments, in a pair ofC. aureus twice crossed mutually after spawning, functional sex changes occurred in twice, opposite directions, in a single gonad, i.e., from female to male and from male to female. These gonadal aspects in captiveC. aureus seemed to show an intermediate phenomenon between consecutive and simultaneous hermaphroditism. This finding also suggested thatC. aureus had similar sex succession characteristics to those of the serranid,Serranus fasciatus, studied by Hasting and Petersen (1986) and the gobiid,Trimma okinawae, studied by Sunobe (1990). We also established that normal functional hermaphroditism occurred in the four other cirrhitid species, which showed almost identical gonadal characteristics toC. aureus.  相似文献

A testis from an 18-day-old chick embryo was transplanted into the extra-coelomic cavity of 3-4-day-old hosts. The embryos surviving at 17 days were sacrificed and their genital system was examined. Testis grafting produced inhibition of testicular development. Development of the female gonads was also inhibited. A more or less complete modification of sex was associated with this inhibition. The left ovary lost its cortex, but its medulla remained mostly ovarian in structure. The right gonad frequently acquired a typical testicular structure. These results confirm the possibility of obtaining sex reversal in the female chick embryo by testis grafting.  相似文献

The gonads of Lepidogalaxias salamandroides are structurally distinctive. A duct, the surface of which is ciliated, is located on the ventral surface of each ovary and may be a modification of the ovarian tunica. The testes are small and the majority of the testicular structure functions as a sperm storage area. Secondary requisite sex characters are present in the male and the function of a modified scale sheath in copulation is unique among teleosts. Fertilization is internal and a viscous mucus mass secreted during copulation may serve as a copulatory plug. It is postulated that internal fertilization within this species has arisen as a response to sperm competition.  相似文献

All gonads of the Hawaiian dascyllus Dascyllus albisella , irrespective of the final sex of individuals, developed an ovarian lumen and primary-growth-stage oocytes after an initially undifferentiated state. From this ovarian state or from more differentiated ovaries, some gonads redifferentiated into testes. None of 117 individuals examined had a gonad containing degenerating vitellogenic oocytes and proliferating spermatogenic tissue. Eleven individuals had gonads containing degenerating cortical-alveolus-stage oocytes and developing spermatogenic tissue. The size of these individuals overlapped with the female size range in which the majority of the females were still in the middle of the maturation process. They were absent from the larger size range where the majority of females had vitellogenic oocytes. This indicated that the transition toward maleness is likely to have occurred after the onset of cortical-alveolus stage, but before final oocyte maturation and spawning as females. Therefore the protogynous pattern of gonadal development was non-functional. There was no dimorphism in the sperm duct configuration, and all the testes were secondary testes reported for diandric, protogynous species with undelimited gonads. Very early development of an ovarian lumen appeared to have resulted in a secondary-male configuration in all testes, although redifferentiation into males appeared to have occurred before sexual maturity and spawning as females.  相似文献

The sex-specific molecular marker is a useful gene resource for studying sex- determining mechanisms and controlling fish sex. Artificially produced male and female half-smooth tongue sole (Cynoglossus semilaevis) were used to screen sex-specific amplified fragment length polymorphism (AFLPs) molecular markers. The phenotypic sex of 28 tongue soles was determined by histological sectioning of gonads. The AFLP analysis of 15 females and 13 males via 64 primer combinations produced a total of 4681 scorable bands, of which 42.11% and 43.39% of bands were polymorphic in females and males, respectively. Seven female-specific AFLP markers were identified and designated as CseF382, CseF575, CseF783, CseF464, CseF136, CseF618, and CseF305, respectively. One female-specific AFLP marker (CseF382) was amplified, recovered from the gels, cloned, and sequenced (accession no. DQ487760). This female-specific AFLP marker was converted into a single-locus polymerase-chain reaction (PCR) marker of a sequence-characterized amplified region (SCAR). A simple PCR method of using the specific primers was developed for identifying genetic sex of half-smooth tongue sole. PCR products demonstrated that the initial 15 females produced the female-specific band of about 350 bp, but the initial 13 male individuals failed to produce the band. We also investigated the applicability of the PCR primers in other tongue sole individuals. The same female-specific fragment of about 350 bp was found in the additional 59 female individuals, but not in the additional 58 male individuals. This AFLP-based molecular sexing technique may have great application potential in elucidation of sex determination mechanisms and sex control in half-smooth tongue sole.  相似文献

Abstract. Individuals with 46, XY pure gonadal dysgenesis present with a completely female phenotype. These individuals develop bilateral streak gonads and have normal Müllerian structures. The apparent absence of testicular tissue in these individuals suggests a mutation in the initial steps of the male sex-determining pathway. A candidate gene for the primary signal in this pathway was recently cloned ( SRY ) which encodes a protein with a DNA-binding capacity. In a study of 14 XY females with pure gonadal dysgenesis harbouring SRY , we analysed the histology of the gonads and compared it to the presence or absence of mutations in the SRY open reading frame ( SRY -orf). The histological analysis revealed two distinct groups of streak gonads. In the first group, the gonad was composed of exclusively ovarian-like stroma, with sclero-hyaline nodules in some areas. No tubules were observed. The gonads in the second group were composed of undifferentiated stroma harbouring either tubules or a rete structure. This suggests that in the latter group some differentiation (towards testis formation) has occurred, whereas in the first group ovarian differentiation has been interrupted. Individuals with mutations in the SRY -orf were found to have streak gonads of the first group, whereas most of the remaining XY females without detectable mutation in the SRY -orf had streak gonads belonging to the second group.
On the basis of histology, it may be possible to distinguish between mutations in the sex-determining or sex-differentiation pathways. We suggest that SRY may play a role in rete testis formation. We also present arguments favouring the mesonephros as the origin of testicular somatic cells in humans.  相似文献

Identifying the sex of larvae is important in social Hymenoptera. Until now for Polistes wasps it has been necessary to genotype larvae at microsatellite loci, and assign their sex based on homozygosity at these loci. In our study on the paper wasp Polistes dominulus we have found morphological differences between larval sexes that can be used for larvae from the 3rd instar on to easily and cheaply identify larval sex: the external gonopore and the shape and size of larval gonads. The robustness of these indicators was supported by genotype data at four microsatellite loci. Using gonopore and gonad features for sex assignment will assign diploid males as males, unlike techniques based on genetic loci or chromosomes. A total of 141 Polydactylus quadrifilis were sampled from the Kwanza Estuary in Angola ranging in size from 436 to 1360 mm fork-length (FL). Of these, 124 were male, six intersex and 11 female. Female fish were significantly longer (mm, FL) and heavier (kg) than males and had significantly higher gonadosomatic indices (GSI’s) than those of males and intersex fish. Transitional (intersex) gonads were delimited, with testicular and ovarian regions separated by connective tissue. The first signs of ovarian tissue appeared on the outer ventral surface of the gonad. A second layer of ovarian tissue was first noticeable at either end of the initial ovarian region and developed back towards the centre of the ventral wall to form a luminal space. Early-stage oocytes were commonly found in the outer area of male regions and residual late-stage spermatids and spermatozoa were found in the luminal space of ovarian regions, suggesting a process of sex change from the outside inwards. A loss of male function was noted with increased ovarian prevalence. Based on this evidence it is suggested that P. quadrifilis in the Kwanza Estuary are protandrous. Owing to the reliance of P. quadrifilis on large highly fecund females for egg production, it is likely that they will be sensitive to fishing practices that target larger individuals within the population.  相似文献

A primary role of P-glycoprotein (P-gp), encoded by the multidrug resistance type I gene, is to protect against naturally occurring xenotoxics. Recently, the preferential expression of chicken multidrug resistance type I (Cmdr1) was identified in the embryonic gonads during the early periods of development. Here we investigated the expression of Cmdr1 and P-gp in the gonads during embryogenesis, and compared to that in the ovarian follicles of domestic hens (Gallus gallus). As revealed by immunohistochemistry, P-gp was highly expressed in theca cells of mature follicles, whereas the expression was low in immature follicles. Immunohistochemical analysis showed that expression of Cmdr1-type P-gp was very low in embryonic gonads. Cmdr1 mRNA was undetectable in the gonads of 5-day embryos (E5) by RT-PCR, whereas Cmdr1 mRNA was significantly detectable in the developing gonads at E9 and E21. In the testicular tissues, germ cells were distributed along developing seminiferous cords as identified by a specific marker gene, whereas Cmdr1-type P-gp positive cells were observed evenly on testicular tissues. Collectively, it is concluded that Cmdr1 expression is initiated in the chicken ovary and testis after sexual differentiation, but expression of Cmdr1-type P-gp is very low through embryogenesis.  相似文献

Patterns of gonad structure in hermaphroditic gobies (Teleostei Gobiidae)   总被引:5,自引:0,他引:5  
Synopsis Hermaphroditism has been reported for a small number of gobiid fishes, but the extent of this sexual pattern within the family is not known. Gonad structure was examined in one or more species from twenty-one gobiid genera. No evidence of hermaphroditism was found in the species selected from 14 genera. Laboratory studies supported the conclusion of gonochorism for the examined species in four of them:Asterropteryx, Bathygobius, Gnatholepis, andPsilogobius. Currently, the absence of precursive testicular tissues associated with the ovary in females, in conjunction with no retained ovarian features in the testes of males, appear to be reliable indicators of a gonochoristic sexual pattern in gobiid fishes. Evidence for hermaphroditism was observed in seven genera:Eviota, Trimma, Fusigobius, Lophogobius, Priolepis, Gobiodon, andParagobiodon. Protogyny was experimentally confirmed inE. epiphanes, and the gonad structure in another nine of ten species ofEviota suggested either protogyny or protogynous tendencies. With the exception ofGobiodon andParagobiodon, which exhibited similar gonadal structure, ovarian and testicular structure varied considerably among the hermaphroditic genera examined, both with regard to the configuration and to the degree of development of ovarian and testicular tissues, or testicular tissue precursors. Findings of this study indicate that hermaphroditic gonad structure will prove to be a useful trait in determining evolutionary relationships within the Gobiidae.  相似文献
