This study determined how surgical removal of the stem terminal, with indole-3-butyric acid (IBA) treatment, influenced concentrations and partitioning of carbohydrates in Pinus banksiana Lamb, cuttings during propagation. Seedlings and cuttings that originated from 90-day-old stock plants were untreated or treated by removing the stem terminal, followed by application of IBA to the severed apical or basal (cuttings only) stem. Fresh and dry weights of the basal 1-cm stems of cuttings were determined daily for the first 10 days of propagation (i.e., before roots were visible). In addition, basal 1-cm stems, upper (ca 9-cm) stems and needles of seedlings and cuttings were analyzed for sucrose, soluble reducing sugar and total non-structural carbohydrate. Net concentrations of each carbohydrate in cuttings were obtained by subtracting corresponding concentrations for similarly treated seedlings, yielding data directly related to only the physiology of rooting. Data for cuttings indicated that presence of the stem terminal combined with applied IBA positively influenced rooting through processes that increased basal stem fresh and dry weights before root emergence. Removal of the stem terminal influenced accumulation of net total carbohydrate in cuttings, but the major effect was on carbohydrate partitioning. Either type of IBA treatment after removal of the stem terminal usually resulted in different net carbohydrate concentrations in each tissue source of cuttings, compared with only removal of the terminal. Neither basal nor apical IBA treatment of cuttings without stem terminals yielded results for carbohydrate accumulation and partitioning like those obtained with intact cuttings. Removal of the stem terminal, even if followed by IBA treatment, may have lessened rooting potential of cuttings because it resulted in greater reducing sugarstarch concentration ratios in basal stems compared with those in intact cuttings.  相似文献

Tests were conducted to identify possible relations between carbohydrates and callusing-rooting of Pinus banksiana Lamb, cuttings. Terminals, upper stems, and basal (1 cm) stems of 90-day-old untreated seedlings and seedling cuttings were analyzed for sucrose, total soluble reducing sugar, starch and total non-structural carbohydrate during propagation. Seedlings were evaluated in order to determine whether data for cuttings alone properly described carbohydrate-callusing-rooting relations under conditions where stock plants and cuttings were propagated in different environments. Results indicated that seedling terminals and upper stems, but not basal stems, accumulated the measured carbohydrates much like cuttings, though to lesser concentrations. Thus, carbohydrate accumulation by cutting terminals and upper stems would have been overestimated, based on cutting data alone. In terms of rooting, results indicated that: 1) Total carbohydrate accumulation in cutting basal stems was related to callusing-rooting, but a cause-effect relation was not established; 2) The positive relation between callusing-rooting and total carbohydrate accumulation was primarily due to accumulation of reducing sugar and starch, with reducing sugar predominant. 3) Reducing sugar/starch concentration ratios were the most sensitive and convenient indicators of specific carbohydrate differences within and between seedlings and cuttings.  相似文献

Rooting ability was studied for cuttings derived from stock plants of wild type pea seedlings and seedings of two mutants deficient in photosystem II activity and chlorophyll. Stock plants were grown at 15, 20, 25 or 30°C at 38 W m-2. Cuttings were rooted at 20°C and at an irradiance of 16 or 38 W m-2. The rooting ability seemed to be correlated with the initial carbohydrate content only at 38 W m-2. Based on the findings of the present study it may be concluded that for pea seedlings the growth temperature is more important than photosynthesis as regards accumulation of extractable carbohydrates. During the rooting period carbohydrates are necessary for root formation, but the effect of the iradiance on the number of roots formed is not mediated by the carbohydrate content. Under specific rooting conditions it is possible to correlate the initial carbohydrate content with the rooting capacity of the cuttings within a phenotype, but not always when different phenotypes are considered. The results indicate a connection between the metabolic activity of the cuttings and their ability to form adventitious roots.  相似文献

BACKGROUND AND AIMS: The aim of this study was to determine the role of nitrogen- and storage-affected carbohydrate availability in rooting of pelargonium cuttings, focusing on the environmental conditions of stock plant cultivation at low latitudes, transport of cuttings, and rooting under the low light that prevails during the winter rooting period in Central European greenhouses. METHODS: Carbohydrate partitioning in high-light-adapted cuttings of the cultivar 'Isabell' was studied in relation to survival and adventitious root formation under low light. Effects of a graduated supply of mineral nitrogen to stock plants and of cutting storage were examined. KEY RESULTS: Nitrogen deficiency raised starch levels in excised cuttings, whereas the concentrations of glucose and total sugars in leaves and the basal stem were positively correlated with internal total nitrogen (Nt). Storage reduced starch to trace levels in all leaves, but sugar levels were only reduced in tissues of non-nitrogen deficient cuttings. Sugars accumulated in the leaf lamina of stored cuttings during the rooting period, whereas carbohydrates were simultaneously exhausted in all other cutting parts including the petioles, thereby promoting leaf senescence. The positive correlation between initial Nt and root number disappeared after storage. Irrespectively of storage, higher pre-rooting leaf glucose promoted subsequent sugar accumulation in the basal stem and final root number. The positive relationships between initial sugar levels in the stems with cutting survival and in leaves with root formation under low light were confirmed in a sample survey with 21 cultivars provided from different sources at low latitudes. CONCLUSIONS: The results indicate that adventitious rooting of pelargonium cuttings can be limited by the initial amount of nitrogen reserves. However, this relationship reveals only small plasticity and is superimposed by a predominant effect of carbohydrate availability that depends on the initial leaf sugar levels, when high-light adaptation and low current light conditions impair net carbon assimilation.  相似文献

Abstract: Diurnal courses of gas exchange were measured throughout one year in fully expanded current-year leaves in the uppermost canopy (sun leaves, 18 m above ground) and in the lower canopy (shade leaves, 12 m above ground) of Myrica faya Ait., a dominant component of the Canarian laurel forest in Tenerife, Canary Islands, Spain.
M. faya showed large differences between sun and shade leaves in gas exchange characteristics (about 50 % of maximum carbon assimilation rate (Amax) reduction in shade leaves, but this reduction can be higher on specific days) that were modulated by strong light attenuation and high leaf area index (LAI) of the stand. This species presented low Amax, about 10 μmol m-2 s-1, high maximum transpiration (E, 8 mmol m-2 s-1) and stomatal conductance (gs, 750 mmol m-2 s-1) and very low instantaneous water use efficiency (WUE, mean maximum 1.1 mmol mol-1) and A/gs (mean maximum 23.5 μmol mol-1). M. faya responded to high air vapour pressure deficit (VPD), decreasing its gs but maintaining relatively high values of A and E during the studied period. Stomatal response to VPD showed a higher sensitivity than its congeners, M. cerifera, and Laurus azorica, tree species co-occurring in the Canarian laurel forest. In general, all these gas exchange characteristics lead us to consider this species more similar to subtropical plants of humid regions than to species of the Mediterranean region.  相似文献

N-phenyl indoryl-3-butyramide (NP-IBA) and phenyl indole-3-thiolobutyrate (P-ITB) are new synthetic auxins that enhance rooting of bean ( Phaseolus vulgaris cv. Top Crop) hypocotyl cuttings and jack pine ( Pinus banksiana Lamb.) seedling cuttings. In comparison with an equal concentration of indole-3-butyric acid (IBA), NP-IBA treatment resulted in greater rooting of bean and jack pine seedling cuttings. IBA and P-ITB treatment at equal concentration were equally effective in inducing rooting of bean cuttings. However, in comparison with an equal concentration of IBA, P-ITB treatment promoted greater rooting of jack pine seedling cuttings, but P-ITB treatment was less effective than NP-IBA treatment. Application of crystalline chemical to jack pine seedling cuttings indicated lesser toxic effects and earlier and greater root-promoting effects of P-ITB and NP-IBA treatments, in comparison with IBA treatment. The earlier rooting of NP-IBA-treated jack pine seedling cuttings was positively related to increase in fresh weight of basal 1 cm stem before primordia were initiated. Weight of basal steins of NP-IBA-treated cuttings initially increased during propagation at day 2 in comparison with the control, and at day 3 in comparison with IBA-treated cuttings. Results that were obtained with seedling jack pine cuttings were generally confirmed for cuttings from 7-year-old trees.  相似文献

Rooting ability was studied for cuttings derived from pea plants ( Pisum sativum , L. cv. Alaska) grown in controlled environment rooms. When the cuttings were rooted at 70 μmol m−2 s, 1 (photosynthetic photon flux density) or more, a stock plant irradiance at 100 μmol m−2 s−1 decreased rooting ability in cuttings compared to 5 μmol m−2, s−1, However, cuttings rooted at 160 μmol m−2 s−1 formed more roots compared to 5 (μmol m−2 s−1. Although a high irradiance increased the number of roots formed, it could not overcome a decreased potential for root formation in stock plants grown at high irradiance. Light compensation point and dark respiration of cuttings decreased by 70% during the rooting period, and the final levels were strongly influenced by the irradiance to the cuttings. Respiratory O2 uptake decreased in the apex and the base of the cutting from day 2 onwards, whereas a constant level was found in the leaves. Only the content of extractable fructose, glucose, sucrose and starch varied during the early part of the rooting period. We conclude that the observed changes in the cuttings are initiated by excision of the root system, and are not involved in the initiation of adventitious roots.  相似文献

Clones of Cryptomonas phaseolus Skuja , Cryptomonas rostratiformis ( Skuja ) Skuja in Huber-Pestalozi, and Cryptomonas undulata Gervais were isolated from the deep chlorophyll maximum near the oxic/anoxic boundary layer of the mesoeutrophic lake Schlachtensee, Germany. Different autecological features of these species were studied in batch culture experiments . Cryptomonas cf . ovata Ehrenberg and Chroomonas sp. that never dominated in the deep chlorophyll layer were also isolated from Schlachtensee to study their light-dependent growth in comparison to the deep-living species . Cryptomonas undulata, C. cf . ovata, and C. phaseolus had a very low light compensation point ( 5–7 μmol.m-2.s-1 ), whereas the growth rate of Chroomonas sp. and C. rostratiformis was positive above 16 and 24 μmol.m-2.s-1 . Cryptomonas phaseclus and Chroomonas sp. became photoinhibited above photon flux densities of 92 and 116 μmol.m-2.s-1 . Cryptomonas rostratiformis, C. cf . ovata, and C. undulata reached a maximum growth rate at a considerably higher photon flux density (198–250 μmol.m-2.s-1 ). Cryptomonas phaseolus grew fastest under light-limiting conditions . Chyptomonas phaseolus and C. undulata were best able to suruive prolonged periods of darkness . Cryptomonas phaseolus, C. rostratiformis, and C. undulata did not show any uptake of fluorescent latex beads. When labeled glucose was provided in naturally occurring concentrations, carbon uptake by C. phaseolus, C. rostratiformis, and C. undulata was negligibly small in comparison to cellular carbon content. I suggest that the adaptation to a low-light environment is an important preadaptation for the dominance of C. phaseolus and C. undulata near the freshwater chemocline .  相似文献

研究了日本落叶松母株年龄、插穗内源激素含量与生根之间的关系,以及外源IBA对插穗内源激素含量的影响及其对插穗生根的促进作用。结果表明:不同株龄插穗生根性状及插穗茎和叶中激素含量差异均达极显著水平,叶中激素含量对插穗生根力没有直接影响;插穗茎中生根抑制激素(ABA)含量随株龄增长而增加,生根促进激素与抑制激素的比值(IAA+GA+ZR)/ABA却随株龄的增长而递减,与生根力随株龄的变化趋势一致,且该比值与生根性状紧密相关,因此可作为评价母株(无性系)生根力的指标;插后13~32d是插穗愈伤组织形成和不定根诱导的关键期,此期生根促进激素消耗量大,茎中含量大幅度降低,进入根伸长生长阶段,含量上升;外源IBA促进插穗生根的机制在于通过外源激素的刺激,在不定根诱导期,插穗茎中ABA含量大幅度降低,从而有利于不定根的发生和发育。  相似文献

Stockplants of the tropical hardwoods Terminalia spinosa Engl, and Triplochiton scleroxylon K. Schum were grown in a controlled environment under red:far-red (R:FR) ratios ranging from 0.5-3.1. In both species, rates of shoot height increment were higher (P < 0.05) under the low R:FR ratios as a result of increased internode elongation. In Terminalia spinosa , specific leaf area (SLA) was also significantly higher under lower R:FR ratios, values ranging from 175 to 210 cm2 g-1 in the 3.1 and 0.9 treatments respectively. No effect of R:FR ratio on SLA was recorded in Triplochiton scleroxylon. Pre-severance photo-synthetic rate, stomatal conductance and water-use efficiency were increased under the higher R:FR ratios in Terminalia spinosa , rates of photosynthesis ranging between 2.68-4.59 μmol m-2 s-1 in the 0.5 and 3.1 R:FR treatments respectively. Gas exchange rates of Triplochiton scleroxylon were unaffected by R:FR treatment. These contrasting responses to variation in R:FR ratio were associated with differences in rooting ability. In Terminalia spinosa , significantly higher percentage rooting was recorded in the cuttings from the 3.1 R:FR treatment than from 0.5, with values of 93.7% and 77.5% recorded respectively. R:FR ratio also affected rooting percentages of Triplochiton scleroxylon , but in this case, higher rooting percentages were recorded in the lower R:FR ratios, values ranging from 31.1–54.1% in the 3.0 and 0.5 R.FR treatments respectively. This difference in rooting response is attributed to the contrasting effects of R:FR ratio on the leaf and stem morphology of the two species. The implications of these results for stockplant management are discussed.  相似文献

Abstract. A field study was conducted to determine the relationship of solar-excited chlorophyll a fluorescence to net CO2 assimilation rate in attached leaves. The Fraunhofer line-depth principle was used to measure fluorescence at 656.3 nm wavelength while leaves remained exposed to full sunlight and normal atmospheric pressures of CO2 and O2. Fluorescence induction kinetics were observed when leaves were exposed to sunlight after 10 min in darkness. Subsequently, fluorescence varied inversely with assimilation rate. In the C4 Zea mays , fluorescence decreased from 2.5 to 0.8 mW m-2 nm-1 as CO2 assimilation rate increased from 1 to 8 μmol m-2 s-1 (r2= 0.520). In the C3 Liquidambar styraciflua and Pinus taeda , fluorescence decreased from 6 to 2 mW m-2 nm-1 as assimilation rate increased from 2 to 5 or 0 to 2 μmol m-2 s-1 (r2= 0.44 and 0.45. respectively). The Fraunhofer line-depth principle enables the simultaneous measurement of solar-excited fluorescence and CO2 assimilation rate in individual leaves, but also at larger scales. Thus, it may contribute significantly to field studies of the relationship of fluorescence to photosynthesis.  相似文献

The marine alga Heterosigma carterae Hulburt (Raphidophyta) was grown in N-limiting batch cultures using either nitrate or ammonium as the N source, at photon flux densities (PFDs) of 50, 200, and 350 μmol·m-2·s-1 in a 12:12 h LD cycle. Carbon content could be estimated from biovolume (μg C = 0.278 × nL; R = 0.98) but not reliably from pigment content. During exponential growth, ammonium-grown cells (in comparison with nitrate-grown cells at the same PFD) attained higher growth rates by at least 20%, contained more N, and had a lower C:N ratio, higher concentrations of intracellular free amino acids, and higher ratios of glutamine: glutamate (Gln: Glu) and asparagine: aspartate (Asn:Asp). Growth was nearly light-saturated on ammonium at 200 μmol·m-2·s-1 (cell-specific growth rate of 1.2 d-1) but probably not saturated in nitrate-grown cells at 350 μmol·m-2·s-1. PFD did not affect Gln: Glu or Asn: Asp for a given N source. These results indicate that the nitrate-growing cells were more N-stressed than those using ammonium (which in contrast were relatively C-stressed) and that this organism would show an enhanced competitive advantage against other species when supplied with a transient supply of ammonium rather than nitrate .  相似文献

Paclobutrazol (PB) only slightly stimulated the rooting of mung bean cuttings but, interestingly, the number of adventitious roots formed was dramatically increased when PB was used together with indole-3-butyric acid (IBA). Application of PB in the first phase of root formation, when root initials are induced, caused the greatest enhancement of the promotive effect of IBA on rooting. Investigation of the effect of PB on uptake, transport and metabolism of [5-3H]-IBA in mung bean cuttings revealed some changes in the rate of metabolism of IBA in comparison with control cuttings. PB was found to be involved in the partitioning of carbohydrates along the cuttings. Application of sucrose, like PB to the base of IBA-treated cuttings enhanced the effect of IBA. The patterns of the effects of PB and IBA, separately and together, on rooting were similar in defoliated and intact cuttings, however the number of roots was much lower in the defoliated cuttings, which lacked a source of assimilates. PB counteracted the effect of GA3 in the upper regions of the cuttings and seemed to increase the sink capacity at the base of the cuttings. The results of the present study clearly demonstrated the enhancing influence of PB on IBA stimulation of the rooting of mung bean cuttings. It is suggested that PB may affect the rate of metabolism of IBA during rooting and the status of the local sink, in the base of the cuttings, thus partially contributing to the enhancement of the rooting-promotive effect of IBA.  相似文献

Seedlings of Pinus sylvestris were grown for 6 weeks under natural light conditions in a temperature controlled environment room. Cuttings from these plants were rooted in tap water or in indolebutyric acid (IBA) solutions for 60 days at an irradiance of 16 W m-2. Experiments were performed at 3-week intervals during two growth seasons. — Seasonal changes in root formation were found in control cuttings as well as in IBA treated cuttings. The number of roots and the percentage of cuttings that rooted were high during early spring and autumn. During the summer period hardly any roots were formed. Stimulation of root formation by IBA occurred manily during spring and autumn when cuttings already possessed the ability to form roots. — The influence of photoperiod during stock plant growth was also investigated. Shorter photoperiod resulted in an increase in the number of roots and rooting percentage. The period during summer where rooting was inhibited under natural light conditions was considerably shortened when stock plants were grown at a photoperiod of only 4 h. The results demonstrate the importance of the growing conditions for stock plants for subsequent root formation. The results are discussed with special reference to the role of irradiance.  相似文献

The effect of fruit removal on gas exchange, water relations, chlorophyll and non-structural carbohydrate content of leaves from mature, field-grown plum trees ( Prunus domestica L. cv. Stanley) was determined over 2 consecutive growing seasons. Removal of fruits during stage II of fruit development decreased CO2 assimilation rate within 24 h from 12.6 to 8.5 μmol m-2 s-1 in 1986, and from 12.1 to 10.2 μmol m-2 s-1 in 1987. Depression of net photosynthesis persisted for at least 5 days and was greatest in the early afternoon. Recovery of the CO2 assimilation rate to pretreatment levels coincided in defruited trees with vegetative growth that was more than 5-fold that of fruiting trees in the first 6 weeks after fruit removal in 1986. Estimated photorespiration was similar in both fruiting and defruited trees. The stomatal contribution to the decrease of CO2 assimilation rate, calculated from assimilation/intercellular CO2 curves, ranged from 31 to 46%. Defruiting did not affect leaf water potential, but decreased leaf osmotic potential. Leaf levels of chlorophyll, fructose, glucose, sorbitol and sucrose were not affected by defruiting, whereas starch content increased up to 51% in leaves of defruited trees within 24 h after fruit removal. However, because of the small starch pool present in plum leaves (<1.9% dry weight) it is unlikely that starch accumulation was responsible for the observed decline in CO2 assimilation rate after fruit removal. The decrease of CO2 assimilation rate is discussed in relation to the hypothesis of assimilate demand regulating photosynthesis through a feedback mechanism.  相似文献

Indole-3-butyric acid (IBA, 10−4 M ), spermine (7 × 10−5 M ) and vitamin D2 (6.3 × 10−5 M ), all of which enhance rooting in mung bean cuttings ( Phaseolus aureus Roxb. cv. Berkin), influence RNA metabolism. Total and poly (A)+-RNA synthesis within the hypocotyl is inhibited by each of these chemicals within 24 h. These changes precede induced cell division and are therefore associated with the so-called inductive period of regeneration during which some cells in the hypocotyl undergo dedifferentiation. However, following subsequent transfer of cuttings to borate, which is an essential prerequisite for development of root primordia in these cuttings, RNA synthesis is enhanced by pretreatments with IBA, spermine or vitamin D2. Furthermore, IBA inhibits synthesis and turnover of protein within the hypocotyl.  相似文献

The apple rootstock A2 can be readily propagated in vitro both in the juvenile and in the adult growth phase. Shoots were produced by meristem tip culture from the apple rootstock A2 in different growth phases. The influence of growth phases and different concentrations of PG and IBA was investigated as to rooting percentage, survival percentage, number of roots per rooted shoot, root length, shoot length and formation of callus. IBA at 15 μ M without PG gave a significantly lower rooting percentage than 5 and 10 μ M IBA. PG together with IBA stimulated rooting, the optimum concentrations of PG being, however, not the same for the different growth phases. For the adult growth phase, 10−4 M PG promoted rooting, whereas 10−3 M PG markedly inhibited rooting. In the juvenile growth phases, both 10−4 and 10−3 M PG stimulated rooting. PG at 10−4 M also increased the number of roots. The longest roots were obtained at 10−3 M PG and 5 μ M IBA. PG at 10−3 M reduced callus formation at all IBA concentrations used. Neither shoot length nor root length influenced the survival percentage.  相似文献

Cold storage of cuttings is frequently applied in the vegetative propagation of ornamental plants. Dianthus caryophyllus was used to study the limiting influences of auxin and sugars on adventitious root formation (ARF) in cuttings stored at 5°C. Carbohydrate levels during storage were modulated by exposing cuttings to low light or darkness. The resulting cuttings were treated (or not) with auxin and planted, and then ARF was evaluated. Carbohydrate levels in the cuttings were monitored and the influence of light treatment on indole-3-acetic acid (IAA) and zeatin (Z) in the basal stem was investigated. Dark storage for up to 4 weeks increased the percentage of early rooted cuttings and the final number and length of adventitious roots, despite decreased sugar levels in the stem base. Light during cold storage greatly enhanced sugar levels, particularly in the stem base where the Z/IAA ratio was higher and ARF was lower than observed in the corresponding dark-stored cuttings. Sugar levels in nonstored and dark-stored cuttings increased during the rooting period, and auxin application enhanced the accumulation of sugars in the stem base of nonstored cuttings. Auxin stimulated ARF most strongly in nonstored, less so in light-stored, and only marginally in dark-stored cuttings. A model of auxin-sugar interactions in ARF in carnation is proposed: cold storage brings forward root induction and sink establishment, both of which are promoted by the accumulation of auxin but not of sugars, whereas high levels of sugars and probably also of cytokinins act as inhibitors. Subsequent root differentiation and growth depend on current photosynthesis.  相似文献

Abstract. A behavioural test was used to determine the light sensitivity of the nocturnal mosquito Anopheles gambiae Giles s.s. to low intensities of 'white' light (tungsten filament), 'red' light (white light filtered by a darkroom safelight filter) and 'infra-red' light) of two types (white light filtered by a λ>700 nm filter, and light-emitting diodes with λ>900 nm). Mosquitoes were placed in a 20 cm diameter flight-tunnel and their 'optomotor' response to a pattern of stripes moving across their visual field (at 14.5 cm s-1) was recorded with infra-red-sensitive video. In free-flight, with ample light, the mosquitoes controlled their flight speed and direction in relation to the stripe movement, so that the stripes always appeared to move across their visual field from front to back. They did this by flying either with the moving stripes fast enough to overtake them (19.5 ± 0.7 cm s-1), or against them more slowly (10.3 ± 0.7 cm s-1)- The net ground speed of the mosquitoes was thus c. 4–5 cm s-1. This response was significant down to 10-5 W m-2 in 'white' light, and 10-3 W m-2 in 'red' light. At light intensities below threshold and in infra-red light, however, they appeared to fly at random with respect to the stripe movement. The assumption commonly made, that mosquitoes do not 'see' in red light, may thus have to be revised.  相似文献
