The present study examined intraspecific interference and searching behavior of Chrysopa phyllochroma Wesmael (Neuroptera: Chrysopidae) for Aphis gossypii Glover (Homoptera: Aphididae) nymphs under laboratory and greenhouse conditions. The results were shown as follow: 1) In four different arenas (i.e. Petri dish, glass vessel, glass vessel with barriers in it, and cage with potted cotton plant), the predaceous efficiency of C. phyllochroma larvae varied with the predator density, the hunt constant (Q) and the intraspecific interference (m)increased with the prey density but decreased with the space heterogeneity; 2) In cage with potted cotton plant, the first- and second-instar green lacewing larvae consumed 13.6 and 29.4 cotton aphids/day respectively. The number of cotton aphids consumed by C. phyllochroma on lower leaves was significantly less than that on upper leaves; and 3) In cage with potted cotton plant, the percentage of the first- and second-instar green lacewing larvae located on upper leaves was significant less than that on lower leaves.  相似文献

Nineteen kinds of spiro enol ether analogues were screened with larvae of Pieris rapae for antifeedant activity. The results showed that the antifeedant activity of compounds No.20 and No.12 was higher than others. In non-choice test, AFC50 values within 24 h of compounds No.20 and No.12 against 3rd instar larvae of P. rapae were 226.93ug/mL and 370.00ug/mL, and that in choice test against 4th larvae were 280.54 ug/mL and 398.88 ug/mL, respectively. Compd. No.20 could prolong the eggs hatch time and reduce the haemolymph content and the protein content in haemolymph of 4th instar larvae obviously. Compd. No.20 could protect tested leaves and control larvae of P. rapae effectively.  相似文献

The effects of four rice varieties highly susceptible ( Xiang-Zao-Xian31 and Jia-Yu 164 ), moderately susceptible ( Wei-You402 ), and resistant ( Zhu-Liang-You02 ) to the whitebacked planthopper Sogatellafurcifera on the nymphal development, reproduc- tion and survival of Microvelia horoathi were investigated through a food chain using SogateUafurcifera as herbivore prey. Significant longer nymphal developmental time were observed in the 1 st, 4th, 5th instar and whole nymphal stage of M. horvathi raised on S. furcifera fed ZLY02 and WY402 than those of the other two varieties. M. horvathi raised on S. furcifera fed resistant ZLY02 showed a significant lower survival of whole nymphal stage than those fed other three susceptible varieties. Moreover, M. horvathi raised on S. furcifera fed resistant ZLY02 had shorter female longevity and lower lifetime fecundity than those of on susceptible XZX3l. Our results suggest that the S. furcifera- resistant rice variety ZLY02 had a negative effect on the growth and development of the predator M. horvathi through a food chain.  相似文献

Feeding and oviposition preference studies of Helopeltis theivora Waterhouse, on leaves and stems of tea, Camellia sinensis L.(O)Kuntze, revealed that the insect mostly preferred the second leaf for feeding in comparison to the first, and the third leaf and stems; while the fifth instar nymphs were the most voracious feeders among the life stages, producing themost and largest feeding lesions. The insect preferred to oviposit mainly on stems and very few eggs are found on leaves.  相似文献

An invasive new biotype of the tomato/potato psyllid (Bactericera [Paratrioza] cockerelli [Sulc.]) (Homoptera: Psyllidae) recently has caused losses exceeding 50% on fresh market tomatoes in western North America. Despite these extensive losses, little is known regarding the threshold levels at which populations must be suppressed in order to prevent economic losses. A series of experiments were therefore designed using combinations of two common tomato cultivars (QualiT 21 and Yellow Pear), five pest-densities (0, 20, 30, 40 and 50 nymphs/plant), and three feeding-duration (5 days, 10 days, and lifetime) treatments to test the relative importance of pest density, feeding period, and cumulative psyllid-days to establish economic threshold levels for psyllids. The cultivars differed considerably in their response to the toxin injected by the psyllid nymphs. ‘Yellow Pear' plants could recover from feeding by up to 40 nymphs for as long as 10 d, whereas ‘QualiT 21' plants were irreparably damaged by densities of 20 nymphs feeding for only 5 days. On ‘Yellow Pear', all plant measurements such as the number of yellow leaves and plant height were significantly better correlated with cumulative psyUid-days than with either pest density or feeding duration. On ‘QualiT 21 ', all plant measurements other than the number of yellow leaflets and leaves were significantly better correlated with pest density than with feeding duration or cumulative psyUid-days, and pest density was a better predictor of psyUid damage. Potential reasons for the variable responses between cultivars and the implications for psyllid sampling and integrated pest management are discussed.  相似文献

The geomagnetic field(GMF)is well documented for its essential role as a cue used in animal orientation or navigation.Recent evidence indicates that the absence of GMF(mimicked by the near-zero magnetic field,NZMF)can trigger stress-like responses such as reduced body weight,as we have previously shown in the brown planthopper,Nilaparvata lugens.In this study,we found that consistent with the significantly decreased body weight of newly emerged female(—14.67%)and male(—13.17%)adult N.lugens,the duration of the phloem ingestion feeding waveform was significantly reduced by 32.02%in 5th instar nymphs reared under the NZMF versus GMF.Interestingly,5th instar nymphs that exhibited reduced feeding had significantly higher glucose levels(+16.98%and+20.05%;24 h and 48 h after molting),which are associated with food aversion,and expression patterns of their appetite-related neuropeptide genes(neuropeptide F,dow regulated overall;short neuropeptide F,dowregulated overall;adipokinetic hormone up-regulated overall;and adipokinetic hormone receptor,down-regulated overall)were also altered under the absence of GMF in a manner consistent with diminishing appetite.Moreover,the expressions of the potential magnetosensor croptochromes(Crys)were found significantly altered under the absence of GMF,indicating the likely upstream signaling of the Cry-mediated magnetoreception mechanisms.These findings support the hypothesis that strong changes in GMF intensity can reduce adult body weight through affecting insect feeding behavior and underlying regulatory processes including appetite regulation.Our results highlight that GMF could be necessary for the maintenanee of energy homeostasis in insects.  相似文献

Population dynamics of Chaoborus flavicans larvae of various instars was studied from November 1986 to December 1987 in a eutrophic, fish-free pond, Japan. First and 2nd instar larvae were observed from late April to late October, indicating a reproductive period of about half a year. C. flavicans overwintered in the 4th instar larvae. In water column samples, total density of all instars was 680–23680 m-2, and pupal density 0–2600 m-2; larvae of the 1st, 2nd, and 3rd instars showed 5–6 density peaks in 1987, suggesting that 5–6 generations occur during a year (peaks of the 4th instar larvae were not clear, probably due to their longer development than those of younger instars). In sediment samples, no 1st and 2nd instar larvae were found, 3rd instar larvae were found occasionally but density of the 4th instar larvae was 280–18600 m-2, and pupal density varied between 0–502 m-2. Fouth instar larvae accumulated in sediment in the cold season and in the water column in the warm season; high temperature and low oxygen concentration were the most important factors limiting the distribution of larvae in the sediment in summer in the NIES pond. The dry weight of total C. flavicans larvae was 0.08–4.2 g m-2 in sediment samples and 24–599 μg l-1 (0.10–2.40 g m-2) in water column samples. Comparisons of maximum densities in the NIES pond in different years and in waters of different trophic status show that density is generally higher in eutrophic than in oligotrophic habitats. Complementary sense promoter from cotton leaf curl virus (CLCuV) is a novel plant promoter for genetic engineering that could drive high-level foreign gene expression in plant. To determine the optimal promoter sequence for gene expression, CLCuV promoter was deleted from its 5' end to form promoter fragments with five different lengths, and chimeric gus genes were constructed using the promoterdeletion. These vectors were delivered into Agrobacterium and tobacco (Nicotiana tabacum L cv. Xanthi) plants which were transformed by leaf discs method. GUS activity of transgenic plants was measured. The results showed that GUS activities with the promoter deleted to -287 and -271 from the translation initiation site were respectively about five and three times that of full-length promoter. There exists a c/s-element which is important for the expressing activity in phloem from -271 to -176. Deletion from -176 to -141 resulted in a 20-30-fold reduction in GUS activity in leaves with weak activity in leaves and  相似文献

We used brightfield electron microscopy (BEM), differential interference contrast microscopy (DICM), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and confocal laser scanning microscopy (CLSM) to investigate the stylet pathways of Bemisia tabaci during nymphal feeding behavior in cotton leaves beginning with penetration of the abaxial leaf surface and ending with stylets in sieve tubes in phloem tissues. Most nymphal stylets within salivary sheaths penetrating leaf tissues made complex turns and developed more than one salivary sheath branch before ending in sieve tubes. The external morphology of the salivary sheaths and their routes between and through leaf cells are described during the present study. Results showed the presence of the stylet within the sieve tubes. B. tabaci nymphs may remove stylets and feed in different sieve tubes. Ten short movies showing the progression of the stylet penetrations from adaxial surface to the sieve tubes are attached to Figures 8-15. The report and movies can be viewed from the internet. The topography and wettability of the underside of English weed (Oxalis pes-caprae) leaves and of their biomimetic replicas are investigated. Polyvinyl siloxane molds were cast from the leaves and then filled with an epoxy pre-polymer to produce replicas. The particular topographical structures of leaves and replicas were evaluated by optical microscopy and Scanning Electron Microscopy (SEM) analysis. The static wettability of leaves and replicas was assessed by contact angle measurements, while the dynamic wettability was characterized by estimating contact angle hysteresis and studying the dynamic behavior of impacting water droplets. A smooth glass slip and its replica were used as control surfaces. The replica moulding method used was able to transfer the characteristic pattern of irregular 100 μm - 200 μm × 60 μm convex papillae interspersed with stomata of the original leaf to the epoxy replicas. The static contact angle of 143°± 3° and the contact angle hysteresis of 2~ indicate that the underside of the English weed leaf is close to superhydrophobic. The lower contact angles (130° ± 4°) and higher hysteresis (31°) observed for the replica when compared with the original leaves were associated to an inaccurate replication of the chemistry and structures of the three-dimensional wax projections covering the plant surface. Also, trichomes in the original leaves could not be accurately reproduced due to their flexibility and fragility. Differences in wetting behavior were also evident from droplet impact experiments, with rebound regimes prevailing in the original leaves and regimes characterized by higher adhesion and larger dissipation predominating in the replicas. Nevertheless, the morphological features of the leaf transferred to the replica were sufficient to promote a clear hydrophobic behavior of the replica when compared with the smooth epoxy reference surface.  相似文献

The silverleaf whitefly (Bemisia argentifolii Bellows and Perring) is a widely distributed pest of cotton (Gossypium hirsutum L.) and the population levels may be affected by rates of nitrogen fertilization and planting date. Field experiments were conducted to investigate the impact of cotton planting date and nitrogen fertilization on silverleaf whitefly population dynamics. Cotton was planted on 26 April and 8 June, for the early and late plantings, respectively. Nitrogen treatments consisted of soil applications of 0, 112, 168 and 224 kg of nitrogen per hectare. The population levels of adult whiteflies were much higher on early-planted cotton than on late planting. Also, increased numbers of adult whiteflies on both early and late plantings occurred with increasing amounts of applied nitrogen.Applied nitrogen increased seed cotton yields of early plantings but had no effect on the yields of late plantings.  相似文献

Summary Cardiac myocytes cultured over microfabricated extracellular recording devices can be used to assay bioactive compounds. However, electrophysiological signals recorded from these devices vary in amplitude with time. Theoretically, changes in signal amplitude arise from myocytes being moved over recording sites by cocultured fibroblasts. To test this, neonatal rat cardiac myocytes were cultured at high densities and low densities on fibronectin-coated glass. After 36.5 h, myocytes were identified by their rhythmic contractions and then time-lapse-recorded for 3.5 h. Length, width, and angle of orientation was then determined every 30 min for five cells in low density and five cells in high-density culture. Low-density cells had mean lengths of 65.3 μm and widths of 35.1 μm, whereas cells in high-density culture had greater mean lengths of 74.2 μm and lower mean widths of 24.3 μm. Length, width, and angle of orientation of cells in low- and high-density culture changed by 4.1%, 11.8%, and 2.7 degrees, and 6.4%, 10%, and 4.6 degrees, respectively, every half hour. We found no evidence of myocyte-fibroblast interactions influencing cell position or shape in low density, but in high density, we found evidence that fibroblast-myocyte interactions could transiently influence cell shape. We conclude that fibroblast-independent changes in cell shape are largely responsible for the changes in signal amplitude recorded from cardiac myocytes cultured on microfabricated extracellular recording devices. However, there is some evidence that myocyte-fibroblast interactions may augment this process in high-density culture. The implications of these findings for bioassay development are discussed.  相似文献

Effects of entomopathogenic fungus Verticillium lecanii on biological characteristics and life table of Serangiumjaponicum, a predator of whiteflies against five different conidial concentrations (1 × 10^4, 1 × 10^5, 1 × 10^6, 1 × 10^7, and 1 × 10^8 conidia/mL) were studied under laboratory conditions. The developmental periods for all immature stages (from eggs, 1 st, 2nd, 3rd, 4th instar nymph and pupae up to emergence) among the treatments were significantly different when compared to that of control, and the longest development period was observed as treated with 1 × 10^8 spore/mL. However, no significant difference on the percent survival of all immature stages was observed among the treatments and control. Also, there were no significantly different effects of V. lecanii on mean generation time, intrinsic rate, the finite rate of increase and longevity of S. japonicum among the treatments and control.  相似文献

Solitary and gregarious locusts differ in many traits, such as body color, morphometrics and behavior. With respect to behavior, solitary animals shun each other, while gregarious animals seek each other's company, hence their crowding behavior. General activity, depending on the temperature, occurs throughout the day but is much lower in solitary locusts. Solitary locusts occasionally fly by night while gregarious locusts fly regularly during daytime (swarming). In search of new assays to identify substances that control or modify aspects of (phase) behavior, we designed a simple activity assay, meant to complement existing behavioral measurement tools. The general activity is reflected in the number of wall hits, that is, the number of contacts between the locust and the vertical walls of a small arena. Using this single parameter we were able to quantify differences in total activity of both nymphs and adults of isolation-reared (solitary), regrouped- and crowdreared (gregarious) locusts under different conditions. Furthermore, we demonstrate that there are inter- and intra-phase dependent differences in activities of 5th instar nymphs afar injections of the three different adipokinetic hormones.  相似文献

Differences in anatomy and morphology of the kiwifruit leaves and leaf petioles might play a considerable role in the sex-determination. Three months after bud break (June), the kiwifruit leaves of both male and female plants, grown on the vegetative and generative shoots showed different leaf area (128.6 ± 13.45 cm2 in male and 104.5 ± 4.02 cm2 in female plants) and shape. The most frequently leaf shape was determined as "folium cordatum" and "folium rotundato-cordatum". Higher values of total leaf thickness of the female leaves (190 ± 3.84 μm) in comparison to male leaves (174 ± 3.52 μm) were estimated, resulting in the thicker adaxial leaf epidermis and especially in thicker palisade parenchyma in female leaves (136 ± 2.76 μm in comparison to 104 ± 1.61 μm in male leaves). Typically bifacial leaves were observed in both male and female leaves. Anomocytic stomata in hypostomatic leaves were found. The reticulate venation appears to be the main type of leaf venation. Stalked stellate multicellular trichomes on the abaxial leaf side were frequently observed in the leaves of both sexes. No important differences between male and female plants were found in the structures of vascular system in leaves and leaf petioles. Thus leaf thickness and surface morphology of adaxial leaf epidermis can be considered as important structural parameters in the sex determination. Presence of potyvirus in single garlic (Allium sativum L.) cloves from the same bulb, and in five single leaves excised from commercial field-grown individual plants was studied using ELISA. It was found that the viruses were not present in all organs of the same plant, since some cloves of the same bulb were infected with potyvirus but some others were potyvirus-free. Analyzed leaves from a given plant also exhibited irregular distribution of potyvirus. This study also aimed to obtain potyvirus-free plants from two commercial garlic cultivars (Taiwan and Chileno) using cloves subjected to thermotherapy, chemotherapy or meristematic dissection followed by in vitro culture. Thermotherapy (sequential treatment at 32°C for a week, 36°C for 2 weeks, and 38°C for 3 weeks) was found to affect survival of explants and 36.5% cloves from Taiwan and 26.8% from Chileno cultivars were recovered after the treatment. ELISA tests showed that 63% of the cloves of Taiwan that survived the treatment and 70.9% of Chileno explants were potyvirus-negative. Regarding chemotherapy (205 μM Ribavirin solution), the explants (cloves) survived, but only an average of 27.0–34.8% were negative for the presence of potyvirus. When meristematic dissection was applied, an average of 41.7% explants of Taiwan and 34.2% of Chileno survived the treatment, and approximately 64% of these explants from both cultivars were potyvirus-negative. Potyvirus-free garlic plants grown in field conditions showed longer stems with a major fresh and dry weight per bulb, and also exhibited a higher yield than non-treated plants.  相似文献

The plant genome possesses a large number of microRNAs(miRNAs)mainly 21-24 nucleotides in length.They play a vital role in regulation of target gene expression at various stages throughout the whole plant life cycle.Here we sequenced and analyzed~10 million non-coding RNAs(ncRNAs)derived from fiber tissue of the allotetraploid cotton(Gossypium hirsutum)1 days post-anthesis using ncRNA-seq technology.In terms of distinct reads,24 nt ncRNA is by far the dominant species,followed by 21 nt and 23 nt ncRNAs. Using ab initio prediction,we identified and characterized a total of 562 candidate miRNA gene loci on the recently assembled D5 genome of the diploid cotton G.raimondii.Of all the 562 predicted miRNAs,22 were previously discovered in cotton species and 187 had sequence conservation and homology to homologous miRNAs of other plant species.Nucleotide bias analysis showed that the 9th and 1 st positions were significantly conserved among different types of miRNA genes.Among the 463 putative miRNA target genes,most significant up/down-regulation occurred in 10-20 days post-anthesis,indicating that miRNAs played an important role during the elongation and secondary cell wall synthesis stages of cotton fiber development.The discovery of new miRNA genes will help understand the mechanisms of miRNA generation and regulation in cotton.  相似文献

Aphis gossypii Glover shows obvious host specialization, with cucurbit- and cotton-specialized biotypes or host races in many regions. Because its annual natal hostcrops senesce earlier the cucurbit-specialized biotype may suffer food deficiency. The method this biotype uses to overcome this challenge is still poorly understood. In orderto understand the potential of the cucurbit-specialized biotype aphids in host shift and usage, the performance of this biotype on cotton (Gossypium hirsutum), a common butpoor quality host plant, was explored in this study. The cucurbit-specialized aphids could establish populations on cotton only when these plants had at least nine leaves, and subsequent populations developed rather slowly. The presence of whitefly populations on cotton improved the success rate of cucurbit-specialized aphids. The cucurbit-specialized aphidswere mainly distributed on the older leaves of cotton, with only a few settling on the upper leaves. The cucurbit-specialized aphids reared on cotton for 40, 54 and 61 days stillmaintained strong preference for their natal host plant, cucumber (Cucumis sativus), rather than cotton, and their net reproductive rates and intrinsic rates of natural increase weredramatically lower when they were transferred onto new six-leaf cotton plants or detached leaves. Therefore, we concluded that the cucurbit-specialized aphids have the potentialto utilize mature or whitefly-stressed cotton plants, but that this feeding experience on cotton did not alter their specialization for cucurbits. Some cotton plants could act as atemporary host for the cucurbit-specialized aphids to overcome food deficiency arising from senescing cucurbits.  相似文献

Nestmate recognition codes show remarkable chemical complexity, involving multiple biochemical pathways. This complexity provides the opportunity to evaluate the ecological and social conditions that favor the evolution of complex signaling. We investigated how the chemical signatures of three populations of the social paper wasp Polistes biglumis differed in terms of concentration of hydrocarbons, proportions of branched hydrocarbons and overall variation. We tested whether the variation in chemical signatures among populations could be explained by the prevalence of social parasites or whether this was just an effect of local abiotic conditions which influenced the composition of the hydrocarbon cuticular layer. We studied the chemical signa- ture in three populations in which obligate social parasites differed in the selection pressures they imposed on host populations. Within each population, we restricted our analyses to non-parasitized hosts, to avoid potential short-term effects of parasite pres- ence on the host chemical signatures. We found that host colonies in parasitized populations had more diverse profiles than the parasite-free population. Moreover, the overall concentration of hydrocarbons and the relative proportion of branched hydrocar- bons were larger in the parasitized populations, relative to the non-parasitized one. This is to our knowledge the first evidence in favour of the hypothesis that different traits in the host chemical signatures as a whole undergo evolutionary changes resulting from directional or balancing selection imposed by social parasites. We conclude that obligate social parasites act as 'engines of diversity' on host chemical signatures and operate in favor of a geographic mosaic of diverging communication codes  相似文献
