In the present study, two cellulose-degrading bacteria (CDB-5 and CDB-12) were isolated from mangrove soils of Mahanadi river delta, based on halo zone formation in Congo red agar medium and evaluation for cellulase production in CMC broth medium. Based on morphological, biochemical and 16S rRNA gene sequencing, the two strains, CDB-5 and CDB-12, were identified as Brucella sp. and Bacillus licheniformis, respectively. The gene bank accession number of the strains CDB-5 and CDB-12 are KR632646 and KR632645, respectively. The strain Brucella sp. and B. licheniformis showed an enzyme activity of 96.37?U/ml and 98.25?U/ml, respectively, after 72?h of incubation period. Enzyme production was optimized under different growth conditions such as pH, temperature, agitation rate, carbon source, sodium chloride (NaCl), and nitrogen sources. Maximum cellulase production by both the strains was obtained in the same parameter condition such as pH (7.0), rpm (150), and NaCl (2%, w/v) which varies for other parameters. The strain, CDB-5, produced maximum cellulase at 35?°C temperature, maltose as a carbon source, and yeast extract as a nitrogen source where as the strain CDB-12 produces maximum cellulase at 45?°C temperature, carboxyl methyl cellulose (CMC) as carbon source and trypton as a nitrogen source. The bacterial crude enzyme was purified by ammonium sulfate precipitation followed by overnight dialysis. SDS-PAGE analysis of the partially purified cellulase enzyme exhibited band sizes of approximately 55 and 72?kDa.  相似文献

The effect of inorganic and organic nitrogen compounds on the synthesis of biomass and extracellular lipase by Oospora lactis was studied. Among the inorganic nitrogen sources ammonium sulphate and ammonium secondary phosphate and among the organic nitrogen sources yeast autolysate proved to be most beneficial for the lipase synthesis. Lipase activity and biomass accumulation in the medium containing yeast autolysate were greater than in the media containing the above ammonium salts. Lipase synthesis reached maximum in the nutrient medium containing yeast autolysate (0.7%) and ammonium sulphate (0.3%).  相似文献

以L-苏氨酸生产菌TRFC为供试菌株,研究了氮源对L-苏氨酸发酵的产量和糖酸转化率的影响。首先通过摇瓶实验确定发酵的最佳无机氮源和有机氮源分别为硫酸铵和酵母粉,进一步利用10L罐补料分批发酵确定硫酸铵和酵母粉的最佳用量,继续优化培养条件,采用发酵中后期流加硫酸铵和糖氨混合补料等措施,L-苏氨酸产量得到进一步的提高。在最优发酵条件下,通过10L罐补料分批发酵36h,产酸可达118.9g/L,糖酸转化率为47.6%。  相似文献

The psychrotrophic, dimorphic yeast Candida humicola, isolated from Antarctic soil, secretes an acidic protease into the medium. The secretion of this protease by C. humicola was found to be dependent on the composition of the medium. In YPD or yeast nitrogen base medium containing either amino acids or ammonium sulfate as the nitrogen source, the activity of the protease in the medium was low (basal level). However, when yeast nitrogen base medium was depleted of amino acids or ammonium sulfate and supplemented with proteins, the activity of the enzyme increased. The secretion of the enzyme was greater during exponential growth at low temperatures than during growth at higher temperatures. The purified protease had a molecular mass of 36,000 Da and was inhibited by pepstatin, iodoacetamide, and sodium dodecyl sulfate. Despite the prevalent cold temperatures in Antarctica, this extracellular protease of the psychrotrophic yeast C. humicola was active at temperatures ranging from 0 to 45 degrees C, with an optimum activity at 37 degrees C.  相似文献

选用5种不同的白葡萄酒活性干酵母,以硫酸铵、氯化铵、硝酸铵、尿素、酵母粉等5种物质为氮源,观察其生长量并称量菌体重,以此分析其对氮源利用情况及不同氮源对酵母生长的影响。研究表明:供试菌系在不同氮源中均能生长。不同氮源对酵母的生长速度和生长量有不同影响;不同酵母菌种对不同氮源的利用也有差异。在以酵母粉为氮源的培养基中生长最好。在实验提供的氮源中,酵母粉为供试菌最优氮源,其次是硫酸铵,氯化铵与硫酸铵基本相当,而硝酸铵最差。8#菌种对各种氮源的利用能力相对较强。17#菌种对各种氮源的利用能力最弱。  相似文献

Batch fermentations of glucose to ethanol by Z. Mobilis.(ATCC 10988) were examined in several semidefined nutrient media. The measurement of acid produced by the microorganism was used to study its transient fermentation characteristics. Limitation of nitrogen source in the semidefined medium of Rogers and coworkers(2) was found to limit the growth of this microorganism in the late stages of batch fermentations, when the initial glucose concentration was 75 g/L and higher. The microorganism exhibits a preference for inorganic nitrogen over preformed organic nitrogen provided by yeast extract. The microbial growth occurs exponentially in the presence of ammonium sulfate and yeast extract. However, in the absence of ammonium sulfate, the growth occurs in a linear fashion. The "linear" growth phase is characterized by poor cell-mass yields, and during this phase, growth and ethanol production are decoupled. An improved semi-defined growth medium is established which supports better growth rate and cellular yield, without affecting the ethanol yield.  相似文献

Nineteen bacterial isolates were grown in shaken cultures in media containing chitin as carbon source and different additional nitrogen sources such as yeast nitrogen base (YNB), yeast extract (YE), corn steep liquor (CSL) and ammonium sulfate. Strain BM17 showed the highest activity (200 U/l) in medium containing Chitin (1%) and YNB (0.5%). Molecular analysis of the 16S rRNA gene showed that strain BM17 belongs to the species Paenibacillus pabuli (99.72% homology). The enzyme activity started after 12-24 h; exponential enzyme production was recorded from the 24th h and lasted till the 96th h of incubation when activity peaked to decrease thereafter. Medium optimisation was carried out by Response Surface Methodology (RSM) considering the effects of chitin, corn steep liquor and yeast extract. BM17 chitinolytic activity was induced by chitin but the increase of its concentration did not have significant effects on the enzyme activity. By contrast, the nitrogen source, particularly YE, strongly affected the enzyme production.  相似文献

考察一株新分离的毛栓菌静置产纤维素酶的条件,通过单因素试验和正交设计试验分别对培养基初始pH、培养基中的碳源、氮源及诱导物进行研究。利用DNS(3,5-二硝基水杨酸)法测定CMC-Na(羧甲基纤维素钠)纤维素酶活。结果表明:该毛栓菌静置发酵产纤维素酶的最适pH为5.0,最适碳源、氮源和诱导物分别是淀粉、酵母粉和干麸皮,且质量浓度分别为淀粉1%、酵母粉0.4%、干麸皮1.5%,在此条件下,进行静置发酵3d,可获得CMC-Na(羧甲基纤维素钠)纤维素酶活力最高为56.62U/mL。此研究开创了纤维素酶生产菌的新发现,与以往所报道的好氧产纤维素酶菌种黑曲霉、木霉等相比,具有发酵时间短、耗能小等优点,具有一定的潜在应用价值。  相似文献

The rate of transport of L-amino acids by Saccharomyces cerevisiae epsilon 1278b increased with time in response to nitrogen starvation. This increase could be prevented by the addition of ammonium sulfate or cycloheximide. A slow time-dependent loss of transport activity was observed when ammonium sulfate (or ammonium sulfate plus cycloheximide) was added to cells after 3 h of nitrogen starvation. This loss of activity was not observed in the presence of cycloheximide alone. In a mutant yeast strain which lacks the nicotinamide adenine dinucleotide phosphate-dependent (anabolic) glutamate dehydrogenase, no significant decrease in amino acid transport was observed when ammonium sulfate was added to nitrogen-starved cells. A double mutant, which lacks the nicotinamide adenine dinucleotide phosphate-dependent enzyme and in addition has a depressed level of the nicotinamide adenine dinucleotide-dependent (catabolic) glutamate dehydrogenase, shows the same sensitivity to ammonium ion as the wild-type strain. These data suggest that the inhibition of amino acid transport by ammonium ion results from the uptake of this metabolite into the cell and its subsequent incorporation into the alpha-amino groups of glutamate and other amino acids.  相似文献

高产纤维素酶枯草芽胞杆菌S-16的筛选及其发酵工艺优化   总被引:1,自引:0,他引:1  
利用刚果红鉴别培养基及基础液体筛选培养基进行菌种筛选,从新疆盐碱地分离得到的16株菌株中筛选获得一株产纤维素酶活力较高的菌株S-16,对该菌株进行16SrDNA鉴定,确定该菌为枯草芽胞杆菌(Bacillus subtilis)。对S-16发酵产纤维素酶的主要影响因素进行研究,分别考察了碳源、氮源、培养基初始pH和接种量等因素对发酵产纤维素酶的影响。结合单因素影响实验得到优化后的培养基配方为:羧甲基纤维素钠1.5%,酵母粉1%,NaCl 1%,MgSO_4·7H_2O 2‰,KH_2PO_4·3H_2_O 1‰。优化后的发酵条件为:初始pH为8,接种量1%,种龄8h,培养时间48h。经过发酵工艺优化,S-16产生的羧甲基纤维素酶活(CMCase)和滤纸酶活(FPase)分别达到4.64IU/mL和0.46IU/mL,与初始培养条件下的酶活相比分别提高了3.14倍和1.30倍。本研究得到的枯草芽胞杆菌S-16及其优化发酵工艺为秸秆的快速腐熟和高产纤维素酶的应用奠定了基础。  相似文献

以沪农灵芝1号为供试菌株,葡萄糖作为碳源,用硫酸铵、氯化铵、鱼粉蛋白胨、胰蛋白胨和酵母粉作为氮源,研究不同种类氮源对灵芝菌丝体液态深层发酵过程的影响。首先,确定了N-10酵母自溶粉作为发酵的氮源,降低了发酵的复杂性和不确定性;其次,考察N-10酵母自溶粉不同浓度对灵芝菌丝体发酵合成灵芝三萜过程中菌丝体的生物量、葡萄糖消耗、灵芝三萜产量等方面的影响,确定了N-10酵母自溶粉的适宜添加浓度。在此基础上,采用响应面中心组合设计,对4因素最佳水平范围进行研究,结果表明,葡萄糖、N-10酵母自溶粉、磷酸二氢钾和七水硫酸镁的含量分别为31.06g/L、2.76g/L、1.77g/L和1.99g/L时,灵芝三萜的理论产量为21.166g/kg干菌丝体,实际发酵产量提高到21.153g/kg干菌丝体。与原工艺相比,新工艺的灵芝三萜产量提高了6.22%。  相似文献

Strain IVIC-Pb9, unlike other strains ofParacoccidioides brasiliensis, cannot grow on a simple basal medium and requires the addition of casein hydrolyzate or yeast extract. The present study shows that this requirement is limited to very low concentrations of methionine and that methionine concentrations above 0.01% inhibit growth. The levels of glucose and organic nitrogen required for maximum rate of growth of strain IVIC-Pb9 on both basal medium and GGY medium composed of glucose, glycine and yeast extract were also determined. An evaluation of the suitability of the GGY medium revealed that its composition, as commonly used to grow dimorphic fungi, is not adequate to obtain a maximum rate of growth with strain IVIC-Pb9 ofP. brasiliensis.  相似文献

The biomass yield, cellulolytic activity, and protein recovery using Aspergillus terreus GN1 with alkali-treated sugarcane bagasse was studied using different levels (250-600 mg of N/L of broth) of organic and inorganic nitrogen sources. e.g., cattle urine, urea, cornsteep liquor, ammonium sulfate, ammonium nitrate, ammonium iron sulfate, ammonium chloride, and sodium nitrate. Among different levels of alkali-treated bagasse substrate concentrations (0.5-4.0% w/v) tested, 1.0% substrate yielded the highest crude protein content, protein recovery, and cellulolytic activity. The biomass recovery with 1.0% substrate ranged from 290-380 mg/500 mg bagasse substrate in a 50-mL broth with a nitrogen level of 250-600 mg of N/L in all the sources except ammonium iron sulfate, which yielded 402-439 mg/500 mg bagasse substrate. However, crude protein content of biomass obtained with an ammonium iron sulfate nitrogen source was the lowest. Cornsteep liquor nitrogen source at the rate of 600 mg of N/L yielded the maximum crude protein of 32.9%, protein recovery of 22.2 g/100 g of bagasse, and carboxymethyl cellulase and filter paper enzyme activities of 1.1 and 0.09 units/mL, among the organic and inorganic nitrogen sources studied. In general, the organic nitrogen sources and inorganic nonammonium nitrogen sources were utilized preferentially for protein production over the inorganic ammonium nitrogen sources. The fermentation time required under optimum cultural and nutritional conditions for A. terreus GN1 was also evaluated. The crude protein content of the biomass increased gradually up to the seventh day of fermentation, but the protein recovery rate was high up to two or three days. It was observed that the cellulose utilization rate increased after an initial lag of one day up to the third day and gradually increased further, which corresponded positively with protein content, biomass protein recovery, and cellulase enzyme activity. On the seventh day of fermentation, the crude protein content, biomass protein recovery, water-soluble carbohydrate, bagasse cellulose utilization, CMCase, and FPase activities were 32.8%, 20.1 g/100 g of bagasse, 6.2%, 82.7%, 1.0. and 0.08 U/mL, respectively. The final biomass recovered contained 32.8% crude protein content and had an in vitro rumen digestibility (IVRD) coefficient of 68.8%. The biomass contained almost all the essential and nonessential amino acids and was comparable with FAO reference protein. It is concluded that a fermentation time of 72 h gave a faster rate of protein production of 16.9 g/100 g of bagasse with 69.8% bagasse cellulose utilization with 76.0% IVRD. and contained almost all the essential and nonessential amino acids.  相似文献

The effect of sugar and ammonium nitrogen content in the initial nutrient medium as well as the cultivation temperature on the respiration activity and respiratory quotient of yeast used in the champagne manufacture was studied. An addition of ammonium nitrogen stimulated respiratory processes in the yeast cell. The respiration activity increased with an increase of pH to 4.0. With an elevation of the sugar content of the substrate the respiratory quotient increased resulting in an uneconomic utilization of sugar. The maximum respiration activity and minimum respiratory quotient occurred at a temperature of 20 degrees.  相似文献

Rhodotorula glutinis, an oil producing strain, can utilize monosodium glutamate (MSG) wastewater as a raw material for lipid production. The effects of ammonium-N in the MSG wastewater (ammonium 15,000–25,000?mg/L, COD 30,000–50,000?mg/L) on cell growth, lipid accumulation and malic enzyme activity of R. glutinis have been studied. Four initial ammonium sulfate concentrations in the medium were set, which were 20, 60, 100, and 140?g/L. With an increase in the ammonium sulfate concentration, the uptake of ammonia nitrogen and lipid accumulation increased while the biomass decreased at 72?h. The maximum value of ammonia nitrogen consumption reached 5.77?g/L for an initial ammonium sulfate concentration of 140?g/L at 72?h. In addition, 60?g/L ammonium sulfate concentration may be an appropriate concentration for R. glutinis cultivation. The activity of the malic enzyme was measured and the results showed that there was a linear relationship between the intracellular lipid content and the total malic enzyme activity.  相似文献

The effect of inorganic nitrogen source on population growth of Euglena mutabilis, an acidophillic benthic protozoa colonizing on the sediment of acid mine drainage, was investigated. Sodium nitrate, ammonium chloride, and ammonium sulfate were tested as nitrogen sources. The population density of E. mutabilis at equilibrium density cultivated in ammonium chloride‐ and ammonium sulfate‐containing media was 9–11 times higher than that in sodium nitrate‐containing medium at the optimal salt medium concentration. The population growth of E. mutabilis in ammonium sulfate‐containing medium was rapid and reached half of the equilibrium density after ca. 228 h, which was ca. 77 h earlier than that in ammonium chloride‐containing medium. Culture medium with ammonium sulfate as the nitrogen source achieved the highest maximum population density and the fastest growth rate among the three nitrogen salts used as nitrogen sources.  相似文献

Growth of Desulfovibrio desulfuricans (API strain) was found to take place in an atmosphere of hydrogen on the agar surface of complex media, including yeast extract (Difco), and Trypticase Soy Agar (BBL) without any added reducing agents. For growth on a 2% yeast extract-agar surface in the absence of hydrogen (nitrogen atmosphere), sodium lactate was required in the medium. Growth on the surface of Trypticase Soy Agar (TSA) under nitrogen took place readily in the absence of an added hydrogen donor. A medium (TSA plus salts) is described based upon the addition of sodium lactate (4 ml per liter), magnesium sulfate (2 g per liter), and ferrous ammonium sulfate (0.05%) to TSA, which appears suitable for the isolation and growth of Desulfovibrio on the surface of agar plates in an atmosphere of hydrogen. Sodium lactate does not appear to be essential in this medium for good growth and sulfate reduction in a hydrogen atmosphere, but is essential in a nitrogen atmosphere. Growth of Desulfovibrio (hydrogen atmosphere) on the agar surface of media commonly used for its cultivation as well as on an inorganic medium containing bicarbonate as a source of carbon is poor and erratic unless inoculated (Desulfovibrio) plates of TSA plus salts are incubated in the same container with plates of these media. This stimulatory effect of incubation with inoculated plates of TSA plus salts medium appears to be due to as yet unidentified volatile material produced by D. desulfuricans when growing on this medium. Another volatile material, or possibly the identical material, appears to act similarly to a hydrogen donor.  相似文献

Toxic waste generated by Jatropha seed cake after utilization of biodiesel on one hand has stimulated the need to develop new technologies to treat the waste and on the other, forced us to reevaluate the efficient utilization of its nutritive potential for production of various high-value compounds and its conversion to non-toxic forms which could be used as animal feed stock. In this study, Jatropha seed cake was used for production of cellulases by new isolate of Thermoascus aurantiacus under solid-state fermentation. The interaction of nitrogen source concentration, moisture ratio, initial pH of the medium and inoculum size was investigated and modelled using response surface methodology (RSM) using Box-Behnken Design (BBD). Under optimized conditions endo-β-1,4-glucanase, β-glucosidase and filter paper activities were found to be 124.44, 28.86, 4.87?U/g of substrate, respectively. Characterization of endo-β-1,4-glucanase, β-glucosidase was done after partial purification by ammonium sulfate fractionation followed by desalting. The endo-β-1,4-glucanase and β-glucosidase showed maximum activity at 70?°C and pH 4. Saccharification studies performed with different lignocellulosic substrates showed that sugar cane bagasse was most susceptible to enzymatic hydrolysis. The study suggests that Jatropha seed cake can be used as a viable nutrient source for cellulase production without any pretreatment under solid-state fermentation by T. aurantiacus.  相似文献
