蒌蒿对镉的富集特征及亚细胞分布特点   总被引:2,自引:0,他引:2  
以镉(Cd)富集植物蒌蒿(Artemisia selengensis)为试材, 采用超声波细胞破碎处理和超速离心的方法, 对蒌蒿根和叶中亚细胞水平的Cd分布状况进行研究, 同时测定Cd在蒌蒿不同器官中的富集效果。结果表明, 在30 mg·kg–1Cd胁迫下, 蒌蒿叶片中Cd的富集浓度是根和茎中的2–3倍, 但因叶片所占植株的生物量比例较小, 其对Cd的积累量远小于茎和根; Cd在蒌蒿叶片细胞壁、胞液和细胞器中含量比为16:5:1。细胞壁固定是叶片对Cd的主要防御机制。随着Cd处理浓度的增加, 细胞壁和胞液中的Cd含量大幅上升, 但细胞器中Cd含量仍维持在较低水平。长时间和高浓度的Cd胁迫可使细胞壁解毒机制失活并诱导细胞器中的Cd含量增加, 导致植株死亡。根中液泡的Cd贮存量较大, 解毒效果显著。  相似文献

丛枝菌根真菌(Arbuscular mycorrhizal fungi,AMF)能提高植物抗重金属毒性。为评价镉(Cd)胁迫条件下AMF对烟草生长和抗Cd毒性的影响,对烟苗(Nicotiana tabaccum L.)进行0、5、20、40 mg/kg Cd处理,接种和不接种AMF玫瑰红巨孢囊霉Gigaspora rosea的双因子试验。培养90 d后测定植株的营养生长量、渗透调节物质和叶片保护酶活性。0~5 mg/kg Cd处理下,无论接种或不接种AMF,烟草的营养生长、渗透调节物质含量、叶片保护酶活性都上升。5 mg/kg Cd处理下,接种与不接种AMF的烟草各项指标均达到各处理的最高值,并显著高于0 mg/kg的对照组;Cd浓度继续升高时,烟草的营养生长、渗透调节物质含量、叶片保护酶活性均下降。在0~40 mg/kg Cd浓度处理范围内,各项检测指标的总体趋势是先升后降;在相同Cd浓度处理中,接种AMF烟草,其营养生长、渗透调节物质含量和叶片保护酶活性均高于不接种的烟草。结果表明:低于或等于5 mg/kg Cd浓度处理,能促进烟草的营养生长和抗重金镉毒性能力的提高,高于5 mg/kg Cd浓度处理时,Cd对烟草植株产生毒害,且随着浓度的升高毒性加重,用AMF Gigaspora rosea接种烟苗,可以减轻烟苗受Cd胁迫的毒性。  相似文献

该研究采用盆栽控制试验,测定不同浓度(0、5、50、100、150、200 mg/kg)土壤Cd胁迫下小报春幼苗生长及生理生化指标,以探究小报春(Primula forbesii Franch.)对重金属镉(Cd)污染的抗性和敏感性,为新型香花地被植物应用于Cd污染土壤提供理论依据。结果表明:(1)低浓度(5 mg/kg)Cd胁迫能促进小报春株高和根长的伸长,高浓度(≥150 mg/kg)Cd胁迫下株高、根长和生物量明显降低。(2)随Cd胁迫浓度增加,小报春幼苗叶片光合作用、叶绿素含量、SOD和CAT活性先上升后下降,而其超氧阴离子产生速率、过氧化氢、丙二醛含量、叶相对电导率和POD活性持续升高。(3)在Cd胁迫条件下,小报春体内K、Zn含量降低,叶和根中Ca、Mg含量显著增加,体内Cd含量明显升高,且根系Cd含量远远高于叶和叶柄。研究发现,小报春幼苗对Cd胁迫具有一定的耐性,低浓度(5 mg/kg)Cd胁迫对小报春生长影响较小,但高浓度(≥150 mg/kg)Cd胁迫对小报春产生明显的毒害作用,影响其正常生长;小报春可能通过增强体内抗氧化酶系统活性、增加叶和根中Ca、Mg含量和根系截留镉来减轻镉胁迫的伤害,提高自身耐受镉胁迫能力。  相似文献

通过盆栽研究了Cd胁迫下德国鸢尾的生长状况、生态效应、生理特性及吸收和富集Cd的能力.结果表明:德国鸢尾对小于5 mg/kg的Cd有较强的耐性,适用于城区土壤修复;Cd浓度大于5 mg/kg时抑制德国鸢尾生长,降低了其生态效应.随着Cd浓度的增大,德国鸢尾根系活力、叶绿素含量和含水量逐渐降低,游离脯氨酸和可溶性糖含量先升高后降低,细胞膜透性逐渐升高.Cd在德国鸢尾体内分布为根系>地上部分,随着Cd浓度的增大,德国鸢尾根系和地上部分Cd积累浓度逐渐升高、富集系数和转运系数逐渐降低;Cd浓度为20 mg/kg时德国鸢尾对Cd的积累量最大,为2.122 mg/plant.  相似文献

采用盆栽试验,研究了不同镉(Cd)浓度胁迫下忍冬科荚蒾属珊瑚树(Viburnum odoratissimum)和地中海荚蒾(V.tinus)2种园林树种3年生苗木叶片受伤害形态特征及生理生化指标(叶绿素、丙二醛、可溶性糖及可溶性蛋白含量、电解质渗透率、过氧化物酶活性)的变化,并分析了植株各营养器官(根、茎、叶)Cd的富集水平。结果表明:Cd对珊瑚树叶片生长及生理生化指标均未产生显著影响;地中海荚蒾在Cd胁迫浓度为100mg.kg-1时出现伤害症状,电解质渗透率增加,丙二醛含量增加,叶绿素含量减少;而当胁迫浓度提高到400mg.kg-1时,植株叶片枯黄,植株萎蔫,生长势差;2种植物地下部富集量均大于地上部,可以推断珊瑚树和地中海荚蒾对土壤中Cd具有一定的富集能力,在土壤镉污染严重地区栽植具有重要意义。  相似文献

镉在槐叶苹叶片中的蓄积及其生态毒理学分析   总被引:4,自引:0,他引:4  
以分布广泛的浮水蕨类植物--槐叶苹为研究对象,用含不同浓度Cd(2.5,5,7.5 mg/L和10 mg/L)的10% Hoagland营养液培养7d,旨在分析Cd在蕨类植物中的蓄积及其诱导产生的毒理学作用.随着培养液中Cd浓度的增加,(1) 槐叶苹叶片中Cd含量极显著上升;(2) Cd处理造成槐叶苹叶片产生明显的矿质营养失衡症状,主要表现为显著增加了对Ca和降低了对P,K和Na的吸收;(3)叶绿素含量和叶绿素荧光Fv/Fm比值都逐渐降低;(4)抗氧化防御系统活性或含量紊乱,其中超氧化物岐化酶(SOD)活性下降最突出,过氧化物酶(POD)、过氧化氢酶(CAT)、还原型谷胱甘肽(GSH)、抗坏血酸(AsA)和类胡萝卜素(Car)也都先升后降,膜脂过氧化产物MDA含量则显著增多;(5)可溶性蛋白含量一直下降;(6)透射电镜观察发现细胞超微结构损伤明显,主要为叶绿体膨胀和解体、线粒体嵴突减少和空泡化、细胞核核仁解体、核膜破裂、核质消失,并有晶体出现.结果表明槐叶苹虽然对水体Cd有一定的净化作用,但Cd浓度与毒害之间的剂量-效应关系明显;MDA含量的明显上升以及叶绿素和可溶性蛋白含量的显著下降标志着槐叶苹遭受到明显的氧化损伤;Cd破坏了槐叶苹进行正常生理活动所必需的生理和结构基础;SOD可以作为Cd诱导产生氧化压力的分子生物标志物和水体Cd污染的灵敏生态毒理学指标,Cd的半效应浓度为2.41mg/L,最大允许浓度为0.241mg/L.  相似文献

通过温室盆栽试验,研究珊瑚树对土壤中镉(Cd)的耐受和富集特征。研究结果表明,珊瑚树对污染土壤中Cd具有较强的耐受能力。培养56 d内,土壤中Cd含量对珊瑚树生物量影响不明显;随着培养时间的延长(105—203 d),珊瑚树的生长明显受到土壤中Cd的抑制作用。与对照处理(土壤中Cd含量为3.6 mg/kg)相比,培养154 d后,土壤中Cd含量为24.6 mg/kg处理下珊瑚树叶片中叶绿素a、叶绿素b、类胡萝卜素和丙二醛(MDA)含量没有明显变化;培养203 d后,土壤中Cd含量为24.6mg/kg处理下珊瑚树叶片中类胡萝卜素和MDA含量无明显变化,但叶片中叶绿素a和叶绿素b含量明显受到抑制,从而导致珊瑚树叶片的生长明显受到抑制(P0.05)。培养203 d后,珊瑚树对土壤中Cd的富集系数和转运系数均大于1,表明珊瑚树对土壤中Cd具有一定的富集和转运能力。上述结果表明,珊瑚树对Cd污染土壤具有一定的生态修复潜力。  相似文献

研究了在梯度浓度Hg2+和Cd2+胁迫下,满江红(Azolla imbricata (Roxb.) Nakai)的叶绿素含量、叶绿素a/b比值、光合放氧速率、呼吸速率、抗氧化酶系(超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD))和细胞超微结构受Hg2+和Cd2+的毒害影响.结果显示:随着胁迫程度的增大,叶绿素含量、叶绿素a/b比值、光合放氧速率明显下降,呼吸速率均在2 mg/L浓度下达到峰值,尔后下降; SOD、CAT、POD的活性均出现不同程度的应激性升高(除POD在Cd2+处理时下降),尔后下降.电镜观察发现,随着污染物浓度的增加和胁迫时间的延长,叶绿体出现膨大、破损和解体;线粒体嵴突膨胀和线粒体变形及空泡化;核染色质凝集,核仁消失,核膜破裂.实验结果表明: Hg2+和Cd2+污染不仅损害植物的生理活性,而且也破坏细胞的超微结构,最终导致植物死亡;随着Hg2+和Cd2+胁迫的增大,细胞超微结构的损伤程度和植物的生理变化是同步的;植物受毒害的程度表现出明显的剂量效应关系;在同一处理时间和浓度下,Cd2+对满江红的毒性大于Hg2+.Hg2+对满江红的致死浓度为3.5~4.0 mg/L,Cd2+为3.0~3.5 mg/L.对满江红鱼腥藻(Anabaena azollae Strasburger)细胞的超微结构变化观察表明,满江红鱼腥藻对Hg2+和Cd2+的耐受性明显高于满江红.  相似文献

为探讨镉(Cd)胁迫条件下蒌蒿对促生细菌(PGPB)的响应机制,以芽孢杆菌T3(Bacillus sp.)菌株为对象,通过盆栽试验,研究不同Cd处理水平下芽孢杆菌T3菌株对蒌蒿生长、生理、Cd富集转运以及根际微生物的影响。结果表明:(1)在Cd胁迫条件下,芽孢杆菌T3菌株可显著促进蒌蒿地上部、地下部及总干物质的积累,但对株高和根冠比的影响不明显。(2)芽孢杆菌T3菌株能显著促进Cd胁迫条件下蒌蒿对Cd的富集与转运,相比单一Cd胁迫,蒌蒿地上部生物富集系数(BCF)增加了8.3%~29.3%,地下部BCF减少了6.6%~11.1%,同时转运系数(TF)增加了20.8%~38.3%。(3)在Cd胁迫和芽孢杆菌T3菌株的共同作用下,蒌蒿叶绿素a、叶绿素b和总叶绿素含量明显增加。(4)芽孢杆菌T3菌株使Cd胁迫条件下蒌蒿体内丙二醛(MDA)含量显著减少,超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性明显增加,可有效抵御由Cd胁迫所产生的氧化伤害。(5)芽孢杆菌T3菌株能显著增加Cd胁迫条件下蒌蒿根际土壤细菌、放线菌数量及微生物总量,可减少蒌蒿根际土壤真菌的数量,但与对照差异不显著。研究表明,在Cd胁迫条件下芽孢杆菌T3菌株对蒌蒿生长、生理和根际土壤微生物环境可产生积极影响,可增强蒌蒿植物对Cd胁迫环境的适应性,从而提高其对Cd的耐受能力。  相似文献

为探究中国沙棘对土壤镉(Cd)胁迫的性别响应差异,该研究以中国沙棘2年生幼苗为材料,利用盆栽试验研究在不同浓度Cd处理下(0(CK)、25、50、100和200 mg·kg-1)雌、雄株幼苗的生长、叶片生理特性以及Cd富集特征的差异。结果表明:(1)Cd处理下中国沙棘幼苗雌、雄株的株高和基径生长以及各器官生物量均表现出低浓度(<50 mg·kg-1)促进,高浓度(> 100 mg·kg-1)抑制的现象;低浓度Cd处理下雌株的株高、基径增长率和生物量的增幅均高于雄株;高浓度Cd处理下(200 mg·kg-1)雄株株高增长率、叶生物量和总生物量分别较CK显著降低,而雌株均未显著下降。(2)随着Cd浓度升高,雌、雄株叶片光合色素含量和抗氧化酶活性呈先升后降的变化趋势,丙二醛(MDA)和渗透调节物质含量呈上升趋势;Cd浓度为50~200 mg·kg-1时,雌株叶片的光合色素含量、抗氧化酶活性和渗透调节物质含量均高于雄株,而MDA含量始终低于雄株。(3)随着Cd浓度升高,雌、...  相似文献

Plant tolerance is one of the preconditions in soil phytoremediation. The physiological responses and tolerance threshold of centipedegrass (Eremochloa ophiuroides) were investigated under eight different Cd concentrations (0, 60, 120, 180, 240, 300, 360, and 420 mg Cd kg(-1)) in a sand culture system. The results showed that turf quality, leaf relative water content (RWC), leaf electrolyte leakage (EL), leaf osmotic potential did not show significant changes under 180 mg Cd kg(-1) compared with the control, and relative growth rate (RGR), turf density, leaf chlorophyll content, photochemical efficiency (Fv/Fm) did not show significant changes under 240 mg Cd kg(-1) compared with the control throughout the whole experiment. Regression analysis was used to determine the threshold Cd concentrations for each physiological parameter and the most sensitive parameter occurred by RWC of 197 mg Cd kg(-1) which was chose as Cd tolerance threshold in centipedegrass because under this concentration the plant did not show any significant difference with the control in all growth and physiological parameters measured in this experiment. Greenhouse hydroponic experiments were conducted using Cd-sensitive (Dong 17) and tolerant (Weisuobuzhi) barley genotypes to evaluate genotypic differences in response of photosynthesis and ultrastructure to Cd toxicity in the presence of exogenous glutathione (GSH). Addition of 20 mg L(-1) GSH in 5 μM Cd culture medium (Cd?+?GSH) significantly alleviated Cd-induced growth inhibition and reduced Cd concentration in leaves and roots especially in the sensitive genotype Dong 17. Exogenous GSH greatly ameliorated Cd-induced damages on leaf/root ultrastructure, e.g., compared with Cd alone treatment, chloroplasts in plants treated with Cd?+?GSH become better or in relatively normal shape with well-structured thylakoid membranes and parallel pattern of lamellae and unfolded more starch grains but less osmiophilic plastoglobuli; nuclei of root cells were better formed and chromatin distributed more uniformly in both genotypes, and number of plastids and mitochondria cristae in Dong 17 resumed to control level. The examination of photosynthetic performance revealed GSH dramatically increased net photosynthetic rate (P(n)), stomatal conductance (G(s)), and transpiration rate (T(r)) in the both genotypes and strongly stimulated Cd-induced decrease in the maximal photochemical efficiency (F(v)/F(m)) especially in the sensitive genotype.  相似文献

Twenty-four male rats of the Wistar strain divided into four groups were injected sc with a dose of 0.8, 1.5, and 3.0 mg Cd/kg body wt as CdCl2 in saline, and saline alone to the control rats, three times a week for 3 wk. Cadmium levels of whole kidney homogenate, supernatant (cytosol), precipitate, and metallothionein (MT) fraction were measured. Histological changes of the renal proximal tubules were investigated by optical and electron microscopy. In the kidneys, Cd levels were increased with the increment of Cd dosage; 80–90% of Cd was contained in cytosol, and 55–75% was in MT fraction. Non-MT-Cd reached a maximum in the 1.5 mg Cd group, whereas that of the 3.0 mg Cd group showed some decline. With increasing Cd doses, the size of nuclei and nucleoli in the cells of proximal tubule showed significant enlargement and also an increase in the number of nucleoli on light microscopy. At higher doses, chromatin condensation of the tubular nuclei and vacuolar degeneration of the tubular cells were evident. On electron microscopy, perichromatin granules of the proximal tubular nuclei were increased in number, especially in the rats of Cd 0.8 mg and 1.5 mg/kg groups. As the Cd doses increased, ring-shaped nucleoli were increased in number and nucleolar segregation was observed more clearly. Moreover, in the 3.0 mg/kg Cd group, nuclear indentation and nucleoli containing compact dense granules were observed. In the cytoplasm, there was an increase of lysosomes, myelin bodies, ring-shaped mitochondria, and vesiculation; ultimate changes were degeneration and cell necrosis. The injured cells were heterogenously distributed in each nephron and this heterogeneity was attributed in the difference in Cd content and cell cycle in each cell of the nephron.  相似文献

利用盆栽试验,研究了龙须草对Cd的耐受能力和富集特征.结果表明:低浓度Cd处理(5 mg·kg-1)能促进龙须草的生长,增强其生理活性,生物量、净光合速率等12个指标比对照提高1.0%~15.5%;高浓度Cd处理(>5 mg.kg-1)对龙须草的生长产生抑制作用,但当Cd浓度达到100 mg.kg-1时,龙须草仍能完成正常的生理周期,生物量、净光合速率仅比对照分别下降27.0%和25.6%.龙须草各器官的Cd含量随Cd污染程度的上升而大幅度增加,根的Cd含量为350~500 mg.kg-1,茎叶为15~35 mg·kg-1.在Cd浓度<50 mg·kg-1时,吸Cd量随添加浓度的增大而增大,茎叶吸Cd量占总吸Cd量的15.7%~38.4%,茎叶与根的吸Cd量比值最高可达0.62.龙须草对Cd污染的耐受性、富集和转运能力均较强,是一种潜在的Cd超富集植物.  相似文献

Remediation of heavy metal contaminated sites using hyperaccumulators presents a promising alternative to current environmental methodologies. In the pot-culture experiment, the effects of Cd, and Cd-As on the growth and its accumulation in the Cd-hyperaccumulator (Solanum nigrum L.) were determined. No reduction in plant height and shoot dry biomass was noted when the plants were grown at Cd concentration of 1.0. The plant can be classified as a Cd-hyperaccumulator. Growing in the presence of 10 mg/kg Cd and 50 mg/kg As, the plant height and shoot dry matter yields did not decrease significantly (p>0.05) compared to that at 10 mg/kg Cd, however the stem Cd content increased by 28%. It was also observed that S. nigrum used exclusion strategy to reduce As uptake in the roots and restricted translocation into the shoots, resulting in As contents of the plant being root>leaf>stem>seed. The Cd accumulation capacity coupled with its relatively high As tolerance ability could make it useful for phytoremediation of sites co-contaminated by Cd and As.  相似文献

The cadmium (Cd) tolerance and metal-accumulation characteristics of 29 species (18 families) of weed were studied by using outdoor pot-culture experiments. The results of this screening showed that Bidens pilosa and Kalimeris integrifolia (both Asteraceae) expressed some properties that are characteristic of Cd hyperaccumulators. In 10 mg/kg Cd-spiked soil, they accumulated a good deal of Cd in shoots (28 and 25 mg/kg DW, respectively) with high Cd enrichment factors (EFs; concentration in plant/soil). Cd accumulations in shoots were greater than those in roots (translocation factor (TF) >1, concentration in shoot/root) and the shoot biomasses did not decreased significantly compared to the unspiked control. The other weed species showed little accumulation of Cd, Pb, Cu, or Zn. In a concentration-gradient experiment, the Cd accumulation potentials of B. pilosa and K. integrifolia were examined further. Cd concentrations in leaves of B. pilosa growing in soils spiked with 25, 50, and 100 mg/kg Cd were up to 145, 160, and 192 mg/kg, respectively, and the Cd content in stems in the 100 mg/kg Cd-spiked soil was 115 mg/kg, all greater than the 100 mg/kg notional criterion for Cd hyperaccumulation. The Cd EFs and TFs were all greater than 1. The shoot biomasses did not decrease significantly compared to the controls. B. pilosa was thus shown to have some characteristics of a true Cd hyperaccumulator plant.  相似文献

以不同浓度Cd2+处理6d的浮萍为材料,从细胞核超微结构损伤、nDNA一级结构变化及抗氧化酶系活性改变等方面分析重金属的植物毒理学效应。透射电镜观察发现,5~7mg/L Cd2+处理的细胞核膜完整,染色质凝聚并趋边化;10mg/L Cd2+处理后则核仁解体,且DNA原位末端标记(TUNEL)检测发现DNA的3′-OH断端可被特异标记,表现出典型的凋亡细胞特征。而20mg/L Cd2+已导致细胞坏死。同时,较低浓度的Cd2+胁迫可刺激抗氧化酶(SOD,POD,CAT)活性升高,以清除体内活性氧,而随金属浓度的进一步增高,三种抗氧化酶活性都急速下降。研究发现,活性氧和抗氧化酶系密切参与了重金属Cd2+诱导的浮萍体细胞凋亡过程。  相似文献

This work was part of a project designed to assess whether organic selenium (Se) can protect against the toxic effects of cadmium (Cd). A total of 300 1-day-old, as hatched, broilers were randomly distributed in four dietary treatments with five replicate pens per treatment. In T1 treatment, broilers were fed a diet with 0.3 mg/kg added Se, as Se-yeast, without added Cd; in T2, broilers were fed a diet with 0.3 mg/kg Se and 10 mg/kg Cd; in T3, broilers were fed a diet with 0.3 mg/kg Se and 100 mg/kg of Cd; and in T4 treatment broilers were fed a diet with 3 mg/kg Se and 100 mg/kg Cd. The Cd was added to diets T2, T3 and T4 as CdCl2. On the 4th and 6th week, two broilers per replicate pen were killed in order to obtain whole blood, liver, kidney and breast samples. Body mass, feed conversion ratio and mortality were assessed and haematological analyses were performed. Se and Cd levels in tissues were analysed by inductively coupled plasma mass spectrometry. Broilers supplemented with 0.3 mg/kg Se can tolerate low levels of Cd added to the diets, as there were no significant negative effects on the examined performance parameters, whereas addition of excess Cd led to an impairment of broilers’ performance. Mortality of broilers did not differ between the four dietary treatments at any interval point or the whole period. The examined haematological parameters such as haematocrit, total blood protein concentration, and leukocytes types ranged within physiological values, revealing no negative health effects after simultaneous Cd and Se addition. The present study indicated that Se can help against the negative effects of Cd, but cannot counteract all of its negative effects.  相似文献
