刺芹侧耳生长条件和栽培特性的研究   总被引:2,自引:0,他引:2  
对刺芹侧耳[Pleurotus eryngii(DC.ex Fr.)Quel]生长条件与栽培特性进行了分析.结果表明:刺芹侧耳菌丝生长的适宜pH值为pH 5~pH 7,最适pH值为pH 6;黑暗环境有利于菌丝生长,光照对菌丝生长具有抑制作用;菌丝生长适宜的碳源为可溶性淀粉、葡萄糖和蔗糖,适宜的氮源为蛋白胨和钼酸铵,葡萄糖和蛋白胨的适宜浓度分别为50和1 g·L-1;在马铃薯黄豆粉培养基、黄豆粉玉米粉培养基和马铃薯蛋白胨培养基中可较好地形成菌丝球.采用液体菌种栽培,菌丝的生长速度比使用玉米固体菌种快,其中用马铃薯黄豆粉液体菌种栽培,菌丝生长最快;使用马铃薯蛋白胨液体菌种在木屑棉籽壳混合培养料中栽培,可获得菌柄长菌盖大的子实体,而且产量高.  相似文献

用正交试验法对樟芝菌 (AntrodiacamphorataZang&Su)的发酵培养基进行了研究。选取马铃薯淀粉、葡萄糖、麦芽糖、酵母膏、蛋白胨、麸皮、KH2 PO4 、MgSO4 ·7H2 O等作为试验因素 ,采用L2 7(31. 3)正交表设计五因素、三水平的正交试验 ,对所选各因素的添加量分别作了试验 ,对菌丝干重和发酵液多糖含量结果作了方差分析 ,确定了合适的发酵培养基。试验结果表明 :樟芝菌的深层发酵适宜培养基是 :马铃薯淀粉 1 .0 0 % ,葡萄糖2. 0 0 % ,酵母膏 0 . 10 % ,蛋白胨 0 . 10 % ,麸皮 1 .0 0 % (或 0. 5 0 % ) ,KH2 PO4 0 . 10 % ,MgSO4 ·7H2 O 0 . 0 5 %。  相似文献

竹荪(Dictyophora rubrovalvata)在PDA琼脂培养基上培养,菌丝体生长很慢,若在PDA培养基上分别添加酵母膏、生啤酒、天冬酰胺、核糖核酸,可促进菌丝体的生长,菌落直径生长和菌丝线性生长都加快;添加蛋白胨、椰子汁、硫胺素和肌醇效果不明显。菌丝体生长的pH值在4.4—5.4之间,最适pH值为4.7;pH值在6.7以上,菌丝体不生长。在木屑培养基中分别添加麦芽汁、玉米、高粱和黄豆粉可以增加子实体的鲜重,其中黄豆和玉米粉、黄豆和高梁粉混合使用的效果比单用一种的更好,菇产量比对照增加1—2倍。播种后基质中含水量控制在50—60%,当原基形成时,含水量提高到55—70%,空气相对湿度在95%以上,菌蕾才能发育成子实体。在自然温度条件下栽培竹荪,出菇高峰期出现在八月份,基质内平均温度为22.4℃,子实体生长的最适温度在20—22.5℃之间,在此温度下产菇率占总产值的85%。  相似文献

探讨了不同营养基质与条件对灰树花[Grifola frondosa(Fr.)S.F.Gray]生长的影响。试验结果表明:菌丝生长的最适pH为5-6;最适氮源为蛋白胨、牛肉膏和谷氨酰胺,葡萄糖和蛋白胨最适浓度分别为100和1g/L;菌丝在黑暗环境下生长良好;使用马铃薯蛋白胨培养基进行振荡培养,菌丝球生长较好;母种采用马铃薯麦麸培养基,栽培种选用棉子壳麦麸石培养料,可缩短制种周期,获得质量较高的菌种;菌丝含有较多的钙、铁和锌等元素。  相似文献

在哈尔滨地区使用液体菌种栽培红平菇技术初探   总被引:3,自引:0,他引:3  
从福建三明真菌研究所(福建省三明市375000)购入红平菇优良菌株,在哈尔滨地区对其进行了用液体菌种代替固体原料的栽培技术研究。包括一级种培养基的筛选、液体菌种培养基的筛选、栽培种制作与培养以及出菇管理技术。结果表明:对于一级种,红平菇在PDA和马铃薯半组合培养基上都生长良好,菌丝洁白、粗壮、浓密,7~8d长满斜面;红平菇在玉米粉葡萄糖蛋白胨和麦麸葡萄糖蛋白胨液体培养基上生长较好,7d时菌丝长满培养液,菌丝量较多;用与平菇相似的常规方法制作栽培种、养菌处理与出菇管理,红平菇的产量达100%~150%。由于采用了液体菌种的栽培模式,从制种到采收的整个过程共需要50~60d,比二级种为固体菌种的常规方法缩短26~31d,大大地缩短了生产周期,提高了经济效益。  相似文献

蜜环菌属Armillaria真菌具有较高的食药用价值。由于蜜环菌的生长发育过程较复杂,还未完全实现商业化栽培,野生资源的供应受到季节性和地域性的影响。本研究以采自东北地区蜜环菌属的3个菌株为研究对象,通过培养物的形态特征及分子标记确定菌株JG19016为奥氏蜜环菌A. ostoyae,菌株JG19017为高卢蜜环菌A. gallica,菌株JG19018为中国蜜环菌生物种C。奥氏蜜环菌JG19016最适生长温度为25 ℃,高卢蜜环菌JG19017的最适生长温度为22 ℃,中国蜜环菌生物种C JG19018则在22-25 ℃时菌丝生长速度最快;3个菌株最适pH为5-6。奥氏蜜环菌JG19016对葡萄糖和蔗糖利用率较好,高卢蜜环菌JG19017对葡萄糖利用率较好,中国蜜环菌生物种C JG19018对葡萄糖和淀粉利用率较好;蛋白胨对3个菌株促进作用最强,为最适氮源。培养基中加入VB1,对3个菌株的菌丝生长均有明显的促进作用。奥氏蜜环菌JG19016菌丝生长的最优培养基配方为:葡萄糖20 g,蛋白胨3 g,磷酸二氢钾2 g,硫酸镁1.5 g,VB1 10 mg,琼脂20 g,水1 L。在木屑基质中培养,其配方的最优碳氮比为38:1,最佳木屑粗细比为3:1以上。出菇条件探索结果显示,菌丝及菌索长满菌袋(17 mm×33 mm×5 mm丝聚乙烯袋)需要50-60 d,之后在18 ℃、60%湿度和12 h散射光的环境中,10 d左右可观察到原基产生。增加菇房湿度到90%-95%,2-3 d可观察到1-3 cm的幼子实体,7 d左右菌柄和菌盖完全分化,10 d左右观察到菌盖展开。  相似文献

研究了四种碳源(蔗糖、乳糖、葡萄糖、可溶性淀粉)和九种氮源(玉米面、麸皮、马铃薯、大豆粉、酵母粉、蛋白胨、硝酸钾、硝酸铵、尿素)对黑盖木层孔菌菌丝生长的影响。从不同代数的菌丝体中提取多糖,并测定多糖含量、分子量分布范围及单糖组成。结果表明:黑盖木层孔菌菌丝生长的最佳碳源为可溶性淀粉,最佳氮源为玉米面,最优碳氮组合为蔗糖和玉米面的组合。不同代数的菌丝体多糖性状基本稳定。  相似文献

培养基质碳氮比显著影响食用菌菌丝生长及子实体生长发育。广叶绣球菌是一种珍贵的食药用真菌,前期研究发现其生长发育过程中对碳源的需求较多。本研究采用松木屑、复合氮源作为栽培原料,研究基质碳氮比对绣球菌菌丝生长及子实体生长发育的影响。结果表明:6种不同碳氮比(C/N)的栽培基质对菌丝生长速度无显著影响,但是随着C/N比值的增加,子实体形成期逐渐延长,子实体形成率呈先增大后减小的趋势。当基质C/N在48.8-55.9之间时,采收率较高,在80-90d内,达到采收标准的菌袋比例分别为83.7%和81.4%。当基质C/N为43.4时,鲜菇产量最高,但外观品质受影响。综合考虑生产成本及效率,采用松木屑和复合氮源作为栽培原料、适宜的基质C/N为48.8时,绣球菌子实体形成期为64d,子实体形成率为95%且较为集中,鲜菇平均产量为231g/袋(76%集中于200-260g/袋),整个生产周期为84d。本研究结果表明绣球菌具有“高碳低氮”的营养生理特性。  相似文献

本文报道了不同碳源、氮源、微量元素及维生素培养基对干巴菌菌丝生长的影响。试验表明,干巴菌菌丝利用最好的碳原是葡萄糖,其次是蔗糖;利用最好的氮源是硝酸钙,其次是硝酸铁和硫酸饮,对蛋白际和尿素的利用效果差。缺少联源或氮源时菌丝生长细弱、稀少,不形成原基。微量元素和维生素均对干巴菌菌丝生长有促进作用,其中以锰、铜和维生素B2的作用尤为突出。  相似文献

为实现漏斗多孔菌资源化利用,设计单因素试验,以菌丝生物量、菌球密度和菌球直径为指标,获得漏斗多孔菌Polyporus arcularius液体菌种培养基配方为马铃薯(去皮)200g、玉米粉20.0g、蛋白胨5.0g、KH2PO4 3.0g、K2HPO4 1.0g,MgSO4·7H2O 1.5g,初始pH 5.0,并优化培养条件为装液量250mL/500mL,接种量2%,培养温度32℃,转速170r/min,种龄6d;依上述条件制备液体菌种,以固体菌种为对照,进行出菇试验,调查农艺性状,试验结果表明液体菌种接种后,菌袋满袋时间提前7d,子实体原基产生时间提前8d,头潮菇鲜重提高22.5%;配方试验结果表明,以细木屑为主料的配方产量最高,头潮菇鲜重为270.6g/袋。  相似文献

从碳源、氮源、酸碱度及生长温度等方面对引起马铃薯干腐病的硫色镰孢Fusarium sulphureum的生物学特性进行了研究。结果表明,该病原菌在不同发育阶段对营养和环境条件的要求存在差异。在固体培养基上,菌落生长最佳碳源为葡萄糖、麦芽糖,最佳氮源为蛋白胨,pH8?为最佳;在液体培养基中,菌丝体生长以麦芽糖为最佳碳源,以硝酸钠为最佳氮源,pH6?为最佳;在分生孢子萌发阶段,在以羧甲基纤维素钠、蛋白胨和谷氨酸为碳、氮源的营养液中,分生孢子萌发率最高,最适pH 6–8。该病菌最适生长温度为25℃,分生孢子致死  相似文献

Summary The production of protease and mycelium byNeurospora sitophila cultured on solid and liquid potato dextrose media was studied. Maximal activity of protease extracted from 4-day-old cultures occurred at pH 6.5 when an unfractionated peanut (groundnut) protein substrate was used. The greatest protease activity and mycelium production occurred during the first day of the 4-day test period. Potato dextrose media containing more than 0.2 M NaCl resulted in decreased protease and mycelium production, while tapioca starch was without affect at concentrations up to 1.4%. Addition of up to 0.3 M sucrose to growth media greatly stimulated protease production and mycelial growth. Maximal proteolytic activity was observed in extracts from mycelium cultured in potato dextrose media adjusted from pH 6.0 to 7.5. Activity was greater when soluble peanut protein was used as a substrate, compared to unfractionated or globulin protein substrates.  相似文献

采用 5种不同的氮素营养源 ,配制合成液体培养基 ,测定各种氮源对樟芝菌丝液体深层培养的影响。结果表明 ,樟芝对 5种氮源的利用表现明显的差异性 ,对有机氮利用较好 ,尤其是酵母膏 ,无论是菌球数量还是菌体收率均高于蛋白胨。酵母膏的菌球数量达 1 2 5 8个 /dL,比蛋白胨多出 63 8个 ;菌体干重为 7 2 1 6g/dL比蛋白胨高出 1 45 0g/dL。樟芝对无机氮的利用不如有机氮 ,其中对硫酸铵能够利用 ,而对硝酸铵的利用较差 ,对碳酸铵基本不能利用。樟芝菌丝在液体深层培养过程中 ,pH值变化幅度不大 ,基本处于平缓状态。有机氮处理的发酵液起始pH值与终止pH值呈上升趋势 ,而无机氮处理的发酵液pH值呈下降趋势 ,但下降的幅度不大 ,尤其是碳酸铵 ,pH值一直处于偏碱性状态 ,不适合樟芝菌丝的生长。樟芝在马铃薯基础培养基中 ,不同氮源的各个处理 ,菌球生长都比较均匀 ,表面光滑 ,没有分枝 ,圆形或肾形 ,且发酵液有浓郁的果香味。  相似文献

本文初步研究了不同类型的凝固剂对水稻花药培养愈伤组织形成的影响。结果发现,用Gelrite、马铃薯淀粉、甘薯淀粉、可溶性淀粉代替琼脂可明显促进水稻花药培养愈伤组织的产生而尤以5.0%马铃薯淀粉为最佳。出愈率比琼脂增加5.2倍,达液体培养水平。以8个不同基因型,为材料研究发现,5.0%马铃薯淀粉作凝固剂,有7个材料出愈率高于对照,最高的BC 163比对照增加7.75倍,平均增加1.15倍。另外,以5.0%马铃薯淀粉作凝固剂代替0.8%琼脂可降低成本30%。因此,用马铃薯淀粉作凝固剂在水稻花药培养中可能具有潜在的应用价值。  相似文献

The effect of addition of peptone to serum-free and serum supplemented media for the growth of hybridoma cells in various systems was studied. Supplementation of defined medium with either proteose peptone or meat peptone resulted in significant increases in cell number and specific monoclonal antibody production in batch culture system. Other peptones were either inactive or less effective. In continuous culture, using medium supplemented with new born calf serum, the addition of peptone resulted in 125% and 150% increases in cell and antibody concentrations respectively. Similar increase in cell number (128%) was also obtained in spin-filter perfusion culture when medium was supplemented with peptone. By comparison, the substitution of a defined 1xMEM amino acids mixture resulted in only a 50% increase. At higher perfusion rates the cell number maintained in steady state using peptone supplement could be increased to 1.3×107 cells ml–1 while the serum concentration was reduced from 5% to 1% at a perfusion rate of 2.5 volumes per day.  相似文献

Summary For mycological fat production,Aspergillus nidulans andPenicillium lilacinum were grown on media made up of crushed and boiled sweet potatoes. Mycelial mats containing a fair amount of fat were obtained, namely, 15 % in case ofAspergillus and 24 % in case ofPenicillium. Media containing an external supply of glucose gave rise to increase in percentage fat in mycelium, but the amount of mycelium and consequently the total fat content decreased.Addition of sodium nitrate gave heavier mycelial mats but the total fat content could not be raised.Addition of potassium phosphate or magnesium sulphate or both to the sweet potato medium gave rise to heavier mycelial mats but with lower fat content.  相似文献

Two fungi, namely,Aspergillus oryzae andAspergillus terreus, have been grown on enzyme-hydrolyzed sweet potato medium either as such or supplemented with an external source of nitrogen in the form of ammonium carbonate, sodium nitrate, or asparagine. Hydrolysis alone promoted carbohydrate uptake, growth as well as fat formation to a remarkable extent in both fungi.Addition of an external supply of nitrogen to the hydrolyzed media increased further carbohydrate absorption which continued until the potato media were almost completely depleted; on unhydrolysed potato media, appreciable amounts of carbohydrates always remained unutilized. It also accelerated the rate of building up so that the dry mycelium increased by more than 4 times in some cases. Fat formation was consequently remarkably enhanced specially in the presence of ammonium carbonate or asparagine.  相似文献

Cell cultures of sweet potato grown in media containing sucrose, glucose, maltose, or starch secreted amylase into the growth medium. The growth rate of cells was not appreciably affected by the carbon source employed for growth, although cells grown on sucrose had a slightly longer lag period before exponential growth occurred. Amylase levels inside the cells were not affected by carbon source, but the amount of amylase released into the medium was drastically affected. Maltose-grown cells released the most amylase while sucrose-grown cells released the least. Cells grown in the light released about twice as much amylase as cells grown in the dark when grown on glucose, maltose, or starch.Three amylase electrophoretic forms were found in the storage root tissue from which all cultures were derived. Cells grown in culture exhibited either two or three amylase forms, depending on the carbon source. The slowest migrating root amylase was found only in cells grown on starch. The root amylase having intermediate mobility was present in all cultures, as was a form having higher mobility than the most mobile root form.
